Dispharynx nasuta (Nematoda) in California quail (Callipepla californica) in western Oregon.

Seventy-six California quail (Callipepla californica) were collected during a 22-mo period from the E. E. Wilson Wildlife Area near Monmouth, Oregon. Dispharynx nasuta occurred in 38% of the birds with a mean intensity of 4.9 +/- 5.1. In one of 2 yr, host age was significantly associated with prevalence, with immature males showing the highest prevalence (73%). Although C. californica has been the subject of several parasitological surveys, this is the first record of D. nasuta in this host.

Effect of chronic centrifugation on mouse breeding pairs and their offspring.

Fetuses of chronically centrifuged mice are smaller than 1g fetuses, and have reduced areas of long bone ossification. Fewer pregnancies occurred in centrifuged mice. In this study, exposure of chronically centrifuged females to male bedding 48 hours prior to mating induced estrus as determined by vaginal smears. Females were paired with males on the centrifuge or at 1g, and were sacrificed at 18 days gestation. Fetuses were weighed, measured, and stained with alizarin red and methylene blue. This new mating method increased centrifuge pregnancies, but not to control levels. There were no pregnancies at 3.5 g. Weights and crown rump lengths of centrifuged 18-day fetuses were less than those of controls, significantly so at 2.6 and 2.9g. There was no effect on litter size, number of resorptions per litter or male/female ratio. Bones of centrifuged fetuses were found to be smaller and shaped differently from controls. Effects were mitigated by matings with 1g males. Results confirm our previous reports of decreased fetal size and areas of ossification in hypergravity, and demonstrate a reliable method for providing timed-pregnant mice in hyper-g conditions.

Using PC/GENE for protein and nucleic acid analysis.

This paper describes a series of protein analyses using the molecular biology software package PC/GENE, which runs on an IBM or compatible microcomputer. A nucleic acid sequence was first edited and then translated into an amino acid sequence. The amino acid composition, isoelectric point, molecular weight, and other properties of the sequence were determined. Programs to predict secondary structure, alpha helix membrane associations, hydrophobic and hydrophilic regions, and surface and antigenic sites from the amino acid sequence were also used. A search was made in a data base for sequences containing a region similar to a region in the protein sequence. Sequence alignments and queries of data bases can also be performed.

Mini-dose intrathecal morphine for the relief of post-cesarean section pain: safety, efficacy, and ventilatory responses to carbon dioxide.

To determine the safety, efficacy, and the ventilatory responses to carbon dioxide (CO2) of mini-dose intrathecal morphine, 33 healthy women who underwent cesarean section with spinal anesthesia using 0.75% bupivacaine in 8.25% dextrose were studied. Patients were randomly assigned to receive, in a double-blind fashion, either morphine 0.25 mg (group I, n = 11), morphine 0.1 mg (group II, n = 10), or saline (group III, placebo group, n = 12) in 0.5 ml volume mixed with the bupivacaine. In both groups I and II excellent postoperative analgesia with long duration was obtained (27.7 +/- 4.0 and 18.6 +/- 0.9 hours, respectively, X +/- SEM). All patients in group III required an analgesic (8 mg subcutaneous morphine) within 3 hours of spinal anesthesia. Seven patients in group I and four patients in group II developed mild pruritus that did not require treatment. Ventilatory responses to CO2 showed no evidence of depression attributable to either the 0.25 or 0.1 mg of morphine, but significant depression of the CO2 responses was observed in group III patients after administration of subcutaneous morphine. It is concluded that a dose as low as 0.1 mg of intrathecal morphine gives excellent analgesia with minimal to no side effects and that subcutaneous morphine is associated with marked depression of the ventilatory variables.

In vivo 31P nuclear magnetic resonance spectroscopy of bone mineral for evaluation of osteoporosis.

The mineral content of stationary bone samples can be quantified by 31P nuclear magnetic resonance (NMR) spectroscopy. The assay can be performed in regions of the anatomy that pose problems for absorptiometric techniques, because the mineral content is measured within a selected volume without concern for the geometry of the bone. In vivo 31P NMR spectra of the bones in human fingers and wrist are reported. Soft tissue such as marrow and skeletal muscle contributes little to the 31P NMR spectra of human fingers and wrist and thus should not seriously affect the accuracy of the mineral assay. 31P NMR spectrometry should prove helpful for confirming rapid bone mineral loss in those at risk and for monitoring response to treatment.

Clinical and pharmacologic effects of high dose single agent busulfan with autologous bone marrow support in the treatment of solid tumors.

A Phase I-II clinical trial of high dose single agent busulfan (16-20 mg/kg) administered over a 4-day period was undertaken. Pharmacokinetic measurements reveal that steady state total plasma busulfan levels between 2 and 10 microM were achieved by the second day and maintained through the remaining treatment period. Urinary excretion of mutagenic activity monitored by the Salmonella mutagenesis assay persisted for up to 48 h following the last dose of busulfan. The treatment showed specificity for myelocytic precursors as evidenced by selective depression of granulocytes with relative sparing of lymphocytic elements, and by differences in DNA damage as measured by a nucleoid sedimentation assay. Dose limiting toxicity was mucositis, anorexia, and hepatic toxicity. Transient autoimmune disorders were observed in three of the six patients. Partial responses were seen in two of five patients with melanoma, but these lasted for only 2 and 3 months. High dose busulfan represents an alkylating agent with marked myelocytic selectivity and may be useful for inclusion in intensive combination regimens.

Comparison of the efficacy of a two-day and a five-day schedule for infusing intravenous gamma globulin in the treatment of immune thrombocytopenic purpura in adults.

The standard schedule for treating immune thrombocytopenia purpura in adults with intravenous immunoglobulin G infusion (IVIG), 400 mg/kg per day for five days, was compared with a shorter schedule using 1,000 mg/kg per day for two days. Both schedules were found to be effective in correcting thrombocytopenia. Eleven of the 17 patients treated with the five-day regimen and nine of 10 patients treated with the two-day regimen had significant responses. Patients with an initial platelet count of less than 20,000 platelets/mm3 or with an estimated in vivo platelet survival in excess of 90 hours were less likely to have a response than were other patients. There were no serious side effects in either group, but thrombophlebitis was observed in some patients receiving the five-day regimen when a single intravenous catheter was used for more than three days. Headaches and, less commonly, low-grade fever were noted in some patients receiving the two-day regimen when infusions were given at flow rates in excess of 0.04 ml/kg/minute. Since the two-day regimen requires shorter hospitalization and corrects thrombocytopenia slightly faster than the five-day course, it may be particularly useful in correcting thrombocytopenia in hospitalized patients requiring splenectomy or other surgery.

Non-cytocidal natural variants of human immunodeficiency virus isolated from AIDS patients with neurological disorders.

To understand the mechanism of HIV-mediated neuropathology five viral isolates were obtained from four AIDS cases with central nervous system manifestations as the primary involvement. The isolates were identified as HIVs by antigenic cross-reactivity and nucleic acid hybridizations to HIV-specific antibodies and DNA probes. The replication and cytopathic properties of these isolates were studied and compared with lymphadenopathy-associated virus (HIVLAV). All isolates had replication competence equivalent to LAV, but four isolates did not kill T4 (CD-4) cells. This isolation of non-cytocidal natural variants of HIV raises the possibility that in some AIDS cases the neurological disorders might be due to HIV variants that are non-cytocidal to T4 cells. The results also indicate that virus replication and cytotoxicity are not always concordant functions in HIV.

Ookinete antigens of Plasmodium berghei: a light and electron-microscope immunogold study of expression of the 21 kDa determinant recognized by a transmission-blocking antibody.

The expression of a 21 kDa transmission-blocking determinant on the malarial parasite Plasmodium berghei was studied by using the immunogold method at the light, scanning-electron and transmission-electron microscope levels. The determinant was shown to be expressed exclusively on the macrogamete and its immediate progeny the zygote, ookinete and oocyst. It is first detected on the plasmalemma two hours after the escape of the parasite from the red blood cell, reaches a maximal density on the young ookinete some ten hours later, and is still found on the oocyst after six days. The antigen is distributed evenly over the entire surface of the zygote and ookinete, but is readily shed from the parasite surface. The general applicability of the silver-enhanced immunogold method in parasitological research is emphasized.

Occurrence of multiple myeloma three years after successful renal transplantation.

A living-related renal transplant recipient developed premature progressive allograft dysfunction 3 years after transplantation. Histopathologic examination of a transplant biopsy specimen demonstrated kappa light chain nephropathy, whereupon the diagnosis of multiple myeloma was subsequently confirmed. A variety of tumors have been reported to develop in the successful renal allograft recipient, although multiple myeloma is decidedly uncommon. This may be due to the relatively young age of transplant recipients. Histopathologic examination of renal tissue in the evaluation of premature or unexpected renal failure in the transplant recipient is underscored.

Dialysis-induced ascites treated with peritoneal dialysis.

Dialysis-induced ascites is an uncommon complication that occurs in some chronic hemodialysis patients. Attempts at treatment have included intensive hemofiltration, bilateral nephrectomy, severe dietary fluid and salt restriction, and renal transplantation. We have described a patient successfully treated with continuous ambulatory peritoneal dialysis, and have reviewed other reports of successful therapy with peritoneal dialysis.
