Differential translation of virogenic and oncogenic sequences in malignant lymphoproliferative diseases and transfection of coding DNAs into NIH 3T3 cells.

The expression of oncoviral p30 polypeptides and onc gene-specific proteins has been examined in different human lymphoid malignancies. The distribution of antigen(s) related to the p30 of BaEV lacked any specificity. Antigen(s) related to the main core polypeptide of GaLV could be detected mainly in B- and O-cell malignancies. The myc-encoded protein was translated at higher levels in malignant than in normal lymphoid cells. An active src gene was identified in three acute lymphoid leukaemias and in one non-Hodgkin lymphoma of T-cell origin. Human DNAs coding oncoviral antigens or onc gene-specific proteins could be transfected into NIH 3T3 cells. These data suggest that the synergistic effect of the myc and src genes would operate in malignant transformation of some progenitors of T-cell lineage.

Bidirectional neuronal connections between the cerebellar interpositus nucleus and the brainstem (an autoradiographic study).

Ortho- and retrograde axonal labelings were demonstrated in several brainstem nuclei of cat after [3H]-leucine and [3H]-lysine microinjections into the cerebellar interpositus nucleus. Retrogradely labeled perikarya in the lateral reticular and solitary tract nuclei, locus coeruleus, substantia nigra and the raphe nuclei (midbrain raphe, dorsal, pontine and magnus) indicate the existence of possible aminergic afferents to the interpositus nucleus. The anterograde labeling of the neuropil in the above brainstem regions suggests the termination of axons or axon-collaterals originating from the interpositus nucleus.

Epidemiological features of biliary tract cancer incidence in Slovakia.

This descriptive study is based on detailed data of biliary tract cancer incidence in Slovakia in the period 1968-1977. Age-adjusted incidence rates of biliary tract cancer in the given decade decreased in females while in males they showed a slight increase. The study of geographic distribution revealed diminishing incidence rates from west to the east in both sexes with extremely elevated rates in towns and suburban districts in males. Different shapes of age-specific incidence curves together with the identified histological types suggested different epidemiological characteristics of individual subsites within this site. Relatively high incidence rates of biliary tract cancer in Slovakia as well as in whole Czechoslovakia, in comparison with other countries or areas of Europe, could be related to the excessive prevalence of gallstones and high number of surgical interventions upon biliary tract in this country while the decreasing incidence rates in women coincided with elevated number of cholecystectomies. The significance of detailed descriptive data from cancer registries for further analytic and etiologic study of biliary tract cancer according to subsites for a better delimitation of high risk groups and primary prevention strategies is stressed.

Epidemiologic aspects of malignant skin melanoma in Slovakia.

The trends of age-adjusted incidence rates of malignant skin melanoma in Slovakia from 1968 to 1977 showed important increase with an annual increment of 5.8% in males and 3.2% in females. The mortality rates during this period remained lower and their increase was less expressed. The study of geographic patterns of incidence revealed great variation with highest rates in urban districts. A marked predominance of the primary lesions on trunk in males and on lower limbs in females was observed. The differences in anatomic distribution are demonstrated also in age-specific incidence rates of malignant skin melanoma by main subsites. Intermittent overexposures to the sunlight in connection with rising standard of life and leisure seem to be the most suitable explanation for the rise and subsite distribution and dynamics, as well as the geographic patterns and international position of incidence rates of malignant skin melanoma in this country.

Cell production and morphological pattern formation in primary brain cell cultures. I. Pattern formation within the basal layer(s).

Spontaneous pattern formation within the basal cellular layer of primary brain cell cultures were studied. Basal cells were found to be organized into a limited number of morphologically well distinguishable types of cell-arrangements. Phase contrast microscopy was used to characterize the different cell-assemblies morphologically. Under standard culture conditions the time of appearance and specific changes of relative frequency in time were also characteristic of different patterns. The distribution of mitotically active cells among the different morphological patterns within the basal layer was also investigated by recording [3H]thymidine uptake of cells. Reduction of mitotic activities of cells in the basal layer was found in the vicinity of overlying cells. In a given area of cultures the reduction of mitotic activity was proportional to the number of overlying cells. The influence of cell proliferation on the morphological pattern formation in primary cultures is discussed.

Serotoninergic endings on VIP-neurons in the suprachiasmatic nucleus and on ACTH-neurons in the arcuate nucleus of the rat hypothalamus. A combination of high resolution autoradiography and electron microscopic immunocytochemistry.

A combination of immunocytochemistry with transmitter specific autoradiography at the electron microscopic level was used to identify vasoactive intestinal polypeptide (VIP)- or ACTH-containing neurons and serotoninergic nerve terminals in the suprachiasmatic (SCN) and arcuate nuclei (AN). We observed nerve terminals showing selective uptake of [3H]serotonin forming synaptic contact with perikarya or dendrites where the postsynaptic structures exhibited VIP-like immunoreactivity in the SCN and ACTH-like immunoreactivity in the AN. The findings provide ultrastructural evidence that serotoninergic nerve fibers terminate on VIP-neurons in the SCN and on ACTH-neurons in the AN.

Antibodies to primate retrovirus antigens in circulating immune complexes of patients with acute myeloid leukemia.

Circulating immune complexes were isolated from sera of 8 patients with acute myeloid leukemia (AML) in relapse, and 20 healthy blood donors. F(ab')2 fragments were prepared from the isolated complexes. Using a radioimmunoassay (RIA), these F(ab')2 fragments, the undigested complexes and the original sera were examined for the presence of antibodies against a panel of primate retrovirus antigens: gp70, p15 and p30 of gibbon ape leukemia virus (GaLV) and baboon endogenous virus (BaEV). F(ab')2 fragments derived from the immune complexes of all patients reacted with one or more of the antigens tested, whereas no antibody activity was found in the sera or undigested immune complexes of the same patients. By a competitive RIA, antigens related to GaLV and/or BaEV were found in the serum of 7 out of 8 patients. No markers of these retroviruses were detected in the F(ab')2 preparations, in immune complexes or in sera of any of the 20 control subjects. Our results indicate that a part of the circulating immune complexes in AML contain antigens related to primate retroviruses and specific antibodies to these antigens.

Studies on the specificity of human antibodies reacting with GP70 and P15 antigens of baboon endogenous (BaEV) and gibbon ape leukaemia (GaLV) viruses.

Antibodies reacting with gp70 and p15 antigens of baboon endogenous virus (BaEV) and gibbon ape leukaemia virus (GaLV) were detected by radioimmunoprecipitation (RIP) in blood plasma samples of patients with chronic granulocytic leukaemia, acute myeloid leukaemia and with potentially preleukaemic haematological disorders. Anti-gp70 antibodies were found more frequently than anti-p15 antibodies. Digestion of the carbohydrate part of gp70 antigens by glycosidase treatment abrogated the precipitation mediated by IgM antibodies, whereas that mediated by IgG antibodies was not markedly affected. Data suggest that antibodies detected in human plasma samples may have oncovirus specificity, but in considerable part of cases they can be of heterophil nature.

Effect of transganglionic degenerative atrophy on opiate receptors in the dorsal horn of the spinal cord.

We investigated alterations in opiate-binding sites in the upper dorsal horn after transection of the related peripheral sensory nerve in rats. The binding of (3H)diprenorphine was measured autoradiographically. The findings indicated a shift of the binding sites, rather than a degenerating disappearance, with a decrease in nerve fibers but an increase in nerve cells. This may be due to latent opiate-binding sites becoming manifest.

Syndrome of inappropriate secretion of antidiuretic hormone (SIADH) after head injury.

The authors report a review of 1808 patients admitted for the treatment of craniocerebral injuries. Eighty-four (4.6%) developed the syndrome of inappropriate secretion of antidiuretic hormone (SIADH). Two types of SIADH (severe and mild) were defined on the basis of laboratory findings and clinical signs. SIADH occurred in 0.6% of the patients with mild head injury, 10.6% of those with moderate head injury, and 4.7% of the patients with severe head injury. Regular measurements of serum electrolytes, osmolality, and urinary sodium depletion should be made during the treatment of patients suffering from head trauma because unexpected clinical deterioration may often have a reversible cause: SIADH.

Brain water accumulation after the central administration of vasopressin.

The intraventricular administration of vasopressin or DDAVP (desmopressin acetate) increased the brain water content from 78.2% to 79.2-79.5%. This was achieved without an accompanying water load. The applied water load alone did not increase the water content of the brain. There was no significant difference in the water content of the brain between animals treated with intraventricular vasopressin and intravenous water load and animals receiving only intraventricular vasopressin. The water content of the olfactory bulbs of the control animals was 3.8% higher than that of the hemispheres. While the water content of the hemispheres increased by 1.3%, that of the olfactory bulbs did so by 1.7% subsequent to the intraventricular administration of DDAVP. Measurement of the brain electrolyte content was not conclusive as to the mechanism of water permeability changes. The possible mechanism is discussed. Although no tissue or cerebrospinal fluid concentrations of vasopressin enabling comparison with clinical pathological conditions have been measured, it is suggested that increased secretion of vasopressin into the cerebrospinal fluid in conditions such as subarachnoid hemorrhage or intracranial hypertension of various origins might play a role in edema formation.

Studies on antibodies reactive with glycoproteins of primate type C viruses in patients with myeloid leukemias and with potential preleukemia.

Human blood plasma from patients with myeloid leukemias, potentially preleukemic disorders and healthy individuals contains antibodies which react with purified glycoproteins of the baboon endogenous virus and the gibbon ape leukemia virus. Experiments are presented which illustrate characteristic distributions of antiviral antibodies in the plasma of different investigated groups. The presence of high-titer antibodies is associated with remission of acute myeloid leukemia and longer survival of patients with preleukemia.
