Retrovirus-neutralizing antibodies in AML and AMMoL patients: stage-specific distribution.

Plasma samples of patients with AML or AMMoL were tested for antibodies reacting with gp70 antigens of BaEV and GaLV as well as for antibodies neutralizing BaEV or GaLV. Both frequency and titer values of antibodies were higher in remission than in blastosis. Neutralizing activity could be detected only in those plasma samples which contained antibodies to the appropriate gp70 antigen. The data suggest the presence of retroviruses in humans as antigenic stimuli for the immune system in AML and AMMoL.

Trends and patterns in cancer incidence in Czechoslovakia, 1968-1985.

Cancer incidence rates and trends in Czechoslovakia investigated in the present study are based for the first time on valid data derived mainly from population-based cancer registries. All rates were age-adjusted using world standard population as a reference. The results confirmed first of all a real and apparent increase of the whole cancer incidence trends which was more expressed in males. The increase in the overall cancer incidence among males could be attributed to the steadily continuing growth of lung cancer, responsible for one-quarter of all new cases in recent years, and to a lesser extent, also to the rising rates of "other skin", prostatic, colonic and rectal cancer. The rates and trends among females were influenced by increase and dominant position of breast and "other skin" cancer, responsible together for one-third of new cases. Increasing trends were observed for the incidence of the uterine body, ovary, and for the great majority of digestive tract and other cancer sites. A reduction of incidence was seen only for stomach in both sexes, for the uterine cervix among females, and lip cancer among males. The actual rates and trends of cancer incidence in Czechoslovakia could be compared with those observed in other developed countries. The usefulness and unconditional necessity of adequately reliable data on cancer incidence for epidemiological research and administration of cancer control is emphasized.

Demonstration of serotoninergic axon terminals on somatostatin-immunoreactive neurons of the anterior periventricular nucleus of the rat hypothalamus.

Using a combination of electron microscopic autoradiography and immunocytochemistry, the connections between serotoninergic axons and somatostatin neurons of the anterior periventricular nucleus of the rat hypothalamus were examined. The serotoninergic elements were identified after selective uptake of tritiated serotonin and the somatostatin neurons with immunocytochemistry. Synaptic connections between labeled serotoninergic nerve endings and somatostatin-immunoreactive neurons were observed. This finding provides morphological evidence for a direct influence of serotoninergic elements on somatostatin neurons of the anterior periventricular nucleus projecting to the median eminence of the hypothalamus.

Effect of ammonium chloride on the multiplication of infectious pancreatic necrosis virus.

The multiplication of IPNV in CHSE cells is inhibited by ammonium chloride. This inhibition is complete if NH4Cl is added early in the infective cycle. Immunofluorescence analysis shows that NH4Cl reduces the number of virus producing cells and thus suggests it may act by inhibiting virus internalization. In addition, NH4Cl can produce partial inhibition of IPNV multiplication if added later in the replicative cycle. Analysis of 3H-uridine incorporation into viral RNA shows that this later effect could be an inhibition of viral RNA synthesis.

Immunohistochemical localization of cells containing nerve growth factor receptors in the different regions of the adult rat forebrain.

Conditions have been optimized for an immunohistochemical procedure for the localization of nerve growth factor receptor-containing cells in the brain. Using this immunohistochemical procedure, the normal morphology and distribution of the nerve growth factor receptor-containing neurons of the adult rat forebrain have been studied, and the findings compared with observations on the choline acetyltransferase-positive neurons present either in the immediately-adjacent sections or in the medially-divided half of the same section. Unlike in the peripheral nervous system, only neurons showed immunoreactivity to the nerve growth factor receptor in the brain. Both the nerve growth factor receptor- and choline acetyltransferase-immunoreactive cells appear to form a continuous anteroposterior band, which includes the olfactory tubercle, the medial septal nucleus, the vertical and horizontal limbs of the diagonal band and the basal nucleus. In each subdivision of the basal forebrain, the topographic organization, the localization, the intensity of the immunoreaction and the total cell number of nerve growth factor receptor- and of choline acetyltransferase-immunoreactive neurons were strikingly similar, indicating that nearly all nerve growth factor receptor-containing cells were cholinergic neurons. However, in the striatum, only about half the number of the choline acetyltransferase-positive cells showed immunopositive reactions to the nerve growth factor receptor, and, also, in the nerve growth factor receptor-containing neurons the intensity of the reaction product was much less than the choline acetyltransferase immunoreactivity. In the neurons of the basal forebrain nuclei, the choline acetyltransferase immunoreaction product was uniformly distributed on the cell bodies, while the nerve growth factor receptor immunoreaction product was present also as intensely stained granules on the cell somata and the dendrites. The mean diameter and the mean cross-sectional area of the nerve growth factor receptor-containing neurons were least in the medial septal nucleus and were greatest in the basal nucleus, and showed a gradation in cell size going from the medial septal nucleus through the nucleus of the diagonal band and extending more posteriorly to the basal nucleus.

Interferon production in myelo- and lymphoproliferative diseases. I. Spontaneous interferon production in acute and chronic leukaemias.

Blood plasma interferon (IFN) of patients with acute myeloid (AML) and chronic granulocytic (CGL) and with acute and chronic lymphoid leukaemia (ALL and CLL) was measured by bioassay and characterized by neutralization with anti-human-IFN-alpha treatment. Elevated IFN-alpha level (60-125 IU/ml) was found in the quiescent phase of CGL as compared to the control samples (15 +/- 10 IU/ml). No significant differences could be found between plasma IFN of patients suffering from the blastic crisis of CGL and control persons. Analogous results were obtained in experiments with plasmas of AML patients, regardless the stage of the disease. Elevated IFN-alpha production (60-100 IU/ml) was found in patients being in the remission phase of T-cell ALL but only in one case in the progressive phase of the disease. No significant elevation of plasma IFN level was demonstrated in patients with O-cell ALL and B-cell CLL, as compared to the control samples.

Cytotoxic activity of lymphocyte subpopulations against autologous tumour cells in patients with myeloid leukaemias and preleukaemic disorders.

Autologous lymphocyte populations from different phases of chronic granulocytic leukaemia (CGL), acute myeloid leukaemia (AML) and preleukaemic disorders were compared for cytotoxic activity. 51Cr-release tests showed that T lymphocytes from the quiescent phase of CGL and from the remission phase of AML exerted cytotoxic activity against autologous tumour cells. Such activity was also found in patients with potentially preleukaemic haematological disorders characterized by cytopenia, but not in polycythaemia vera patients. In the majority of cases cytotoxic activity of T lymphocytes could be blocked by native gp70 antigens of gibbon ape leukaemia virus (GaLV) and baboon endogenous virus (BaEV). Blocking effect of carbohydrate-free gp70 as well as p15(E) antigens could be observed less frequently. The role of cell-mediated immune response to oncovirus antigens in the course of myeloproliferative diseases is discussed.

Synaptic connections between serotoninergic axon terminals and tyrosine hydroxylase-immunoreactive neurons in the arcuate nucleus of the rat hypothalamus. A combination of electron microscopic autoradiography and immunocytochemistry.

A combination of electron microscopic autoradiography and immunocytochemistry was used to examine the connections between serotoninergic axons and dopaminergic neurons of the arcuate nucleus of the rat hypothalamus. The serotoninergic elements were identified after selective uptake of tritiated serotonin and the dopaminergic neurons with tyrosine hydroxylase cytochemistry. Synaptic junctions between labeled serotoninergic nerve endings and tyrosine hydroxylase-immunoreactive neurons were observed, providing a structural basis for a direct influence of serotoninergic elements on tuberoinfundibular dopaminergic neurons.

Ependymal lining of infundibular recess in perinatal rats: relationships with portal capillaries and permeability.

Structure and permeability of the ependymal lining the infundibular recess were studied in perinatal rats with silver impregnation, electron microscopy, radioautography, and tracer techniques. According to our data basal processes of ependymal cells reach the primary portal plexus linking the 3rd ventricle and the hypophysial portal system all through the perinatal period. After birth, some of the processes penetrate into the perivascular space of the primary portal plexus and abut there on the endothelium of capillaries. Ependymal cells of fetuses and neonates are joined by specialized junctions (tight junctions, gap junctions and zonulae adhaerentes). Intraventricularly injected ionic lanthanum crosses the ependymal lining of fetuses both trans- and extra-cellularly everywhere in the infundibular recess. By postnatal day 9 only the rostral portion of the recess remains readily permeable. Caudally, extracellular leakage becomes highly restricted, apparently due to the appearance of circumferential tight junctions. Finally, [3H]dopamine seems to penetrate through the ependymal lining in the same way as ionic lanthanum entering the portal capillaries. These findings suggest that the adenohypophysiotropic neurohormones can penetrate from the cerebrospinal fluid into the portal circulation from the very beginning of the establishment of the hypothalamo-hypophysial functional relationships during ontogenesis.

Cytotoxicity of lymphocytes and antibodies against autologous tumor cells in patients with myeloid leukaemias and preleukaemic disorders. III. Stage-dependence of oncovirus-specific immune response.

Lymphocyte and plasma samples from the quiescent and blastic phase of chronic granulocytic leukaemia (CGL) and from the blastosis and remission of acute myeloid leukaemia (AML), were compared for cytotoxic activity. Target cells were collected from the blastic phases of diseases. 51Cr-release tests showed that the lymphocytes and plasma samples from blastic crisis of CGL had no cytotoxic activity for autologous blast cells. In contrast, cryopreserved lymphocytes and plasmas from the quiescent phase of CGL proved to be cytotoxic for the autologous tumor cells, and their effect could be blocked by native gp70 antigens of gibbon ape leukaemia virus (GaLV) and baboon endogenous virus (BaEV). A blocking effect was less frequently exerted by carbohydrate-free gp70 and p15(E) antigens. A similar relationship was found between the blastosis and remission stage of AML, however, out of the antigens of BaEV only the native gp70 showed a marked blocking effect.

Demonstration of serotoninergic axons terminating on luteinizing hormone-releasing hormone neurons in the preoptic area of the rat using a combination of immunocytochemistry and high resolution autoradiography.

The synaptic relationship between serotoninergic terminals and luteinizing hormone-releasing hormone-containing neurons was investigated in the medial preoptic area using a combined technique. Axon terminals selectively taking up 5-[3H]hydroxytryptamine were labelled autoradiographically and luteinizing hormone-releasing hormone-containing neuronal elements were identified by means of immunocytochemistry. Synaptic contacts were observed between tritiated 5-hydroxytryptamine-labelled boutons and luteinizing hormone-releasing hormone-immunoreactive dendrites. About 5% of the boutons which formed synapses with luteinizing hormone-releasing hormone-immunoreactive dendrites were found to be labelled by the tritiated indolamine. Luteinizing hormone-releasing hormone-immunoreactive axon terminals occurred as presynaptic elements in contact with unidentified dendritic spines, shafts or perikarya. These observations provide morphological basis for the idea that 5-hydroxytryptamine-containing neurons can act directly on luteinizing hormone-releasing hormone release. Further, they support the assumption that luteinizing hormone-releasing hormone is not only a neurohormone but may also function as a neurotransmitter or neuromodulator.

Detection of main core proteins of simian C-type viruses and human retrovirus HTLV and antibodies to them in patients with lymphoid malignancies.

Peripheral leukocytes or lymph node cells and blood plasma samples from patients with lymphoid malignancies were investigated for immunological markers of BaEV, GaLV and HTLV. Antigens and antibodies were shown with radioimmunoassay. Antigen related to the p30 core protein of BaEV could be detected in each cell type of leukaemias and lymphomas. Antigen related to the GaLV p30 was found mainly in B- and O-cell forms, while that related to the p24 protein of HTLV could be detected only in two T-cell malignancies. Antibodies reactive with these antigens showed a similar distribution.
