Changes in the structure and properties of corpus vitreous and the preservation of enzymatic composition of rabbit antioxidative system under the influence of sulphur hexafluoride gas.

PURPOSE: To evaluate clinical, physiochemical and biochemical changes in rabbit vitreous body caused by local injection of sulphur hexafluoride gas. MATERIAL AND METHODS: The volume of fluid vitreous fraction was measured with Na+, K+, Mg2+, Ca2+ levels and full proteins concentration in both vitreous fractions in 24 New Zealand rabbits at 2, 7 and 14 day after SF6 injection. The activity of superoxide dismutase, catalase and malonyl dialdehyde were used to evaluate the activity of antioxidative enzymatic system. Control group consisting of 6 New Zealand rabbits had no experimental procedures. RESULTS: In the investigated group, the fluid vitreous fraction volume was increased while gelatous one was diminished from 0.08 ml in control group to 0.32 ml in the study group (on day 14). The level of Na+, K+, Mg2+, Ca2+ in the fluid fraction was unchanged. On day 7, we noticed statistically significant increase in protein concentration in comparison with the control group and the study group on 14 day. The activity of superoxide dismutase and catalase as well as the level of malonyl dialdehyde were increased in the fluid vitreous fraction compared to the gelatous one in the control group. After the SF6 injection we did not observe any changes of superoxide dismutase and catalase activities in the gelatous part of vitreous body while in the fluid one there was statistically significant decrease in the enzymatic activity and the MDA level in the whole observation time. CONCLUSIONS: The injected sulphur hexafluoride gas caused the damage of the gelatous vitreous fraction with the increase in the fluid one. The oxygen free radicals might trigger these pathological processes.

Moderate systemic hypothermia and cold crystalloid cardioplegia influence on myocardial ischemic and revascularisative injury.

The number of granulocytes, their ability to generate superoxide anion (O2-) and the activities of Cu, Zn--superoxide dismutase (SOD-1), glutathione peroxidase (GSH-Px), catalase (CAT) as well as malonyldialdehyde (MDA) concentrations in erythrocytes in the blood extracted from the venous sinus and aorta under coronary artery bypass were examined with the use of St. Thomas Hospital cardioplegic solution. Specimens at the peak of ischemia of the right atrium for ultrastructural examination of the endothelial cells of capillary vessels and sarcomers were taken. The blood was obtained during cardiopulmonary bypass (CPB) before the aorta clamping and immediately after aorta declamping (peak of ischaemia) between 1-3 minute and 10-13 minute of reperfusion. Increase of the number of granulocytes both in the coronary sinus and aortal blood at all examined intervals as well as decrease in the number of ones in sinus compared with aortal blood was noted. The ability to produce superoxide anion radical decreased at the peak of ischemia and during reperfusion. The activity of SOD-1 was lower both after the period of ischemia and reperfusion. The increase in aortal blood activity during reperfusion was characteristic of GSH-Px; the activity was higher in the blood sample from the coronary sinus taken during ischemia and initial reperfusion. With the exception of the initial reperfusion the activity of CAT diminished in all observed cases. MDA concentration did not demonstrate any significant changes with the exception of the initial reperfusion in the aortal blood and later towards the end of reperfusion in the blood from the coronary sinus. Ultrastructural studies indicated overhydration of the cells both in the endothelium and the intercellular space. The obtained data demonstrate that the applied cardioplegic solution protects the myocardium from harmful effects of reactive oxygen species produced as a result of ischemia and reperfusion.

[The effect of suloctidil and acetylsalicylic acid on eicosanoid synthesis in human platelets].

The study aimed at comparing an effect of suloctidil and acetylsalicylic acid on malonyldialdehyde levels resulting from the arachidonic acid metabolism in blood platelets. It was shown that inhibitory effect of suloctidil is more potent than that of acetylsalicylic acid. Therefore the first is more inhibitor of an enzymatic metabolism of arachidonate in blood platelets than the latter.

[Effect of gamma radiation on selected indicators of oxygen metabolism in erythrocytes of patients with multiple sclerosis].

The aim of the work was to study certain parameters of aerobic metabolism in the erythrocytes of multiple sclerosis patients and healthy controls after exposure of the erythrocytes to ionising radiation. The group of patients included 32 cases (17 women and 15 men) aged 19-54 years. The control group comprised 60 healthy subjects (28 women and 32 men) aged 19-56 years. The erythrocytes were exposed at +20 degrees C in contact with air during 50 minutes. The source of gamma radiation was 60 Co of 1.33 MeV energy and 10 Gy/min yield. In the erythrocytes of the patients the activity of the antioxidative enzymatic system was reduced, with decreased activity of superoxide dismutase (SOD-1) and glutathione peroxidase (GPx) and with increased level of malonyl dialdehyde (MDA) at the same time gamma radiation as an additional source of free radical generation increased the changes of these biochemical parameters which suggested a greater susceptibility of multiple sclerosis erythrocytes to oxidative stress.

[Superoxide dismutase (SOD-1) activity in erythrocytes of patients with multiple sclerosis].

The purpose of the study was determination of the activity of superoxide dismutase (SOD-1) in the erythrocytes of multiple sclerosis patients taking into consideration certain aspects of the disease, such as its duration, clinical form and course, age and sex of the patients. The study included 34 patients (17 males and 17 females) aged 19 to 56 years. The control group comprised 64 healthy subjects (25 females and 38 males) aged 19-59 years. A significant reduction was found of SOD-1 activity in the erythrocytes of the patients which may suggest a lower enzymatic defense mechanisms against oxidative stress. No difference in the activity of SOD-1 was found between clinical forms and different courses of the disease, although the activity changed in relation to age, sex and disease duration.

The influence of haloperidol and propranolol on behavior and biochemical changes in the brain of mice treated with LSD.

The influence of haloperidol and propranolol on aggressiveness, motility exploration and general behavior, and the activity or level of adenylate cyclase, cyclic AMP, and protein kinase in the brain of mice treated with LSD was tested. Haloperidol evidently, and propranolol slightly less, inhibited the behavioral and biochemical changes induced by LSD. It is suggested that psychotomimetic effects of LSD depend on complex action of this compound on aminergic receptors in the central nervous system, and the antipsychotic effectiveness of haloperidol and propranolol is related to interaction of these drugs and LSD with the receptors for monoamines participating in the central neuromediation.

[Production of superoxide free radicals by peripheral blood leukocytes in patients with psoriasis treated by the Re-PUVA-C method].

The investigations were carried out in 20 patients with severe forms of psoriasis, and in 18 healthy subjects. The generation of free superoxide radicals by polymorphonuclear leukocytes of peripheral blood was evaluated by Bellavite et al. (1983) method by use superoxide dismutase of Sigma firm. The obtained results may confirm alterations in PMNL functions in patients with psoriasis observed by many authors. It may confirm the probability of damage of membrane structures and alterations in their bactericidal functions.

[Effect of the treatment with adrenocorticotropic hormone on the 2,3-diphosphoglycerate level in the erythrocytes of patients with multiple sclerosis].

The reported study was carried out in 20 patients with multiple sclerosis (12 women, 8 men) aged 23 to 54 years, with disease duration from 3 to 20 years. The patients were treated with the preparation ACTH-Polfa 2000 mg for one treatment course. The patients were divided into subgroups considering the age, disease duration, clinical form and clinical course of the disease. The control group comprised 22 healthy subjects (10 women and 12 men) aged from 22 to 41 years. The concentration of 2,3-diphosphoglycerate in the erythrocytes was determined by Bartlett's method twice: before and after ACTH treatment. Before the treatment the concentration of 2,3-DPG in the erythrocytes of the patients was not different from that in the controls, but after the treatment this concentration was significantly increased in all groups. It was demonstrated, furthermore, that 2,3-DPG concentration in the erythrocytes could change before ACTH treatment depending on the age of the patients and the clinical course of the disease.

The effect of acoustic stimulus of different continuity pattern on plasma concentration of catecholamines in humans during submaximal exercise.

The purpose of the study was determination of adrenaline and noradrenaline concentrations in peripheral blood in 13 men aged 20--22 years subjected to acoustic stimulation of continuous, intermittent and pulse pattern during graded submaximal exercise. The studied subjects performed physical exercise on a cycle ergometer at workloads of 2 W/kg of body mass. The noise produced during the exercise had peak acoustic pressure of 100--120 dB, at frequencies ranging from 63 Hz to 4000 Hz. The exposure to noise and exercise lasted 15 minutes. Catecholamines in the peripheral blood were determined by spectrofluorometry by the method of Anton and Sayer. The obtained results showed that the concentrations of adrenaline and noradrenaline increased considerably in the peripheral blood during exposure to this acoustic stimulation in relation to the initial concentrations. The highest increases were observed during exposure to impulse noise.

Exercise effect on the circadian rhythm of left ventricular systole in healthy men with higher and lower physical fitness.

The purpose of the present work was to investigate the effect of exercise on the diurnal changes of electromechanical systolic time (QS2) in healthy men with different physical fitness (N = 20, PWC150-804.3 +/- 114.6 kpm/min), and with higher (N = 12, PWC150-881.3 +/- 71.8 kpm/min) and lower (N = 8, PWC150-688.8 +/- 46.0 kpm/min) physical fitness. The study on 20 male volunteers aged 19-21 at 6 times of 24-hours period in intervals (4-hours) was performed. The policardiograms according to Blumberger at rest and after 10 minutes of exercise were obtained. The individual level of exercise at test PWC150 was determined. The values of QS2 from policardiograms according to Weissler's method were read and for heart rate were corrected. The mathematical analysis by the method of Halberg was made. It was found that in healthy men at rest the circadian rhythm of QS2 existed. The effect of exercise on the circadian rhythm of QS2 was dependent on level of physical fitness. After exercise in human with higher physical fitness the circadian rhythm of QS2 was presented, whereas in human with lower physical fitness it was not evident.

Adenine nucleotides and 2,3-dophosphoglycerate in the erythrocytes during physical exercise and restitution in healthy subjects.

The reported investigations were carried out in 6 young (20 years old) healthy males who performed a 10-minute exercise on a Monark cycle ergometer at workloads increasing the heart rate to 170/minute. Before the exercise, after its termination, and after 30 minutes of restitution the concentrations of adenosine-5' triphosphate, adenosine-5' diphosphate, adenosine-5' monophosphate, 2,3-diphosphoglycerate, nicotinamide-adenine dinucleotide and nicotinamide-adenine dinucleotide phosphate were assayed in erythrocytes. After the end of exercise a non-significant increase in ATP concentration and a non-significant decrease of 2,3-DPG were found in the erythrocytes. The concentrations of ADP and AMP were lower than at rest, both, after the end of exercise and after 30 minutes of restitution (p less than 0.05). The concentrations of nicotinamide-adenine dinucleotide and nicotinamide-adenine dinucleotide phosphate decreased after exercise (p less than 0.05) fell even more after 30 minutes of restitution (p less than 0.05).
