[Certain parameters of platelets oxygen metabolism in patients with multiple sclerosis].

The hypothesis of Mickel (1975) on multiple sclerosis suggested disturbances of lipid peroxidation in that disease. The purpose of this report was an analysis of oxygen metabolism in platelets in that disease, taking into account the peroxidation of lipids and antioxidant enzymatic protection against oxygen stress. In the reported study the concentration of malonyl dialdehyde (MDA) and the activity of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD-1) were determined in multiple sclerosis patients. The studied group comprised 20 non-smoking patients (9 men and 11 women) aged 21-61 years during disease exacerbation. The criteria of disease diagnosis were those suggested by Poser et al (1983). The obtained results may suggest abnormal acceleration of lipid peroxidation in platelets in this disease.

Effect of morphine and naloxone on oxidative metabolism during experimental renal ischemia and reperfusion.

During experimental renal ischemia and reperfusion in rabbits, morphine as well as naloxone significantly inhibited the increased superoxide anion (O2-) generation by resting and opsonized zymosan-stimulated phagocytes in renal venous blood. Morphine with naloxone in combination inhibited O2- generation to a lesser extent than that observed when these drugs were used separately. Morphine and/or naloxone did not significantly affect erythrocyte superoxide dismutase (SOD-1), glutathione peroxidase (GPx) and catalase activities or malonyldialdehyde (MDA) concentrations in venous blood during renal ischemia. During reperfusion there was a tendency to a slight reduction of erythrocyte catalase activity in morphine-treated animals, and to slight diminutions of erythrocyte SOD-1 and GPx activities and erythrocyte MDA concentrations in rabbits treated with naloxone and morphine in combination. These results indicate that opioid receptor agonists and antagonists modify the response of the kidney to acute injury. These effects may have relevance to the pattern of oxidative stress seen in patients with acute ischemic renal failure.

The effect of coronary angioplasty on superoxide anion generation and lipid peroxidation. Propriety of pharmacological intervention.

In patients with unstable angina pectoris, subjected (n = 20) or not subjected (n = 12) to percutaneous transluminal coronary angioplasty (PTCA), baseline superoxide anion (O.2-) generation by neutrophils in the coronary sinus blood was significantly higher than that found in the basilic vein blood of control healthy subjects (n = 12). During reperfusion following effective PTCA, neutrophil counts in the coronary sinus blood tended to decrease, an effect accompanied by a significant decrease in the neutrophil O.2- generation and enhancement of blood plasma lipid peroxidation as reflected by increased malonyldialdehyde concentrations.

Effect of the H2 histamine receptor antagonist on oxygen metabolism in some morphotic blood elements in patients with ulcer disease.

BACKGROUND/AIMS: Our investigations was carried out in order to examine the effect of cimetidine, ranitidine and famotidine on the generation of free radicals, lipid peroxidation and enzymatic antioxidative defense in the blood of patients with peptic ulcer disease, clinically diagnosed as gastric or duodenal ulcer. METHODOLOGY: 124 non-smoking males (aged 20-51 years), were randomly divided into 4 groups: 28 patients received intravenously 200 mg of cimetidine; 26 patients intravenously 50 mg comprised of ranitidine; 25 patients received intravenously 20 mg of famotidine; and 45 healthy men served as the control group. Superoxide dismutase activity, malonyldialdehyde concentration in blood platelets and superoxide anion generation in granulocytes were determined in all examined men. An assay of superoxide dismutase activity and malonyldialdehyde concentration were performed before drug administration and after 2 and 72 hours. Superoxide anion generation was estimated before drug administration and after 2 hours. RESULTS: Our data indicate that all examined H2 receptor antagonists stimulate superoxide dismutase activity, but after 72 hours a distinct increase was observed, in addition to a decrease of malonyldialdehyde concentration. No differences have been observed in superoxide anion generation in patients with ulcer disease or in healthy subjects before and after ranitidine and famotidine administration. Only after 2 hours of cimetidine administration was a significant increase in superoxide anion generation observed. CONCLUSION: We concluded that H2 receptor antagonists have a beneficial effect on antioxidative processes.
