[Pharmacokinetics and effects of xylazine (Rompun) in dogs].

Six beagle dogs were treated with xylazine hydrochloride (1 mg/kg i.m.). The plasma xylazine concentration was measured by HPLC. Additionally, clinical effects were registered (cardiac rate, respiratory activity, electrocardiogram, body temperature, motoric activity, attention, analgesia). Maximum plasma concentrations were measured after 15 minutes (476 ng/ml). The plasma half-life was 24 minutes. Sedation was registered over one hour (xylazine concentration of more than 150 ng/ml). Within the first 30 minutes after treatment (xylazine concentration of more than 300 ng/ml), a low-grade analgesia was observed. In contrast, cardiac and respiratoric depression and also significantly diminished body temperature were registered over 2 to 3 hours.

[Pharmacokinetics and tolerance of an orally administered combination preparation containing phenylbutazone and prednisolone in the dog].

Tolerance and pharmacokinetics of a combination of phenylbutazone and prednisolone (tablets) were studied in Beagle dogs. For two weeks, the drug was administered orally (3.33 mg/kg phenylbutazone combined with 0.1 mg/kg prednisolone and 6.66 mg/kg phenylbutazone combined with 0.3 mg/kg prednisolone twice daily). The dosage was reduced to 50 per cent after one week. No signs of intolerance were found. After treatment with 3.33 mg/kg phenylbutazone, a plasma concentration of 10 micrograms/ml was reached within 1 to 2 hours. No prednisolone concentration above 2 ng/kg was measured. Additionally, a synergistic antiinflammatory effect of the two ingredients was obvious (hind paw dextran edema in rats). In addition to results of ENGELKE et al. (1995) who studied the clinical efficacy of the combination, the presented data demonstrate a good tolerance.

[The treatment of pseudopregnancy in the bitch with prolactin inhibitors metergoline and bromocriptine].

In the present study, the prolactin inhibitor Metergoline was compared with Bromocriptine and tested against a placebo in 63 pseudogravid bitches. Bromocriptine has already been tested successfully in numerous investigations on the therapy of canine pseudogravidity, but--probably because of its high price and vomitus as a frequent side effect--it has not been really introduced as a therapeutical device in canine practice. It can be deduced from the results presented herein that prolactin is essential for maintaining the pseudogravidity, but keeping up the lactation process--especially galactopoiesis--can probably not be ascribed solely to prolactin. However, prolactin definitely plays an essential role in the hormonal scenario, the detailed regulating mechanisms of which are not known until today. Thus, no statistically convincing therapy outcome could be achieved by the prolactin inhibitors compared to the placebo group. A tendency towards earlier regression of the symptoms "mammogenesis", "behavioural change" and "galactorrhea" was however present in the treated animals. A striking difference was the much more lively behaviour of the bitches with 53% being more lively in the Metergoline group, 37% in the Bromocriptine and 10% in the placebo group. There were also clear differences in the compatibility of the drugs; in the Bromocriptine group, 30% of the animals vomited, in the Metergoline only 6.3%. This however did not lead to termination of the therapy in any case. In two cases of the Metergoline group (6.3%), the medication was ended due to extreme restlessness.

[Scintigraphic study for the diagnosis of portosystemic shunts in dogs].

The technique of scintigraphy of the liver is presented by two case examples of dogs with portosystemic shunts. 99mtechnetium-pertechnetate was used as a marker. Comparison of velocity and height of the activity reaching liver and heart and lung allows a statement about the existence of a portosystemic shunt.

[Kidney scintigraphy in dogs and cats using 99mTc-MAG3].

Renal scanning is one of the imaging techniques used to investigate the upper urinary tract. In the present examination, the principles of performing renal scanning in the dog and car are described. Eleven animals with healthy kidneys (six dogs and five cats) were included in the study for the determination of normal scintigraphic findings as well as eight animals (seven dogs and one cat) with renal damage. The following nephropathies were investigated scintigraphically: nephrolithiasis, hydronephrosis, polycystic renal degeneration, renal aplasia, renal hypoplasia, obstruction of the ureter and renal trauma. Renal scanning proved to be easy to perform with acceptable effort in the dog and cat. It allows the simultaneous assessment of renal function on both sides during the phases of renal perfusion and tubular se- and excretion and represents a valuable adjunct to the morphologically orientated radio- and sonography. The nephrogram curves are comparable to those found in man, although the time until reaching the maximum of the curves and elimination half times are shorter. Also, the sequential scintigrams can be divided into the corresponding phases as in man. Through permanent technical development, today there are gamma processors at disposal that allow a faster, comprehensive and sophisticated evaluation. In the present investigation, this was demonstrated by a comparison of two different calculators. However, due to the financial and technical expenditures and the special radiological safety precautions required, renal scanning will be a technique reserved to larger clinics.

[Validity of diagnostic methods for kidney function tests in the cat].

The diagnosis of kidney disease is difficult in the stage of compensation and impossible based solely on the routinely performed laboratory tests on blood and urine. For this reason, more sensitive methods are required. In the present study, three special techniques are compared with regard to their validity in the early diagnosis of kidney disease in the cat: 1. the molecular-weight related separation of urine proteins with the sodium-dodecyl-sulfate-polyacrylamide-gradient gel electrophoresis (SDS-page) in the PhastSystem, 2. measurement of the glomerular filtration rate (GFR) with the renalyzer PRX90 using an iodine containing contrast medium and 3. kidney scintigraphy. The results of this comparison demonstrate that these procedures are important adjuncts to common laboratory investigations in the testing of renal function. The SDS-page allows an early qualitative assessment on alterations of specific functional compartments of the kidney. However, it is not possible with this method alone to evaluate the degree of renal disturbance and it does not give information concerning the severity of renal functional impairment. Measurement of the GFR is also a valuable procedure which gives a quantitative result on the global renal function within a few hours. It is of special importance when subclinically disturbed kidney function is present. In the cat however it is until now not possible to give a correct prognosis in high grade nephropathies. Only scintigraphy allows unilateral assessment of renal function, which is most important in cats with morphologically altered kidneys, such as kidney cysts, hydronephrosis or tumours.

[Maintenance of the cranial cruciate ligament rupture in the dog using an intra-articular ("over-the-top") and an extra-articular ("fascia-doubling") method].

The present study compares the results of an intraarticular ("over-the-top") and an extraarticular (fascia lata imbrication) surgical method to treat the rupture of the cranial cruciate ligament in dogs. In a period of 18 months, 132 stifle joints of 125 dogs with a partial or complete rupture of the cranial cruciate ligament were examined according to a systematic protocol at the Clinic for Small Animals of the Hannover School of Veterinary Medicine, Hannover, Germany. Surgery was performed with either one or the other of the two surgical techniques. In all cases a " cleaning up " was carried out. If the medial meniscus was damaged, it was resected and the lesion was investigated. The imbrication technique of the lateral aspect of the fascia lata was easier and faster to perform. Using the "over-the-top" method, there were difficulties to guide the fascial strip through the intercondylar notch. After six months 88 patients were re-examined clinically and radiologically according to the same protocol as mentioned above. In the present study it was demonstrated that patients treated with the extraarticular method showed a higher rate (76.5%) of normal limb function after six months and walked earlier without lameness compared with dogs operated with the "over-the-top" technique (41.5%). The same results were valid for patients of giant breeds. Patients operated with the imbrication technique showed more arthrosis of the patellar tip. After six months stability, evaluated with the cranial drawer test and the tibia compression test was satisfying with both techniques.

Investigations of platelet aggregation and platelet counts from stored canine whole blood.

The effects of the long term storage of canine blood at 4 to 6 degrees C in PVC-bags containing CPDA-1 as a stabiliser on platelet aggregation and platelet counts were investigated. Aggregation induced by collagen, adenosine diphosphate or a calcium ionophore was preserved well during the first six hours, but there was then a decrease of 24 to 46 per cent in the ability of the platelets to aggregate, after which during the next three to four weeks of storage there was no further decrease in their aggregation properties. The formation of aggregates of platelets reduced the numbers of platelets counted as single thrombocytes by more than 30 per cent during the first four days. The numbers of platelets recorded varied widely with the counting method used (counting chamber or automatically) and with the ethylenediamine tetra-acetic acid content of the dilution fluid.

Long-term treatment of diseased, FIV-seropositive field cats with azidothymidine (AZT).

The long-term effectiveness of Azidothymidine (AZT, Zidovudine) was evaluated in FIV-seropositive cats with clinical symptoms (n = 9; Group 1) compared with conventional symptomatical therapy (n = 5; Group 2). The oral administration of Azidothymidine at a dosage of 5 mg/kg BW yielded a mean peak plasma concentration of 4.59 microM one hour after application (median: 3.74 microM). Elimination half time was 1.5 h. The permanent oral application of Azidothymidine at a dosage of 5 mg/kg BW TID led to a total recovery from clinical symptoms in six of nine FIV-seropositive cats (Group 1) 4-6 weeks after the onset of therapy. One cat clinically improved with only sporadical recurrence of disease; therapy with Azidothymidine was not effective in two cats. All the FIV-seropositive cats treated symptomatically responded well to antibiotics and immunomodulators within 10-14 days (n = 5; Group 2). Recurrence of clinical symptoms was noticed in three of five patients within 2 years after therapy and one cat died. During the treatment with Azidothymidine hyperproteinemia and abnormal albumin-to-globulin ratio became normal within 6 months in four FIV positive cats (Group 1), whereas hyperproteinemia did not change in Group 2. The following adverse effects were noticed in Group 1: a transient decrease of red blood cell count (RBC), packed cell volume (PCV) and haemoglobin until the 4th week of application of Azidothymidine (5/8), but the haemogram was within the normal range after 3 months. In one cat with hyperglycemia the anaemia remained until the administration of Azidothymidine was terminated. Heinz (Schmauch) bodies in the erythrocytes appeared in two FIV-positive cats 2 weeks after the onset of the therapy.

[Effect of acetylsalicylic acid on platelet aggregation and capillary bleeding in healthy cats].

The influence of acetylsalicylic acid (ASA) in vitro and in vivo on several parameters of the hemostatic system was evaluated in healthy cats. The aggregation response of cat platelets in vitro was not influenced by 10 micrograms ASA/ml plasma, whereas ASA concentrations of 35, 70, 100 micrograms/ml and of 1 mg/ml consistently inhibited the platelet aggregation induced by 0.1 (n = 4 cats) and 0.25 micrograms collagen/ml (n = 6 cats). The aggregation response to ADP and thrombin was partially (n = 2) or completely (n = 8) inhibited by an ASA concentration of 5 mg/ml. Administration of ASA at a dosage of 10 and 25 mg/kg BW orally and intravenously yielded a plasma concentration of salicylic acid of 30-42 micrograms/ml (10 mg/kg BW) and 50-70 micrograms/ml (25 mg/kg BW). All the dosages of ASA produced an inconsistent inhibition of the collagen-induced platelet aggregation (1/5 and 2/5 for the respective groups), whereas the response to ADP and thrombin was not altered. Capillary bleeding time was not significantly altered after the ASA administration. Platelet count and screening tests were not influenced by ASA application. According to the results of this study, ASA as a platelet aggregation inhibitor is not useful for the prophylaxis of thromboembolism in cats.

[Measurements of the activity of the coagulation factors VIII:C, IX,XI, and XII in cats].

Using an optimized test, reference ranges were established for the activity of the coagulation factors VIII:C (72-124%), IX (81-130%), XI (69-134%) and XII (51-142%) in the cat (n = 58 cats, 2.5-97.5% quantile). Compared to a reference curve prepared by using a cat-pool plasma (n = 50), the factor VIII:C activity in humans (commercial human reference plasma) was 7.6%, i.e. the factor VIII:C activity in cats was 13 fold higher than in humans. The values for the factors IX, XI and XII depended distinctly on the dilution step of the human plasma. The activity of the cat in relation to humans was approximately 90% (factor IX), 170% (factor XI) and 135% (factor XII). Two clinically healthy cats who were conspicuous pre-operative because of a distinctly prolonged, activated partial thromboplastin time showed a decreased factor XII activity.

[Applicability of activated partial thromboplastin time (APTT) as a screening test of milk to medium deficiencies of coagulation factors in cats].

The present investigation examined if the aPTT shows sufficient sensitivity for single factor activities also in cats when measured with the test optimized for humans using a commercial reagent. Comparative measurements were done with different modifications of the aPTT (sample predilution, addition of fibrinogen). Measurements of the aPTT using different methods and the activity of the coagulation factors II, V, X, VIII, IX, XI, and XII were performed in 42 healthy cats in order to determine the reference ranges. The same measurements were done on 21 cat plasmas where at least one of the coagulation factors was diminished in relation to the corresponding reference range. The conventional aPTT reflected the decrease in coagulation factor activity in each of the 21 plasmas by prolongation above the reference range (14.6-24.4 s). The test thus possesses high sensitivity and is a suitable screening test also for the cat. In contrast, in tests with sample predilution the sensitivity was lower (3-4 false negative results). This was probably caused by the distinct increase of the range of the reference values.

Diagnostic value of indirect pancreatic function test in serum of anuric patients with chronic renal failure.

Although patients with chronic renal failure have a high incidence of chronic pancreatic disease, the condition is frequently overlooked. We have modified the pancreolauryl test--an indirect pancreatic function test--for anuric patients. The test permitted good discrimination between patients with chronic pancreatic disease and those with a normal pancreas when serum levels of fluorescein were measured 10 h after administration with a standard meal. The sensitivity at this time interval was 80% and the specificity 83%. We conclude that the pancreolauryl test with serum measurements provides a simple, noninvasive, and reliable diagnostic test for chronic pancreatic disease in anuric patients with chronic renal failure.

A study on the efficacy of a phospholipid solution in dogs with aspirin-induced thrombocytopathy.

The aim of the present study was to test the haemostyptic properties of a phospholipid complex with platelet factor-3 (PF-3) activity in platelet-dependent haemostatic disorders. The substance was investigated in an animal model comprising six investigational groups (five dogs each). A thrombocytopathy was created in the dogs belonging to groups 1-5 by intravenous administration of 20 mg/kg body weight (BW) acetylsalicylic acid (ASA). Two hours later a human albumin solution (5%) (control group) or a phospholipid complex with PF-3 activity was injected or infused intravenously in different dosages. In dogs pre-treated with ASA, injection of 2 ml/kg BW of the phospholipid complex shortened capillary bleeding time, which was prolonged as a consequence of ASA-treatment. This effect lasted for 4 h at least. The capability of the platelets to aggregate increased 5 min after intravenous injection of the phospholipid without differences in the respective groups. At the same time platelet counts dropped to approximately 50%, but increased again distinctly after 30 min. When the phospholipid complex was administered to clinically healthy dogs who had not been treated with ASA, this resulted in prolongation of the capillary bleeding time as well as a significant platelet drop in the peripheral blood. Although these were only short-term effects after administration of the phospholipid complex, a disadvantageous effect cannot be excluded in patients suffering from haemorrhagic diathesis. For this reason, a phospholipid complex with PF-3 activity cannot be recommended as a therapeutic agent for platelet-dependent coagulation disorders in the dog.

Histological and immunohistological classification of canine glomerular disease.

A histological and immunohistological study of the kidneys of 115 dogs, with and without clinical signs of spontaneous renal disease, was performed to prove the applicability of the WHO criteria for the classification of human glomerulopathy. Aside from the morphological investigation of paraffin and resin semithin sections, deposits of immunoglobulins, the complement component C3, and fibrinogen were observed immunoenzymatically in paraffin-embedded tissue specimens. From this, eight different types of glomerular lesions with various frequencies were identified: minor glomerular abnormalities (28 cases), focal and segmental hyalinosis and sclerosis (12 cases), focal glomerulonephritis (GN; 18 cases), diffuse membranous GN (nine cases), diffuse mesangial proliferative GN (2 cases), diffuse endocapillary proliferative GN (five cases), diffuse mesangiocapillary GN (25 cases), diffuse sclerosing GN (11 cases) und unclassified GN (two cases). In one case, renal dysplasia was diagnosed and two dogs did not present glomerular alterations. The results are discussed with regard to human glomerular diseases and pathogenic mechanisms.

Evidence that metacentric and submetacentric chromosomes in canine tumors can result from telomeric fusions.

We have hypothesized that metacentric and submetacentric chromosomes frequently observed in malignant canine tumors are a result of telomeric fusions. Therefore cells from a canine mammary pleomorphic adenoma were transformed with a plasmid containing the SV40 'early region', known to cause telomeric associations. Compared with non-transformed adenoma cells, the cells had a higher proliferative capacity and expressed the large SV40-T-antigen. Karyotype studies showed the conversion from a normal to an aberrant karyotype with an increase of bi-armed chromosomes resulting from fusions of acrocentric chromosomes. In addition, the length of the telomeric repeats (TTAGGG) was determined for an early and a late passage of the transformed cells by Southern hybridization. The length of the telomeric repeats was apparently longer in the 5th than in the 38th passage. In situ hybridization with a telomere-specific DNA revealed interstitial telomeric repeats in the bi-armed chromosomes. We have concluded that these findings reflect the clonal expansion of head-to-head-telomeric fusions of canine acrocentric chromosomes leading to dicentric chromosomes with a very short distance between the two centromeres. Our results support the idea that the apparent centric fusions that have been described in some canine tumors may in fact be the cytogenetic products of head-to-head-telomeric fusions.

[Optimized methods for measuring coagulation factors II, IV, VII and X in cats].

Since the single factor activity of the cat differed in part distinctly from humans (factor II: 94%, factor V: 500%, factor VII: 125%, factor X: 62%) the method had to be optimized, above all to exclude an influence of a reactive increased factor V-activity on the results of the other coagulation factors. Dilution series of cat pool plasma (CPP) (n = 50 cats) showed, that a commercial human placenta thromboplastin was more suitable as activating reagent than rabbit brain thromboplastin. Using the required sample predilution (1:40), measurements with the latter reagent were more precise when measured on the relation between the imprecision and the time interval between different CPP-dilution steps. Reference curves were prepared for CPP using the optimized method and human placenta thromboplastin (50 microliters 1:40 prediluted sample and 50 microliters deficient plasma were incubated at 37 degrees C for 1 minute, addition of 200 microliters Ca-thromboplastin) and reference ranges were calculated using the values of 57 cats (factor II: 74-126%, factor V: 42-184%, factor VII 56-150%, factor X: 65-143%).

Different cytogenetic findings in two clinically similar leukaemic dogs.

A 4-month-old German Shepherd puppy and a 7-year-old Boxer dog, in which a clinical diagnosis of acute lymphoblastic leukaemia had been made, were investigated cytogenetically. In the puppy a diploid karyotype was found, but the malignant clone in the Boxer was characterized by an extra metacentric chromosome, apparently formed as the result of a Robertsonian translocation of two chromosomes 1. These findings are discussed against the background of similar cases reported in the literature. Possibly, different types of acute canine leukaemic disease, occurring at different ages, can be distinguished by cytogenetic evaluation.
