Production of 3-acetoxyscirpene-4,15-diol from anguidine (4,15-diacetoxyscirpene-3-ol) by Fusarium oxysporum f.sp. vasinfectum.

Growing cells of Fusarium oxysporum f.sp. vasinfectum (ATCC 7808) formed 3-acetoxyscirpene-4,15-diol from anguidine (4,15-diacetoxyscirpene-3-ol) by way of the intermediates triacetoxyscirpene, 3,4-diacetoxyscirpene-15-ol and 3,15-diacetoxyscirpene-4-ol. The new 3-acetoxy analog was found to be less active than anguidine and the other monoacetoxy derivatives when tested against a series of fungal strains and against HeLa cells in vitro.

Cytomegalovirus strain differentiation by DNA restriction analysis.

The heterogeneity of CMV DNA obtained from standard strains and new isolates, including a vaccination strain (Towne 125), was investigated. The cleavage patterns produced by the restriction endonucleases Eco RI and Bam 1 revealed stable strain specificities of CMV. On the other hand, a remarkable homology of sequence-specific CMV DNA fragmentation was demonstrated. A CMV subtyping relevant to clinical questions seems to be improbable.

[Serodiagnosis of cytomegalovirus -- (CMV) -- infections].

Specific IgM-antibodies to cytomegalovirus had been detected by both immunofluorescence and enzyme immunoassay. Using isolated nuclei instead of whole cells nonspecific reactions could be markedly reduced. -- IgM antibodies to cytomegalovirus were found in 7% of all pregnant women studied. Congenital infections could be demonstrated almost exclusively among children borne to mothers who had specific IgM antibodies during pregnancy.

[Cimetidine in the treatment of duodenal ulcer (author's transl)].

In a double-blind trial of patients with duodenal ulcer at three centres the healing rate after a four-week course of treatment (1 g cimetidine daily) was 79% (30 out of 38 patients), compared with 58% (23 out of 40 patients) on placebos. Cimetidine did not effect subjective symptoms or the need for antacids to relieve pain, except for a minor beneficial effect on diurnal ulcer symptoms during the second week of treatment. Results of long-term administration suggest that cimetidine at 400 mg daily prevent s relapses, but the possibility of an increased relapse rate after cimetidine withdrawal can not as yet be excluded.

Improved detection of virus-specific IgM antibodies. Elimination of non-specific IgM binding.

A non-specific reaction between human IgM and cytoplasmic structures of virus infected cells can often be observed if MgM antibodies to virus antigens are detected by indirect immunofluorescence or by immuno enzyme assays. Formaldehyde selectively inactivates the cytoplasmic receptors for human IgM without affecting the virus structural proteins. Alternatively, receptor-free antigens can be obtained by isolation of nuclei from virus infected cells. Due to reduced background, a more specific and more sensitive detection of IgM antibodies to Epstein-Barr virus, cytomegalovirus or central European encephalitis virus is possible.

Microbial and chemical transformations of some 12,13-epoxytrichothec-9,10-enes.

Resting cells of Streptomyces griseus, Mucor mucedo, and a growing culture of Acinetobacter calcoaceticus when mixed with compounds related to 12,13-epoxytrichothec-9-ene-4beta,15-diacetoxy-3alpha-ol(anguidine) produced a series of derivatives that were either partially hydrolyzed or selectively acylated. These derivatives showed marked differences in activities as assayed by antifungal and tissue culture cytotoxicity tests.

[Chemically induced carcinogenesis of stomach of the rat after vagotomy and resection (author's transl)].

180 male Wistar-rats were daily exposed to 30 and 100 mg MNNG/1000 ccm tapwater. After 7 weeks exposure a truncal vagotomy with pyloroplasty or a Billroth-II-resection was performed in 25 animals of each dosage-group. 80 rats were not operated. The incidence of carcinomas in the Billroth-II-rats of both dosage-groups was evidently higher than in the exposed controls. In the vagotomized rats an increased occurrence of carcinomas could not be observed.

Cytomegalovirus as a frequent cause of Guillain-Barré syndrome.

In 10 patients with Guillain-Barré syndrome a preceeding cytomegalovirus infection could be demonstrated by virus-specific IgM antibodies that were present in high titers in 9 of the 10 patients in the first serum specimen. The IgM antibodies to cytomegalovirus were detected by a sensitive "double" indirect immunofluorescence technique. In most of our cases (8 of 10) the complement-fixing antibody titers had already reached high levels on admission into a hospital, and significant titer changes were not observed. Cytomegalovirus was isolated from the urine of five patients.

IgM antibodies to cytomegalovirus during pregnancy.

IgM antibodies to cytomegalovirus (CMV) could be detected in about 7 per cent of 629 pregnant women whereas in 225 nonpregnant control women of similar age distribution only 2.6 per cent showed CMV IgM antibodies. Intrauterine CMV infections were almost exclusively detected among the CMV IgM positive gravides. The high incidence of CMV IgM antibodies in pregnant women can be possibly explained by an increased rate of CMV reactivations during pregnancy. We were able to show that during CMV reactivation an intrauterine infection might occur.

Solid-phase enzyme immunoassay for immunoglobulin M antibodies to cytomegalovirus.

A sensitive, solid-phase enzyme immunoassay for the detection of immunoglobulin M antibodies to cytomegalovirus is described. The results of the enzyme immunoassay correlated well with those obtained by an indirect immunofluorescence method. Horseradish peroxidase proved to be a more sensitive label than alkaline phosphatase. Nonspecific reactions, occurring with commercially available cytomegalovirus antigens, could be avoided by using a nuclear antigen prepared from sonically disrupted nuclei of cytomegalovirus-infected cells.

Simple mathematical deductions in the seroepidemiology of viral infections. I. Herpesvirus group (herpesvirus hominis, varicella-zoster virus, cytomegalovris, Epstein-Barr-Virus).

Large samples of nonselected persons collected in South-West Germnay were investigated for the prevalence of serum antibodies to the human Herpesviruses HSV, VZV, CMV and EBV. According to "catalytic models", which compare the infection spread to simple chemical reactions of molecules, a mathematical approximation of the serum surveys was performed. Through a new deduction of the exponential function y = k (l - e-r+) a simple way was found to estimate the annual attack rates in percent of the susceptible, seronegative people. While it was possible to represent the prevalence of serum antibodies to VZV by a continuous curve, a biphasic curve to the antibody prevalence rates in the epidemiology of the other Herpesviruses proved to be more adequate indicating changes in hormonal balance and social behaviour. The use of the epidemiologic parameters k and r for the characterization of the test sensitivity was examined for CMV. By evaluation of the mean antibody titres to CMV, HSV, and EBV throughout different age groups, information about the reactivation of the Herpesvirus diseases could be obtained.
