Fibrinolytic activity in periodontal disease. The relationship between fibrinolytic activity and severity of periodontal disease.

IN RECENT YEARS, evidence has evolved that suggests a relationship between fibrinolytic phenomenon and various inflammatory diseases. Plasmin activity and the response of the fibrinogen and fibrin degradation products latex test (FDPL test) in gingival fluid, along with plasminogen activator activity in gingival tissue were assessed. Samples were taken from periodontally involved persons and compared with healthy individuals. The following conclusions could be drawn: (1) The fluid from inflamed gingival crevices possessed more plasmin activity than that from clinically healthy crevices. The reaction of the FDPL test was stronger in the former. (2) Plasminogen activator activity in gingival tissue showed no significant difference between periodontally involved and healthy subjects.

Resistance to immune cytolysis induced by 4NQO and MNNG: a possible mechanism involved in chemical carcinogenicity.

L1210 cells were cultured for 20 hr in the presence of 4NQO or MNNG. Both agents rendered L1210 cells resistant to immune cytolysis by rabbit antibody and guinea pig complement. This resistance to immune cytolysis was not due to a loss or a distribution change of antigenic determinants to rabbit antibody, since the treated cells retained the capacity to consume guinea pig C4 in the presence of rabbit antibody to the same extent as untreated L1210 cells. Because acyltransferase activity to incorporate free fatty acids into phospholipids and neutral lipids of L1210 cells increased after the 4NQO and MNNG treatments, it was suggested that there might be a close relationship between the increased resistance of L1210 cells to immune cytolysis and the increase in their acyltransferase activity.

Sendai virus infected cells are readily cytolysed by guinea-pig complement without antibody.

HeLa cells infected with Sendai virus (SV) acquired the ability to activate the alternative pathway (AP) of guinea-pig complement without antibody reaction. For induction of complement activating ability (CAA), at least 2 hr incubation of the infected cells was required. However, ultraviolet (UV)-irradiation (8400 erg/mm2) of SV did not impair the inducibility of CAA and 51Cr-labelled HeLa cells infected with UV-irradiated SV (UV-SV) became readily cytolysed by C4-deficient guinea-pig complement without antigen-antibody reaction. This phenomenon may represent a primary in vivo defence mechanism against SV infection by eliminating the virus-infected cells via activation of complement on the cell surface. Although SV-infected HeLa cells activated the AP of guinea-pig complement, they did not activate the AP of human or mouse complement. The species restriction in the CAA induced by SV infection may result in different pathological manifestations in virus-infected animals and may affect their susceptibility to this type of infection.

[Fundamental and clinical studies on cefotaxime in the field of obstetrics and gynecology (author's transl)].

Fundamental and clinical studies were made on cefotaxime (HR-756, CTX), a new cephalosporin antibiotic, with the following results. 1. Cefotaxime was given to 22 patients. Efficacy was excellent in 3 cases, good in 17 cases and poor in 2 cases. No side effects were observed in any cases. 2. Following a single intravenous injection of 1.0 or 2.0 g, the transfer of CTX to the internal genital organs was found to be good. The transfer of CTX to exudate of the dead space of pelvis was also good. 3. The above data demonstrated that CTX is a safe and effective drug.

[Clinical evaluation of pivmecillinam in the maintenance therapy of chronic urinary tract infection (author's transl)].

In order to evaluate the efficacy and safety of pivmecillinam (melysin tablet, PMPC), PMPC was administered to 78 chronic UTI cases in the field of obstetrics and gynecology (posthysterectomy infection, chronic cystitis, chronic pyelonephritis and etc.). In principle, daily 400 mg of PMPC was administered for 2 weeks. (1) Overall clinical efficacy judged by doctor was evaluated in 78 cases and the result was; excellent in 17, good in 37, fair in 10, poor in 13 and unknown in 1 case with the effectiveness rate of 69.2%. (2) Overall clinical efficacy judged by 'criteria for clinical evaluation in complicated UTI' recommended by UTI study member was evaluated in 54 cases and the result was; excellent in 15, good in 20 and poor in 19 cases with the overall efficacy rate of 64.8%, the result of which was similar to that of doctor's judgement. (3) Efficacy on pyuria was evaluated in 72 cases and it was cleared in 27, decreased in 25, unchanged in 20 and unknown in 6 cases. Efficacy on bacteriuria was evaluated in 72 cases and it was eliminated in 44, decreased in 9, replaced in 8, unchanged in 8 and unknown in 9 cases. (4) Side effect, considered by doctors to be caused by PMPC administration, was noticed in 3 out of 78 cases (3.8%), all of which was mild gastrointestinal disturbance and the administration of PMPC was continued. Abnormal change of laboratory finding considered by doctors to be caused by PMPC administration was noticed in 1 out of 78 cases, which was slight elevation of GOT and GPT values. It is therefore considered that PMPC appear to be useful drug for the maintenance therapy of chronic UTI in the field of obstetrics and gynecology.

[Clinical studies on cefoperazone in gynecological and obstetrical field (author's transl)].

Fourteen patients suffering from female genital organ infections including mastitis and urinary tract infections were treated with cefoperazone (CPZ). CPZ was administered intravenously or by drip infusion at a dose of 2.0-4.0 g/day for 3-7 days. Clinical results obtained were as follows: Of 8 patients with genital organ infections, excellent responses were seen in 2 patients, good responses in 5 patients. Of 3 patients with mastitis, good responses were seen in all the patients. Of 3 patients with pyelonephritis, excellent and good responses were seen in all the patients. No adverse effects and abnormal laboratory findings were observed. From the above results, we concluded that CPZ was an effective and safe antibiotic against infections in the field of obstetrics and gynecology.

[A case report: Osler-Weber-Rendu disease with cerebral aneurysm, cerebral arteriovenous malformation and pulmonary arteriovenous fistula (author's transl)].

Osler-Weber-Rendu disease (hereditary hemorrhagic telangiectagia) is a rare vascular disorder which is characterized clinically by inherited telangiectasia of skin, mucous membrane and recurrent hemorrhage. However, it is not rare that various organs, for instance, lung, liver, intestine are inflicted by vascular disorder in this disease, and central nervous system is not an exception. A 60-year old man was examined for repeated epistaxis and an episode of subarachnoid hemorrhage. In this case, left middle cerebral artery aneurysm, arteriovenous malformation fed by left posterior cerebral artery and pulmonary arteriovenous fistula were revealed by angiography and computerized tomography. It is interesting that various vascular disorders were found in one case.

[Traumatic middle meningeal arterio-venous fistula--a case of bilateral occurrence (author's transl)].

A case of traumatic middle meningeal arterio-venous fistula was reported. The present case reported here appears to be unique because of its bilateral occurrence. As for traumatic middle meningeal arterio-venous fistula, we could not find any case with bilateral occurrence in the literature. Bilateral carotid angiograms revealed extravasation in the extradural spaces. Draining veins were found pouring into the superior sagittal sinus and also into the transverse sinus. This was also thought to be rare. We discussed to some extent on the etiology, diagnosis, angiographical findings and treatment of this disease.

Macromolecular derivatives of NAD and their application.

The effects of the properties of polymer chain and NAD density on the cofactor activity of polymeric NAD derivatives were investigated. NAD-N6- [N-(N-acryloyl-l-methoxycarbonyl-5-aminopentyl)-propioamide] (monomeric NAD derivative) was copolymerized with different kinds of vinyl monomers. The resulting polymeric NAD derivatives, except for that prepared with methacrylamide, had similar cofactor activities. These results show that the size and charge of the vinyl monomers do not have a great effect on the cofactor activity of polymeric NAD derivatives. The polymeric NAD derivative prepared by copolymerization with methacrylamide had the highest cofactor activities, and the activities for lactate and yeast alcohol dehydrogenases decreased with an increase in the NAD density of the polymer. These NAD density effects were mainly due to the inhibition by the polymeric NAD derivative with a high NAD density.

Plasmid control of 6-aminohexanoic acid cyclic dimer degradation enzymes of Flavobacterium sp. KI72.

Flavobacterium sp. K172, which is able to grow on 6-aminohexanoic acid cyclic dimer as the sole source of carbon and nitrogen, and plasmid control of the responsible enzymes, 6-aminohexanoic acid cyclic dimer hydrolase and 6-aminohexanoic acid linear oligomer hydrolase, were studied. The wild strain of K172 harbors three kinds of plasmid, pOAD1 (26.2 megadaltons), pOAD2 (28.8 megadaltons), and pOAD3 (37.2 megadaltons). The wild strain K172 was readily cured of its ability to grow on the cyclic dimer by mitomycin C, and the cyclic dimer hydrolase could not be detected either as catalytic activity or by antibody precipitation. No reversion of the cured strains was detected. pOAD2 was not detected in every cured strain tested but was restored in a transformant. The transformant recovered both of the enzyme activities, and the cyclic dimer hydrolase of the transformant was immunologically identical with that of the wild strain. All of the strains tested, including the wild, cured, and transformant ones, possessed identical pOAD3 irrespective of the metabolizing activity. Some of the cured strains possessed pOAD1 identical with the wild strain, but the others harbored plasmids with partially altered structures which were likely to be derived from pOAD1 by genetic rearrangements such as deletion, insertion, or substitution. These results suggested that the genes of the enzymes were borne on pOAD2.

A radioimmunoassay for a highly active luteinizing hormone-releasing hormone analogue and relation between the serum level of the analogue and that of gonadotropin.

Antisera to an LH-RH analogue, des-Gly10-[D-Leu6]-LH-RH-ethylamide (TAP-144) were produced in 10 rabbits. By using these antisera, a specific and sensitive radioimmunoassay for TAP-144 was established. Sensitivity of the radioimmunoassay ranged from 5 to 100 per assay tube with these antisera. Lh, FSH, TRH, LH-RH, and the 1-6 fragment of TAP-144 did not practically cross-react with LH-RH analogues, though the degree of the cross-reactivity differed among individual sera. The average cross-reactivity of the 10 antisera showed low specificities to TAP-144 analogues which are altered at positions 2, 3, 4, 5, and 6, but showed high specificities to those altered at positions 8 and 9. The antisera also showed low cross-reactivities to LH-RH analogues replaced at position 6 by D-amino acids and those altered at position 10 by alkylamines, but they showed fairly high cross-reactivities to analogues which are altered simultaneously at both positions 6 and 10. When TAP-144 was administered intraperitoneally to rats on the diestrous day, serum concentrations of TAP-144 increased dose-dependently but maximal serum concentrations of both LH and FSH were attained in response to higher doses of TAP-144. The peak LH and FSH concentrations appeared 70 to 110 min after the peak TAP-144 concentration had been reached. Similar delays in reaching the peak LH and FSH levels were also observed when TAP-144 was administered intravenously, subcutaneously and intramuscularly. When TAP-144 was administered intravaginally, a low but constant serum level of TAP-144 was maintained from 5 to 300 min after the administration, but serum LH and FSH levels declined to a low level from 180 min after the TAP-144 administration.
