Microcirculatory responses to estradiol benzoate in chronic liver damage induced by carbon tetrachloride in the rat.

Microcirculatory responses to estradiol benzoate (ES) under condition of chronic liver damage induced by long-term administration of carbon tetrachloride (CCl4) was investigated in the rat. One hundred and five male rats were divided into the following groups receiving 0.1 mg/100 g body weight ES injected intraperitoneally 5 times per week: controls, exposed to CCl4 alone; rats treated with ES from the fourth week of CCl4 exposure; animals treated with ES from the 11th week of CCl4 exposure. In rats receiving CCl4 alone, liver cirrhosis was induced by 10 consecutive weeks of exposure. Microangiograms of the liver demonstrated conspicuous rarefaction of the vascular tree. On the other hand, animals treated with ES had neither atrophic liver nor rarefaction of the intrahepatic vascular tree. ES produced also intrahepatic neovascular proliferation in the cirrhotic liver. After long-term CCl4 administration, ES treated rats had extremely enlarged nodules with tumor-stain like findings, giving rise to a structure differing from hepatocellular carcinoma which latter generally displays a broom-swept appearance. It is concluded that in providing potent angiogenesis in the liver, ES protects the liver against microcirculatory dearangement and parenchymal damage induced by CCl4.

Immunohistochemical and ultrastructural demonstration of keratin in epi-myoepithelial islands of autoimmune sialadenitis in man.

Immunohistochemistry of salivary gland suggested that keratin is a specific marker of duct-epithelial cells and myosin for myoepithelial cells. Epi-myoepithelial islands found in some autoimmune sialadenitis, such as Sjögren's syndrome, were mainly composed of keratin-positive cells. Keratin filaments, with bundles and desmosome-attachments were invariably found in epithelial cells with electron microscopy. The islands contained no cells with myoepithelial characteristics. The findings show that there is no obvious participation of myoepithelial cells in the growth of the epi-myoepithelial islands.

A comparative study on the immunolocalization of keratin and myosin in salivary gland tumors.

Surgical specimens of the salivary gland tumor were studied by immunohistochemical techniques using the anti-keratin antibody and the anti-myosin antibody. In the normal tissue, keratin was localized predominantly in the duct epithelial cells and myosin in the myoepithelial cells. According to the immunohistochemical staining patterns, the tumors were able to be divided into two groups: one group consisted of pleomorphic adenoma, adenoid cystic carcinoma, and adenocarcinoma which showed a mixture of keratin- and myosin-positive cells, respectively, mimicking the structures of the intercalated duct; the other comprised monomorphic adenoma and mucoepidermoid tumor which disclosed keratin-positive cells predominantly, resembling the constituent of the excretory duct. These results were mostly consistent with the "bicellular theory" that the salivary tumors generate from the intercalated duct reserve cells and the excretory duct reserve cells.

Sulfated glycoprotein biosynthesis in human gastric mucosal biopsies.

Sulfated mucus glycoprotein biosynthesis by human gastric mucosal biopsies was studied. Human gastric healthy specimens and cancer specimens were obtained from antral and corpus normal mucosal areas by biopsy. A comparison was made of the biosynthesized sulfated mucus glycoproteins assessed by the incorporation of 35S-sulfate and 14C-glucosamine into the specimens using an organ culture technique. A significant uptake of 35S-sulfate or 14C-glucosamine into the mucus glycoproteins was demonstrated in all specimens. 14C-glucosamine incorporation into the glycoproteins was almost the same for all biopsies. The synthesis of sulfated glycoprotein in healthy antral mucosa was about four times that in corpus mucosa. Significantly raised levels in sulfated glycoprotein biosynthesis were found in cancer specimens, being about 30 and 13 times higher in antrum and corpus mucosa than that of healthy specimens, respectively.

Automated measurement of amylase isoenzymes with 4-nitrophenyl-maltoheptaoside as substrate and use of a selective amylase inhibitor.

We automated a kinetic procedure for determining amylase isoenzymes in serum and urine samples. We used 4-nitro-phenylmaltoheptaoside as substrate and a selective amylase inhibitor with the Abbott-VP bichromatic system. By use of the maximum differences between pancreatic (P) and salivary (S) amylase activities remaining after inhibition by the selective inhibitor and by use of the linear range, a one-point standard method for calibration is proposed for determining amylase activities between about 50 and 1500 U/L when the P/S ratio exceeds 0.2. Results correlated well with those by electrophoresis and the Phadebas method (r = 0.99 for both pancreatic and salivary amylase). Reproducibilities (CVs) were 1.5% to 5.5% for pancreatic amylase and 1.4% to 3.3% for salivary amylase in serum, 0.8% to 2.0% for pancreatic amylase and 0.8% to 2.3% for salivary amylase in urine.

[Cytofluorometric analysis of cell kinetics in relation to tumor growth of advanced gastric cancers].

Correlated studies on both cell kinetics in association with nuclear ploidy pattern and pathological morphology of advanced gastric cancers were carried out using DNA-RNA cytofluorometry (NIKON SPM-RF1-D) with AO stain. The results showed that advanced gastric cancers could be divided into two main groups. Group 1 (12 cases) was the diploid cell population, commonly found in Borrmann type 4 showing diffuse infiltration of cancer cells, and group II (14 cases) was characterized by polyploidization, usually found in Borrmann type 2 or 3 showing adenocarcinomas composed of markedly pleomorphic cells. In addition, we further differentiated from group II four cases of cancers which included aneuploid cells with more pronounced pleomorphism than group II, and classified them as group II'. These results suggest that cell kinetics and nuclear ploidy pattern of the advanced gastric cancers are closely related to pathological characteristics, i.e., groups I and II to undifferentiated (gastric type) and differentiated (intestinal type) carcinomas, respectively.

[DNA-RNA cytofluorometry for a primary cancer of the stomach and choledochus].

We standardized DNA-RNA cytofluorometry with AO stain using a NIKON SPM-RFI-D instrument for cell smears. We applied this technique in a 72-year-old woman with primary double cancer of the stomach and choledochus. We first suspected malignancy of the choledochus and examined the desquamated cells in the bile cytofluorometrically to obtain by X-Y plotting the irregularly increased polyploid cells with a high percentage of cells in S phase. These polyploid cells and S phase cells were found to have an increased RNA content. Before operation, the presence of another malignancy of the stomach was also ascertained. The surgical specimens were diagnosed histopathologically as well differentiated, tubular adenocarcinoma of the stomach and moderately differentiated, tubular adenocarcinoma of the choledochus. By cytofluorometric analysis similar results were obtained, i.e., polyploidization with increase of S phase cells. Based on these results, we concluded that our method is useful for the diagnosis of alimentary tract malignancy, and that the criteria of malignancy appear to be unusual polyploidization with increased fractions of S phase cells.

Changes of enzyme activities recognized in lymphocytes from patients with carcinoma of the gastrointestinal tract.

Adenosine triphosphatase (ATPase) activity and acid phosphatase activity in lymphocytes from patients with carcinoma of the gastrointestinal tract were determined in order to investigate whether or not changes in these enzyme activities has any relation to the immune reactivity of carcinoma-bearing patients. In patients with a performance status of more than 60%, the mean value of the total ATPase activity in lymphocytes differed little from that in controls, but the mean value of the oligomycin-sensitive ATPase activity decreased as compared to that in the controls. On the other hand, the mean values of the activities of both free and total acid phosphatase in lymphocytes increased as compared to those in controls. The mean values of the activities of both ATPase and acid phosphatase in lymphocytes from patients whose performance status was less than 50% decreased as compared to those from controls and patients whose performance status was more than 60%. The change of the activities of both ATPase and acid phosphatase in lymphocytes has relation to that of the immunological parameters of the patients with carcinoma of the gastrointestinal tract. These results indicate that both ATPase and acid phosphatase in lymphocytes may play an important role in the immune mechanism.

Inhibitory effects of autologous hepatic tumor suspension on the development of liver cell carcinoma in rats.

Inoculation with a crude suspension prepared from autologous liver cell carcinoma inhibited the development of liver tumor in cirrhotic rats exposed to 3'-methyl-4-diethylaminoazobenzene (3'-Me-DAB). The suspension had also an inhibitory effect on the growth of tumor in rats bearing liver cell carcinoma. The results obtained suggest that a close relationship exists between tumor-associated antigens and onco-development.

[Oral amelanotic melanoma--report of a case with desmoplastic changes at the site of lymph node metastasis].

Amelanotic melanoma of the gingiva in a 59-year-old Japanese woman is presented. The histologic diagnosis of melanoma was confirmed by the concurrent lentigo maligna in the palatal mucosa and desmoplastic malignant melanoma (DMM) in the metastatic foci of a cervical lymph node. Electron-microscopical observations revealed that DMM cells exhibited mesenchymal metaplasia or myofibroblastic differentiation. Accordingly, collagen synthesis of the tumor cells was suspected. A review of 31 reported DMM cases is also presented.
