The protective effect of cysteine on chemical-induced liver injury in rats.

The possible protective effect of cysteine on chemical-induced liver injury was studied in rats in vivo and in vitro. There was no increase in the activity of serum glutamic oxaloacetic transaminase (GOT) of rats pretreated with cysteine (1.2 g/kg, p,o.) followed by 0.25 ml/kg carbon tetrachloride (CCl4), d-galactosamine (GalN) or alpha-naphthylisothiocyanate (ANIT). However, rats pretreated with cysteine followed by 0.5 ml/kg CCl4 were not protected. The content of cytochrome P-450, activity of aminopyrine N-demethylase or serum ratio of 5,5-dimethyl-2,4-oxazolidinedione (DMO) to trimethadione (TMO) (DMO/TMO ratio) after CCl4, GalN or ANIT were not altered by pretreatment with cysteine. However, pretreatment with cysteine prevented changes in the content of cytochrome P-450, activity of aminopyrine N-demethylase and DMO/TMO ratio in serum as well as the activities of serum GOT and GPT when the rats were treated with bromobenzene (BZ). The degree of lipid peroxidation from CCl4 was markedly reduced by the presence of 10(-4)M cysteine. These results suggest that cysteine has a protective effect on chemical-induced liver injury produced via epoxide metabolites.

[Effect of electroacupuncture on the sympathetic nervous, endocrine and metabolic functions].

Electroacupuncture (1.5 volts, 0.13 msec duration, 5 Hz for 10 min) was applied to the ten sites of lumbodorsal region in women aged 20 to 47. Immediately after the acupuncture plasma noradrenalin concentration significantly increased 9 out of 20 subjects, while it decreased in 11. Plasma noradrenaline returned to the intitial level in both groups 30 min after the acupuncture. In the noradrenaline elevated group plasma insulin concentration and plasma insulin/glucagon molar ratio significantly decreased until 30 min after the acupuncture. Such changes were not observed in the noradrenaline decreased group. Plasma glucagon, dopamine-beta-hydroxylase, free fatty acids, glycerol, creatine kinase and blood glucose were not changed in both groups.

[Changes in plasma 2-hydroxyestrone levels in pubertal females and correlation with sex hormones].

Plasma levels of 2-hydroxyestrone (2-OHE1) were measured by specific radioimmunoassay during puberty to elucidate the physiological role of this hormone on female sexual development. Plasma levels of FSH, LH, PRL and estradiol (E2) were also measured with an RI-kit. 67 girls between aged 6 and 16 were selected for this study. Blood samples were collected into tubes containing 0.1W/V% EDTA and 0.1W/V% ascorbic acid and immediately centrifuged. 2-OHE1 in plasma was extracted and separated with a Sephadex LH-20 column. RIA was applied using anti 2-OHE1-17-CMO-BSA. Plasma 2-OHE1 levels before menarche were low (6-7 pg/ml) until 10 years of age. The levels started to increase from 11 and reached 11.2 +/- 5.8pg/ml at 16 years of age. The levels of 2-OHE1 in girls after menarche were significantly higher than that before menarche (p less than 0.01). There was a significant correlation between 2-OHE1 and E2 in girls before menarche (p less than 0.001, r = 0.5416). However, the 2-OHE1 to E2 ratio decreased significantly from 9 to 10 years of age. These results indicate that during this period, the E2 increase is more predominant than that of 2-OHE1. There was a significant negative correlation between 2-OHE1 and PRL after menarche while no correlation between 2-OHE1 and FSH, LH was noticed. These results suggested that 2-OHE1 may play a role in sexual development after menarche rather than in the initiation of menarche.

Karyotype evolution in B-cell lymphoid malignancy with an 8;14 translocation.

To evaluate the significance of karyotypic evolution of tumor cells with an 8;14 translocation [t(8;14)(q24;q32)], we examined the clinicopathologic features and immunologic phenotypes of nine Japanese patients with various types of B-cell malignancy with the translocation. All these patients had structural rearrangements of the long arm of chromosome No. I (Iq) in a stem line or the subline of tumor cells with a t(8;14). The rearrangements were composed of a translocation involving Iq with other chromosomes and a tandem duplication of Iq, and they were exclusively associated with a partial trisomy for Iq. Two patients with diffuse large-cell lymphoma, whose tumor cells did not express surface immunoglobulins (s-Ig), had the Iq translocation in their highly complex karyotypes. Tumor cells from the other seven patients expressed s-Ig and the karyotypes were relatively simple. Among these patients, the Iq translocation was found in two patients with Burkitt's lymphoma, and the Iq duplication were observed in a stem line or the sublines from four patients with Burkitt's lymphoma-leukemia and one each with small non-cleaved-cell or diffuse large-cell lymphoma. Except for one patient in the stage of IE, these patients had a poor prognosis because of the clinical conversion of extranodal metastases in the earlier disease phase. These findings are compatible with those of Western patients with a t(8;14). Therefore, tumor cells marked primarily with a t(8;14) could have "major routes" in the karyotypic evolution, for which potentials should be recognized as clinical risk factors, and the morphologic presentation and the expression of surface immunoglobulins may be associated with the process of karyotypic evolution.

Independent induction of sister-chromatid exchanges by 3-aminobenzamide and ultraviolet radiation in HeLa cells.

The effect of 3-aminobenzamide in HeLa cells was examined in several aspects. 10(-3) M 3-aminobenzamide inhibited poly(ADP-ribose) polymerase activity to more than 95% in disrupted nuclei of HeLa cells. The mode of inhibition of the enzyme was competitive inhibition with NAD at a Ki value of 2.5 microM 3-aminobenzamide. The combined treatment of HeLa cells with 3-aminobenzamide and ultraviolet radiation revealed the independent induction of SCEs by these 2 agents.

Structure of high-temperature oxidation zones of gold alloys for metal-porcelain bonding containing small amounts of In and Sn.

Changes in the morphology of the oxidation zone and the quantities of oxide for gold alloys containing two base metals (In and Sn) were investigated by exposing alloys at 1000 degrees C for one h in air. The contents of In and Sn were varied in the range of from 0-1.5 wt%. In the range of from 1.5-0.35 wt% In (from 0-1.15 wt% Sn), In 2O3 was formed predominantly at the alloy surface. The electron probe x-ray micro-analyzer detected no uniform external oxidation zone. A mixture composed of In2O3 and SnO2 precipitated internally in the alloy matrix. An external oxidation zone composed of SnO2 was formed on the alloys containing over 1.2 wt% Sn, and no internal oxidation zone was observed.
