A novel cell line, designated KIS-1, was established from a patient with Ki-1-positive diffuse large cell lymphoma. Multiple phenotypic analysis of the KIS-1 cells was carried out with a total of 22 monoclonal antibodies defining hematopoietic cell subsets and lineages. The KIS-1 cells were positive for Ki-1, B4, HLA-DR, and 2D1 (common leucocyte) antigens, but were negative for the antigens reportedly specific for T cells, natural killer cells, granulocytes, monocytes, interdigitating reticulum cells and dendritic reticulum cells. The genomic analysis of the KIS-1 cells showed not only the rearrangement of JH and J kappa genes but also the probable rearrangement of C lambda genes. Moreover, the cells produced immunoglobulin lambda chains. Thus, KIS-1 was considered to be of B-cell lineage. The lymphoma-cell derivation of KIS-1 was based on the following facts. The cytochemical, immunologic, cytogenetic properties and the results of the molecular genomic analysis in the KIS-1 cells were essentially the same as those of the original tumor cells, and the KIS-1 cells were negative for Epstein-Barr virus-associated nuclear antigen. KIS-1 is the only known B-cell line derived from Ki-1-positive diffuse large cell lymphoma, and should be useful for defining the biological implications of Ki-1 antigen.
The purpose of this report was to investigate the actual need, as opposed to patient demand, for treatment of TMJ dysfunction syndrome in adolescents. For this investigation, 2198 Japanese schoolchildren, aged 10-18 yr (1095 boys and 1103 girls), were examined by questionnaire. In addition, each subject was given a clinical examination. The results of the subjective evaluations were compared with the frequency of actual occurrence of TMJ dysfunction syndrome. The results were: 1) Subjective recognition of TMJ dysfunction syndrome symptoms involved 253 students (11.5%): 129 boys (11.8%) and 124 girls (8.2%), with no significant difference in number between the sexes, 2) by comparing subjective assessment with clinical examination, the junior high school age was found to be the proper age for the introduction of preventive medical care, because these pupils were able to recognize the symptoms of TMJ dysfunction syndrome as accurately as adults, 3) the results obtained from the questionnaire and the clinical examination showed evidence of higher frequency of accurate recognition on the part of girls.
Acute biphenotypic leukemia composed of lymphoblasts and myeloblasts developed in a patient with T lymphoblastic lymphoma (T-LBL) who had an anterior mediastinal mass. A novel myeloid cell line, termed TK-1, has been established from his peripheral blood after the leukemic conversion. The identical rearranged pattern of T cell receptor gamma-chain gene was observed among the DNAs derived from lymph node cells in the lymphoma phase, the myeloid cell line TK-1, and the subclones with different karyotypes (TK-1B and TK-1D), which showed that myeloid cells had been derived from the T-LBL of the same patient. This finding demonstrates that phenotypic conversion occurs in the clonally propagating tumor cells and suggests that some hematopoietic cells retain the capacity to adopt either lineage.
Blowout fractures most frequently involve the orbital floor. The contour of the orbit has been postulated as one of the factors responsible for this phenomenon, but only a few studies have been carried out. The present study was planned to determine the effect of the contour of the orbit when intraorbital pressure is raised to simulate the hydraulic mechanism of blowout fracture. The profile of the orbit was estimated from five dry human skulls, and the measurements were approximated to frame models. The deformation of these models by raised intraorbital pressure was calculated by computer simulation using the finite element method. In all of the orbital models, the deformation of the orbital floor was significantly greater than that of the roof. These findings verified that the orbital roof has a higher resistance than the floor against raised intraorbital pressure. We suspect that this resistance is due to the arched shape of the orbital roof, whereas the floor is rather flat.
The effects of flutropium bromide (Ba598Br), a new antiasthma drug possessing the quarternary ammonium structure of atropine derivatives, on mediator release from mast cells and on actions of leukotriene (LT) D4 and serotonin were investigated. Flutropium bromide (3 and 10 mg/kg, i.v.) showed an inhibitory action on the 48 hr homologous PCA in guinea pigs. Atropine showed no inhibitory effect. Flutropium bromide also inhibited the release of histamine from isolated rat mast cells stimulated by antigen, although the inhibitory action was weaker than that of disodium cromoglycate. Atropine also had no inhibitory action in this case. Flutropium bromide and atropine showed no antagonistic action against LTD4-induced contraction of isolated tracheal smooth muscle of guinea pigs. Inhalation of flutropium bromide (0.3%) also showed no antagonistic action against serotonin-induced bronchoconstriction in dogs. From the above results, it is indicated that flutropium bromide has a weak mast cell stabilizing action, but no antagonistic action against LTD4 and serotonin.
The alkaline solution method for detecting uterine implantation sites (metrial glands or spots after delivery), which indicate the position of the embryo, was compared with conventional methods using Turnbull blue (Salewski's technique) and benzyl benzoate (Orsini's technique) in rats. When rat uteri were immersed directly in 2% sodium hydroxide [NaOH], potassium hydroxide [KOH] solution or one of several other alkaline solutions for one hour, the uteri were cleared. Then the metrial glands could be observed without further staining, and the spots after delivery with further staining. After alkaline treatment, unstained metrial glands appeared as yellowish-white degenerated masses and stained spots after delivery appeared as yellowish-brown patches covered with yellowish-white lumps. These yellowish-brown or yellowish-white sites were probably stained by the iron in hemosiderin deposits in the placental scar or in a vestige of the metrial glands left over from pregnancy. Formalin-fixed uteri were not cleared by immersion in a high concentration of NaOH, KOH or other alkaline solutions, but the spots after delivery were stained by this treatment. Turnbull blue did not stain the metrial glands, which contain no iron. Benzyl benzoate cleared fixed or unfixed uteri and allowed observation of the metrial glands and spots after delivery although the entire process required a few days and the specimens became hard and shriveled. The best and most convenient solution for clearing the rat uterus and for staining implantation sites was 2% NaOH or KOH.
An EBV-negative B-cell lymphoma cell line designated as HBL-1 was established from the pleural effusion of a patient with malignant lymphoma, diffuse, large cell. Surface marker studies revealed that HBL-1 cell possessed the same phenotypes of a mature B-cell as the original lymphoma cell from pleural effusion. HBL-1 closely resembled morphologically to the original lymphoma cells observed in the lymph node biopsy and pleural effusion. Chromosome analysis revealed that HBL-1 cells showed chromosomal aberrations of a 14q+ marker chromosome as the result of a translocation between chromosomes 14 and 16. Heterotransplantation to subcutaneous tissue of an athymic nude mouse has succeeded, but intraperitoneal heterotransplantation has failed. Tumors were localized in the subcutaneous tissues and no metastasis was found. This newly established cell line may facilitate immunological and oncogenic studies for human B-cell lymphomas.
We synthetically derived protoheme (oxygen-binding site of hemoglobin) to an amphiphilic heme compound and embedded it in the phospholipid bilayer of liposomes, not in an aqueous inside region of liposomes. This liposome/heme transports oxygen efficiently under physiological conditions. Characteristics of our totally synthetic liposome/heme as an artificial oxygen carrier, are summarized as follows. (i) Oxygen-binding is reversible and very rapid. (ii) The oxygen volume dissolved in the fluid is similar or superior to that of blood. (iii) The oxygen-binding affinity is close to that of blood. (iv) The particle size is smaller than 0.1 micron. (v) It is physically and mechanically stable under shear stress and storage. (vi) The components are synthetic derivatives of protoheme and phospholipid originated in nature. The advantages (i)-(iii) and brilliant red color of the solution are derived from the fact that our totally synthetic liposome/heme, transports oxygen in the same principle of red blood cells. When the liposome/heme solution was mixed with human blood, the liposome/heme delivered oxygen to blood in the mixture system. The oxygen-exchanging of the liposome/heme with blood was also tested ex-vivo using an artificial lung apparatus. The oxy liposome/heme was circulated through A-V of a dog leg, and the oxygen concentration was kept at a high level. This demonstrates the fact that the synthetic liposome/heme transports and delivers oxygen to the muscle tissue. Application to extracorporeal circulation and transfusion test will also be reported.

The effects of 10-week physical training on both the activity and concentration of gamma-glutamyltransferase (gamma-GT) in plasma were investigated on 7 sedentary healthy male students. The training consisted of running over 5 km, 6 times/week. The maximal oxygen uptake (Vo2max) and 12-min field performance increased significantly after training, from 43.5 and 2,683.7 to 48.1 ml.kg-1.min-1 and 2,931.1 m, respectively. After training the resting plasma gamma-GT concentration decreased to half, whereas the gamma-GT activity did not vary substantially. The training did not affect the response to the Vo2max test of either concentration or activity of gamma-GT. These results may suggest that plasma gamma-GT concentration is useful as an index of the extent of physical training.

In eight normal mothers the effects of pyridoxal 5'-phosphate (PLP) addition on the plasma activities of aspartate aminotransferase isoenzymes (AST-s and AST-m) during labor and following delivery were investigated. The AST-s activities with and without reactivation by PLP appeared to increase immediately after delivery and they were even higher on the 4th day postpartum. On the other hand, there were significant elevations in both the AST-m activities immediately and at 2 h after delivery, but not on the 4th day postpartum. Of AST isoenzymes measured in the nonstress test, only the relative activation rate of AST-m by PLP added was significantly higher than the control value. The present study may come to the following conclusions: 1) The relative activation rate of plasma AST-m activity by PLP may be a reliable index of vitamin B6 nutritional status during pregnancy. 2) The increases in AST-m activity with and without PLP added during labor suggest a minimal damage of mitochondria in skeletal, cardiac and uterine muscle cells. 3) The AST-s isoenzyme determinations with and without PLP may be especially useful as sensitive indication of erythrocyte and/or liver damage after delivery.

Karyotype evolution of t(14;18)-positive lymphoma was studied in 13 Japanese patients. The extra 18q- chromosome, found in six of ten patients with complex karyotypes, was the most common change subsequent to a t(14;18)(q32;q21) chromosome translocation. The additional change was interpreted as being a duplication of an 18q- derived from a t(14;18). The six patients had transformed histology of follicular small cleaved cell lymphoma or diffuse large cell lymphoma, and five of them had extranodal expansion associated with a poor prognosis. These findings indicate that the extra 18q-, together with other chromosome abnormalities, is closely associated with the advanced grade disease of t(14;18)-positive lymphoma, and the extra chromosome is evolutionally comparable with the second Philadelphia (Ph1) chromosome often found in the blastic phase of chronic myelocytic leukemia carrying a t(9;22)(q34;q11). In addition, since the extra 18q- is rarely found in American patients with t(14;18)-positive lymphoma, there appears to be a difference in the karyotype evolution between Japanese and American patients.
