The c-yes-1 and bcl-2 genes have been independently mapped at the same position of the chromosome band, 18q21.3, which is a breakpoint in the t(14;18) chromosomal translocation. High-molecular-weight genomic DNAs and mRNA isolated from a follicular lymphoma cell line FL-18 and an Epstein-Barr virus harboring FL-18EB cell line, which carried the t(14;18), were analyzed. Although there was no detectable c-yes rearrangement, the chromosome 18-specific probe b (bcl-2) revealed one rearranged fragment after EcoRI, BamHI, PstI and SstI digestion. Therefore, breakage of chromosome 18 in the t(14;18) occurred at the SstI fragment containing the probe b sequence on the bcl-2 gene. The transcript of c-yes was undetectable, whereas active transcription of bcl-2 was observed in the FL-18 and FL-18EB cell lines at a higher level than in other types of lymphoma cell line. These data suggest that the c-yes-1 is not involved in the t(14;18), and the 2 genes located at 18q21.3 are independent sequences.
Plasma corticotropin-releasing factor (CRF), corticotropin (ACTH) and cortisol levels were simultaneously determined by radioimmunoassays at 0600 h, 1200 h, 1800 h and 2200 h in six normal subjects, in order to examine whether the diurnal rhythm in plasma CRF exists and how it correlates to the diurnal rhythm in plasma ACTH and cortisol concentration. The highest CRF level was observed at 0600 h (7.0 +/- 1.2 pg/ml) and significantly lower levels (p less than 0.01) at 1800 h (1.7 +/- 0.2 pg/ml) and 2200 h (1.9 +/- 0.4 pg/ml). A clear diurnal rhythm was demonstrated in plasma ACTH and cortisol levels, with the highest values at 0600 h (44.6 +/- 8.1 pg/ml and 15.9 +/- 2.0 micrograms/dl, respectively) and the lowest at 2200 h (12.3 +/- 2.8 pg/ml and 4.6 +/- 1.0 micrograms/ml, respectively). These results suggest that the diurnal rhythm in ACTH and cortisol is under the regulation, at least in part, of the diurnal rhythm in CRF secretion.
The dissociation behavior of several species of proteoglycan aggregates (PGA) has been studied quantitatively by monitoring the changes in particle size by dynamic laser light-scattering. Firstly, studies of the thermal dissociation of reconstituted PGA from bovine nasal septum and bovine fetal epiphysus are described. The effect of link protein in stabilizing reconstituted PGA against changes in temperature has been demonstrated. It was also confirmed that, upon prolonged heating at 70 degrees, the dissociation of link-containing, reconstituted PGA is effectively irreversible. Secondly, the dissociation characteristics of native PGA isolated from chick chondrocyte and rat chondrosarcoma cell-cultures were investigated in aqueous solvents containing increasing concentrations of guanidine hydrochloride. It was found that the more densely packed, native, aggregate structure has a higher susceptibility to thermal disruption than the reconstituted, link-containing material. Considerable loss of subunits from the native aggregates occurs at temperature where the link protein retains its activity. The dissociation of native PGA is irreversible, in the sense that reconstituted PGA always exhibits smaller sizes than the native PGA, reflecting a smaller degree of aggregation of the subunits.

A 69-year-old woman underwent successful thrombolysis for total occlusion in the right coronary artery using urokinase. One week later, the patient developed reinfarction in the area supplied by the right coronary artery, followed by ventricular rupture. She was resuscitated with drainage of the pericardial effusion. Cardiac catheterization confirmed that the site of the right coronary artery reocclusion was identical to that in the acute phase. A false aneurysm developed over the true aneurysm located in the inferior portion of the left ventricle as demonstrated by a ventriculogram at the convalescent stage and at surgery. The orifice of the false aneurysm has closed by suture.

gamma-Glutamyl cyclotransferase activity is assayed in tissues by a colorimetric method using gamma-glutamyl alanine as a substrate coupled with alanine dehydrogenase from B. sphericus, to measure the formation of NADH. In order to avoid interference by the reaction catalyzed by gamma-glutamyl transpeptidase, anthglutin, a specific inhibitor of the transpeptidase was included in the reaction mixture. The Km value of rat kidney gamma-glutamyl cyclotransferase with respect to gamma-glutamyl alanine appeared to be the same when determined by either the colorimetric or the radiometric method. This assay presents a reliable alternative to the use of radiolabeled substrate and is used for the assay of gamma-glutamyl cyclotransferase in a variety of physiological and experimental samples.
In 15 normal mothers the stress effects of labor on zinc and copper distributions in erythrocytes and plasma were investigated. Lower levels of total zinc and of zinc derived from both carbonic anhydrase isoenzymes (CA-I and CA-II), especially from CA-I, in erythrocytes were obtained immediately after delivery. On the other hand, there was a significant increase in total plasma zinc concentration only on admission to hospital in labor, although albumin-bound zinc concentration remained elevated throughout the entire labor. These results suggest that there are unexplained disparities between the changes in zinc levels in maternal erythrocytes and plasma during labor.
The electroencephalographic (EEG) effects of 7-chloro-5-(o-fluorophenyl)-1,3-dihydro-1-(2,2,2-trifluoroethyl)-2H-1, 4-benzodiazepine-2-thione (Sch 161, quazepam) were investigated in unanesthetized rats and rabbits with chronic electrode implants, and the effects were compared with those of flurazepam and diazepam. In freely moving rats, quazepam at doses of 10 to 30 mg/kg, p.o., induced an increase in drowsy EEG pattern periods: high amplitude slow waves and spindle bursts in the cortical EEG and desynchronization of the hippocampal theta waves. Quazepam at doses of 10 to 30 mg/kg significantly decreased the onset of the drowsy EEG pattern after p.o. administration of rats in a dose-dependent manner, while flurazepam and diazepam reduced it only at higher doses of 20 to 30 mg/kg, p.o. In rabbits, quazepam at doses of 0.5 to 5 mg/kg, i.v., evoked a drowsy EEG pattern similar to that of flurazepam (1 to 5 mg/kg) or diazepam (1 to 2 mg/kg). Quazepam at higher doses caused sedation and a slight muscle relaxation in both rats and rabbits, which were weaker than those of flurazepam or diazepam. In both rats and rabbits, quazepam and flurazepam depressed the EEG arousal response to auditory stimulation but not to electrical stimulation of the midbrain reticular formation or the posterior hypothalamus, while the arousal response to either auditory or brain stimulation was markedly suppressed by diazepam. The recruiting response was not altered by quazepam, flurazepam and diazepam. Quazepam has no effect on the photic driving response to flash light in the occipital cortex of the rabbit, while the response was suppressed moderately by flurazepam and markedly suppressed by diazepam. Quazepam (2 mg/kg) suppressed either the hippocampal or amygdaloid afterdischarges. These results suggest that the EEG effect of quazepam is different to those of flurazepam and diazepam in qualitative aspects and that quazepam is likely to be an effective sleep-promoting drug with slight adverse reactions.

Effects of acute cold exposure (10 degrees C for 60 min) on the concentrations of adrenocorticotropin (ACTH) and zinc in plasma were investigated in seven healthy male students. There were no significant changes in total zinc, albumin-bound zinc, and alpha 2-macroglobulin-bound zinc concentrations throughout the experimental period. On the other hand, ACTH concentration increased markedly during cold exposure. In addition, a statistically significant inverse relationship existed between the changes in ACTH and albumin-bound zinc values during the experiment. These results suggest that acute cold exposure produces elevated plasma ACTH levels, with resulting zinc redistribution in the human body.
A novel cell line, FL-18, was established from the pleural effusion of a patient with follicular small cleaved cell lymphoma. At the same time, an Epstein-Barr virus (EBV) nuclear antigen (EBNA)-positive cell line, FL-18-EB, was established from the EBV-infected culture of the same pleural effusion cells. Both cell lines had the same monoclonal surface immunoglobulin (IgG kappa), and they had the same karyotype as that of the fresh pleural effusion cells in which a reciprocal translocation between the long arm of chromosomes 14 and 18 [t(14;18)(q32;q21)] was detected. Gene rearrangement analysis of immunoglobulin heavy-chain gene (JH) and kappa light-chain gene (J kappa) showed the same rearranged configuration in the two cell lines; however, some morphological and phenotypic differences were found. The FL-18-EB cells, which were morphologically similar to common EBNA-positive lymphoblastoid cell lines of normal B cell origin at the initial phase of culture, were larger than the FL-18 cells and contained multinucleated giant cells. The FL-18 cells lacked cytoplasmic immunoglobulin and were positive for common acute lymphoblastic leukemia antigen (CALLA), whereas the FL-18-EB cells had cytoplasmic immunoglobulin and were negative for CALLA. Thus, the phenotype of FL-18-EB seems to be a result of a shift by EBV infection to a more mature stage in the B cell differentiation pathway than that of FL-18. The paired availability of EBV-free and EBV-infected cell lines of a neoplastic clone is unique and valuable in considering EBV infectibility of neoplastic B cells and resultant phenotypic changes.

We established an interleukin 2 (IL-2)-dependent human T cell line, Kit 225, from a patient with T cell chronic lymphocytic leukemia (T-CLL) with OKT3+, -T4+, -T8- phenotype. Southern blot analysis showed that Kit 225 is not infected with human T cell leukemia/lymphoma virus (HTLV) type I or II, and is probably derived from the major clone in the fresh leukemic cells. Kit 225 cells express a large amount of IL 2 receptors constitutively and their growth is absolutely dependent on IL 2. No other stimuli, such as lectins or antigens, are required for maintaining the responsiveness to IL 2. As abnormal IL 2 receptor expression was also seen originally in the fresh leukemic cells, the establishment of this cell line with IL 2 suggests that IL 2-mediated T cell proliferation is involved in the leukemogenesis of some cases of HTLV-negative T-CLL.
