A novel 15 kDa Ca2+-binding protein present in the eggs of the sea urchin, Hemicentrotus pulcherrimus.

A novel Ca2+-binding protein, different from calmodulin, has been purified to homogeneity from the soluble cytoplasmic protein fraction of the egg of the sea urchin, Hemicentrotus pulcherrimus. This protein, designated as 15 kDa protein, shows a Ca2+-dependent mobility shift upon SDS-gel electrophoresis and has Ca2+-binding ability. This protein did not resemble the sea urchin egg calmodulin in either molecular mass or amino acid composition. The 15 kDa protein could not activate cyclic adenosine 3',5'-monophosphate-dependent phosphodiesterase from bovine brain and did not bind to fluphenazine-Sepharose 6B. Antibodies against the 15 kDa protein did not react with sea urchin egg calmodulin. These results suggest that the 15 kDa protein is a novel Ca2+-binding protein in the sea urchin egg.

Acquired von Willebrand disease and storage pool disease in chronic myelocytic leukemia.

Platelet function was evaluated in 20 patients with chronic myelocytic leukemia (CML), all Ph positive. Seven showed abnormal epinephrine-induced aggregation, while four had impaired both ADP- and collagen-induced aggregation. The platelets of all patients aggregated with arachidonic acid, thus ruling out cyclooxygenase or lipoxygenase deficiency. The intracellular concentrations of ATP and ADP were significantly below normal, and the ratio of ATP/ADP was greater than normal in all 12 patients. ATP released from platelets by Lumi-aggregometer was reduced. In four patients with abnormal ristocetin-induced aggregation, vWF:Ag, RCoF, and FVIII:C were all reduced. No significant inactivation of factor VIII was induced in normal plasma by incubation with patient's plasma. The crossed immunoelectrophoretic analysis revealed that vWF:Ag in these patients was mainly composed of more anodic component as compared with that of normal plasma. The ratio of vWF:Ag/RCoF was significantly greater than normal. A marked increase of factor VIII and a rapid return of vWF:Ag and RCoF to the baseline after the 1-deamino-8-arginine vasopressin (DDAVP) infusion were observed. Transient increase in vWF:Ag after the infusion of DDAVP appeared with less anodic forms and in the same relative proportion as that in normal plasma. The present study shows that in some patients with CML storage pool disease occurs with acquired von Willebrand disease.

Purification of human sperm by a discontinuous Percoll density gradient with an innercolumn.

Human sperm were highly purified through the use of a discontinuous Percoll density gradient placed in an inner column of a centrifuge tube. Six ml of 80% Percoll solution were poured into a centrifuge tube with an inner column containing successive 1.0-ml layers of 70, 60, and 40% Percoll solutions. Diluted semen was placed on top of the gradient, and the tube was centrifuged at 600 X g for 30 min using a swing-out rotor. After centrifugation, the majority of the progressive motile sperm were isolated in the sediment; they had a mean motility of 93 +/- 4.1% (n = 10). Other cellular components, including bacteria, remaining in the inner column. The level of bacterial contamination in the purified sperm fraction was below detection for most of the species quantified. The purified sperm were found to be more than 92 +/- 3.2% viable, as judged by dye exclusion, and abnormal sperm were reduced to 5.2 +/- 1.4%. Because of the use of the inner column, the contamination by seminal plasma was negligible in the purified sperm, as estimated by residual protein, fructose, and acid phosphatase activity.

Characterization of rabbit sperm by equilibrium sedimentation in Percoll during frequent ejaculation.

Qualitative and quantitative changes of rabbit sperm were observed during frequent ejaculation. Semen volume, sperm concentration, percentage of motility, and fertility index of Farris (FI) decreased with frequent ejaculation, and these values recovered to initial levels after a 24-day abstinence period. A remarkable change was found in the sperm distribution profile in Percoll density gradients after equilibrium sedimentation. The main peak of sperm was shifted from fraction number 4 on the first day of ejaculation to number 7 on the fifth day. Fraction numbers 1-4 showed a high percentage of motility, whereas the sperm motility from fraction numbers 7-10 was quite low. Motile and morphologically mature sperm had higher apparent density than immotile or immature sperm. This suggests that rabbit sperm motility might be related to the apparent density of the sperm cell.

[Surgical approach to abdominal aortic aneurysm with malignant alimentary tract tumor: report of three cases].

The clinical courses of three cases with various alimentary tract malignant lesions coincidental with abdominal aortic aneurysm were reported. Of those three patients, a simultaneous resection of the malignant lesion and aneurysm was carried out in two patients, while an secondary abdominal aneurysmectomy following the resection of the malignant lesion was done in one patient. A 70-year-old man with cancer of the cecum and an infra-renal abdominal aneurysm, was diagnosed preoperatively, and a simultaneous right hemicolectomy and aneurysmectomy were carried out. In the other patient, a 77-year-old man, presence of the gastric cancer was incidentally found at laparotomy and a 75 percent gastrectomy and an aneurysmectomy were carried out. In the third patient, both gastric cancer and an abdominal aneurysm were detected preoperatively. Distal partial gastrectomy was performed first because of severe epigastralgia and an asymptomatic aneurysm. The abdominal aneurysmectomy was carried out six months later. All patients were treated by daily administration of Cefazolin sodium or cefalotin sodium (4-10 g) and Dibekacin sulfate (200 mg) for seven to ten days postoperatively. In the case of second look operation, however, Fosfomycin 2-4 g/day was added to the above mentioned drug following the aneurysmectomy. All tolerated surgery well without any signs of infections. The first patient died on the 57th postoperative day from panperitonitis carcinomatosa following an episode of intestinal obstruction. Selection of the operative approaches for patients having both an alimentary tract malignant tumor and an abdominal aortic aneurysm was difficult, although the initial surgical intervention for the more life threatening lesion would be better justified.(ABSTRACT TRUNCATED AT 250 WORDS)

[Methods of suppression of a myonephropathic metabolic syndrome after infra-renal aortic occlusion].

Acute arterial occlusion of the extremities may result in severe and complex metabolic derangement. In order to evaluate the development and means of therapeutic control of metabolic derangements following the acute arterial occlusion of extremities, 32 adult mongrel dogs weighing between 7 and 15 kg underwent acute arterial occlusion by cross-clamping the infrarenal aorta. The experimental animals were divided into two groups: an untreated group, and treated group. The former was divided into three groups--12, 24 and 48 hour's arterial occlusive groups and the latter into two groups with 48 hour's arterial occlusion--tris (hydroxymethyl) aminomethane (THAM) and perfusion groups. Biochemical and electrolyte analyses were measured before occlusion, immediately before, and 1, 3, 12, 24 and 48 hours after the release of the occlusion. The SGOT, CPK, aldolase, creatinine and blood urea nitrogen levels rose after the release of the occlusion and were significantly higher in the 48 hour's group than in the 12 and 24 hour's occlusive groups. Among these enzymatic changes, the creatinine and urea nitrogen levels were high 48 hours after the release of the occlusion, though the others decreased with time after the occlusion release. The blood pH level fell after the occlusion in the untreated groups and these levels increased slowly after the release of the occlusion. However, there were no significant differences in the blood pH among the untreated groups. The acute arterial occlusion by cross-clamping the infra-renal aorta caused severe renal damage among the various organs. In the groups treated with THAM and perfusion, the SGOT, CPK, aldolase, creatinine and blood urea nitrogen levels remained almost at preocclusion levels after the release of the occlusion. There were statistically significant differences in these enzymatic changes between the treated group and the 48 hour's occlusive group without treatment. The blood pH levels in the treated groups showed minimal changes after the release of the occlusive, although there were no significant differences in the blood pH between the treated groups and the 48 hour's occlusive group without treatment. It was concluded that the intravenous administration of THAM and peripheral washing were effective against untoward metabolic changes occurring in the ischemic extremities.

Gravimetric, histological and cytotoxic changes of lymphoreticular tissue after implantation of tumor cells in mice.

In inbred C3H/He male mice, gravimetric, histologic and cytotoxic changes in the lymphoreticular tissue after implantation of the tumor cells were studied. Gravimetric changes found in the thymus and the spleen suggest that the thymus plays an essential role until death in a tumor-bearing host and that the thymus and the spleen have inverse functions in relation to tumor growth. In this study, no association was found between the large pyroninophilic cells in the deep cortex of the lymph nodes and the cytotoxicity against the tumor cells. However, enlargement of the deep cortex of lymph nodes may be associated with cytotoxicity against these tumor cells.

Aneurysms of the descending aorta: surgical experience with three methods of adjunct procedures.

Thirteen patients were surgically treated for the repair of aneurysms of the descending aorta, using three different types of adjunct procedures--an external temporary bypass with a vascular prosthesis, a tridodecylmethylammonium chloride (TDMAC) or a partial cardiopulmonary bypass. There was no operative death, though one patient died 73 days following surgery. Significant intraoperative morbidity occurred in 3 patients: one had ventricular fibrillation and the other two massive hemorrhages. There was no instance of paraplegia or renal failure. The only significant complication that developed was pulmonary insufficiency in two patients with a pump bypass. The mean operative time and the mean aortic occlusion time in patients with the TDMAC shunt were shorter than the times in patients with the vascular prosthetic shunt or the pump bypass. TDMAC shunt required no special equipment and cannulation was simpler and safer.

Effects of coenzyme Q10 added to a potassium cardioplegic solution for myocardial protection during ischemic cardiac arrest.

To evaluate effects of coenzyme Q10 added to a potassium cardioplegic solution for myocardial protection, 17 mongrel dogs underwent 60 minutes of ischemic cardiac arrest under cardiopulmonary bypass. Cardiac arrest was induced by infusing the cardioplegic solution into the aortic root every 20 minutes. Experimental animals were divided into three groups according to the cardioplegic solution used. In Group 1, we used our clinical potassium cardioplegic solution (K+, 22.31 mEq/L); in Group 2, potassium cardioplegic solution with coenzyme Q10 added (coenzyme Q10, 30 mg/500 ml of solution); and in Group 3, cardioplegic solution with coenzyme Q10 solvent. Exogenous coenzyme Q10 in the cardioplegic solution provided significantly high myocardial stores of adenosine triphosphate and creatine phosphate and a low level of lactate during induced ischemia and reperfusion. Furthermore, percent recovery of the aortic flow in Group 2 was significantly higher than that in the other two groups. Ultrastructures of the ischemic myocardium in Group 2 were better preserved than those in Group 1. Addition of coenzyme Q10 to potassium cardioplegia resulted in improved myocardial oxygen utilization and accelerated recovery of myocardial energy metabolism after reestablishment of circulation.

A quantitative description of flagellar movement in golden hamster spermatozoa.

Flagellar movement of golden hamster spermatozoa obtained from the testis and the caput and cauda epididymides was observed by a light microscope while holding them at their heads with a micropipette. Flagellar movement of capacitated spermatozoa and of reactivated spermatozoa demembranated with Triton X-100 was also observed. Testicular and caput epididymal spermatozoa showed weak movement in Tyrode's solution, whereas cauda epididymal spermatozoa showed vigorous movement. The flagellar bends of the cauda epididymal spermatozoa were almost planar. Capacitated spermatozoa moved with waves of a large amplitude. Demembranated spermatozoa reactivated with ATP only had a latent period before the initiation of flagellar movement, and beat at low frequency, whereas demembranated spermatozoa reactivated with both ATP and cAMP began to move immediately at high frequency. Thrust and hydrodynamic power output were calculated using the parameters for the typical waveforms of cauda epididymal spermatozoa before and after capacitation. The possible role of the large amplitude beat in capacitated spermatozoa is discussed. A comparison of the 'principal' and 'reverse' bends in golden hamster sperm flagella as defined by Woolley (1977) with those in sea urchin sperm flagella suggests that the so-called 'principal' bend in golden hamster sperm flagella corresponds to the reverse bend in sea urchin sperm flagella and vice versa.

Human X- and Y-bearing sperm differ in cell surface sialic acid content.

Analysis of cell surface charge of human sperm by means of free-flow electrophoresis revealed that the electrophoretic mobility of X-bearing sperm was faster than that of Y-bearing sperm, indicating that X-bearing sperm have a higher net negative charge on the cell surface than do Y-bearing sperm. Sialidase treatment of X- and Y-bearing sperm caused an obvious and progressive reduction in the electrophoretic mobilities of the two types of cells, until they finally merged into a single peak. This observation suggests that the difference in net negative charge between X- and Y-bearing sperm is due mainly to sialic acid content on the cell surface.
