Improved method of determination of biologically important C10:0-C22:6 fatty acids as their 2-nitrophenylhydrazides by reversed-phase high-performance liquid chromatography.

Fatty acids were separated by reversed-phase high-performance liquid chromatography after derivatization with 2-nitrophenylhydrazine hydrochloride in the presence of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride. The separation of a mixture of fourteen kinds of biologically important fatty acid hydrazides (C10:0-C22:6) was achieved within 15 min. Using margaric acid (C17:0) as internal standard, each fatty acid could be quantitated over the range of 2.5-5000 pmol per injection. Analytical recoveries ranged from 98.1 to 102.6%. The intra- and inter-assay coefficients of variation were less than 2.5 and 3.2%, respectively. For the determination of esterified fatty acids in fats and oils, the saponified mixture was directly derivatized without extraction. This method was compared gave similar fatty acid profiles to those obtained with the conventional liquid-liquid extraction method. It is simple, rapid and accurate for routine analyses of esterified fatty acids in biological materials.

Transformation of chenodeoxycholic acid to ursodeoxycholic acid in patients with Crohn's disease.

In vivo 7 beta-epimerization of chenodeoxycholic acid to ursodeoxycholic acid and the role of 7-ketolithocholic acid as an intermediate in this biotransformation were studied in 11 patients with Crohn's disease and in 5 healthy volunteers. The incorporation of deuterium into biliary ursodeoxycholic acid and 7-ketolithocholic acid was determined by computed gas chromatography-mass fragmentography after ingestion of a dideuterated chenodeoxycholic acid, chenodeoxycholic-11,12-d2 acid. The incorporation of deuterium into ursodeoxycholic acid increased to a peak level at 48 h in the patients with Crohn's disease, but was delayed in healthy volunteers. In 8 patients and 2 healthy controls there were small amounts of 7-ketolithocholic acid in bile. The incorporation of deuterium into 7-ketolithocholic acid was confirmed in only 2 patients and the peak level was noted at 48 h. These observations suggest that 7-ketolithocholic acid is an intermediate of this biotransformation in patients with Crohn's disease.

A computer model for simulating reflected ultrasound signals.

This letter describes a model for simulating an ultrasound signal reflected from a medium composed of randomly distributed scatterers, as typified by soft biological tissue, such as liver. The model is specified in terms of the effective transducer beam radius B and the mean scatterer spacing S. The novel feature of the model is that the transducer field is partitioned by packing cylinders, called microbeams, into concentric annular regions that lie parallel to the transducer axis. The radii of the microbeams and the annuli are related to S. An independent reflector sequence is generated for each microbeam, the microbeam sequences in each annulus are summed and convolved with the impulse response of a point reflector. The reflected waveform sequence is then generated by summing the annular contributions and convolving with a band-limited pulse waveform. Simulated signals were generated for different values of B and S and compared with actual signals reflected from two in vivo livers and a tissue-equivalent phantom. Estimates of the kurtosis for the simulated signals indicate the range of signals that can be generated by varying the values for S and B.

Cytochemical, immunologic, chromosomal, and molecular genetic analysis of a novel cell line derived from Hodgkin's disease.

A novel cell line, KM-H2, was established from the pleural effusion of a patient with Hodgkin's disease of mixed cellular type. Multiple phenotypic studies were carried out with this cell line. Acid phosphatase and nonspecific esterase activities were detected. Rosette formation with T lymphocytes and the receptors for C3b and Fc portion of IgG were positive. Among the antigens tested with a total of 22 monoclonal antibodies defining hematopoietic cell subsets or lineages, Ki-1, Leu-M1, MCS1, HLA-DR, and OKT9 antigens were found to be positive. The other antigens reportedly specific for T cells, B cells, natural killer (NK) cells, monocytes, interdigitating reticulum (IR) cells and dendritic reticulum cells were negative. These phenotypic features were identical to those of the Sternberg-Reed (SR) and Hodgkin (H) cells in the fresh materials reported by other researchers. Moreover, the KM-H2 cells and the parental pleural effusion cells shared several structural chromosome anomalies. These findings indicated that the KM-H2 cells are derived from the SR and H cells. Molecular genetic analysis of the KM-H2 cells disclosed that the human immunoglobulin JH gene was rearranged but not the JK gene, and that the human T cell receptor beta chain gene was of the germline type. Based on these properties of the KM-H2 cells, Hodgkin's disease may be derived from a cell lineage other than T cell or B cell.

[Clinical effects of TA-077 in non-Hodgkin's lymphomas].

Ten patients with non-Hodgkin's lymphomas (NHL), six untreated and four with previous chemotherapy, were treated with TA-077, a new derivative of nitrosourea. Partial remission was observed in three untreated cases (30%) of NHL [Case 1: 71-year-old female with B cell lymphoma/diffuse small cell type, Case 2: 79-year-old male with T cell lymphoma/diffuse large cell type, and Case 3: 64-year-old female with adult T cell leukemia lymphoma (ATLL)]. Remission durations were as follows: Case 1; 33 days, Case 2; 38 days and Case 3; 14 days. Side effects were transient anorexia (40%), nausea & vomiting (30%), liver dysfunction (10%) and delayed hematological toxicities (80%). Hematological toxicities consisted of leukocytopenia (80%), thrombocytopenia (60%) and anemia (20%). Our study suggests that TA-077 is a useful agent as one of the drugs used in combination chemotherapy against NHL, since it was effective for refractory T cell malignancies such as ATLL.

Coronary thrombolysis with urokinase infusion in acute myocardial infarction: multicenter study in Japan.

The efficacy of intracoronary administration of urokinase was evaluated in 514 patients with acute myocardial infarction (anterior, 296 patients; inferior, 195; lateral or posterior, 18; and anterior and inferior, five). The time between onset of chest pain and coronary arteriography was 0.5 to 81.0 hr with an average of 5.0 hr. Initial administration of nitrates resulted in recanalization of the coronaries in 9.3%. Subsequently, urokinase was infused into the coronary arteries, and coronary thrombolysis was successfully achieved in 66.8%. The success rate was low in a group with average infusion speed of more than 30,000 units/min or with a total dose of urokinase of 480,000 units or less. Complications, mainly arrhythmias, were present in 111 patients (33.2%) of the 334 who had successful thrombolysis and in 18 patients (10.8%) of the 166 with unsuccessful thrombolysis, but serious hemorrhage was rare and no fatal case was reported. Patients who had successful thrombolysis had less in-hospital mortality than those who did not (6.3 vs 13.3%). Thus, coronary thrombolysis can be achieved effectively and relatively safely with a sufficient amount of intracoronary urokinase administration in acute myocardial infarction.

Nonlinear parameter tomography system using counterpropagating probe and pump waves.

In this paper, a novel tomographic system for imaging the nonlinear parameter (B/A) of biological objects is described. This parameter is closely related to the detailed properties of tissue, and may well provide a new powerful tool for ultrasonic tissue characterization. In our new system, an impulsive, relatively high power (10 mW/cm2), low frequency pump wave is applied from the opposite direction of a cw low intensity probe wave of high frequency (5 MHz) so that the phase of the probe wave is modulated sequentially by the product of the nonlinear parameter (B/A) along the beam (x axis) and the pressure of the impulsive pump wave. This modulated probe wave is detected and demodulated to derive the distribution of (B/A) along the x axis. Many responses are averaged to increase the S/N ratio. Inverse or other filtering operations are applied to widen the frequency bandwidth of the pump wave. The entire system is realized in hardware. The counterbeam orientation makes the imaging system compact, with easy access to many parts of the human body. Its resolution is two times that of the perpendicular system proposed previously by us and the attenuation of the pump wave can also be compensated for easily. A practical system aimed at breast and liver diagnosis is described. The principle of the method and the system construction are described. B/A images of several objects are given.

Effects of dietary butter fat on fecal bile acid excretion in patients with Crohn's disease on elemental diet.

The excretion rate of fecal bile acids was determined by gas-liquid chromatography in nine patients with Crohn's disease and six healthy volunteers under two dietary conditions with different amounts of fat: during nasoduodenal tube feeding with a fat-restricted elemental diet containing approximately 1.5 g/day of fat, and during the addition of 50 g/day of butterfat by peroral administration. The fecal bile acid excretion rate on the elemental diet was significantly greater in the patients with Crohn's disease than in the healthy controls. With ingestion of the additional fat, the excretion rate was significantly increased in the patients with Crohn's disease, but not significantly changed in the healthy controls. The bile acid excretion rate in Crohn's disease correlated with fecal fat excretion, but not with either fecal weight or disease activity. These studies show that the amount of dietary fat represents an important consideration in the evaluation of bile acid malabsorption in Crohn's disease.

Antitumor effects of bacterial lipopolysaccharide and tumor necrosis factor in mice.

The effects of lipopolysaccharide (LPS) against tumor growth and on cellular immunity were studied in comparison with those of tumor necrosis factor (TNF). LPS and TNF, which were administered into mice with tumors, induced hemorrhagic necrosis within 48 hours after the initiation of the treatment. LPS and TNF significantly inhibited the tumor growth, as compared with findings in the controls. There was no significant difference in inhibitory effect on tumor growth between mice treated with LPS or TNF once or twice. Both LPS and TNF prevented the decrease of delayed type hypersensitivity in the tumor bearing mice.

Identification and prognostic implications of tumor infiltrating lymphocytes--a review.

Cancer patients who have many tumor infiltrating lymphocytes (TIL) tend to have better prognoses. A relationship between prognosis and TIL or regional lymph node response is present in several malignant diseases. TIL are mainly T lymphocytes, as ascertained by immunological methods. Results of studies on T-lymphocyte subsets comprising TIL using monoclonal antibodies (OKT series and Leu series) are summarized in this review.

Antitumor effect of bacterial lipopolysaccharide (LPS) alone and in combination with lentinan on MH-134 tumors in C3H/He mice.

Using C3H/He mice, the antitumor effect of lipopolysaccharide (LPS) alone and in combination with Lentinan extracted from Lentinus edodes was studied. The influence of LPS on cellular immunity and the antitumor effect of the tumor necrosis factor (TNF) were also examined. LPS, which was administered into mice with tumor, induced hemorrhagic necrosis of the tumor within 48 h, demonstrating a high antitumor effect. When LPS was used in combination with Lentinan, the tumor growth was significantly inhibited as compared to that in the control mice. The combination of LPS and Lentinan prevented a decrease in the delayed type hypersensitivity in tumor bearing mice. Application of rabbit serum containing TNF resulted in hemorrhagic necrosis of the tumor within 48 h, as with LPS.
