Deletion of 3' untranslated region of human BSF-2 mRNA causes stabilization of the mRNA and high-level expression in mouse NIH3T3 cells.

Most lymphokine genes have an mRNA-destabilizing signal in their 3' untranslated sequences. In this study we demonstrated the usefulness of deletion of this sequence for lymphokine production. By expression of B cell stimulatory factor 2 (BSF-2) cDNAs with and without this region in mouse NIH3T3 cells, it was shown that mRNA of BSF-2 with the 3' destabilizing sequence was very unstable, and that by deletion of this region, the mRNA was stabilized. More than 10-fold BSF-2 productivity was observed in cells harboring the plasmid without the 3' region than in those with it.

Minimum enzyme unit for Na+/K+-ATPase is the alpha beta-protomer. Determination by low-angle laser light scattering photometry coupled with high-performance gel chromatography for substantially simultaneous measurement of ATPase activity and molecular weight.

The oligomeric state of canine renal NA+/K+ -ATPase solubilized by octaethylene glycol n-dodecyl ether (C12E8) was studied by means of low-angle laser light scattering photometry coupled with high-performance gel chromatography (HPGC). At around 0 degree C the solubilized enzyme was separated into the (alpha beta)2-diprotomeric and alpha beta-protomeric protein components with Mr values of 302,000 +/- 10,000 and 156,000 +/- 4,000, respectively, in approximately equal quantities. As the temperature of chromatography was increased toward 20 degrees C, the two protein components converged into a single major component. The Mr of this component depended on the monovalent cation included in the elution buffer, and was 255,000 or 300,000 in the presence of 0.1 M NaCl or 0.1 M KCl, respectively. A computer simulation technique showed that the solubilized enzyme was in a dissociation-association equilibrium of 2 protomers = diprotomer at 20 degrees C, and the difference in apparent Mr of the solubilized enzyme between the two species of monovalent cation was interpreted by an association constant (Ka) in the presence of 0.1 M KCl that was about 50-fold larger than in the presence of 0.1 M NaCl. In order to measure ATPase activity and Mr of the solubilized enzyme simultaneously, a TSKgel G3000SW column had been equilibrated and was eluted with an elution buffer containing 0.30 mg/ml C12E8 and 60 microgram/ml phosphatidylserine (bovine brain) as well as the ligands necessary for the enzyme to exhibit the activity at pH 7.0 and 20 degrees C. The solubilized enzyme was always eluted as a single protein component irrespective of the the amount of the protein applied to the column, ranging between 240 and 10 microgram. The Mr of the protein component, however, decreased from 214,000 and 158,000 with the decrease of the protein amount. The specific ATPase activity, however, remained constant at a level of 64 +/- 4% of that of the membrane-bound enzyme even in the range of protein concentration sufficiently low as to allow the enzyme to exist only in the protomeric form. Thus, the alpha beta-protomer is concluded to be the minimum functional unit for the ATPase activity. The value of Ka obtained from the concentration-dependent dissociation curve was 5 . 10(5) M-1 for the enzyme turning over, and 1.1 . 10(7) M-1 for the enzyme inhibited with ouabain. It was discussed, based on the values of Ka obtained, that the enzyme would exist as the diprotomer or the higher oligomer in the membrane.

Alpha-secondary tritium kinetic isotope effects for the hydrolysis of alpha-D-glucopyranosyl fluoride by exo-alpha-glucanases.

alpha-Secondary tritium kinetic isotope effects ranging from 1.17 to 1.26 were measured for the hydrolysis of alpha-D-glucopyranosyl fluoride (forming beta-D-glucose) catalyzed by several glucoamylases and a glucodextranase. These results indicate that cleavage of the C-F bond is slow and that the enzymic transition state has significant oxo-carbonium ion character. Strong support for this conclusion is provided by the agreement found in the case of Rhizopus niveus glucoamylase (alpha-TV/K 1.26; Km 26 mM) between measured values of the alpha-secondary deuterium kinetic isotope effects (alpha-DV/K 1.16; alpha-DV 1.20) and those calculated from the tritium isotope effect. The data are consistent with the promotion of an intramolecular elimination of fluoride by the present exo-alpha-glucanases based on their ability to stabilize, perhaps with a counter ion, the development of a carbonium ion-like transition state. Although the oxo-carbonium ion is formally denoted as an intermediate it could represent a transition state along a reaction pathway to a covalent glucosyl intermediate.

Localization of the fourth membrane spanning domain as a ligand binding site in the human platelet alpha 2-adrenergic receptor.

The human platelet alpha 2-adrenergic receptor is an integral membrane protein which binds epinephrine. The gene for this receptor has been cloned, and the primary structure is thus known [Kobilka et al. (1987) Science 238, 650-656]. A model of its secondary structure predicts that the receptor has seven transmembrane spanning domains. By covalent labeling and peptide mapping, we have identified a region of the receptor that is directly involved with ligand binding. Partially purified preparations of the receptor were covalently radiolabeled with either of two specific photoaffinity ligands: [3H]SKF 102229 (an antagonist) or p-azido[3H]clonidine (an agonist). The radiolabeled receptors were then digested with specific endopeptidases, and peptides containing the covalently bound radioligands were identified. Lysylendopeptidase treatment of [3H]SKF 102229 labeled receptor yielded one peptide of Mr 2400 as the product of a complete digest. Endopeptidase Arg-C gave a labeled peptide of Mr 4000, which was further digested to the Mr 2400 peptide by additional treatment with lysylendopeptidase. Using p-azido[3H]clonidine-labeled receptor, a similar Mr 2400 peptide was obtained by lysylendopeptidase cleavage. This Mr 2400 peptide corresponds to the fourth transmembrane spanning domain of the receptor. These data suggest that this region forms part of the ligand binding domain of the human platelet alpha 2-adrenergic receptor.

ATL-derived factor (ADF), an IL-2 receptor/Tac inducer homologous to thioredoxin; possible involvement of dithiol-reduction in the IL-2 receptor induction.

HTLV-I transformed T cells not only express a large number of interleukin-2 receptors [IL-2R/p55(Tac)], but also produce a factor named ATL-derived factor (ADF) that augments the expression of IL-2R/p55(Tac). Based on a partial N-terminal amino acid sequence, complementary DNA (cDNA) clones for human and mouse ADF were isolated and sequenced. Recombinant ADF produced by COS-7 monkey kidney cells showed IL-2R/Tac inducing activity on YT cells, which are sensitive for ADF. ADF mRNA was strongly expressed in HTLV-I(+) T cells lines, but not in inactivated cells (THP-1, unstimulated PBMC). Furthermore, in normal human peripheral blood mononuclear cells, the expression of ADF mRNA was enhanced by mitogens or phorbol myristate acetate, suggesting a possible involvement of ADF in the lymphocyte activation. Homology analysis revealed an unexpected relationship between ADF and dithiol-reducing enzyme, thioredoxin, involved in many important biological reactions such as the conversion of ribonucleotides into deoxyribonucleotides, or the stabilization of glucocorticoid receptors. The biological significance of the generation of a redox potential in lymphocyte activation, and the possible involvement of dithiol reduction in the induction of IL-2R/Tac are discussed.

Demonstration of different regional distributions of calcineurin subunits using monoclonal antibodies.

Immunohistochemical localizations of calcineurin subunits A (60 KDa) and B (20 KDa) were examined in a rat brain using subunit specific monoclonal antibodies. The immunoreactivity of the subunit A was abundant in the hippocampus, in the striatum, and in the thalamus, but weak in the neocortex. On the contrary, the immunoreactivity of the subunit B was more abundant in the cortex, and it was more ubiquitous than subunit A. The distribution of subunit A in the rat brain very well agreed with that of zinc, which is an intrinsic metal ion and a potent inhibitor of calcineurin phosphatase.

Isolation and characterization of low- and high-metastatic clones from murine RCT (Radiological, Chiba, and Toyama) sarcoma.

We have established low- and high-metastatic clones, named RCT(-) and RCT(+) cells, respectively, from the RCT (Radiological, Chiba, and Toyama) sarcoma spontaneously developed in a C3H/He male mouse by the limiting-dilution method in vitro or by the combination of the lung passages and limiting-dilution methods. After 20 serial passages in vitro, the metastatic potential of each clone did not alter. Morphologically, both cells were spindle-shaped, but RCT(+) cells were slightly thicker and larger than RCT(-) cells. The organization of actin-containing filaments was slightly poorer in RCT(+) cells than that in RCT(-) cells. Marked differences were observed in their growth characteristics and adhesiveness to plastic or collagen-coated surfaces, that is, RCT(+) cells grew more slowly but could adhere more rapidly and firmly to the surfaces than RCT(-) cells. RCT(+) cells were agglutinated by all lectins used but several lectins could not agglutinate RCT(-) cells. These results could be a reflection of the difference in oligosaccharide residues on the surface of each cell and, in part, might reflect the difference in organization of the actin-containing filaments that regulate the mobility of lectin receptors. No significant difference between these cell clones was noted in their sensitivity to natural-killer-cell-mediated cytotoxicity in vitro. RCT(-) and RCT(+) cells are considered to be the most useful experimental model for the study of the certain sarcomas.

Pharmacokinetics of famotidine in elderly patients with and without renal insufficiency and in healthy young volunteers.

The pharmacokinetics of the H2-receptor antagonist famotidine, after oral administration of a 20 mg tablet, has been studied in 10 elderly patients with normal renal function (CLCR greater than or equal to 59 ml.min-1, Mean = 80 ml.min-1), 5 elderly patients with renal insufficiency (CLCR less than or equal to 38 ml.min-1, Mean = 15 ml.min-1), and 6 healthy young volunteers. Elimination half-life in the elderly patients with renal insufficiency was significantly prolonged compared to the elderly patients with normal renal function and the young volunteers. The correlation coefficient between creatinine clearance and the elimination rate constant of famotidine was 0.672. Mean urinary recovery of unchanged drug up to 24 h in the young volunteers was 44%. The mean renal clearance of famotidine in the young volunteers (270 ml.min-1) was substantially greater than the creatinine clearance, 128 ml.min-1, which suggests the possibility of tubular secretion of famotidine.

Effect of the administration of cycloheximide to thioacetamide-primed rats or partial-hepatectomized rats on the RNA polymerase I activity in isolated nuclei of liver.

1. Twenty-four hours after administration of thioacetamide to normal rats, the activity and amount of RNA polymerase I in isolated liver nuclei were almost doubled. 2. When cycloheximide was administered to the drug-treated rats and normal rats 1 or 2 hr before death, the reduction in the activity was of the same degree, that is, about half of the activity of liver nuclei from normal rats. 3. Partial hepatectomy also caused about 2-fold increase in the RNA polymerase I activity in isolated liver nuclei after 16 hr, but the increased activity was almost completely abolished by injection of cycloheximide 1-3 hr before killing.

Single active site mechanism of rabbit liver acid alpha-glucosidase.

An acid alpha-glucosidase was purified from rabbit liver by fractionation with ammonium sulfate, and chromatographies on Sephadex G-100, CM-Toyopearl, Toyopearl HW-55F, and Toyopearl HW-65F column. The resulting preparation showed a single band on polyacrylamide disc gel electrophoresis. The molecular weight was estimated to be 1.03 X 10(4) by SDS-disc electrophoresis. The optimum pH was found to be 4.7. The alpha-glucosidase showed relatively high activity not only toward maltose but also toward alpha-glucans, such as shellfish glycogen, soluble starch, beta-limit dextrin, amylopectin, and amylose. The Km values for maltose and shellfish glycogen were 2.1 and 16 mM (the concentration of non-reducing glucose units), respectively, and the ratio of maximum velocities of hydrolysis of the two substrates was 100:133. The nature of the active site catalyzing the hydrolyses of maltose and shellfish glycogen was investigated by electrophoresis in the presence of urea and by kinetic methods. The purified enzyme was not separated into two components, maltase and glycogen hydrolase, in the electrophoretic gel containing 3 M urea, contrary to the report by Belenki and Rosenfeld ((1972) Biochem. Biophys. Res. Commun. 46, 443-448). In experiments with mixed substrates of maltose and glycogen, the kinetic features agreed very closely with those theoretically predicted for a single site mechanism. The essential ionizable groups, 1 (on the acidic side) and 2 (on the alkaline side), were identified as -COO- and -COOH for the hydrolysis of both substrates. Cations, Na+, K+, Mg2+, were about equally effective for the stimulation of enzyme action on maltose and glycogen.(ABSTRACT TRUNCATED AT 250 WORDS)

Location of signal sequences for membrane insertion of the Na+,K+-ATPase alpha subunit.

To study the membrane insertion of the Na+,K+-ATPase (EC 3.6.1.37) alpha subunit with six to eight transmembrane segments, mRNAs encoding the entire alpha subunit and its four different domains were prepared and translated in rabbit reticulocyte lysate with rough microsomal membranes. On the basis of the resistance of the membrane-inserted products to alkali extraction and the failure to insert the translation products into N-ethylmaleimide-treated membranes, it is suggested that at least two signal sequences are contained within the four transmembrane segments from the amino terminus of the alpha subunit.

Sarcolemmal Ca2+ transport activities in cardiac hypertrophy caused by pressure overload.

To examine Ca2+ transport activities in sarcolemmal membrane in cardiac hypertrophy caused by pressure overload, rats were subjected to aortic banding for 28 days. Heart-to-body weight ratio was increased by 46% in aortic-banded animals in comparison with the sham-operated rats. Ouabain-sensitive Na+-K+-ATPase activity in sarcolemma (SL) from hypertrophied hearts was not different from that in the control preparation. The initial rate of Na+-dependent Ca2+ uptake in SL vesicles from the hypertrophied hearts was stimulated by 53% compared with the control vesicles. ATP-dependent Ca2+ uptake and Ca2+-stimulated adenosinetriphosphatase (ATPase) activities in SL from hypertrophied hearts were increased by 35%. The number of Ca2+ channels estimated by [5-methyl-3H]nitrendipine binding was decreased by 33% in SL from hypertrophied hearts. Total and individual phospholipid contents in the hypertrophied heart preparations were not different from those in the control, except that phosphatidylcholine and phosphatidylethanolamine contents were significantly increased. Sarcolemmal preparations from hypertrophied hearts from the 22-wk-old spontaneously hypertensive rats exhibited changes in Na+-Ca2+ exchange and Ca2+-pump activities (similar to those observed in banded hearts), whereas the Na+-K+-ATPase activity decreased, [3H]nitrendipine binding increased, and phospholipid contents were not different. Thus, although differences were defined between two types of hypertrophy, these results suggest alterations in the sarcolemmal Ca2+ transport activities that may serve as an adaptive mechanism for the removal of Ca2+ from the myocardial cells during the development of cardiac hypertrophy.

Wall motion asynchrony prolongs time constant of left ventricular relaxation.

To evaluate the effects of wall motion asynchrony on left ventricular (LV) relaxation, we performed atrioventricular sequential pacing with the second stimulation at six epicardial sites in open-chest anesthetized dogs. Myocardial segment lengths in the basal, mid, and apical LV free wall were measured by ultrasonic crystals. The standard deviation of interval from the onset of the QRS complex to that of elongation in each segment length was used as a quantitative index for asynchrony (asynchrony index, AI). The AI increased significantly in all sequential pacing modes compared with the control right atrial pacing. The time constant (T) of LV relaxation derived from exponential fit with zero-asymptote was prolonged significantly in all sequential pacing modes except for pacing at the LV base. In each dog there was a good correlation between changes in AI and T [r = 0.61 - 0.98 (mean = 0.84)]. Since the regional inactivation process of the myocardium is considered to be unchanged during these interventions, we concluded that asynchronous wall motion plays an important role in the impairment of LV relaxation.

Pharmacokinetics of cefpiramide in healthy volunteers, patients with various degrees of renal dysfunction, and patients on hemodialysis.

The elimination kinetics of cefpiramide, a parenteral semisynthetic cephalosporin derivative, were studied in 6 healthy volunteers and 31 patients with renal dysfunction after a single 500-mg intravenous injection. Cefpiramide concentrations in serum and urine were determined by an agar-well diffusion method. The serum levels in patients with renal impairment were slightly higher than those in volunteers during the period from 30 min to 24 h after the administration. The mean serum half-life during the beta-phase was 4.06 h in the volunteers and was slightly prolonged in the patients. The mean urinary recovery during 24 h amounted to 21.5% in the volunteers and decreased with reduced renal function. Hemodialysis did not enhance the clearance of cefpiramide.

Pathophysiology and evaluation of severity of congestive heart failure on the basis of venous characteristics.

To elucidate the possibility of grading the severity of congestive heart failure by using venous characteristics, we constructed venous pressure-volume curves (PVR) and calculated venous stiffness constants (K). In addition, effects of vasoactive drugs on venous distensibility were studied. A venous pressure-volume curve could be fit well by an exponential curve (r = 0.98 +/- 0.01). The PVR was shifted to the left with an increase in the clinical severity of congestive heart failure. The exponent of these curves, K, increased as the PVR was shifted to the left. K correlated with heart rate (r = 0.52, p less than 0.01), right atrial pressure (r = 0.54, p less than 0.02) and mean pulmonary arterial pressure (r = 0.47, p less than 0.04). Nitroglycerin and amrinone dilated veins and decreased K by 19.6 +/- 6.9% (p less than 0.03) and 14.0 +/- 4.3% (p less than 0.02), respectively. Changes in K (delta K) during the nitroglycerin and amrinone infusions correlated closely with the baseline K (delta K = -0.41 K + 0.22, r = 0.92, p less than 0.01). Therefore, the venodilating effects of these drugs were greater in patients with more severe congestive heart failure. The venous stiffness constant could be useful to grade the severity of congestive heart failure.

[Determination of dibutyltin compounds in polyvinyl chloride resin by capillary gas chromatography].

Dibutyltin compounds (DBTC) in polyvinyl chloride resin (PVC) were examined by capillary gas chromatography using a flame photometric detector (FPD). DBTC was extracted with a carbon tetrachloride-methanol (2:1) mixture under reflux. The extract was mixed with a hydrobromic acid solution. Dibutyltin dibromide was converted to dibutyldipentyltin (DBDPeT) by the Grignard reaction with pentylmagnessium bromide and then subjected to gas chromatography. A linear correlation between the amount of tin and peak height was observed in the range of 7.8 to 250 pg of tin on log-log paper. Recoveries of DBTC added to PVC were 92.6-108.1%. The results show that this procedure is more useful and sensitive (the detection limit is 3-4 pg, as tin) than previous methods reported.
