Monoclonal antibodies which preferentially bind to 22 K human growth hormone rather than its 20 K variant.

We have established 13 hybridoma cell lines which secrete mouse IgG1 monoclonal antibodies (McAbs) to human growth hormone (hGH). Binding affinity and binding specificity of McAbs were analyzed by competitive radioimmunoassay. Among these McAbs, CL. B1 showed a high affinity of 9.8 x 10(8) l/mol, and all McAbs so far tested showed very weak cross-reactivity or none at all with human prolactin (hPRL) and human chorionic somatomammotropin (hCS; human placental lactogen). Analysis of binding sites of McAbs using hGH variant and fragments in both ELISA and RIA demonstrated that McAbs could be classified into two groups. All the McAbs obtained in this study bound to plasmin-digested fragment S2 (hGH 1-134 and 141-191) and fragment alpha 3 (hGH 1-134 and 147-191). However, five (such as 1D2) out of 13 McAbs bound to fragment F1 (hGH 1-134) and others (such as CL. B1) did not. The McAb CL. B1 in the latter group showed low affinity with 20 K hGH (residue 32-46 deleted in native 22 K hGH) in contrast to high affinity with hGH (22 K). This suggests that the former McAbs recognize an epitope located at the N-terminal two-third part of hGH. In contrast, the McAbs of the latter group are likely to recognize three-dimensional structure of native 22 K hGH.

[Simultaneous determination of tributyltin and its metabolites, dibutyltin and monobutyltin, in biological materials by capillary gas chromatography].

Determination of tributyltin and its metabolites, dibutyltin and monobutyltin, in biological materials was made by capillary gas chromatography (C-GC) using a flame photometric detector (FPD). Butyltin compounds (BuTC) were extracted (as bromides) from tissue homogenates with hydrobromic acid and ethyl acetate. These compounds were converted to pentyl derivatives with pentyl Grignard reagent and then analysed by C-GC. The recoveries of each BuTC added to tissues were 96-99% for monobutyltin, 87-93% for dibutyltin and 90-93% for tributyltin. The detection limit of BuTC was 4-5 pg as tin. This method was applied to the analysis of BuTC in the liver and kidney of rats orally administered tributyltin chloride. Time course of three BuTC showed that the maximum value appeared 24 h after administration of the tin compound, which was followed by a rapid decrease. The order of the concentration of BuTC in both organs was dibutyltin greater than tributyltin greater than monobutyltin. The rate of dealkylation was more rapid in liver than in kidney.

[The effects of the viewpoints and observers' traits on the observational learning of altruistic behavior].

This study was to examine the changes of altruistic responses through observational learning (OL) in elementary school children (4th graders) with the conditions of the different viewpoints and observers' traits. The OL was the symbolic modeling of altruistic behavior by presentation of pictures with narration. All the stories had the same situation, but differed whether the actor behaved himself in an altruistic fashion to the object, or not. Observers' viewpoints were determined by the instruction: they were told to observe the story as if they were one of the characters and to pay attention to emotional responses or behavior of the character. The emphasis of each story corresponded to the viewpoint. Out of these eight combined experimental conditions six were performed. Traits were concerned with observers' impression, empathy, and evaluation of the story. The response measure was the change of rating between pre- and post-questionnaires through observation. The effect of OL was higher significantly when the viewpoint directed to the characters' emotion. Observers who reacted strongly to empathy and impression items showed higher altruistic changes.

[Correlation of serum gamma-seminoprotein and serum prostate-specific antigen in patients with prostate cancer].

Enzyme immunoassays (EIAs) using rabbit anti-gamma-seminoprotein (gamma-Sm) antibody and rabbit anti-prostate-specific antigen antibody (PA) for measurements of gamma-Sm and PA were developed in order to determine the correlation between serum gamma-Sm levels and serum PA levels in patients with prostate cancer. Each EIA for measurement of gamma-Sm or PA was revealed to be sensitive and reproducible by fundamental analysis of the quality of EIAs. A significant correlation was established between serum gamma-Sm levels and serum PA levels which were measured by these assays and expressed as protein concentration of seminal plasma (r = 0.99). This result coincided with our previous observation that gamma-Sm and PA are immunologically identical materials. We conclude, therefore, that gamma-Sm and PA should be regarded as tumor markers identical to each other for serodiagnosis of prostate cancer.

[Analysis of antigens recognized by anti-gamma-seminoprotein antibody and anti-prostate-specific antigen antibody by means of Ouchterlony method and western blotting method].

Antigens recognized by anti-gamma-seminoprotein (gamma-Sm) antibody and anti-prostate-specific antigen (PA) antibody were analyzed immunologically by Ouchterlony method and Western blotting method. The double immunodiffusion test demonstrated that anti-gamma-Sm antibody and anti-PA antibody reacted against an identical antigen in seminal fluid or prostate homogenate. In the immunoblotting examination, anti-gamma-Sm antibody and anti-PA antibody formed a single and identical band, which gave a molecular weight of 33,000, in seminal fluid and prostate homogenate. This study demonstrated that anti-gamma-Sm antibody and anti-PA antibody recognized a single and identical antigen in seminal fluid and prostate homogenate and suggested that an identical material could be measured in sera of patients with prostate cancer by immunoassay systems using anti-gamma-Sm antibody and anti-PA antibody.

Inhibition by triphenyltin chloride of cytosolic free calcium mobilization in human neutrophils stimulated by a chemotactic factor.

We examined the effects of triphenyltin chloride (TPTCl) on the signal transduction in cells, especially the rise of cytosolic free calcium in fura-2-loaded neutrophils stimulated by N-folmyl-L-methionyl-L-leucyl-L-phenylalanine (FMLP). The peak concentration of cytosolic free calcium in neutrophils stimulated by FMLP (10(-7) M) after treatment with 4 mM EGTA was inhibited by TPTCl in a dose-dependent manner and completely blocked in the concentration range of 2.5 to 10 microM in the absence of extracellular calcium. TPTCl also had dose-dependent inhibitory effects on the superoxide anion production and the secretion of beta-glucuronidase in neutrophils stimulated by FMLP (10(-7) M). These results indicate that TPTCl is potent inhibitor of cytosolic free calcium mobilization in FMLP stimulation, associated with superoxide anion production and the secretion of beta-D-glucuronidase.

[Characteristics of high- and low-metastatic clones isolated from murine undifferentiated sarcoma (RCT sarcoma)].

We have established low- and high-metastatic clones from the undifferentiated RCT (Radiological Science-Chiba Cancer Center-Toyama Medical and Pharmaceutical University) sarcoma spontaneously developed in a C3H/He male mouse. Metastatic lesions of RCT sarcoma at the lung were inoculated into the subcutaneous tissue which was then cultured and cloned. Clones potential of low- and high-metastatic were named RCT(-) and RCT(+), respectively. RCT(-) cells were cloned by the limiting dilution method in vitro and RCT(+) cells were cloned by the combination of lung passages and limiting dilution methods. After 20 serial passages in vitro, the metastatic potential of each clone remained unchanged. Significant differences were noted in their growing property and adhesiveness to plastic or collagen-coated surfaces; RCT(+) cells grew more slowly, adhered more rapidly and firmly to the surfaces than those of RCT(-) cells. RCT(+) cells were agglutinated by all types of lectins used in this study, while several types of lectins could not agglutinate RCT(-) cells. These results would indicate that there was the difference in oligosaccharide residues on the surface of each cell. No significant difference between these cell clones in their sensitivity to natural killer cell-mediated cytotoxicity in vitro. We believe that RCT(-) and RCT(+) cells are very useful experimental model to study the mechanism of sarcoma metastasis.

Steric course of the hydration of D-gluco-octenitol catalyzed by alpha-glucosidases and by trehalase.

Crystalline Aspergillus niger alpha-glucosidase and highly purified preparations of rice alpha-glucosidase II and Trichoderma reesei trehalase were found to catalyze the hydration of [2-(2)H]-D-gluco-octenitol, i.e., (Z)-3,7-anhydro-1,2-dideoxy-[2-2H]-D-gluco-oct-2-enitol, to yield 1,2-dideoxy-[2-2H]-D-gluco-octulose. In each case, the stereochemistry of the reaction was elucidated by examining the newly formed centers of asymmetry at C-2 and C-3 of the hydration product. The C-1 to C-3 fragment of each isolated [2-2H]-D-gluco-octulose product was recovered as [2-2H]propionic acid and identified by its positive optical rotatory dispersion as the S isomer, showing that each enzyme had protonated the octenitol (at C-2) from above its re face. 1H NMR spectra of enzyme/D-gluco-octenitol digests in D2O showed that the alpha-anomer of [2-2H]-D-gluco-octulose was exclusively produced by each alpha-glucosidase, whereas the beta-anomer was formed by action of the trehalase. The trans hydration catalyzed by the alpha-glucosidases was found to be very strongly inhibited by the substrate; the cis hydration reaction catalyzed by the trehalase showed no such inhibition. Special importance is attached to the finding that in hydrating octenitol each enzyme creates a product of the same anomeric form as in hydrolyzing an alpha-D-glucosidic substrate. This result adds substantially to the growing evidence that individual glycosylases create the configuration of their reaction products by a means that is independent of donor substrate configuration, that is, by a means other than "retaining" or "inverting" substrate configuration.

Stereochemical studies of D-glucal hydration by alpha-glucosidases and exo-alpha-glucanases: indications of plastic and conserved phases in catalysis by glycosylases.

Alpha-Glucosidases from Aspergillus niger, pig serum, ungerminated rice, buckwheat, and sugar beet seeds (but not from brewers' yeast or honeybee) were found to catalyze the hydration of D-glucal. Each reactive alpha-glucosidase, incubated with D-glucal in D2O, was shown to protonate (deuteriate) this prochiral substrate from above its re face, i.e., from a direction opposite that assumed for protonating alpha-D-glucosidic substrates. At the same time, D-glucal hydration catalyzed by three of the alpha-glucosidases that acted rapidly enough in D2O to determine product configuration was found to yield 2-deoxy-D-glucose of the same specific (alpha-) configuration as the D-glucose produced from alpha-D-glucosidic substrates. These findings substantially extend those reported earlier for the hydration of D-glucal by one (Candida tropicalis) alpha-glucosidase preparation. Together with other recent results, they suggest that the process of catalysis by alpha-glucosidases (and perhaps glycosylases in general) may comprise two separate and separately controlled parts, namely, a "plastic" phase concerned with substrate protonation and a substrate-unrelated "conserved" phase concerned with the creation of product configuration. In contrast to the alpha-glucosidases, three "inverting" exo-alpha-glucanases (Arthrobacter globiformis glucodextranase; Rhizopus niveus and Paecilomyces varioti glucoamylase) were found to protonate D-glucal from below its si face. Further, whereas the catalysis of D-glucal hydration by the alpha-glucosidases was intensively inhibited by excess substrate, that promoted by the exo-glucanases showed no detectable substrate inhibition.

Absence of antiviral activity in recombinant B cell stimulatory factor 2 (BSF-2).

Recombinant B cell stimulatory factor 2 (rBSF-2) did not display any detectable level of antiviral activity when using human diploid fibroblasts, DIP-2, FS-4, FS-7, amnion-derived WISH and FL cells with vesicular stomatitis virus (VSV) and Sindbis virus as challenging agents (less than 2.5 X 10(2) IU/mg). Furthermore anti-IFN-beta could not neutralize the immunoglobulin-inducing activity of BSF-2. Moreover anti-BSF-2 could not neutralize the antiviral activity of IFN-beta. The data indicate that BSF-2 is functionally and immunologically not related to IFNs.
