Ultrastructure and function of interdigitating cells in the guinea pig thymus.

Electron micrsocopic investigation of the thymus of normal guinea pigs of both sexes, approximately 3 months of age, have shown that the organ contains at least two types of non-lymphoid cells: epithelial reticulum cells and so-called interdigitating cells. The latter have been observed in the inner part of the cortex and are morphologically characterized by an irregularly shaped or kidney-shaped nucleus, many tubulovesicular cytoplasmic structures accumulated mostly within the cytocentre as well as by finger-like protrusions of the cytoplasm. These interdigitating cells are in close contant with neighbouring lymphocytes with partly destroyed pycnotic nucleus.

[Contribution to the epithelial tumors of the ciliary body (clinical, histopathological and electronmicroscopical observation) (author's transl)].

An adenoma of the non pigmented epithelium of the ciliary body was supposed to be an irisciliary body melanoma (32P-test positive) and was treated by irido-cyclectomy. Vision acuity remained 0,4 one year after operation. The tumor formed adenoid tubes lined with a cubical or cylindrical epithelium. The cytoplasm of all tumorcells was threaded with compactly arranged tonofilaments. The histopathological examination showed unequivocal the benignant character of this tumor. Nevertheless the tumorcells were infiltrating the deep scleral layer in one place. Also the lens was deformed by the tumor and developed a local opacity in the periphery. It is refered to the difficulty of a clear histopathological distinction between adenomas, adenocarcinomas and pseudo-adenomatous hyperplasia of the ciliary body (Fuchs adenoma).

[Electron microscopic representation of the pili structure of Neisseria gonorrhoeae (author's transl)].

The technique of negative staining and ultra-thin section has been used for investigations of 30 Neisseria gonorrhoeae strains in order to represent the structure of pili (fimbriae) electron microscopically. The staining of the gonococci was effected by phosphotungstic acid (0,5%). The pili ascertained were 30 to 60 A thick. In course of in vitro passages up to 10. subculture morphological changes of the pili have been observed. The application of trisbuffer or solution of Hylase (hyaluronidase) showed not any improved results in comparison with buffered NaCl-solution as suspension medium. The investigation of ultra-thin sections showed that the structure of the pili could be exhibited not clearly. Therefore, these technique seems to be not suitable for qualitative representative of the pili.

[New studies on the pathogenesis of lupus erythematosus].

The pathogenesis of the lupus erythematosus results from co-operation of three principles: (1) genetical disposition, (2) increased reactivity of the immune system, (3) different exogenic influences. The genetical disposition is confirmed by family investigations, metabolic disorders and immune anomalies as well as by parallels to animal models. The reaction manner of the immune system is genetically determined. Exogenic factors influence the immune system (behaviour) either as starter or by modifying the genetical material. The most striking humoral immune phenomenon is an immense number of (auto-)antibodies. Investigations of xeroderma pigmentosum as well as with DNA of different antigenity have shown that an increase of the antigenicity is unlikely for this phenomenon. In the serum of patients there were established and partially characterized factors (mitogens, granulocytic adherence-factor) for increasing the immune reactivity. The increase of such factors may be the sequence of a T-suppressor cell-defect. Like-wise no interdigitating cells in systemic lupus erythematosus have been found, cells which may be responsible for the terminal differentiation of T-lymphocytes. The mode of action of exogenic factors is represented and discussed with the example of the UV provocation.

[Non-secreting plasmacytomas].

In non-secernent plasmocytomas there are no characteristic changes of serum protein. Diagnostic difficulties may be overcome by cytomorphological examinations, immunofluorescence and electron microscopy. By referring to 4 own observations the value of the electron microscopic characteristics of plasma cells is demonstrated. Subtile investigations in protein diagnostics are required for avoiding incomplete monoclonal immunoglobulins which may greatly enter the kidneys to be overlooked. Generally the prognosis of non-secernent plasmocytomas is not worse than that of other forms.

[A contribution to the ultrastructure of malignant melanomas of the iris (author's transl)].

After a short description of clinical facts, electron microscopic findings on four malignant melanomas of the iris are described. Though all tumors had been classified as being of the spindle cell type, the ultrastructure of the tumor cells shows remarkable differences between the four cases. This can be seen especially by comparison of the nucleoli, the chromatin, and the cytoplasmatic structures. All four tumors have the same spherical type of melanosomes. No morphologic features pointing to relative clinical harmlessness of iris melanomas were found.

[Immunomorphological and ultrastructural findings on the musculature in progressive scleroderma].

Muscle biopsies taken from 30 patients with generalized scleroderma were studied immuno-histologically as well as by light and electron microscopy. The immunological investigations have demonstrated that an increase of IgM and/or complement depositions in capillaries and in the walls of small arteries have been involved in most cases. Similar alterations were found in the subsarcolemmal zone. Vascular Ig-G precipitates were observed in the later stage of the disease only, whereas a positive reaction of fibrinogen have been proved in the most cases. By light and electron microscopy a variety of pathologic alterations have usually been found, but the most prominent changes involved were thickening of the capillary basement membrane and interstitial fibrosis. Scattered cellular infiltrates consisting of lymphocytes, histiocytes and plasmocytes were also seen either perivascularly or throughout the interstice. Around fibroblasts and capillaries, fine protocollagen fibrils and glycosaminoglycan filaments have been observed. They may partly represent fibrogenesis. Filamentous structures could be seen within the cytoplasm of numerous fibroblasts.

[Experience with a locally applicable antirheumatic (author's transl)].

Clinical trials with 2-(2-hydroxyethoxy)-ethyl-N-(a,a,a-trifluoro-m-tolyl)-anthranilate (etofenamate, active principle of Rheumon Gel) were carried out on a total of 69 patients suffering from extra-articular rheumatic complaints and painful spinal syndromes as well as from post-operative and post-accident complaints. Criteria for assessment included spontaneous pain, pressure pain and pain on movement and also limitation in joint mobility. The new gel formulation proved to have a good analgesic efficacy both in a double blind trial against placebo gel and the open study. Those patients treated with Rheumon Gel showed improvement in the symptoms spontaneous pain and pain on movement, in a large number of which patients showing improvement, in a large number of which patients showing improvement the spontaneous pain was completely eliminated. The differences in therapeutic effect compared to the placebo group as well as those of the final clinical assessment were statistically significant. Particularly good therapeutic results were observed in extra-articular rheumatic complaints and in activated arthroses. The skin tolerance of Rheumon Gel was found to be excellent. The gel form permits good and rapid penetration through the skin. The nursing staff also willingly administered the product because of its pleasant protective properties for skin and clothing.

Ultrastructure of macrophage-lymphocyte interaction in purpura pigmentosa progressiva.

Thin sections of pathologically changed skin lesions of patients suffering from purpura pigmentosa progressiva, a disorder which seems to be mediated by delayed type of allergy, were electron microscopically investigated. In 5 of the 7 cases we found cell infiltrates of the dermis consisting of many lymphocytes frequently in apposition to macrophages (histiocytes). The cell interactions observed occurred either by microvilli-like cytoplasmic protuberances or by local areas of the plasma membrane apposition. No submicroscopical alterations of the cytoplasm were found in the interacting cells although numerous macrophages show abundant microfilaments. The findings observed are interpreted as the ultrastructural substrate of cell cooperation for presenting and transfering antigens from macrophages to T lymphocytes. However, the question concerning the mechanism of the transfer remains open.
