[Electronmicroscopic studies on allergic vasculitis].

Electron microscopic investigations were carried out on cutaneous lesions of different morphological appearances in 8 cases of vasculitis allergica Ruiter. The endothelial damages observed (defoliating, vacuolisation, forming of microvilli and bizarre membrane structures within the lumen, interruption of the endothelial continuity) put the question for participating of a Shwartzman phenomenon. The multilayered formation of the basement membrane or their homogeneous spreading may be a sequence of damage and a compensation phenomenon. It is quite possible that filamentous structures, being within the basal cytoplasm of the endothelial cells, represent immune-complexes. During the necrobiotic phase, the perivascular infiltrates partly consists of leucocytes, fibrin, and debris, and during the later phagocytotic phase, the infiltrates partly consists of lymphocytes and histiocytes (macrophages). The findings are explained as morphological substratum of the Arthus-reaction.

[Hyde's nodular prurigo: an electronmicroscopic study].

Electron microscopic studies were carried out on cutaneous lesions in one case of prurigo nodularis Hyde. The cells of the basal layer partly showed large paranuclear vacuoles, which were sometimes divided by thin membrane structures. The intercellular space of the stratum basale and stratum spinosum was usually dilated, exhibiting acantholytic phenomen. Furthermore, numerous cells of these layers contain vacuolic-like mitochondria and polymorphous granular structures. Melanocytes and Langerhans-cells have not been observed, however, Merkel-cells were found in the basal layer, containing characteristic electrondense granules measuring 80 to 100 nm in diameter. The stratum granulosum frequently showed large intercellular complexes of numerous desmosomes, which sometimes were forming desmosomal bridges crossing the intercellular space. The capillaries in the dermis were usually widened and frequently surrounded by edematous alterations. The cytoplasm of the endothelial cells appeared very electrondens. Numerous unmyelinated axons were observed within the subepidermal zone containing mitochondria and single granules, measuring 80 to 100 nm in diameter.

Electron microscopic examinations of a conjunctival squamous cell carcinoma before and during beta-irradiation.

An electron microscopic study was performed on a conjunctival squamous cell carcinoma in an 82-year-old man. Biopsies were taken before and during beta-irradiation and the morphologic features of nonirradiated tumor cells were compared to those after application of 10,000 rad. There occur striking changes in the nucleus and the cytoplasm after irradiation. The nuclei show hypertrophic and electron-dense nucleoli as well as altered chromatin structures. The cytoplasm of many cells appears destroyed and contains numerous vacuole-like structures, as well as swollen mitochondria. It is of interest that numerous normal lymphoid cells were present within the irradiated tumor.

[Application of morphological and histochemical methods for evaluation of cytostatic effects on organ cultures of malignant tumors].

Histochemical and morphological methods are reported to test the sensitivity of organ culture of human and animal tumors to cytostatics. Topochemical investigations of lactate-dehydrogenase and NADH-diaphorase in organ culture of UV-induced mice tumors have shown that there is a considerably decreased enzyme activity, as compared with the controls, after application of a clinical as well as a tenfold clinical dose, especially after treatment with trenimon and 5-fluorurazil. On the other hand a decreased activity of these enzymes was found in organ cultures of tumors induced by methylcholanthren, following application of vinblastin and 5-fluorurazil. Three out of five of the investigated mammary carcinomas exhibited a markedly decreased enzyme activity when tested with cytostatics. Furthermore, a significant decrease of the enzyme activity was observed in two ovarian carcinomas and in two malignant melanomas of three tumors examined in each case. Following the administration of cytostatics, electronmicroscopical studies showed marked changes in the mitochondria in mammary carcinomas as well as in ovarian carcinomas, whereas no alterations were found in melanomas. The findings are discussed in the light of individual chemotherapy of cancer.
