Cessation of autonomous proliferation of mouse lymphoma EL4 by fusion with a T cell line.

Benzanthracene-induced C57BL/6 (H-2b) mouse T-cell lymphoma EL4 (a thymidine kinase-deficient cell line) was fused by using polyethylene glycol with an Mlsa (Mls for minor lymphocyte stimulatory) antigen-dependent T cell line, which was designated G4 and had been derived from a C3H/He mouse (H-2k), and the fused cells were cultured in HAT medium. Although no growing cells appeared in most of these fusions, we consistently obtained growth-arrested H-2Kb-positive cells from the fused cell populations by the panning method. The cells were tetraploid and were able to proliferate in response to Mlsa antigen. Three H-2Kb-positive clones, isolated by limiting dilution from three different fusions, were shown to be EL4 x G4 hybrids, because (1) they had both H-2k and H-2b antigens; (2) each of the clones had one submetacentric chromosome which was a marker chromosome of EL4, and they were tetraploid with modal chromosome numbers of 74, 78, and 79, respectively; (3) they had 4 isozymes of both parental cells. These results indicate that EL4 lymphoma cells cease to proliferate when fused with T cell line G4. The malignant phenotype of lymphoma EL4 is thus suppressed at the level of cell transformation by the introduction of the G4 cell genome.

The structure of the asparagine-linked sugar chains of bovine brain ribonuclease.

The asparagine-linked sugar chains of bovine brain ribonuclease were quantitatively released as oligosaccharides from the polypeptide backbone by hydrazinolysis. After N-acetylation, they were converted into radioactively-labeled oligosaccharides by NaB3H4 reduction. The radioactive oligosaccharide mixture was fractionated by ion-exchange chromatography, and the acidic oligosaccharides were converted into neutral oligosaccharides by sialidase digestion. The neutral oligosaccharides were then fractionated by Bio-Gel P-4 column chromatography. Structural studies of each oligosaccharide by sequential exoglycosidase digestion in combination with methylation analysis revealed that bovine brain ribonuclease showed extensive heterogeneity. It contains bi- and tri-antennary, complex-type oligosaccharides having alpha-D-Manp-(1----3)-[alpha-D-Manp-(1----6)]-beta-D-Manp -(1----4)-beta-D- GlcpNAc-(1----4)-[alpha-L-Fucp-(1----6)]-D-GlcNAc as their common core. Four different outside oligosaccharide chains, i.e., beta-D-Galp-(1----4)-beta-D-GlcpNAc-(1----, alpha-Neu5Ac-(2----6)-beta-D- Galp-(1----4)-beta-D-GlcpNAc-(1----, alpha-Neu5Ac-(2----3)-beta-D-Galp-(1----4)- beta-D-GlcpNAc-(1----, and alpha-D-Galp-(1----3)-beta-D-Galp-(1----4)-beta-D-GlcpNAc-(1----, were found. The preferential distribution of the alpha-D-Galp-(1----3)-beta-D-Galp-(1----4)-beta-D-GlcpNAc group on the alpha-D-Manp-(1----6) arm is a characteristic feature of the sugar chains of this enzyme.

Increased fibrin/fibrinogen degradation products without increase of plasmin-alpha 2-plasmin inhibitor complex after hepatectomy for hepatocellular carcinoma.

To clarify the meaning of increased serum fibrin/fibrinogen degradation products (FDP) in the postoperative period of hepatectomy, blood coagulation and fibrinolysis were studied using recently devised laboratory assays of a group of 30 patients with hepatocellular carcinoma. Twenty of these cases were associated with liver cirrhosis. As a control group, 15 patients with colorectal carcinoma without liver diseases were also selected. In the early postoperative period following hepatectomy, a hypercoagulable state designated as intravascular thrombin generation was confirmed from the finding of increased plasma levels of fibrinopeptide A (FPA). Fibrinopeptide B beta 15-42 (B beta 15-42) in the plasma also increased immediately after the peak of FPA, followed by a gradual decline in B beta 15-42 levels. On the other hand, FDP in the serum increased significantly rather late in the postoperative period following hepatectomy without increased levels of plasmin-alpha 2-plasmin inhibitor complex. However, postoperative increase of fibrin/fibrinogen degradation products-D (FDP-D) was modest and not different from the colectomy group. Therefore, the relevance of intravascular coagulation in the hepatectomy for the patients with liver cirrhosis seems not to be significant, and then such an increase of FDP in the serum seems to be related to other mechanisms.

Chronic calcifying pancreatitis associated with primary hyperparathyroidism--report of a case and review of the literature.

A 34 year old male was hospitalized because of severe abdominal pain and diarrhea. An abdominal X-ray revealed multiple calculi in the head of pancreas and blood tests showed his serum calcium level to be high. He underwent surgery of the parathyroid gland and a parathyroid tumor was removed. Two months later, resection of the head of the pancreas was also performed. Eighteen months after his operation there has been no recurrence of abdominal pain or diarrhea and his serum calcium level is within the normal range. We report this case herein and also discuss the possible cause and effect relationship between primary hyperparathyroidism and pancreatitis, and the appropriate management, in relation to a review of the literature.

Serum bilirubin fractions in healthy subjects and patients with unconjugated hyperbilirubinemia.

Serum bilirubin fractions were determined by a newly developed, high performance liquid chromatography method in 23 healthy subjects, and in 15 patients with Gilbert's syndrome, five with type 2 Crigler-Najjar syndrome, seven with hemolytic disorders, and 26 with neonatal jaundice. In the healthy subjects, 92.6% of the bilirubin was unconjugated (UCB), 6.2% was bilirubin monoglucoronide (BMG), and 0.5% was bilirubin diglucuronide (BDG). Delta bilirubin (B delta) was not detected. In the patients, the percentage of UCB was significantly higher and that of BMG was significantly lower than in the healthy subjects. The proportion of BDG tended to decrease in Gilbert's and type 2 Crigler-Najjar syndromes; the proportion of B delta tended to increase in hyperbilirubinemia, except in Gilbert's syndrome. In particular, B delta was frequently detected in serum which also contained BDG (mainly in hemolytic disorders) or which presented with high concentrations (above 100 mumol/L) of UCB (mainly in type 2 Crigler-Najjar syndrome and neonatal jaundice). Trace amounts of (Z,E)- and/or (E,Z)-UCB were detected in approximately one fourth of the serum samples analyzed.

A single recessive non-MHC diabetogenic gene determines the development of insulitis in the presence of an MHC-linked diabetogenic gene in NOD mice.

To study the genetic control of insulitis in non-obese diabetic (NOD) mice, we performed breeding studies in crosses of NOD with non-diabetic strains, ICR-L-line Ishibe (ILI), non-obese non-diabetic (NON) and C3H/He mice. The ILI mouse serologically shared the same MHC Class I and Class II as the NOD mouse. Insulitis was defined as islets invaded by lymphoid cells. Periductular, perivascular and peri-insular lymphoid cell infiltrations were often observed in NOD mice and appear to be the initial lesion leading to insulitis. Such lesions, however, were found in 1-year-old ICR, ILI, NON and Cataract Shionogi (CTS) mice of the NOD's sister strain. The lymphoid cells did not invade the islets in ICR, ILI, NON and CTS mice. The incidence of insulitis was 0% in F1 generations and 40% in female backcrosses (BC) [(ILI x NOD)F1 x NOD] at 9 weeks of age, 48 and 50% in BC[(NON x NOD)F1 x NOD] and BC[(C3H/He x NOD)F1 x NOD] at 1 year of age, respectively. Backcross animals were typed for the MHC to investigate correlation between the development of insulitis and MHC haplotypes. Among the backcross females with insulitis, approximately half the animals were heterozygous for MHC(non/nod) in BC[(NON x NOD)F1 x NOD] and MHC(k/nod) in BC[(C3H x NOD)F1 x NOD]. Among the backcross females with no insulitis, approximately half the animals were homozygous for MHC(nod/nod) in BC[(NON x NOD)F1 x NOD] and in BC[(C3H x NOD)F1 x NOD]. The results suggest that a single recessive non-MHC diabetogenic gene determines the development of insulitis regardless of NOD MHC homozygosity or heterozygosity.

Plunging ranula localized in the parapharyngeal space.

Plunging ranulas are a mucous extravasation and usually originate from the sublingual gland. They dissect between the facial planes and muscle of the base of the tongue to the submandibular triangle. We report here a rare case of plunging ranula localized in the parapharyngeal space. The relevant anatomy is reviewed and discussed.

[Studies on quinolone antibacterials. I. Synthesis and antibacterial activity of 7-(2-aminoethoxy)-, 7-(2-aminoethylthio)-, and 7-(2-aminoethylamino)-1-cyclopropyl-6-fluoro-1,4-dihydro-4-oxo qui noline -3-carboxylic acids and their derivatives].

7-(2-Aminoethoxy)-, 7-(2-aminoethylthio)-, and 7-(2-aminoethylamino)-1-cyclopropyl-6-fluoro-1,4-dihydro-4-oxoq uin oline-3- carboxylic acids and their derivatives (11a--f, h, j, k, 12a--f, and 13a--f) were synthesized and their antibacterial activities were tested. Among them, compounds (13a, d) having a primary amino group at the terminal position of alkoxy and alkylthio groups were found to have excellent in vitro and in vivo antibacterial activity comparable to those of ciprofloxacin (5). Structure-activity relationship of these compounds was also stated.

Electrical catheter ablation for drug-refractory sustained ventricular tachycardia with old myocardial infarction--a case report.

We performed electrical catheter ablation (ECA) in a patient who had old myocardial infarction and drug-refractory sustained ventricular tachycardia (VT). ECA using 100J to the earliest activation site and the pace-mapping site failed to prevent the VT. ECA using the same energy to the site at which mid-diastolic potential was recorded during tachycardia could successfully ablate the VT. Therefore the site of energy delivery was important in VT with old myocardial infarction.

A new variant of the Es-4 locus in the rat.

A new variant of kidney esterase in the DK/Nac rat strain is reported. The new esterase was tentatively named ES-4C determined by a third allele of the Es-4 locus of Linkage Group V (LGV). Strain distribution was surveyed using 17 inbred strains, but no strain except for the DK/Nac strain possessed the ES-4C type. Although we surveyed outbred stocks (Jcl: Wistar and Jcl: SD) we could not find rats carrying the ES-4C type. Genetic analysis of the ES-4C type was carried out using mating experiments between DK/Nac and BUF/Nac (ES-4B). The results indicated that the new variant was controlled by the Es-4 locus and it was named the Es-4c allele.

Genetic profile of alloxan-induced diabetes-susceptible mice (ALS) and-resistant mice (ALR).

Two inbred strains from a foundation stock derived from Crj: CD-1 (ICR) mice were established after more than 20 generations of full-sib mating, and by simultaneous selective breeding for developing and not developing diabetes after alloxan administration (45 mg/kg in males, 47 mg/kg in females). To elucidate the genetic background of the two inbred strains, i. e., alloxan-induced diabetes-susceptible (ALS) strain and alloxan-induced diabetes-resistant (ALR) strain, their biochemical genetic markers and immunogenetic markers were examined. 1) For both strains, investigation of biochemical genetic markers at 19 loci and immunogenetic markers at 11 loci revealed no variation in any gene within the same strain, showing to be homogeneous, thus indicating establishment of the inbred strains. 2) The two strains differed from each other at 2 loci of biochemical genetic markers and at 5 loci of immunogenetic markers. 3) The ALS and ALR strains can be regarded as new inbred strains derived from ICR mice. 4) The results show that the marker genes of the two strains are different at 7 loci, but it remains unclear whether or not these genes are involved in the difference between the two strains in susceptibility to alloxan.

[Studies on the effectiveness of adjuvant hepatic arterial chemotherapy after hepatectomy for primary or metastatic liver cancer].

From January 1980 to March 1990, 399 cases of primary liver cancer (hepatocellular carcinoma 357, cholangiocellular carcinoma 42) and 148 cases of metastatic liver cancer were treated in our hospital. Some 222 of H.C.C (hepatocellular carcinoma), 20 of C.C. (cholangiocellular carcinoma) and 42 of metastatic liver cancer were resected; 24 of H.C.C, 2 of C.C and 22 of metastatic cancer received adjuvant hepatic arterial chemotherapy, in which anti-cancer drugs were administered with oily contrast medium Lipiodol in hepatic artery. The relationship between operative findings and postoperative prognosis was studied in 168 resected H.C.C cases and risk factors for recurrence were determined. Risk factors are TW(+), which means that the cancer remains macroscopically within 1 cm of surgical margin; IM(+), which means intrahepatic metastasis exists; more than Vp2, which means tumor embolus exists in the second or more proximal branch of the portal vein; and Fc(-), which means lack of capsule formation. In 132 cases with the risk factors, the survival rate of 19 cases with adjuvant arterial chemotherapy was significantly higher than that of 113 cases without it. In the cases of liver metastasis of colon cancer, resection of metastases and adjuvant hepatic arterial chemotherapy improved the prognosis.

H-2 polymorphisms in outbred strains of mice.

We studied H-2 polymorphisms of five outbred strains, CF1, ddY, ddN, ICR and KUNM, using 15 alloantisera to H-2 class I private antigens. All individuals had at least one private antigen at H-2K and/or H-2D. The number of H-2 class I private antigens in each strain were three in ddY, four in ICR, five in CF1, and seven in ddN and KUNM. The number of H-2 phenotypes were five in ddY, six in CF1 and ICR, twelve in ddN and seventeen in KUNM. Each strain had strain specific antigen(s) which discriminated each strain from the other strains. The CF1 strain had the H-2.23 and H-2.32 antigens controlled by the H-2k haplotype at a frequency of about 70%. The ddN strain had H-2.4 (H-2Dd) at a frequency of 37.8%. The H-2.9 antigen (57%) governed by H-2Df was distributed only in the KUNM strain. Both the ddY and ICR strains possessed H-2.17 and H-2.30 antigens (85-97%) which were controlled by the H-2q haplotype. The H-2 locus will be useful to characterize each strain and distinguish between strains, and for monitoring of outbred strains by H-2 gene frequency.
