Cineangiographic evaluation of diastolic properties of the left ventricle in man.

Diastolic properties of the left ventricle were evaluated cineangiographically in 110 patients with various heart diseases (19 normal cases, 13 mitral stenosis, 19 left ventricular volume overload, 18 coronary artery diseases without myocardial infarction, 27 myocardial infarction, 6 hypertrophic cardiomyopathy, 5 congestive cardiomyopathy and 3 chronic constrictive pericarditis). Early diastolic ventricular filling was assessed by diastolic maximal circumferential relaxation velocity (maxVcf), diastolic maximal dV/dt, volume change during early 1/3 of ventricular filling (V1/3), rapid filling time and isovolumic relaxation time; myocardial stiffness constant K was calculated at end-diastole. Increase in myocardial stiffness constant was not necessarily associated with abnormal indices measured during the early diastolic period but well correlate with isovolumic relaxation time. During the isovolumic relaxation period, the affected left ventricular segments where myocardial infarction or myocardial hypertrophy existed, showed reduced and delayed relaxation resulting in a total increase in the isovolumic relaxation period. Calcium antagonist was effective in improving the delayed relaxation in patients with left ventricular hypertrophy, suggesting a role of calcium on delayed myocardial relaxation in the hypertrophied heart.

Cyclic AMP dependent protein kinase in human placenta.

There have been few reports on cyclic AMP dependent protein kinase in the human placenta. As our first study on this enzyme we here report the confirmation of its existence in human placental chorion and its chemical characteristics and the measured activities of the enzyme in each gestational trimester. 1. Chemical characteristics: (1) pH Profile: Maximum specific activity was found to be between pH7.5 and 8.5. (2) Effects of Mg2+: Magnesium ions activated protein kinase activity. Maximum specific activity was found to be 5mM. (3) Km for ATP: Km was found to be 4.94 microM. 2. Activity of cyclic AMP dependent protein kinase of human placental chorion in each gestational trimester: The specific activities (p moles ATP/mg protein/minute) of each gestational trimester were 44.67 +/- 5.29 for the first, 45.24 +/- 9.13 for the second and 34.19 +/- 4.95 for the third trimester. There was a significantly lower value in the third trimester (t less than 0.001), compared to the first and second trimester.

[Compared studies of antimicrobial agents against E. coli, Klebsiella, Citrobacter and Proteus isolated from urinary tract infections].

In vitro activities of antibacterial agents against E. coli, Klebsiella, Citrobacter and Proteus which were isolated from patients urinary tract infections at 8 hospitals in Japan, were investigated by agar dilution method from July to October in 1979. The summarized results are as follows. 1. Among oral antibacterial agents, MPC and PPA have showed potent antibacterial activities against E. coli and Klebsiella. Among parenteral antibiotics, CTM was the most active against E. coli and Klebsiella. However, ABPC-resistant E. coli and Klebsiella have appeared to occupy about 40% and 96% of bacteria isolated from urinary tract infections, respectively. 2. In vitro activities of antibacterial agents against Proteus and Citrobacter showed not so potent. 3. Causative organisms in female patients with simple urinary tract infections were mainly E. coli and Klebsiella. 4. Among oral antibacterial agents, PPA have shown similar antimicrobial activities against E. coli isolated from simple and complicated urinary tract infections. ABPC and MPC have been influenced in some degree by these factors. However, parenteral antibiotics are not influenced by these factors. On the other hand, in vitro activities of antibacterial agents against Klebsiella isolated from simple and complicated urinary tract infections were similar.

[Collagen in the human placenta].

The aim of this study is to investigate the roles and characteristics of the collagen in the human placenta during pregnancy, measuring it's hydroxyproline contents, solubility of collagen and prolyl hydroxylase activity. The results obtained were as follow: 1) Hydroxyproline contents in the placenta of 1st, 2nd and 3rd trimester were 0.41, 1.38 and 2.17mg/g wet weight respectively. Significant difference was found between the levels in the 1st and the 3rd trimester (p less than 0.01). 2) During the course of pregnancy no significant proportional change in acid-soluble fraction of the collagen, and proportional increase in pepsin-soluble fraction and decrease in insoluble fraction of the collagen were found. 3) Prolyl hydroxylase activity in the placenta of 1st, 2nd and 3rd trimester were 1058.5, 326.2 and 248.4 C.P.M./mg protein respectively. Prolyl hydroxylase activity was found to be higher in the 1st trimester than that in later trimesters (p less than 0.05). It's suggested that the collagen would take part in developmental construction of the placenta from the results obtained, which showed significant changes during the course of pregnancy in it's content and nature and available enzyme for it's synthetic process.

A new fluorometric ultramicro determination of serum cystine aminopeptidase using an aminocoumarine derivative.

A rapid and sensitive fluorometric rate assay of serum cystine aminopeptidase is described using a novel fluorogenic substrate, 7-S-benzyl-L-cysteinyl-4-methylcoumarinylamide. The proposed method enabled us to determine cystine aminopeptidase activity in one minute using 10 microliter of serum. The assay was shown to be free from error due to the adsorption of the substrate to serum albumin. The values obtained by this method gave good correlation with those obtained by the conventional method of Watson [6].

[Studies on prolyl hydroxylase activity, hydroxyproline content and solubility of collagen in the human uterus during pregnancy, delivery and postpartum involution (author's transl)].

Prolyl hydroxylase activity, hydroxyproline content and solubility of collagen in the human pregnant and post-partum uterus were studied. The results were as follows: 1. Prolyl hydroxylase activity in the uterine cervix was slightly elevated during pregnancy, and the highest level was observed on the 4th day post-partum. On the other hand, in the uterine body the highest activity was observed immediately after delivery. 2. Hydroxyproline content in the pregnant uterine cervix was slightly less than that of nonpregnant control and the lowest level was observed immediately after delivery, but on the 4th day post-partum it increased slightly. In the uterine body, there were no remarkable changes in content during pregnancy, but immediately after delivery it decreased remarkably. 3. Pepsin-soluble collagen in the uterine cervix increased significantly from an early pregnancy to immediately after delivery compared with nonpregnant control, but on the 4th day post-partum it decreased significantly compared with that immediately after delivery. In the uterine body, no significant changes in solubility were observed throughout pregnancy. 4. Comparing these data obtained on the cervix and the body at different stages of nonpregnancy, pregnancy and post-partum, enzyme activities in the body were always higher. Hydroxyproline content in the body of nonpregnant uterus showed a lower value than that in the cervix, but no remarkable differences were found between the cervix and the body throughout pregnancy and immediately after delivery. The significant decrease in pepsin-soluble collagen in the body was observed immediately after delivery.

Experimental study on monocrotaline induced pulmonary hypertensive rats. (1) Effect of long-term injection of immunosuppressant.

To investigate the involvement of the immune mechanism in the development of pulmonary hypertension in rats given monocrotaline, pulmonary hypertension was induced in rats and the effect of daily injection of prednisolone was studied. Fifty six Sprague-Dawley rats (4 week-old males, 50-60 g) were divided into four groups as follows: (1) injection of monocrotaline alone (A: 17 rats), (2) injection of monocrotaline + long-term injection of prednisolone (B: 22 rats), (3) long-term injection of prednisolone (C: 7 rats) and, (4) long-term injection of physiological saline solution (D: 10 rats). The survival time was prolonged, and slightly better results in the pressure of the right ventricle, hypertrophy of the right ventricle and lesions of lung tissue were obtained in B than in A. However, the rise in the RV pressure the hypertrophy of the right ventricle in B were more pronounced than in C and D, and findings in the lung tissue closely resembled those of A. Judging from the above, it is certain that long-term injections of prednisolone improve the lung tissue lesions and prolong the life of monocrotaline induced pulmonary hypertensive rats. However, it is unlikely that the immune reaction plays a leading role in the development of pulmonary hypertension in the rats.
