Difference in gene expression between azo-dye-induced primary hepatomas and corresponding transplantable ascites hepatoma lines in rats.

Using RNA blot analysis, we examined the relatedness of gene expression of several rat ascites hepatoma lines (AH) to azo-dye-induced primary hepatomas (PH). Four cDNA clones previously defined as containing sequences abundantly expressed in various tumor lines showed virtually the same pattern on both RNA blots of an AH line, AH60C and of PH, thereby indicating that the tumor-abundant expression of the sequences is also well retained in PH. Expression of ras and myc oncogenes in AH was the same as that in PH. However, liver-specific mRNAs such as albumin and apoprotein A-1 mRNA were expressed in normal liver and PH, but to a much lesser extent in AH60C. alpha-Fetoprotein was expressed in PH, but little in the normal liver and, if any, very little in AH60C. alpha 2u-Globulin was not expressed in either AH60C or PH. Semiquantitative RNA dot blot analyses revealed that the different expressions of certain genes between AH60C and PH observed above also hold true, even in case of other AH lines and a solid tumor line, Morris hepatoma 5123D. Southern blot analyses of the genomic DNAs showed that the different expressions might have occurred, with no major alteration in the gene structure.

Direct regulation of bone resorption of chick embryonic femur in organ culture by the change of calcium or phosphate ion concentration in the medium.

Bone resorbing activity was determined in organ culture by following the change of biological half life of 45Ca (T1/2) incorporated into bone salts of chick embryonic femur. The elevation of calcium or phosphate ion in the medium brought about an increase of T1/2, indicating that the bone resorption was inhibited. Conversely, the decrease of calcium or phosphate content in the medium resulted in the decrease of T1/2. A change of the magnesium ion concentration in the medium had no discernible effect on T1/2. These results suggest that a direct regulating system exists between bone resorption of chick embryonic femur and calcium or phosphate content in the medium and this phenomenon is specific for calcium or phosphate ion.

[Reductive effect on ascites, of neocarzinostatin in patients with ovarian cancer].

Neocarzinostatin (NCS) was administered intravenously by drop infusion to 16 patients with ovarian cancer, and intrathoracically to one patient. In addition to evaluation of the efficacy of the chemotherapy, we mainly investigated its effect on ascites and pleural effusion, and carried out continuous measurements of the abdominal circumference in 3 patients. NCS relieved thoracic effusion in 1 patient. We observed that abdominal circumference decreased in 10 patients after administration of NCS. The results obtained from continuous measurement of abdominal circumference revealed that NCS significantly reduced ascitic retention in ovarian cancer.

[A comparative study of the effects of drug therapy and bladder training therapy].

Treatment of enuresis was studied in 168 patients. The patients were divided into two groups, the drug therapy group which consisted of 88 patients who were treated with drugs only, and the bladder training therapy group which consisted of 80 patients who were treated mainly with bladder training supplemented with drug therapy. In the drug therapy group imipramine was the first choice and was used at bed time. The dose of imipramine was initially 1 mg/kg and was gradually decreased if it worked well, and changed to other drugs if it did not work well. In the bladder training therapy group, bladder training was performed in all patients for 3 months, and 22 patients who cured further bladder training was continued, whereas the rest of the patients (58 patients) drug therapy was started in addition to bladder training. After 3 months, drug therapy was effective for 54 patients (61%) including 10 patients (11%) who were cured, and bladder training therapy plus drug therapy was effective for 55 patients (69%) including 22 patients (28%) who were cured. The number of cured patients in the bladder training therapy group was significantly larger than that of the drug therapy group (p less than 0.01). After 6 months, drug therapy was effective for 56 patients (64%) including 16 patients (18%) who were cured, and bladder training therapy plus drug therapy was effective for 66 patients (83%) including 30 patients (38%) who were cured. The number of effective and cured patients in the bladder training therapy group was significantly larger than that in the drug therapy group (p less than 0.01).(ABSTRACT TRUNCATED AT 250 WORDS)

Evidence of a regulatory role of the level of poly (ADP-ribose) in chromosomal proteins in metallothionein gene expression by glucocorticoids but not by heavy metals.

The induction capacity of dexamethasone, a synthetic glucocorticoid, for the synthesis of metallothionein was about the same as that of 3-aminobenzamide, which is an inhibitor of ADP-ribosylation of chromosomal proteins, in cultured mouse mammary tumor cells. Both inductions of metallothionein were temporally correlated with a decrease in the amount of endogenous poly (ADP-ribose) on nonhistone high-mobility-group 14 and 17 proteins. In contrast, the extent of cadmium-induced metallothionein synthesis was 2-3-times that of dexamethasone or 3-aminobenzamide. However, cadmium had essentially no effect on de-ADP-ribosylation of these proteins.

A new leader exon identified in the rat insulin-like growth factor II gene.

In the rat insulin-like growth factor II gene, we identified an alternative 5'-untranslated sequence, in addition to the two sequences documented recently (1986) J. Mol. Biol. 192. 737-752, (1986) J. Biol. Chem. 261. 17138-17149. This sequence was encoded in an extreme 5' separate exon (E1 exon), the promoter region of which could be distinguished from others by features including lack of the TATA-box and GC-repeat, and heterogeneity in the transcriptional initiation.

Length polymorphism in the 3' noncoding region of rat hepatic alpha 2u-globulin mRNAs.

We detected two size classes in rat liver alpha 2u-globulin mRNAs when analyzing cDNA clones, and named the mRNAs corresponding to the cDNA clones having longer and shorter sizes as L-type and S-type, respectively. When sequencing these cDNA clones, a 25 base insertion was present in the L-type at the 6th exon-intron junction. The extra sequence coincided with the 5' part of the reported 6th intronic sequence that is directly contiguous to the 3' part of the 6th exon. The relative abundance of L- and S-type mRNAs in the steady-state liver was determined by a sensitive S1 nuclease analysis using the probe prepared from the L-type cDNA. The same method was also applied for the determination of the L/S ratios during the course of postnatal development and estrogen treatment, all resulting in similar values, of about 0.03. These findings indicated that L-type and S-type mRNAs are generated by an alternative splicing mechanism in the 3' noncoding region of alpha 2u-globulin transcripts. Sequence analysis also elucidated the presence of at least two active genes showing exactly the same pattern of alternative splicing.

A systematic survey of repetitive sequences abundantly expressed in rat tumors.

A cDNA library made from poly(A)+ RNA of a rat ascites hepatoma cell line, AH60C, was screened with respect to transcribed repetitive sequences by probing with AH60C genomic DNA. Thirty clones, comprising 1.2% of the library, were found to contain repetitive elements. The sequences of 12 clones among them were abundantly expressed in poly(A)+ RNA of AH60C, yet sparse in that of rat normal liver. This differential expression was also noted in case of other tumors and normal tissues tested. These 12 clones could be divided into two classes, depending on whether their sequences were expressed in cytoplasmic poly(A)+ RNA (class I) or not (class II). Rat Alu-like type II (B2) repeats and an unknown repetitive sequence were found in the clones of class I. Adjacent to each of them, we detected two non-repeated sequences, the expressions of which were also activated. The presence of another unknown repetitive sequence was observed in the clones of class II. It is noted that more than one-third of the transcribed repetitive sequences were involved in the predominant expression in tumor cells, whereas the majority of overall transcripts have been found to be common to both transformed cells and their normal counterparts.

Induction of epidermal mucous metaplasia by culture of recombinants of undifferentiated epidermis and retinol-treated dermis in a chemically defined medium.

Epidermal mucous metaplasia of cultured skin is known to be induced by excess retinol. Studies were made on whether retinol affects primarily the epidermis or the dermis during retinol-induced epidermal mucous metaplasia of 13-day-old chick embryonic skin in culture. When recombinants of 13-day-old normal epidermis and retinol-treated dermis were cultured for 7 days in chemically defined medium in the absence of retinol, hormones, and serum, they showed altered epidermal differentiation toward secretory epithelium (mucous metaplasia). Thus retinol acted primarily on dermal cells.

Inhibition by epidermal growth factor of glucocorticoid-induced epidermal alpha-type keratinization of chick embryonic skin cultured in the presence of delipidized fetal calf serum.

When a 13-day-old chick embryonic tarsometatarsal skin was cultured for 4 days in medium containing hydrocortisone (20 nM) and 5% delipidized fetal calf serum (FCS), epidermal growth factor (EGF, 100 ng/ml) decreased epidermal DNA content 42% and inhibited epidermal DNA synthesis 87%. Tonofilament bundles within the basal and intermediate cells of the EGF-treated epidermis were not as conspicuous as those in the glucocorticoid-induced keratinized epidermis, and the upper region of the EGF-treated epidermis did not form either the filament bundles or the electron-dense amorphous masses seen in the cytoplasm of the glucocorticoid-induced keratinized layer. EGF stimulated degradation of glucocorticoid-induced alpha-keratin. Furthermore, EGF caused a twofold increase in glucocorticoid-induced epidermal transglutaminase activity and in the amount of epidermal glucocorticoid receptor. In the absence of FCS, however, EGF did not inhibit steroid-induced alpha-type keratinization and did not affect either steroid-induced epidermal transglutaminase activity or amount of epidermal glucocorticoid receptor. Hence, the effect of EGF on glucocorticoid-induced epidermal transglutaminase activity was observed only in the presence of delipidized FCS and might be supported by an increase in the amount of glucocorticoid receptor.

An NAD:cysteine ADP-ribosyltransferase is present in human erythrocytes.

A novel enzymatic activity, i.e., the catalysis of the formation of ADP-ribosylcysteine, was found in the cytosol of human erythrocytes. The NAD:cysteine ADP-ribosyltransferase was partially purified by sequential chromatographic steps on phenyl-Sepharose, phosphocellulose, and Sepharose CL-6B. The enzyme has an apparent molecular weight of 27,000 +/- 3,000, as determined by gel permeation. The formation of ADP-ribosylcysteine was associated with the stoichiometric release of nicotinamide from NAD. The enzyme was found to be highly specific toward cysteine and cysteine methyl ester as ADP-ribose acceptors. S-Benzoyl-L-cysteine, cystine, histidine, glutamic acid, arginine, arginine methyl ester, and agmatine were ineffective as acceptors for this enzyme.

Multicentric hepatocellular carcinoma following phosphate diethylstilbestrol therapy for prostatic cancer.

This paper reports an autopsy case of a 78-year-old male with multiple nodules in the liver developed after long-termed administration of phosphate diethylstilbestrol (PDES) for prostatic cancer. Large part of these nodules were suspected to be well differentiated hepatocellular carcinoma with high level of serum alpha-fetoprotein (AFP) up to 3,400 ng/ml, but a part of them was evaluated to be a borderline between hepatocellular carcinoma and adenoma with mild cellular atypism. The liver other than the nodules showed liver fibrosis associated with liver cell dysplasia and peliosis hepatis-like change. This is a unique autopsy case of hepatocellular carcinoma closely related to diethylstilbestrol (DES) therapy for prostatic cancer.

Ultrastructural studies of the skin and cultured fibroblasts in I-cell disease.

The skin and cultured fibroblasts from a patient with I-cell disease were examined by electron microscopy. Multiple vacuolations were seen in fibroblast and/or histiocyte-like cells, secretory cells of eccrine glands, and Schwann cells of the skin. Vacuolar inclusions were single membrane-limited, and contained a few reticulo-floccular and vesicular materials, endothelial cells of the dermal capillaries contained other types of inclusions, which were also membrane-limited, more electron dense, and multivesicular. The epidermis and pilosebaceous appendages seemed to be normal. Cultured skin fibroblasts contained prominent inclusions which varied in size and morphology. Acid phosphatase activity was seen in some of those inclusions, indicating their derivation from lysosomes. These findings suggest that the I-cell disease is a type of lysosomal storage disease and that electron microscopic examination of normal-appearing skin in this disease may contribute to the diagnosis.
