[Methods for cytologic and histologic determination of phospholipids in lung tissue].

The demonstration of lipids by light microscopy is one of the most difficult parts of histochemistry. Phospholipids of the surfactant system are a very important part of lung metabolism, but for morphologists not easy to detect. Basing on own experiences, a view is given about different methods for demonstration of phospholipids, in particular of the surfactant system by light microscopy and fluorescent microscopy.

[Immunofluorescence method for the detection of fungi in bronchial secretions].

A method for detecting fungal elements in bronchial secretions with the aid of indirect immunofluorescence is described. Advantages of this method are the white fluorescence of fungal elements on dark background, whereas conventional staining methods (Grocott, PAS) also stain mucus. Further on it is possible to demonstrate with this immunofluorescence test the presence of antibodies against fungi in patients sera. Advantages and disadvantages of this method are discussed.

[Immunohistochemical findings in intrathoracic tumors. II. Demonstration of alpha 1-antitrypsin in tumor tissue].

Immunohistological investigations for demonstration of alpha 1-antitrypsin were undertaken in different intrathoracic tumors. 60% of them showed deposits of alpha 1-antitrypsin in tumor tissue. Among different histological types, bronchial carcinomas showed the highest percentage of positive findings (= 75%). Negative findings were observed in carcinoid tumors and different neurogenic tumors.

[Histiocytosis X--a comparative 2-country study of East Germany and Yugoslavia].

In a retrospective study 25 patients with histologically confirmed histiocytosis X were investigated in the GDR and in Yugoslavia. No differences between the clinical course was observed in the two countries. The disease was found somewhat more frequent in men, and relative to age groups the age group 18-39 prevailed. Only one fourth of the patients were detected by radiophotography. Lung function was impaired in 2/3 of the patients. In the lung radiogramm reticular and micronodular changes predominated at the beginning of the disease while cystic lesions appeared in late stages of the disease. The diagnosis was established on the basis of biopsy (most often transthoracic lung biopsy) and pathohistologic examination. The course of the disease was unfavourable because of the 18 observed patients 3 died and in 6 progression of the disease was noted on average within several years.

[Hamman-Rich syndrome].

An analysis of 29 observations of the Hamman-Rich syndrome was carried out. Histological, histochemical, and electron microscopic studies showed the earliest changes to consist in edema of the interstitial connective tissue of the lungs followed by the development of crude lesions of alveolar elasticity and increase in the number of argirophilic and collagen fibers leading to disorders in the aerohematic barrier. The chronic course of the disease leads to pulmonary microcystosis. Hormone therapy slows down the development of the pathologic process.

Radical radiotherapy in the management of carcinoma of the prostate.

Reasons for the increased role of radical radiotherapy in the management of carcinoma of the prostate are presented. The results of a retrospective survey of 116 patients who received radical external beam radiotherapy to the primary tumour are reviewed. Local tumour control was obtained in 75% of patients with an overall morbidity of 14% mild and 9% severe side effects. The problems of accurate tumour staging, extended field radiotherapy and patient selection are discussed. In selected cases, treatment morbidity may be reduced by using interstitial implant techniques.

[Lung changes induced by copper sulfate. An experimental contribution to the so-called "vineyard sprayer's lung"].

Vineyard sprayer's lung is an occupational lung disease, caused by inhalation of Bordeaux mixture (1-2% aqueous solution of copper sulfate, neutralized with hydrated lime). Experimental studies in mice after a longstanding exposition to copper sulfate aerosol showed focal accumulations of macrophages in alveoli with different cytoplasmatic inclusions, focal interstitial granuloma formation, increased formation of collagenous and elastic fibers, and increased production of surfactant phospholipids, phagocytosed by macrophages. As a conclusion of these results, copper seems to be the responsible agent for the development of lung changes, seen in vineyard sprayer's lung.

[Allogenic lung transplantations following pretreatment of recipients with donor-specific splenic antigens].

Lung transplantations were carried out in 20 not immunologically selected mongrel dogs which were pretreated with donor specific spleno-antigen mixture. The average survival time was double the size (12.3 days) as in a control group of untreated dogs after allogenic lung transplantation. A number of the receivers got Cyclophosphamid in addition to the spleno-antigen mixture. In this group the average survival time amounted to 17 days. A chronic course of the rejection was not to be achieved with this method. The autopsy showed thoracic infectious complications in nearly all receivers. A great number of central pulmonary vein thromboses in the transplant was discovered in this series in contrast to other lung transplantation series. Circumstances refer to a connection with hyperacute rejection courses. The receivers of this series survived the destruction of the transplant longer on an average as such in other experimental series.

[Experimental animal model of airborne influenza infection].

Experiments were performed for analysing the responses of the host to influenza respiratory tract infections. NMRI and Balb/c mice were infected with various doses of mouse adapted influenza virus (A/PR8/34) by aerosol. Clinical symptoms, rate of survival, total cell yield and total protein content in the lung lavage fluid as well as the course of virus titres showed typical changes depending on time and infection dose. Histologically, after moderate infection doses mice developed mononuclear infiltrations as bronchitis and peribronchitis whereas oedema and severe pneumonitis occurred in highly infected animals. The mouse model seems to be a useful basis for analysing the effects of immunization procedure against influenza.

[Light microscopic and histochemical findings on the mouse lung after neutron and 60 cobalt gamma-irradiation].

The morphological changes on lungs of thorax irradiated mice, receiving a single dose of fast neutron and 60 cobalt radiation were investigated. Lungs were studied by histological and histochemical methods 2 weeks up to 10 months. Harvest changes were found after a time of 4-8 weeks. With histochemical methods more detailed damages are seen in contrast to routine histology. Two weeks post irradiation disturbances of phospholipid synthesis in alveolar epithelium cells of type II (AEC II) are found. Impairment of capillary endothelial cells are indicated by decreased activity of adenosine triphosphatase. Increased activity of beta-glucuronidase is seen in damaged bronchial epithelium.

[Stromal reaction on bronchial carcinoma].

The stromal reaction was analysed in histological preparations of 100 patients with bronchial carcinoma (bioptic material). The tumor-stromal reaction in all types of tumors is much greater than the stromal reaction of the surrounding area. A positive correlation exists between the stromal reaction and the histological type of tumor and the differentiation of the tumors. The differentiated tumors have a greater stromal reaction than less differentiated tumors. The stromal reaction should be used in the diagnosis of the types of bronchial carcinoma since it provides information about the tumor type from bioptic material.
