Replication of herpes simplex virus in human T lymphocytes.
The replication of herpes simplex virus (HSV) in cultures of human T lymphocytes was investigated. Virus replication occurred only in lymphocyte cultures prestimulated with mitogen. At least one of the factors responsible for this phenomenon is adsorption of the virus to the cells, which was 5 times less efficient in nonstimulated cells than in stimulated cultures. Growth of virus in infected cultures was restricted to lymphocytes of T origin. This was shown by successful infection of highly purified T-cell cultures and by virus replication in four continuous T-cell lines. No virus production was obtained in cultures of lymphocytes of other than T-cell origin. Virus titers in T-lymphocyte cultures reached 10(7) PFU/ml, which was a four log10 step increase over the input dose. Lymphocytes infected with a syn strain of HSV, HSV-ANG, induced polykaryocyte formation in the cultures and therefore generated a CPE visible by microscopic examination. Infection with syn+ strains of HSV (i.e., strains which do not cause cell fusion) did not lead to such phenomena. Not more than 1% of infected T cells was replicating HSV, as revealed by infectious center assays. In contrast, about 5% of these cells showed positive immunofluorescence with anti-HSV antibodies, indicating the presence of cells which express viral protein but do not actively produce infectious virus.
[Curative radiotherapy in stage C prostatic carcinoma. Experiences with radiotherapy-endocrine treatment].
In a prospective study 209 patients with prostatic cancer stage C who were seen between 1968-1974 received endocrine treatment and radiotherapy (6000 r) of the prostate without inclusion of regional lymph nodes. The patients were seen every 6 months up to 8 years. 247 transrectal biopsies and 7 transurethral resections of the prostate were performed. The analysis of the biopsies revealed that the combined endocrine and radiotherapy is followed by tumor neutralisation of about 70% in highly differentiated prostatic cancers and less effective in lower differentiated tumors. 80% of biopsies from tumors responding to therapy still show tumor cells. The 5-year-survival corresponded with tumor regression and tumor size. However a prognosis on the basis of these findings is not possible for the individual case.

[Diagnostic significance of brain tumor calcifications in the computerized tomogram].
CT investigations of 783 cases of brain tumors, which were covered by histology, were checked with regard to frequency and configurations of calcifications. As expected the calcifications were detected more frequently by CT than by conventional radiology because of higher resolution. Most frequently calcifications were detected in craniopharyngeomas (72%) and in oligodendrogliomas (51%). The calcifications shown by CT can be changed by variation of technical parameters. Certain patterns of calcifications can be differentiated, if constant and best technical conditions are used. One single pattern cannot be specific. A homogeneous calcification of the tumor as a whole has been observed in meningeomas only. For other brain tumors calcifications are useful for the diagnosis, if localisation, density, and reaction of the surrounding tissue after injection of contrast medium and clinical data are known.
[Surgical results of epididymovasostomy and vaso-vasostomy].
In the Urology Clinic of the University of Hamburg 326 epididymo-vasostomies have been done. A follow-up of 241 men was possible. Bilateral anastomosis resulted in a 47% permeability; after unilateral recanalisation a patency-rate of 32% was obtained. Vaso-vasotomy after vasoresection was performed in 56 men. Following a splinted one-layer anastomosis, which we abandoned in the meantime, patency was obtained in 64% of patients. Current techniques in one or two-layers without a splint resulted in a 88% permeability. The double layer anastomosis does not offer an advantage over the one-layer method.
[A prolactin-producing microadenoma as the cause of galactorrhea and delayed puberty in an adolescent boy].
The case history of a 16 years old boy with delayed puberty and the sudden onset of galactorrhea is presented. Prolactin analyses and X-ray studies of the sella turcica lead to the diagnosis of a pituitary tumor. A prolactin producing microadenoma was selectively excised via the transsphenoidal route. Serum prolactin resumed normal concentrations within hours after the operation. During the next months the boy showed the signs of normal puberty with increasing concentrations of serum testosterone. Because serum prolactin concentrations remained within the normal range, therapy with bromocriptine was not necessary. A spermiogram however, performed seven months after the operation, showed signs of a viscosipathy and terato-asthenozoospermia.
[Suitability of current selective media for the detection of Staphylococcus aureus in food].
Three solid media (Baird-Parker-Agar (BP), BP with pig plasma instead of egg-yolk (BPP) and modified BP according to Devriese (BPA) as well as three liquid media (Brain-Heart-Infusion with 7.5% NaCl (BHIS), Liquid Baird-Parker-Medium (LBP) and Giolitti-Cantoni-Medium) were tested with regard to their suitability for the detection of Staph. aureus in foods. The following criteria were assessed: productivity for vital and thermally stressed Staph. aureus; selectivity against undesired microorganisms; practicality in routine examination. The microorganisms used in these studies were: Staph. aureus (vital and thermally stressed), "non-aureus" staphylococci, Streptococcus spp., Pseudomonas spp., Proteus spp., Bacillus spp. Using vital strains of Staph. aureus no significant differences in the productivity of the three solid media could be observed. On the other hand BPA showed a remarkable lower productivity than BP and BPP when thermally stressed cultures of Staph. aureus were examined. LBP was the most productive liquid enrichment medium. The selectivity of BPA was distinctly superior to BP and BPP, whereas no differences could be observed between the latter ones. The selectivity of LBP and GC against coagulase negative staphylococci and gramnegative microorganisms tested was relatively poor. Each culture under examination grew in BHIS. The combination of LBP and BPA yielded better results for enumeration of thermally stressed Staph. aureus from dried milk products than the other possible combinations of media. In view of their practicality dehydrated culture media are commonly preferred in routine examination. The additional time required for the preparation of LBP or BPA (which are not commercially available as dehydrated media) is negligible and is compensated by better productivity and selectivity of these media. At present BPP is not recommendable due to the problems in providing pig plasma. On the other hand this medium has the great advantage that further testing of suspicious colonies is not necessary as it is in the case of BP and BPA.
[Accidental finding: the small, differentiated thyroid carcinoma. Report of 46 thyroid carcinomas of up to 15 mm diameter].
Review of 190 cases of thyroid carcinoma between 1974 and 1981 showed an increasing rate of small papillary carcinoma. The study included 46 tumors which did not exceed 15 mm in diameter. Most of these lesions (3/4) were discovered incidentally at thyroid operations of various types. In spite of the early stage of carcinoma 6 tumors were already associated with cervical nodal metastasis, cases showed multiple lesions within the thyroid gland, homolateral and bilateral. With respect to these cases total thyroidectomy and subsequent radium therapy are generally suggested.
Results of LSA2-L2 therapy in 26 children with non-Hodgkin's lymphoma.
Twenty-six children with non-Hodgkin's lymphoma (NHL), 17 boys and nine girls, were treated with the LSA2-L2 protocol. Seven children had stage I or II, 16 Stage III and three Stage IV according to Murphy's staging system. Eight children had their primaries in peripheral lymph nodes, eight in the abdomen, six in the mediastinum and four in other sites. All tumors were classified histologically according to four different classifications. Overall disease-free actuarial survival is 53.6%. Complete responders show a disease-free survival of 77.8%. Fourteen children survived for 9-56 months. Included are all seven children with Stage I or II who survive irrespective of histologic type of the tumor. Of the remaining 12 children in Stage III or IV three children died in remission and nine of progressive disease. Eight of these nine patients did not attain complete remission. Whereas four of five children with the convoluted type of NHL survive, four of five patients with the Burkitt's type (small noncleaved follicle center cell lymphomas) died of progressive disease. According to Rappaport's classification, four of six children with diffuse undifferentiated lymphoma (DUL) are dead due to tumour progression. Considering the classification of lymphoblastic lymphomas introduced by Nathwani et al., 23 five of seven children suffering from lymphoblastic lymphomas but only two of eight children with nonlymphoblastic lymphomas belong to the survivors. Therefore histologic findings do hold prognostic significance in our series of children with NHL.
[IgD-paraproteinemia in lymphoplasmacytoid immunocytoma].
Laminectomy was performed in a 54-year-old patient who had a hemicauda syndrome. An epidural tumour, extending into the spongiosa of the third lumbar vertebra was found. Histologically it was a lymphoplasmacytoid lymphoma. Subsequent haematological investigation revealed a multilocular involvement (stage IV) and paraproteinaemia of the IgD-kappa-type. There was no Bence-Jones protein in the urine. After irradiation of the affected lumbar vertebra the patient was treated with a combination of chlorambucil and prednisone, later on with COP and VMCP regimens. A rapid downhill course set in 16 months after diagnosis and the patient died from intracerebral expansion of the immunocytoma. The clinical diagnosis was confirmed at autopsy. The generalized lymphoplasmacytoid immunocytoma contained evidence of terminal transformation into the polymorph subtype.
