[Hysterosalpingographic finding tubal sterilization: their importance for the pathologic hysterosalpingogram(author's transl)].

110 tubal sterilizations by Pomeroy's technique were performed in the years 1973--75. 78 cases were examined by hysterosalpingography. In 14 cases the ligature was located close to the intersitial portion of the tube (proximal type), in 52 cases in the isthmic ampullary junction or close to the ampulla (distal type) and in 12 cases on one side proximal type and on the other distal type. In 2 cases of the latter group failure of the ligature was confirmed by contrast medium in the pelvic cavity. The occluded portion of the tube was on the average longer after coeliotomy in comparison with laparotomy. Occluded tubes with smooth distribution of contrast-medium showed good peristaltic activity. Hydrosalpinx never developped, although often reported in the literature. A spill over of contrast-medium into the surrounding tissue was observed only in two cases. This low incidence indicates that the phenomenon is not caused by application of to high pressure.

Uridine 5'-diphosphate galactose: glycoprotein galactosyl transferase activity in exfoliated bladder epithelial cells in rats fed N-(4-(5-nitro-2-furyl)-2-thiazolyl) formamide.

Urine samples of normal male Fischer rats or rats fed 0.2% N-[4-(5-nitro-2-furyl)-2-thiazolyl]formamide for 6,8 or 30 weeks were collected and centrifuged 50 weeks after beginning treatment. After being sonicated and assayed (with purified desialylated ovine submaxillary mucin as acceptor glycoprotein), the exfoliated bladder cells obtained from the urines of treated rats showed uridine 5'-diphosphate galactose:glycoprotein transferase activity. The specific enzymatic activity of the enzyme from cells of 30-week-treated rats was about 10 times higher than from normal rats. The enzyme from cells of hyperplastic rats (treated 6 or 8 weeks) was only slightly higher in specific activity than that of normal rats. A similar was obtained at a later stage of bladder tumor induction, when the urines from 30-week-treated rats contained blood. A correction was made for protein contributed by the blood clot. The possibility that the blood clot contributed galactosyl transferase activity was excluded. Activity of the enzyme was detected in normal rat bladder tissue and in normal human urine.

Glycoprotein biosynthesis by organ cultures of hamster trachea.

Conditions have been developed for maintaining hamster tracheas in organ culture for at least 10 days. Secreted glycoproteins labelled with [14C]glucosamine and [3H]fucose were isolated from the spent medium and digested with papain, and the digest was fractionated on DEAE-Sephadex by stepwise elution with NaCl. The fractions eluted by 0.1 and 0.2 M NaCl and some of the products eluted with 0.4 M NaCl were shown to be derived from epithelial glycoproteins. Glycosaminoglycans were eluted by 0.4 M and by 1.25 M NaCl. Glycopeptides isolated from the epithelium by homogenization, ethanol precipitation and papain digestion, and defined as "intracellular", gave a very similar profile on DEAE-Sephadex. The 0.1 M glycopeptide peak was the major fraction of epithelial origin from both secreted and "intracellular" material; it labelled extensively with both glucosamine and fucose and had a molecular fraction was purified further by chromatography on Sephadex G-75 and DEAE-Sephadex; its amino acid and carbohydrate compositions were determined.

The anomeric configuration of the D-mannosyl retinyl phosphate formed in rat liver microsomes.

Alkaline hydrolysis at 65 degrees converted D-mannosyl retinyl phosphate primarily into D-mannose 2-phosphate which was identified by its behavior on paper chromatography in two solvent systems, as well as by its resistance to acid hydrolysis under conditions that converted authentic alpha-D-mannosyl phosphate to mannose. When the D-mannosyl retinyl phosphate was hydrolyzed in alkali at 100 degrees, the main product was beta-D-mannosyl phosphate and not the alpha anomer (as shown by chromatography with authentic samples on thin layers of silica gel). No evidence for enzymatic hydrolysis of D-mannosyl retinyl phosphate by alpha-mannosidase was obtainable. It is concluded that the compound under investigation is beta-D-mannosyl retinyl phosphate.

[On the cytosic action of N-methyl-N-beta-chloraethyl-hydrazine and its benzaldehydhydrazone (author's transl)].

N-Methyl-N-beta-chloroethyl-hydrazine and its benzaldehydhydrazone are two new cytostatic methylhydrazines. Administered intraperitoneally, they are more effective in inhibiting the ascites tumor growth (Ehrlich's carcinoma in mice and Yoshida sarcoma in rats) than procarbazine in vitro as well as in vivo. The intraperitoneal administration of hydrazone shows a minor effect on the solid tumor. This may be explained by a different pharmacocinetical behaviour. Hydrazone is less toxic than procarbazine.

Stoffwechselprodukte von mikroorganismen. 154. Mitteilung. Nikkomycin, ein neuer hemmstoff der chitinsynthese bei pilzen.

From the fermentation broth of Streptomyces tendae Tü 901 a substance was isolated which inhibits the growth of several fungi. The new antibiotic affects the chitchin biosynthesis. Its structure was identified by mass spectrometry of the products obtained after chemical degradation. Nikkomycin is a nucleoside-peptide antibiotic consisting of uracil, an amino hexuronic acid and a new amino acid containing a pyridin ring.

[The place of plate thermography in the diagnosis of breast cancer (author's transl)].

A report is presented of 319 unselected patients investigated by the method of plate thermography according to Tricoire, which is a comparatively new method in the diagnosis of breast cancer. 281 cases were additionally examined by mammography and those cases in whom malignancy was suspected underwent further cytological and histological investigation. The results of plate thermography and mammography are compared. The high degree of diagnostic accuracy achieved in the present series of cases by the use of plate thermography indicates that this procedure is not only a valable adjunct in the diagnosis of breast cancer but, moreover, seems to be recommendable as a screening method.

[Improved demonstration of normal lymph nodes in lymphangiograms (author's transl)].

Lymphography with Lipiodol ultra fluid was carried out in eight domestic pigs. The purpose of the experiment was to obtain even distribution of the oil in normal lymph nodes in order to improve its diagnostic value. This was attempted by changes in pressure, temperature and by the addition of benzopyrone. By increasing injection pressure from 0.4 to 0.8 atmospheres, using a pump, it was possible to show more of the lymphatic tissue. Whether this would be possible in man is still not certain. The use of a benzopyrone preparation also improved contrast distribution in lymph nodes. Additional pelvic lymphatics were also seen. The improved distribution of oil is indicated by the altered pattern, strikingly even filling of the vacuoles and 10% incfreased filling rate.

[The radiological and scintigraphic findings in psoriatic arthropathy (author's transl)].

The radiological and scintigraphic findings in the skeletons of patients with psoriasis and psoriatic arthropathy are described. The results indicate that is it possible to detect inflammatory disease in joints by scintigraphy in patients who have not yet developed symptoms. The effect of treatment can be satisfactorily followed by serial scans. Further investigations and observations will be required to show whether the paravertebral calcification and syndesmophyte formation which has been described is really pathognomonic for psoriatic arthropathy.

Evidence that vitamin A is not required for the biosynthesis of ovalbumin in chicks.

Four-day-old pullets fed a vitamin A-deficient diet were stimulated daily with 1 mg 17beta-estradiol-3-benzoate/day for 6 to 19 days. The onset of vitamin A deficiency had no effect on oviduct growth in these chicks; even though vitamin A-deficient chicks showed a severe decline in growth rate while controls (fed the same diet supplemented with retinyl palmitate) continued to grow, estrogen stimulated resulted in similar oviduct size. Ovalbumin concentrations of estrogen-stimulated chicks were determined by immunoprecipitation of the soluble protein supernatant fraction of oviduct. The concentration of ovalbumin in oviducts of chicks fed a vitamin A-supplemented diet was similar in the concentration in oviducts of chicks fed a vitamin A-deficient diet. The incorporation of [3H]glucosamine and 14C-amino acids into immunoprecipitable ovalbumin, following the in vitro incubation of minced oviduct, indicated that ovalbumin synthesis was not affected by vitamin A deficiency. The specific activity of incorporated [3H]glucosamine, the 14C-amino acid incorporation into ovalbumin, the relative rate of ovalbumin synthesis, and the relative effiency of [3H]glucosamine incorporation into ovalbumin were each similar between the two diet groups. The relative efficiency of [3H]glucosamine incorporation into sodium dodecyl sulfate and dithiothreitol extractable membranous proteins of oviduct was not affect by vitamin A deficiency.

A plasma glycoprotein depressed in vitamin A deficiency in the rat: alpha 1-macroglobulin.

Plasma glycoprotein synthesis in normal and vitamin A-deficient rats was investigated by injecting the rats with labeled carbohydrate precursors and then fractionating their plasmas on DEAE-Sephadex. Plasma from deficient rats showed a consistent depression of 30% in the uptake of label into a peak eluting with 0.23 M NaCl. The major component of this peak was identified as the rat alpha1-macroglobulin, based on its molecular weight (800,000), its mobility on cellulose acetate electrophoresis and its ability to bind trypsin. Although the alpha1-macroglobulin synthesis appeared to be depressed by 30%, its fractional turnover rate was not affected by vitamin A deficiency (t 1/2 = 18 hours). The trypsin-binding ability of this glycoprotein was used as a comparative measure of its concentration, and the results confirmed that serum levels of this glycoprotein were lower in deficient rats. In severe deficiency, alpha1-macroglobulin levels dropped to between 10% and 20% of normal levels.

[Retroperitoneal lymphangiodysplasia with exudative enteropathy. A case of cystic dilated cisterna chyli and splenic cysts].

In a 16 years old girl we found the syndrome of exsudative enteropathy with intestinal lymphangiectasy which differs from previous reported cases by several malformations of the lymphatic system. Besides changes in the intestinal lymphatics there was a retroperitoneal lymphangiodysplasia, a reduction and hypoplasia of the lymph vessels in the legs and cysts in the spleen. By substitution of albumin followed by a diet with middle chained triglycerids (MCT), serum protein was stablised but a relapse could not be prevented. Surgical treatment was not possible because the changes were too extensive. The value of diagnostic measures for differential diagnosis of the intestinal lymphangiectasy is presented.
