Individual assignments of amide proton resonances in the proton NMR spectrum of the basic pancreatic trypsin inhibitor.

Studies of proton-proton nuclear Overhauser effects were used to obtain individual assignments of 17 amide proton resonances in the 360 MHz proton nuclear magnetic resonance spectrum of the basic pancreatic trypsin inhibitor. First, optimizing the conditions for obtaining selective nuclear Overhauser effects in the presence of spin diffusion in macromolecules is discussed. Truncated driven nuclear Overhauser experiments were used to assing the amide proton resonances of the beta-sheet in the inhibitor. It is suggested that these techniques could serve quite generally to obtain individual resonance assignments in beta-sheet secondary structures of proteins. Combination of nuclear Overhauser studies with spin decoupling further resulted in individual assignments of the gamma-methyl resonances of the two isoleucines and numerous Calpha and Cbeta protons.

Insufficient penile erection due to abnormal drainage of cavernous bodies.

Four men were referred for incomplete erection of the penis. All had abnormal venous outflow from the cavernous bodies: 1 iatrogenic fistula from the cavernous body to the glans, 1 congenital fistula from the cavernous body to the glans, 1 with several congenital large veins connecting the interior of the cavernous body to the outer veins, and 1 who suddenly lost the ability for erection was found to have a venous leak through scar tissue in the tunica albuginea. They were all operated on and the visible leaks closed. Three of them were functionally greatly improved.

The importance of epidemiology in the study of causes of bone tumours.

Research into the causes of disease in human populations makes use of epidemiological methods for recognizing avoidable risk factors which may be active prior to the occurrence of malignant tumours. With the exception of the risks or irradiation, the aetiology of bone tumours remains unclear, and it is particularly striking that in the literature to date and in a survey of research currently in progress there are no epidemiological studies which involve other than radiation-exposed populations. This is the reason why it has only been possible to obtain initial suggestions of potential areas of risk from ecologic correlations based on the evaluation of mortality and morbidity rates with respect to environmental factors. Several results obtained from systematic comparisons of mortality in the countries of the world and two correlative relationships are reported, and the difficulties in interpreting the results are discussed. Population-based registration of bone tumours and case-control studies based on registries are recommended.

8-MOP plasma levels in PUVA problem cases with psoriasis.

8-MOP plasma levels of psoriatic patients poorly responsive to PUVA treatment (PUVA problem cases) (N = 14) and of psoriatic patients with adequate response to photochemotherapy (N = 7) were measured for 8 h after oral ingestion of 0.6-0.8 mg/kg body weight, using a gas chromatographic method. We investigated whether there are any differences in the course of the plasma kinetics between the two groups. Problem patients showed significantly lower 8-MOP plasma levels than the control group. Furthermore, the 8-MOP plasma levels increased more rapidly in the control group than in these problem patients. Deviations in time of the maximum 8-MOP plasma levels from the expected 2 h peak could be observed in 50% of the problem cases compared to only 14% of the control patients. There is no correlation between the dose and the 8-MOP plasma level achieved in the two groups, i.e. higher doses do not result in high levels. In individual cases there is not always a correlation between the plasma maximum of 8-MOP at the time of UV-A irradiation and the response to treatment. Adjustment of the UV-A irradiation to coincide with the maximum plasma levels led to an improvement in therapeutic results for three problem patients.

[Synthetic inhibitors of serine proteinases. Part 19: On the inhibitory effect of amidinobenzylidene derivatives of benzo-condensed cycloalkanones on trypsin, plasmin and thrombin (author's transl)].

Amidinobenzylidene derivatives of benzo-condensed cycloalkanones proved to be potent competitive inhibitors of thrombin and trypsin. On the contrary, the antiplasmin effect of these derivatives is considerably less marked. The conversion of 3-amidinochalcone to derivatives in which the carbonyl group is incorporated into a ring, does not lead to fundamental changes in the inhibitory effect on trypsin and thrombin, whereas the antiplasmin effect decreases. Compared to 4-amidinochalcone, the 2-(4-amidinobenzylidene) derivatives of indanone-(1) and tetralone-(1) exert a stronger inhibitory effect on trypsin and thrombin. The introduction of a hetero-atom to the cycloalkanone component affected the inhibitory effect on trypsin and thrombin but insignificantly. From these results it is concluded that also in amidinobenzylidene derivatives of benzo-condensed cycloalkanone derivatives, the carbonyl function shares in enzyme-inhibitor binding.

[Synthetic inhibitors of serine proteinases. Part 21: Studies on the structure-activity relationships in bisamidines (author's transl)].

The antitrypsin, antiplasmin and antithrombin activities of bis(amidinobenzylidene)- and bis(amidinobenzyl)cycloalkanones are not markedly affected if the amidino group is substituted by an uncharged residue (H, OCH3, Cl, Br, NO2). In contrast, mono(amidinobenzylidene)cycloalkanones exhibit considerably lower inhibitory activities than the compounds of the abovementioned classes of substances. From the results obtained it is concluded that the second aromatic residue and not the second basic amidino group is decisive of the potent inhibitory action of the bisamidino derivatives.
