[Hepatic coma - experimental study with swine (part 1)].

An acute coma hepaticum was produced in 8 pigs by means of one intraperitoneal dose of thioacetamide. Distinct changes in the behaviour of the animals, changes of the biochemical parameters inclusively the enzyme pattern with a typical lesion pattern of the liver parenchyma and a displacement of the plasma amino acids were shown. All pathological changes in comparison with the normal animal characterize the liver insufficiency after 24 hours as a comatose state. This acute toxic model serves as the base for investigations within the physiology of nutrition about parenteral feeding of L-amino acid mixtures in coma hepaticum. The nearly exclusive hepatotropic lesion degree of thioacetamide is suitable as an experimental model for this purpose.

[Contrast echocardiography in congenital cardiopathies].

Besides marking the limits of intracardiac structures, contrast echocardiographic techniques make it possible to recognize a shunt or valvular insufficiency of the left heart. While it is sufficient to use a peripheral venous injection of physiological saline, 5% glucose in water, blood or cardiogreen at the atrial, ventricular or vessel level for the differentiation of a right-to-left shunt, for he diagnosis of a left-to-right shunt it is necessary to inject the left atrium, left ventricle or the aorta. Within the frame of heart catheterizations the diagnosis of mitral or aortic insufficiency can be verified by means of contrast echocardiographic techniques. These conclusions are demonstrated by own results of investigation.

[Lead detoxication effect of a combined calcium phosphate and ascorbic acid therapy in pregnant women with increased lead burden (author's transl)].

In former papers it was stated that the fetus can be exposed a considerable burden by traces of heavy metals and that a combined therapy with calcium phosphate and ascorbic acid has a good detoxifying effect. In fourty lead-burdened mothers who were treated in this way the excretion of 5-aminolevulinic acid in urine decreased by 65%, the lead content of placenta by 90% and the lead content of mother's milk by 15% compared with a control group without treatment. The cadmium content of the placenta was reduced to 4% of the content in untreated mothers.

[Stage Ic cancer of the cervix. A clinical and immunological analysis (author's transl)].

Marked differences in cases of stage I cervical carcinoma, dependent on the extent of tissue involvement, are demonstrated by retrospective, prognostic as well als by prospective, immunological investigations. It seems recommendable to separate cases of a newly-defried stage Ib from stage Ic. The prognosis of these stage Ic cases is even worse than of stage IIa cases. As this subgrouping can be carried out only post-operatively, the preoperative assignment of cases to different stages should not be used for assessing the operative results.

[Mortality data and statistics, importance for health services and epidemiological research (author's transl)].

In most industrialized countries mortality data are the only medically relevant complete statistics for the description of health and disease in a population. Although mortality data reflect just the opposite of the state of health of a population, specific age and sex mortality rates are important indicators of the health status of a population and beyond that for the social system, e.g. for the effectiveness of the subsystem of health services. Validation studies show that the diagnoses on death certificates are much more reliable than generally thought. Scientists must have access to information from the original death certificates because otherwise the increasingly desirable studies on the threat of environmental noxae to the population cannot be carried out.

The detection of carcinogenic effects in man.

Apart from the so-called short-term or in vitro test and the animal experiment, epidemiology represents the third method for detecting carcinogenic effects. Epidemiological methodology constitutes the sole possibility of studying in man cancerous diseases and the influence of noxae suspected to be carcinogenic. The three main approaches of epidemiological research--descriptive, analytic, and experimental epidemiology--are confronted and their importance for tracking down cancer is explained by means of examples.

Vasoactive intestinal polypeptide (VIP) inhibits prostaglandin-F2 alpha-induced activity of the rabbit myometrium.

Seven female rabbits ahd myometrial autografts transplanted into ear-chambers. The electrical activity of the graft was recorded and mechanical activity observed. Vasoactive intestinal polypeptide (VIP) dose-dependently inhibited myometrial activity induced by prostaglandin-F2 alpha. This evidence and the occurrence of "VIPergic" nerve fibres, which seem to innervate the myometrial cells, suggest that VIP may play a physiological role in the local control of the myometrial activity.

Correlations between internal mobility and stability of globular proteins.

The recent work is surveyed which leads to the suggestions that the conformation of globular proteins in solution corresponds to a dynamic ensemble of rapidly interconverting spatial structures, that clusters of hydrophobic amino acid side chains have an important role in the architecture of protein molecules, and that mechanistic aspects of protein denaturation can be correlated with internal mobility seen in the native conformation. These conclusions resulted originally from high resolution 1H nuclear magnetic resonance (NMR) studies of aromatic ring mobility, exchange of interior amide protons and thermal denaturation of the basic pancreatic trypsin inhibitor and a group of related proteins. Various new approaches to further characterize proteins in solution have now been taken and preliminary data are presented. These include computer graphics to outline hydrophobic clusters in globular protein structures, high resolution 1H-NMR experiments at variable hydrostatic pressure and 13C-NMR relaxation measurements. At the present early stage of these new investigations it appears that the hydrophobic cluster model for globular proteins is compatible with the data obtained.

[A new approach to the prophylaxis and therapy of puerperal mastitis (author's transl)].

The prolactin-inhibitor Bromocriptin was examined in an open study in comparison with Hexoestrol, a synthetic estrogen compound, in 31 patients with severe postpartal breast engorgement and in 38 patients with acute puerperal mastitis. In both indications a significantly faster clinical improvement was achieved with Bromocriptin. Also the incision-rate in cases of breast abscess was significantly lower in the Bromocriptin group. The inhibition of prolactin is considered the method of choice, as well in the prophylaxis as in the treatment of puerperal mastitis.
