[Experiences with miconazole in the prevention and therapy of candidiasis].

1. The treatment of 26 children suffering from haemoblastoses and tumors by the antimycotic Miconazol is reported on. 2. The application took place prophylactically as well as therapeutically by an oral or intravenous administration. Mycological examinations (stools, urine, throat swab, blood) have been carried through before, during and after the treatment in weekly intervals. 3. The result of treatment is altogether to be called good, invasive mycoses have been prevented and proved Blastomyces infections successfully treated. 4. Side effects like nausea and vomiting were observed only in one case, damages of the haematopoietic system, the liver and kidneys could not have been demonstrated.

Candidiasis↗

[Structure of mannan of Candida guilliermondii H].

The mannan of Candida guilliermondii was acetolyzed and the seperation by gel chromatography gave five neutral fragments with the characteristic nuclear magnetic resonance spectra, mannose, mannobiose, mannotriose, mannotetraose, and mannopentaose. All oligosaccharide fragments were composed of mannose in alpha-linkage. Methylation analysis combined with gas chromatography-mass spectrometry demonstrated (1 leads to 6)-linked D-mannopyranose main chains with (1 leads to 2)-linked D-mannopyranose side chains.

[Demonstration of alpha-mannosidase and beta-glucosidase in a yeast cell-wall lytic system of Streptomyces spec. 5s].

Streptomyces spec. 5s accumulates in the culture fluid lytic activities against living cells of Candida guilliermondii H. From the culture fluid a freeze-dried dialyzed ammonium sulfate precipitate was prepared. Except beta-glucosidase-activity of alpha-mannosidase which was characterized. Separation of both enzymes by chromatography on polyacrylamide gel was possible.

[Critical remarks on the surgical treatment of syndactylia (author's transl)].

The technique of surgery for syndactylia is described in detail. The author's opinion about the optimal age for surgical correction of equally or unequally long fingers is reported. The importance of early intervention in case of complicated syndactylia is stressed. Until growth has come to an end, continuous follow-up is necessary for early recognizing late contractures.

[Abortive duplication of the gut (author's transl)].

Presentation of a case of intestinal duplication sited in the rectosigmoid colon. Rectosigmoid colon and duplication were coated with gastric mucosa. Diagnosis of heterotopic gastric mucosa was done by biopsy and 99mTc-pertechnetate scintigraphy. The diseased colon was removed by Rehbein pull-through procedure. Complete removal was proved by clinical course and scintigraphy.

[Physiology and biochemistry of streptomycetes. VIII. Esterase activity and production of turimycin in cultures of Streptomyces hygroscopicus JA 6599].

Esterase in cell-free extracts of Streptomyces hygroscopicus JA 6599 has a temperature-optimum of 35 degrees C, a pH-optimum with p-nitrophenylacetate as substrate at pH 7.7--8.1, with alpha-naphthylacetate at pH 7--9. Michaelis constants in cell-free extracts: with alpha-naphthylacetate Km = = 0.71 mM, with p-nitrophenylacetate Km = 0.21 mM. Phenylesters were better hydrolyzed than naphthylesters, phenylacetate was best hydrolyzed; beta-naphthylacetate was better hydrolyzed than alpha-naphthylacetate. Among the naphthylesters the ester of propionic acid was hydrolyzed best. Caprylate, stearate, and 0,0-diethyl-0-(p-nitrophenyl)-phosphate inhibit the splitting of alpha-naphthylacetate. A comparison with esterases of other biological origin shows that the enzyme studied can be a carboxylesterase (E.C.3.1.1.1.). In cultures of JA 6599 V13 and JA 6599-6 the change of esterase activity during the fermentation was determined. We found a carrelation between the enzymatic activity and the antibiotic-concentration in the culture medium.

[Physiology and biochemistry of streptomycetes. X. Biological degradation of paromomycin and alkaline phosphatase activity depending on antibiotic production by Streptomyces albus var. metamycinus nov. var].

After adding 14C-paromomycin to the fermentation broth we observed a varying course of decomposition of the antibiotic, which is dependent on the intensity of paromomycin biosynthesis running simultaneously. At a reduced rate of antibiotic biosynthesis, the activity of alkaline phosphatase is lower than with an increased rate of production. This applies for mycelium as well as for broth.

[Physiology and biochemistry of streptomycetes. XI. Different incorporation of D-glucose-u-14C into the paromomycin isomers and the precursors of paromomycin I].

During application of D-glucose-u-14C paromomycine II is higher labelled and shows a different dependence on the application time than paromomycine I, which is isomer at the paromose part. For the two paromose isomeres different rates of synthesis are supposed that change nonproportionally to each other. The distribution of radioactivity in paromomycine I shows that there is no fragmentation of the glucose chain during the biosynthesis of glucosamine, ribose, and paromose I. As to the 2-deoxystreptamine the result has not been ascertained.

[Anabolic and catabolic enzymes of urea metabolism in a carbohydrate-utilizing strain of Candida guilliermondii].

The yeast "H" of the genus Candida guilliermondii can grow on hydrocarbons as the only source for carbon. Urea can serve as a nitrogen source for this yeast which lacks detectable urease activity. During urea metabolism ammonia has never been accumulated in the culture medium. However, transferring the yeast from complete urea-medium into an urea containing phophate-buffer, the degradation of urea continues and ammonia is accumulated as well as CO2 evolved. In cell-free extracts of the yeast urea amidolyase activity was detected in the presence of ATP, biotin and specific cations. Obviously, the synthesis of urea amidolyase is induced by urea and arginine and repressed by the catabolite ammonia. Similarly the synthesis of arginase is regulated by arginine and ammonia. The analytical data of the arginase action differ significantly in relation to the carbon source of the culture medium. Both the level of arginase and ornithine carbamyl-transferase change in a characteristic way during the batch-culture. From the lower level of arginase in relation to ornithine carbamyltransferase it can be concluded that especially in alkane-metabolizing yeast the arginine catabolism is not very intensive.

[Physiology and biochemistry of streptomycetes. XIII. Biosynthesis of paromomycin by Streptomyces albus var. metamycinus nov. var. supplied with 14C-glucose, 14C-glucosamine, 14C-2-desoxystreptamine and 14C-ribose].

Distribution of radioactivity in paromomycin ascertained after application of 14C-D-glucose, 14C-D-glucosamine, 14C-2-deoxystreptamine, respectively, 14C-D-ribose is taken as basis for a biosynthesis scheme: While ribose bound in the antibiotic originates from glucose by oxidation and following decarboxylation, glucosamine is formed via fructose-6-phosphate. Paromose I arises from glucosamine, but not the cyclohexan derivative 2-deoxystreptamine, whose biosynthesis pathway is directly branching off glucose.

[Recurrence-free phases and survival times in children with cytochemically varying acute lymphatic leukemias. Note on the prognostic evaluation of leukemias].

The recidive-free phases, survival times and the number of long surviging patients were recorded in 64 children with cytochemically differentiated ALL. A significantly more favourable course of ALL of PAS type could be observed as compared with the undifferentiated type and partially with the esterase type, too. According to the present findings the cytochemical examinations of ALL can be recommended in the initial phase of leukaemia as a prognostic evaluation.

[Physiology and biochemistry of streptomycetes. IX. Hydrolysis of cyclohexyl, carbohydrate-and aryl-esters by a carboxyl-esterase from cultures of Streptomyces hygroscopicus JA 6599].

Carboxylesterase in cell-free extracts of Streptomyces hygroscopicus was characterized with cyclohexylacetate and beta-penta-acetyl-D-glucose as as substrates. After concentrating the enzyme, activity with alpha/beta-methyl-4-O-propionyl-L-mycarosid as substrate was also observed. The influence of enzyme effectors was studied. Esterase activity was also found in the culture filtrate of. S. hygroscopicus.
