[Effects of various anesthesia procedures used in forceps delivery on the cardiopulmonary adaptation of the newborn infant. 1. Modification of cardiac adaptation].

Healthy, spontaneously born infants, delivered at the calculated time shows a postnatal tachycardia combined with harrowing of oscillation. But then there is a normalisation in heart action. This phenomenon is nearly the same both in spontaneous delivery and intubation anesthesia for forceps delivery starting with propanidid. Therefore propanidid is of less risk for the neonate. But after hexobarbital anesthesia our results reveal a lingering cardial adaption of the neonate, which is statistical significant. This points to possible cardiac depressive side effects of this drug. Therefore the usage of hexobarbital anesthesia should be well considered in obstetrics.

[Effects of various anesthesia procedures used in forceps delivery on the cardiopulmonary adaptation of the newborn infant. 2. Modification of pulmonary adaptation].

Mature newborns reveal the same pulmonal adaption to extrauterine conditions after forceps delivery in general anesthesia for the mother beginning with propanidid or hexobarbital as newborns after spontaneous delivery. Intervalls of apnoe, being physiological during neonatal adaption up to the fourth life-hour, occurred in all three tested groups in the same frequency. Genuine apnoe in combination with following bradycardia and cyanosis couldn't be observed. Moreover during the whole examination time all mean values of respiration frequency were physiological. Therefore we believe that there is no fundamental deterioration of newborns respiration, i.e. a depression in nerve-centre, caused by propanidid or by hexobarbital, despite of repeatedly contrary description.

[Effect of the oral beta-mimetic drug clenbuterol (Contraspasmin, Spiropent) and orally and intravenously administered Partusisten on the rheobase].

Clenbuterol (Contraspasmin, Spiropent) was used to treat threatened delivery in a dosage of three times 0.02 mg per day because of its beta-mimetic effects and the halflife time for elimination of 34 hours. We examined the effect of Clenbuterol on the maternal rheobase as a factor to estimate the influence of this drug on neuromuscular structures of maternal organism. After the first oral application of Clenbuterol we found a significant decrease in this value and after second and third application a distinct fluctuation. This fact shows besides the positive beta-mimetic properties the side-effects on the vegetative nerve system and on neuromuscular structures of the female body.

[Prognostic significance of lymph node metastases in operable cervix cancer of categories T1 and T2].

In a retrospective long term study the tumorbiological importance of metastases in lymph-nodes of cervix cancer have been examined. The base of this presentation consists of a follow up study of 855 patients suffering from cervix cancer (stage I 557 pat., stage II 298 pat.) in relation to lymph-node-metastases. Metastases in 9,9% in stage I and in 18,4% in stage II could been proved histologically. The 5-year-survival-rate decreased in stage Ib from 89 (T1 No Mo) to 59% (T1 Ni Mo) and in stage II respectively (T2a No Mo) from 73 to 27% (T2a Ni Mo) and from 89 (T2b Ni Mo) to 52% (T2b Ni Mo). About 59% of relapses appeared in the treated region of the pelvis. In cases of adenomatous and dedifferentiated cancer 4% more regional metastases could be found than in squamous cell cancers. By means of post-operative irradiation (especially telecobalt-therapy) the relapse-free interval could be prolonged about one to two years. Recently tumourbiological knowledge is involved in the discussion. The appearance of lymph-node metastases represents the worst prognostic feature for the cure and the life of the patient suffering from cervical cancer.

Glucoprivic responsivity after insulin in hypodipsic and in water deficit rats.

Long Evans rats were made acutely hypodipsic through quinine adulteration of their water supply, and they ate normally in response to 2, 4, and 6 unit insulin injections. Other subjects were made chronically hypodipsic through adulteration; and while their glucoprivic feeding was lowered absolutely, it remained relatively elevated over their control session intake. These data indicate that subjects with reduced drinking abilities can respond to the insulin-induced glucoprivic challenge and that hydrational factors apparently do not serve as critical confounds in brain lesion investigations into the glucoprivic feeding response.

Penicillin acylase from the hybrid strains Escherichia coli 5K(pHM12): enzyme formation and hydrolysis of beta-lactam antibiotics with whole cells.

Penicillin acylase formation by the hybrid strain Escherichia coli 5K(pHM12) was studied under different culture conditions and reached 200 to 250 mumol of 6-aminopenicillanic acid per min per g of bacteria (wet weight) for penicillin G. The Km of whole-cell acylase was determined with 9 to 11 mM for penicillin G at a pH optimum of 7.8 at 45 degrees C. A competitive product inhibition for phenylacetic acid of Ki = 130 mM was found. 6-Aminopenicillanic acid acts as a noncompetitive inhibitor, with a Ki of 131. The temperature optimum of the reaction lies at 54 degrees C. Penicillin G inhibits the reaction at Ki(S) = 1,565 to 1,570 mM. Whole-cell acylase reacts on a wide spectrum of penicillins and cephalosporins, but those substrates with a delta-aminoadipyl rest are not hydrolized. beta-Lactamase activity of less than 1% relative to the acylase activity was found at reaction temperatures between 28 and 45 degrees C. After a comparison of different methods for the estimation of beta-lactamase activity, we found that high-pressure liquid chromatography is to be preferred. During batch fermentation of E. coli 5K(pHM12), problems of plasmid stability in the host strain arose which were overcome by the addition of 4 mg of tetracycline per liter to the medium as a selective marker.

Formation and identification of interfacial-active glycolipids from resting microbial cells.

Resting cells of Arthrobacter sp. strain DSM2567 incubated in the presence of various mono-, di-, or trisaccharides biosynthesized different glycolipids. All eight glycolipids, containing the corresponding carbohydrate moiety and one, two, or three alpha-branched beta-hydroxy fatty acids, were produced when mannose, glucose, cellobiose, maltose, and maltotriose were used as carbon sources in a simple phosphate buffer. The structures of the compounds were elucidated by means of H and C nuclear magnetic resonance spectroscopy and by chemical ionization mass spectroscopy. In high-salinity solution, the substances showed different surfactant properties. Cellobiose and maltose monocorynomycolates reduced the interfacial tension from 42 to 1 mN/m at critical micelle concentrations below 20 mg/liter.

[Cathepsin D activity in human ovarian carcinoma].

In the tissue of 54 ovarian carcinomas and 38 normal human ovaries the activity of cathepsin D was determined by means of the Anson-method. In ovarian carcinomas the mean activity of cathepsin D was 4515 pmol T/s/g tissue, in normal ovarian tissue only 2119 pmol T/s/g. The difference is significant (p less than 0,001). The dependency on age of the activity of cathepsin D suitable for normal ovary, got lost in cancer tissue. There is no relationship between the amount of activity of cathepsin D and the stage of the tumour. Extreme high activities of cathepsin D were found in metastatic ovarian carcinomas. The activity of cathepsin D is higher in endometrioid and papillary than in mucigenous and hypernephroid ovarian carcinomas. The different activities of cathepsin D were discussed with regard to the invasive growth of cancer and to deranged protein metabolism.

Effects of bradykinin on pial arteries and arterioles in vitro and in situ.

The effect of bradykinin on cerebrovascular resistance vessels was investigated by the use of in vitro and in situ preparations. Bradykinin, in the range of 10(-10) to 10(-5) M, elicited a concentration-dependent vasodilatation on both feline and human pial arteries in vitro; the half-maximal response was found to be approximately at 2.8 X 10(-7) M and 1.3 X 10(-8) M (EC50), respectively. This dilatatory effect of bradykinin in vitro was found only in arteries preconstricted with prostaglandin F2 alpha or 5-hydroxytryptamine. In order to determine the effects of bradykinin on the diameter of cat pial arteries in situ, perivascular microapplication was employed. The dose-response curves obtained showed vasodilatation; the EC50 and the maximal response (EAm) were 4.4 X 10(-7) M and 45.5% at 10(-5) M, respectively. Statistically significant (p less than 0.01) reactions were observed at 10(-7) M and higher concentrations of bradykinin. The observed effects were independent of initial vessel size (80-260 microns). These in situ findings are very similar to those found in vitro. The isolated guinea pig ileum was used to check the stability of the bradykinin solutions. In this instance, a concentration-dependent contraction was found when "freshly prepared" or "5 hours stored" bradykinin was applied, indicating no measureable degradation of bradykinin. We conclude that bradykinin is a powerful vasodilator of both human and feline pial arteries.

The choice of contrast agent for myelography in patients with nonpenetrating cervical spine trauma.

Forty patients with neurological deficits from cervical spine trauma had cervical myelography with metrizamide, gas, iophendylate, or a combination thereof. At a low dosage (less than or equal to 1.25 g I), metrizamide was found to be not only the most effective in demonstrating the spinal cord but also safe if used with a C-arm biplane fluoroscopic unit which has tomographic capability.

[Comparison between colposcopic and cytologic findings in relation to clinical and histologic diagnoses in carcinoma in situ].

From 1977 to 1979 95 cases of carcinoma in situ had been detected at the Gynaecological Department of the Medical Academy of Erfurt. In 81 of these women there was a cytological examination before treatment. The accuracy of cytology and colposcopy was opposed the histological diagnosis in these selected cases. An agreement of colposcopy, cytology and histological findings could be found in 70 cases. In 6 cases colposcopy and in 4 cases cytology was wrong. Only in one case both examination methods failed.

The use of cisplatinum for treatment of malignant glioma.

Standard chemotherapy for malignant gliomas has not generally been successful. Recent reports have suggested that cis-diammine-dichloroplatinum is preferentially accumulated into brain tumors compared to surrounding brain tissue. A patient who had failed radiation treatments and multiple other chemotherapeutic agents responded both clinically and radiographically to this new drug. This clinical observation, when combined with the tissue distribution of cis-diamminedichloroplatinum, would indicate that further evaluation of this new agent in the treatment of brain malignancies is warranted.

Formation, Location, and Regulation of Endo-1,4-beta-Glucanases and beta-Glucosidases from Cellulomonas uda.

The formation and location of endo-1,4-beta-glucanases and beta-glucosidases were studied in cultures of Cellulomonas uda grown on microcrystalline cellulose, carboxymethyl cellulose, printed newspaper, and some mono- or disaccharides. Endo-1,4-Glucanases were found to be extracellular, but a very small amount of cell-bound endo-1,4-beta-glucanase was considered to be the basal endoglucanase level of the cells. The formation of extracellular endo-1,4-beta-glucanases was induced by cellobiose and repressed by glucose. Extracellular endoglucanase activity was inhibited by cellobiose but not by glucose. beta-Glucosidases, on the other hand, were formed constitutively and found to be cell bound. beta-Glucosidase activity was inhibited noncompetitively by glucose. Some characteristics such as the optimal pH for and the thermostability of the endoglucanases and beta-glucosidases and the end products of cellulose degradation were determined.

Chemical and Physical Characterization of Interfacial-Active Lipids from Rhodococcus erythropolis Grown on n-Alkanes.

Lipophilic compounds of the culture suspension containing Rhodococcus erythropolis DSM43215 had surfactant properties when the bacteria were cultivated with n-alkanes as the sole carbon source. Thirteen main components from a dichloromethane-methanol extract of the R. erythropolis cultures were isolated and characterized to specify quantitatively their surfactant properties, e.g., minimum surface and interfacial tensions and critical micelle concentrations. The interfacial activity of the organic extract was dominated by alpha,alpha-trehalose-6,6'-dicorynomycolates which reduced interfacial tension from 44 to 18 mN/m. Phosphatidylethanolamines which were also present in the organic extract reduced the interfacial tension below 1 mN/m. The trehalose corynomycolates had extremely low critical micelle concentrations in high-salinity solutions, and the interfacial properties were stabile in solutions with a wide range of pH and ionic strength.
