Repression of endo-1,4-beta-glucanase formation in Penicillium janthinellum and product inhibition of its 1,4-beta-glucanases and cellobiases.

Endo-1,4-beta-glucanase formation of Penicillium janthinellum was repressed by glucose, sophorose, and glycerol. Chromatography on DEAE-Sephadex A-50 was employed to separate the 1,4-beta-glucanases from two cellobiases. The 1,4-beta-glucanases were inhibited competitively by cellobiose and glucose, and the two cellobiases were inhibited by glucose and glucono-delta-lactone.

[LDH activity in human ovarian tissue and in ovarian carcinoma (author's transl)].

Lactate-dehydrogenase activities were measured from tissue homogenates of 34 human ovaries and ovarian carcinomas. Mean activities were 265.2 nmol/s/g tissue in normal ovaries and 695.1 nmol/s/g in ovarian carcinoma tissue. One and the same ratio was obtained from two comparative group of one and the same age. The above difference was significant. No relationship was found to exist between LDH activity in tumour tissue and stage of the disease.

[Effectiveness of surgical and radiotherapeutic action on cervical carcinoma, stages I and II (T1 N0 M0 and T2 N0 M0). First communication].

Reported in this paper are therapeutic results obtained from 855 patients with cervical carcinoma, Category T1 (557 patients) and Category T2 (298 patients), between 1967 and 1976. -- Metastasization of lymph nodes was recorded from 9.9 per cent of the patients in Category T1 and from 18.4 per cent in T2. --Abdominal radical surgery (Wertheim's operation) was applied to 87.3 per cent, while vaginal radical surgery (modified according to Schauta) was performed on 8.2 per cent, and the socalled "minor Wertheim", a modification devised at the authors' hospital, to 4.5 per cent. Complementary postoperative irradiation was used on 497 of 855 patients, with 93.6 per cent of these patients receiving postoperative irradiation under orthovolt conditions and 6.4 per cent under telecobalt conditions. -- Five-year survival rates were 87.5 per cent without postoperative irradiation and 83 per cent with postoperative irradiation in Category T1 or 78.1 per cent without irradiation and 73.2 per cent with irradiation in Category T2. Survival rates declined by about 30 per cent in cases with lymph node metastases. Manifestation of local recurrence was found to be delayed by about one to one-a-half year by postoperative irradiation, an effect, however, which was no longer detectable after five years had passed. Differentiation of radiation doses had no bearings at all on healing. Adjuvant postoperative irradiation of patients in Category p T1 N0 M0 proved ineffective. Mortality following primary therapy amounted to 2.1 per cent. Morbidity accounted for 36.8 per cent of all patients, with due consideration being given to all intercurrent complications.

[Course of pregnancy following cryotherapeutic treatment of the cervix uteri].

Course of pregnancy of 26 women is reported in this paper. All of them had undergone cryosurgical treatment during or prior to one of their pragnancies. The results obtained from such surgical treatment are compared to those recorded from traditional operations on the uterine cervix. Scarless healing of the portio and full retention of its elasticity are points made to claim good functional results of cryotherapy. Suggestions are made for use of cryosurgery during pregnancy.

Formation and Location of 1,4-beta-Glucanases and 1,4-beta-Glucosidases from Penicillium janthinellum.

Formation and location of 1,4-beta-glucanases and 1,4-beta-glucosidases were studied in cultures of Penicillium janthinellum grown on Avicel, sodium carboxymethyl cellulose, cellobiose, glucose, mannose, and maltose. Endo-1,4-beta-glucanases were found to be cell free, and their formation was induced by cellobiose. 1,4-beta-Glucosidases, on the other hand, were formed constitutively and were primarily cell free, but with a small amount strongly associated with the cell wall. Low 1,4-beta-glucosidase activities of periplasmic or intracellular origin were also found. A rotational viscosimetric method was developed to measure the total endo-1,4-beta-glucanase activity of the culture (broth and solids). By this method, it was possible to determine the endo-1,4-beta-glucanase activity not only in the supernatant of the culture but also on the surface of the mycelium or absorbed on residual Avicel. During a 70-liter batch cultivation of P. janthinellum, the adsorption of endo-1,4-beta-glucanases by residual and newly added 10% Avicel was measured. The adsorption of soluble protein and endo-1,4-beta-glucanases by Avicel was found to be largely independent of the pH value but dependent on temperature.

[Results of eclampsia treatment--analysis of 44 cases over 20 years (author's transl)].

Reported in this paper are 44 cases of eclampsia recorded from among 61,809 deliveries at the Gynaecological and Obstetric Hospital of the Medical Academy of Erfurt, between 1960 and 1979. Five mothers died and two more suffered permanent damage. Two infants died perinatally. However, an estimate of eclampsia risk to patients with gestosis cannot be derived from the intensity of the gestosis symptoms and thus from the gestosis index (Goecke).

[Deliveries by caesarean section--variation of indications (author's transl)].

Reported are 657 cases of caesarean section performed in selected periods over the past two decades. An account is given of change in indications and the results accordingly achieved. Particular reference is made to higher risk of infection due to antepartum and intrapartum intensive care. Indications should continue to be based on most stringent criteria.

[Concentration of ascorbic acid in tissue of ovarian carcinoma (author's transl)].

The levels of ascorbic acid were measured in the tissue of 53 ovarian carcinomas and compared to concentrations in normal human ovarian tissue. The mean ascorbic acid concentrations per gram of tissue was 218 microgram in carcinoma tissue and 382 microgram in intact ovarian tissue. The difference was significant, with p less than 0.001. A relationship was found to exist between ascorbic acid concentrations in tumor tissue, on the one hand, stages of the disease, on the other. The importance of ascorbic acid to tumour growth is discussed.

Purification and properties of glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase from a methanol-utilizing yeast, Candida boidinii.

Glucose-6-phosphate dehydrogenase (D-glucose-6-phosphate: NADP oxidoreductase, EC 1.1.1.49) and 6-phosphogluconate dehydrogenase (6-phospho-D-gluconate: NADP oxidoreductase, EC 1.1.1.44) were purified approx. 1700 fold and 330 fold, respectively, from Candida boidinii grown on methanol. The final enzyme preparations were homogeneous as judged by polyacrylamide gel electrophoresis. The molecular weights of the enzymes were estimated to be 118 000 and 110 000, respectively. Both enzymes are composed of two probably identical subunits and the molecular weights of the polypeptide chains were calculated to be 61 000 and 58 000, respectively. From a consideration of enzyme activities and types of inhibition by different metabolites the role of these two enzymes in glucose- and methanol-metabolism is discussed.

Steady-state kinetics of formaldehyde dehydrogenase and formate dehydrogenase from a methanol-utilizing yeast, Candida boidinii.

Initial velocity studies and product inhibition studies were conducted for the forward and reverse reactions of formaldehyde dehydrogenase (formaldehyde: NAD oxidoreductase, EC 1.2.1.1) isolated from a methanol-utilizing yeast Candida boidinii. The data were consistent with an ordered Bi-Bi mechanism for this reaction in which NAD+ is bound first to the enzyme and NADH released last. Kinetic studies indicated that the nucleoside phosphates ATP, ADP and AMP are competitive inhibitors with respect to NAD and noncompetitive inhibitors with respect to S-hydroxymethylglutathione. The inhibitions of the enzyme activity by ATP and ADP are greater at pH 6.0 and 6.5 than at neutral or alkaline pH values. The kinetic studies of formate dehydrogenase (formate:NAD oxidoreductase, EC 1.2.1.2) from the methanol grown C. boidinii suggested also an ordered Bi-Bi mechanism with NAD being the first substrate and NADH the last product. Formate dehydrogenase the last enzyme of the dissimilatory pathway of the methanol metabolism is also inhibited by adenosine phosphates. Since the intracellular concentrations of NADH and ATP are in the range of the Ki values for formaldehyde dehydrogenase and formate dehydrogenase the activities of these main enzymes of the dissimilatory pathway of methanol metabolism in this yeast may be regulated by these compounds.

[Betaglucuronidase activity in ovarian carcinoma (author's transl)].

Beta-glucuronidase activity was recorded from the tissue of 45 ovarian carcinomas and compared to the activity in normal human ovarian tissue. Mean activities were 1442 micrograms Ph./h/g tissue in ovarian carcinomas and 574 micrograms Ph./h/g in intact ovarian tissue. No relationship was found to exist between the amount of beta-glucuronidase activity and the stage of the disease.
