Peripheral blood (PB) and bone marrow (BM) cells were obtained from 12 patients with chronic granulocytic leukaemia (CGL) and cultured in agar for granulocyte macrophage colony formation (GM-CFC). In addition PB and BM CGL cells were co-cultured with T-lymphocytes from patients with immune severe aplastic anaemia (SAA), or with T-cells from healthy donors primed with pokeweed mitogen (PWM). The supernatants of SAA and PWM primed T-cells were also added to CGL cells grown in agar. Normal marrow cells obtained from eight healthy donors were used to set up control cultures and co-cultures. The results of this study indicate that, firstly, T-cells derived from SAA patients and their supernatants suppress GM-CFC growth of normal marrow cells but not of PB nor BM CGL cells, and, secondly, normal T-cells primed with PWM and their supernatants suppress normal marrow GM-CFC but not colony formation of BM nor PB CGL cells. These results provide further evidence that mediators which are effective in regulating normal myeloid progenitor cells fail to inhibit the in vitro growth of GM-CFC from CGL patients.

We describe a method, based on Selye's granuloma pouch, for establishing reproducible Bacteroides fragilis infections in both mice and rats starting from rather low inocula. The infected pouches resemble abscesses and phlegmons. This model is promising for studies of antibiotic therapy in that both bacterial kinetics and antibiotic levels can be followed in the exudates.

The authors refer their experience in fine needle transperitoneal lymphnodes biopsy in 19 prostatic cancer patients with negative or border-line lymphography. This technique improved the diagnosis and may be applied in staging of malignant diseases of the pelvis.

Seven children with chronic or intermittent and six with acute idiopathic thrombocytopenic purpura (ITP) were treated with large intravenous doses of polyvalent, intact immunoglobulin (Ig). In all patients the platelet count rose sharply within 5 days, but the initial response and the subsequent course varied from patient to patient. Among children with chronic ITP the initial response was more marked in splenectomised than in non-splenectomised patients. Among those with acute ITP the two who remained Ig dependent had a smaller initial response than the four patients who required no maintenance treatment. During the 90-110 days of observation five of six patients with chronic ITP could be maintained with Ig alone. No untoward effects of Ig therapy were observed.
The major thrust of the worldwide effort on the disease entity cystic fibrosis is concerned with genetic, diagnostic and therapeutical questions and detection of the primary defect. The authors summarize new aspects presented at the VIIIth International Congress on Cystic Fibrosis in Toronto, Canada, 1980. Their aim is to inform about present knowledge on prenatal diagnosis and neonatal screening, to review clinical problems in the management of children with cystic fibrosis and, finally, discuss the future problems of adolescent and adult cystic fibrosis patients.
Colony formation in agar (CFU-c) was studied in 20 patients with severe aplastic anaemia by three different assays: (1) cultures of light density untreated marrow cells; (2) cultures of marrow cells manipulated in order to enhance colony formation (pretreatment with antilymphocytic globulin, ALG, or 6-methylprednisolone, 6-MPr, T cell depletion, adherent cell (AC) depletion, depletion of both AC and T cells), and (3) co-culture of putative suppressor T cells with autologous T-depleted marrow cells. By the first assay, all patients showed poor colony formation (1 +/- 1.5 colonies/10(5) cells; normal controls 46 +/- 18 colonies/10(5) cells). By the second assay, ALG and 6-MPr had no significant effect on colony formation. Removal of adherent cells proved equally without effect on colony growth. On the contrary, removal of T cells enhanced significantly (P less than 0.001) colony formation in 10 out of 20 patients. By the third assay, colony formation of marrow cells (deprived of T lymphocytes) was inhibited by the addition of autologous T cells in six patients studied. All patients were given high dose bolus 6-MPr as first treatment on admission: only patients who had detectable suppressor T cells in their marrow achieved a complete autologous haematologic reconstitution after 6-MPr or after 6-MPr and ALG. The results of this study indicate the detection of CFU-c/suppressor T cells correlates with responses to immunosuppressive regimens, and may thus help to identify patients with immune mediated aplastic anaemia.
In a patient with thrombotic thrombocytopenic purpura (TTP) severe neurological complications leading to coma were repeatedly reversed by treatment with urokinase. Although the therapeutic modalities for TTP still appear to be a matter of debate, thrombolytic treatment could be considered in TTP patients with severe organ involvement before the corresponding function is completely compromised.
DL 473, a new semisynthetic rifamycin, was 2-10 times more active in vitro than rifampicin (RAMP) against several clinical isolates of Mycobacterium tuberculosis and only slightly less active than RAMP against Gram-positive and Gram-negative bacteria. It showed excellent therapeutic activity in mice in experimental infections caused by Staphylococcus aureus, Streptococcus pyogenes group A, Streptococcus pneumoniae and Klebsiella pneumoniae. In the experimental TB infection in the mouse DL 473 was clearly more active than isoniazide and RAMP, two of the most effective antitubercular drugs in current use. The LD50 in the mouse was significantly higher than that of RAMP and the half-life was about 5 times longer than that of RAMP.
Association cases of tuberous sclerosis (ST) and renal displasia-tumors are infrequently seen in literature. These tumors are bilateral renal cystic displasia (RP) rare hamartomas, p.e. angiomyolipomas, and malignant neoplasms, p.e. renal clear cell carcinomas. There therefor as to the frequency of those pathological associations with ST, AML are frequently seen (50-80%). Reports of polycystic renal disease with ST are rare, whereas occasional associations of renal cell carcinomas with ST are founded out. By extensive literature examination of it's evident that the synchronous association of these pathological lesions is exceptional. This report describes one case of AML, RP and renal clear cell carcinoma in a female, 22 years old, with ST; the pathological, clinical and pathogenetic features are discussed.
