Effect of irradiation on glycosaminoglycans content in rat tissue.

Glycosaminoglycan (GAGs) fractions were determined in tissues (skin, liver, lungs, aortic wall) and blood serum of rats irradiated with a single dose of 500 R. An increase of total GAGs as well as changes in the fractions were found in the tissues and urine of exposed rats.

Studies on the influence of fluoride compounds upon connective tissue metabolism in growing rats. I. Effect of hydrofluoride on collagen metabolism.

Female rats were exposed to hydroxyfluoride (2.7 mg/m3 or 5.0 mg/m3) seven weeks before pregnancy during pregnancy and lactation as well as subsequently with the offsprings up to 1, 2, 4 and 6 months after birth. The offspring was taken for investigations. It was found that serum hydroxyproline and hydroxylysine was elevated in growing rats exposed to hydrofluoride. The elevation was higher in animals inspired to 5.0 mg/m3 than in rats treated with 2.7 mg/m3. One- and two-months old rats were shown to be more suitable to hydrofluoride. Similar changes were observed in urinary excretion of these compounds. Changes in collagen content were found in skin and lungs. A marked decrease total collagen content and changes in collagen fractions were shown.

Effect of sodium fluoride on collagen content in skin and lungs of growing rats.

Adult rats were given water with sodium fluoride (10 mg/l) during 7 weeks before pregnancy, during pregnancy and after the delivery. The born rats were treated in the same manner and were investigated 1, 2, 4 and 6 months after birth. It was found that exposure to sodium fluoride stimulated an increase of hydroxyproline and hydroxylysine concentration in serum and the urinary excretion of these catabolites. A decrease of soluble and insoluble collagen was shown in skin and lungs of exposed animals as compared with controls. The decrease of collagen in tissues was not age-dependent.

Influence of thyroid hormones on collagen content in tissues of guinea pigs.

Collagen fractions content and level of collagen catabolites in body fluids were determined in normal, hypo- and hyperthyroid guinea pigs. An increase of urinary excretion of hydroxyproline and hydroxylysine as well as concentration of these amino acids in blood serum was found in hyperthyroidism, and a decrease was shown in hyperthyroid guinea pigs. Hyperthyroidism stimulated an increase of neutral-salt-soluble and acid-soluble collagen in skin and liver, and a decrease of insoluble collagen in skin as well as increase of all collagen fractions in bone samples. Hypothyroidism induced a decrease of all collagen fractions in skin and liver, and an increase of acid-soluble and insoluble collagen in bone samples.

Morphological changes observed after stimulation or inhibition of adrenic system in rats chronically treated with hydrazinophtalazines.

Morphological studies of the heart muscle were carried out in healthy rats as well as treated with hydralazine or binazine after administration of adrenaline, noradrenaline, isoprenaline, regitine or propranolol. Hydrazinophtalazines (25 mg/kg body wt, daily) were given during 7 months. After discontinuation of this treatment the drugs were administered during 15 days. Evident changes (e.g. inflammatory infiltrations, focal necrosis and fibrosis) were found after administration of noradrenaline, adrenaline or isoprenaline. No differences were found in the reactivity to catecholamines in healthy and hydrazinophtalazine-treated rats.

Increased solubility of collagen. A biochemical defect in the experimental hydralizine-induced collagen disease-like syndrome.

An increase of soluble fractions of collagen and a decrease of insoluble ones was found in the skin of guinea pigs with collagen disease-like syndrome, produced by prolonged treatment with hydralazine. Chromatography studies showed that the collagen alpha chains content in the skin increased and beta-chains decreased under conditions of the development of lupus-like syndrome. The obtained results suggested the inhibition of collagen maturation as one of biochemical defects of connective tissue in drug-induced collagen disease-like syndrome.

Collagenolytic activity in leukocytes isolated from patients with leukemias and Hodgkin's disease.

Collagenolytic activity in leukocytes and plasma concentration of hydroxyproline were estimated in 80 patients with leukemias and Hodgkin's disease and in 20 healthy individuals. An increase of both studied parameters was found in chronic myelocytic leukemia and Hodgkin's disease, and a decrease was shown in acute myelocytic leukemia and acute lymphatic leukemia. The results obtained imply that metabolic changes occur in leukemia leukocytes, and indicate the presence of disturbances in the metabolism of connective tissue in the studied disorders.

Haematological changes in experimental hydralazine-induced collagen-like syndrome in guinea pigs.

Haematological studies were carried out in hydralazine-induced collagen-like syndrome in guinea pigs. 37.5 per cent of animals were found to be LE-positive. It was found that long-term administration of hydralazine caused a decrease of erythrocyte count, a decrease of haemoglobin concentration and a decrease of haemoglobin content in individual red blood cell as well as a decrease of a single erythrocyte volume. A significant leukopenia was shown in LE-positive subgroup of hydralazine-treated guinea pigs. The obtained results confirmed the similarity of hydralazine syndrome to systemic lupus erythematosus.
