Long-term study of the microvascular sleeve anastomosis: an experimental study in the rabbit renal artery.

A long-term study of 5 sleeve anastomoses in rabbit renal arteries was performed. Six months postoperatively angiography, light microscopy and electron microscopy was carried out. Good long-term function of the anastomoses without any stenoses or aneurysm formation was obtained. It was concluded from this study in rabbits that the sleeve technique is suitable for microvascular anastomosis and that good long-term function can be obtained.

Enhanced growth hormone response to clonidine in the spontaneously hypertensive rat.

Plasma concentrations of growth hormone (GH) were measured after administration of clonidine (0.5 mg/kg) to spontaneously hypertensive (SH) rats and normotensive Wistar Kyoto controls. All rats were pretreated with reserpine (10 mg/kg). The GH response to clonidine was significantly higher in SH rats than in controls, indicating that in SH rats the responsiveness of post-synaptic alpha-adrenergic structures in the hypothalamus may be increased.

A comparative analysis of the cell surface properties of activated vs endogenous mouse natural killer cells.

Mouse natural killer (NK) cells can be activated by interferon and by interferon inducers, such as viruses. In the present report we compared the cell surface properties of the nonactivated "endogenous" NK cells from normal mice with NK cells activated in vivo by acute infection with lymphocytic choriomeningitis virus (LCMV) or in vitro by interferon. Several striking differences were found: 1) in vivo LCMV-activated as well as in vitro interferon-activated NK cells were more adherent to nylon wool columns than the endogenous NK cells; 2) activated NK cells were more EA-monolayer adherent than endogenous NK cells; this adherence could be blocked by streptococcal A protein, indicating that activated NK cells expressed greater Fc receptor mediated adherence; 3) LCMV-activated NK cells that passed through nylon wool columns expressed relatively low EA adherence properties; 4) the sensitivity of NK cells to anti-theta serum + complement treatment was increased in spleen cells late after LCMV infection (day 6 to 7) but not in spleen cells early after infection (day 2 to 3); 5) as shown by centrifugal elutriation, LCMV-activated spleens contained a population of large NK cells that were not present among endogenous spleen cells. In contrast to previous reports, these findings show that there are several distinct changes as a result of both long-term in vivo activation or of short-term in vitro activation of NK cells. These changes might functionally be involved in the increased lytic ability of activated NK cells.

Determination of prekallikrein in plasma by means of a chromogenic tripeptide substrate for plasma kallikrein.

A method for plasma prekallikrein determination utilizing a chromogenic tripeptic substrate is presented. The method has a good reproducibility and can easily be automized. Several parameters have been optimized. By using mixtures of deficient plasmas and pooled normal plasma or purified factors it was proved that prekallikrein was the factor determined and that more than 10% (of normal plasma concentration) of FXII and HMW kininogen were essential for the activation of prekallikrein in our method. Further experiments showed that the method was fairly selective and was not influenced by inhibitors present in normal plasma. The later finding was attributed to the high dilution of plasma made possible by using a potent activator and a sensitive substrate.

White blood cell consumption in the microcirculation after a major burn.

The mesenteric microvasculature was observed before and after a distant full-thickness cutaneous burn in rats. Diameters, flow velocities, and white blood cell sticking and emigration were measured. Between 30 and 360 minutes postburn 13 to 16 WBC's were found to stick to the venular walls which had an area of 1.57 x 10(4) microns(2). Therefore, the number of WBC's emigrating from the vascular compartment during the 6-hour observation period could be as many as 0.51 X 10(9)/kg. Based on known values for total stores of WBC's, the data suggest that granulocyte sticking and emigration could account for a consumption of granulocytes that on an acute basis could deplete the body stores, which could explain the apparent decrease in total body granulocytes seen clinically following major thermal trauma.

Dissolution of cobalt from hard metal alloys by cutting fluids.

On grinding hard metal alloys containing cobalt, the dissolution of cobalt during 5 days was studied for nine commercial cutting fluids. After 1 day, a major part of the cobalt liberated from the material ground is found in solution. This percentage decreases when grinding is continued using the same coolant fluid. Only a small fraction of the cobalt is found in particles in the circulating fluid. The bulk must be in the sediment in the storage tank, where it does no harm to the workers' skin. The final concentration of cobalt found in some fluids may be hazardous.

Problems in recording arthroscopy.

Several good combinations of photographic equipment are available today for recording arthroscopies. Some of them are so simple to use that everyone can obtain good still pictures through the arthroscope. It is usually preferable to enlarge the arthroscopic pictures in order to get better slides for presentations during lectures and at scientific meetings. Movie film and video tape offer a more dynamic recording but are both fairly expensive. The use of video tape can cause problems during presentations to larger audiences. Video tape, however, can be transferred to 16 mm. movie film, which is the international standard in the teaching world. Color TV has three different standards in different parts of the world and there are at least four different video casette systems available today.

Ender nailing in fractures of the diaphysis of the femur.

Sixteen fractures of the diaphysis of the femur were nailed according to the Ender technique. Fifteen of these were of types for which Küntscher nailing was contraindicated. In five of the injuries a comminuted fracture of the femoral shaft was present, associated with a fracture of the proximal end of the femur. Most of the nailings were done closed. The time of surgery was short and the blood loss, small. All of the fractures healed and no major postoperative problems were noted.

Immunization aginast tetanus with aluminium free-versus aluminium contain triple vaccine.

In order to discover if elimination of aluminium from the very effective triple vaccine gives an unsatisfactory immunity against tetanus, the following investigation has been performed. 47 children were immunized in 1968 and 1969 during the first year of life with 3 injections of aluminium free triple vaccine given with an interval of 4--6 weeks. The vaccine contained 10 Lf tetanus toxoid per dose. 8 years later, during 1976 and 1977, they were given boosters containing aluminium adsorbed Td with 4 Lf tetanus toxoid. The above mentioned group was compared with 29 children immunized in the 1950's in the same way but with aluminium adsorbed triple vaccine (13--19 Lf tetanus toxoid per dose). The booster with DT given 6 to 7 years later contained 8 Lf aluminium adsorbed tetanus toxoid. The serum content of tetanus antitoxin was determined before and after the booster. The result showed that the tetanus immunity was very satisfactory even though the basic immunization was performed with aluminium free triple vaccine.

Prevention of dermal ischemia after thermal injury.

One percent methylprednisolone acetate was evaluated as a pharmacologic agent in the prevention of dermal ischemia following burning. Standardized partial thickness burns were inflicted on guinea pigs. Burned guinea pigs were separated into five groups; one was treated with topical steroid, one with systemic steroid, one with both, one with emollient base without steroid, and one served as an untreated control. Histology and depth of dermal ischemia were evaluated by india ink perfusion technique. Untreated controls showed progressive dermal ischemia with complete absence of india ink-filled vessels in the dermis by 24 hours. Topical steroid alone improved dermal perfusion as suggested by relative levels of india ink filling. Topical steroid in the dosage used does not potentiate infection in standard burn wound sepsis models. Preservation of dermal appendages was seen secondary to improved dermal microcirculation with a ninefold increase in hair follicles in treated guinea pigs compared with controls.

Experimental flap delay with formic acid.

Using dorsal, cranially based 10 X 3 cm composite pedicle flaps in rats, the effects of an injection of 2 ml of formic acid in different concentrations was studied. Concentrations of 0.2 per cent gave a very significant increase in flap survival over controls. Preoperative penicillin did not influence the outcome.

Hepatitis B immune globulin in prevention of hepatitis B among hospital staff members.

In a double-blind trial of hospital staff members at risk of contracting hepatitis B, 110 subjects received hepatitis B immune globulin or immune serum globulin prophylactically. An additional 125 individuals working in the same locations were unwilling to participate in the trial and received no prophylaxis. During the study period of 28 months, 13 cases of clinical hepatitis B occurred in the untreated group, whereas only three cases occurred among the subjects who received prophylaxis.
