Nature and stability of secondary structures formed from DNA-fragments during electrophoresis in urea containing polyacrylamide-gels were investigated. Duplices and especially hairpin loops were found to be surprisingly stable. Polyacrylamide-gels containing high concentrations of formamide effectively melt these structures. Such gels will improve nucleotide sequence analysis in regions of dyad symmetry that often cause 'band compressions' under standard electrophoresis conditions.
For treatment of spasmodic torticollis (s.T.) microsurgical decompression of the intraspinal-intracranial portion of the accessory nerve (a.N.) has been performed in 11 patients with proved neurogenic lesions of the accessory nerve-dependent muscles. Neurogenic lesions were discovered by meticulous electromyographic (EMG) examination in 26 out of 32 patients with s.T. Based on the EMG findings and a.N. roots were exposed, mostly bilaterally. During operation we found in each case tight adhesions to adjacent structures, in particular the vertebral artery (v.A.), the posterior inferior cerebellar artery (PICA), and spinal arteries. Moreover, various nerve anastomoses were found between the upper dorsal cervical roots and the spinal a.N. roots. After neurovascular lysis and dissection of anastomoses, to C 1 in particular, the a.N. was protected with teflon-foam (Prosthex). Immediately after surgery torticollis had improved in all but one case. Further improvement was achieved by exercises. These first favourable results, and the fact that nerve decompression is less destructive that other surgical procedures in treatment of s.T., make us feel justified in recommending this procedure further to patients suffering from s.T. with proved neurogenic lesions.

Hexobarbital sleeping time was prolonged and ethylmorphine N-demethylation was inhibited after a single dosage or seven administration of 6-SAI to old rats. These effects were independent of the development of arthritis. Changes in cytochrome P-450 concentration after 6-SAI treatment were insignificant and thus not responsible for the decrease in drug metabolism. In vitro 6-SAI inhibited ethylmorphine N-demethylation; the inhibition was of a mixed type. 6-SAI bound to cytochrome P-450 and induced a type II spectrum. The magnitude of hexobarbital-induced type I spectral changes was diminished by 6-SAI. It is concluded that 6-SAI inhibits cytochrome P-450-dependent drug metabolism by binding to cytochrome P-450.
While the health of the general population is not adversely affected by the exposure the pesticides in food and environment, workers, who have during their occupation been intensely exposed to pesticides, exhibit a higher prevalence of slight neurotoxic symptoms. In the present study the electroencephalograms of 3 of a group 10 workmen, who had been continually exposed to hexachlorcyclohexane, show pathological findings. The electromyograms of 8 of these 10 workman demonstrate a disturbance of the peripherical motoneuron. All probands, who exhibit o pathological EEG, also show a polyneuropathy.
The determination of the maximum F-wave conduction speed by means of the photographic superposition technique is a method that can be carried out in a technically simple manner and can give an insight into the peripheral motor neuron up to the anterior horn cell. The values can be determined in a more exact manner than the respective motor conduction speeds. Similar to the motor conduction speed, influencing factors are to be taken into consideration such as the temperature and the age of patients under ten years. The normal values for the N. ulnaris, radialis, fibularis and tibialis are submitted.
The testing of Convulsofin carried out in 220 patients at 14 clinics over a period of six months confirms the international experience gained with valproic acid and sodium valproinate. The main field of application of Convulsofin according to the experience gained will again be the treatment of fits in generalised primary epilepsy for which it permits to carry out a monotherapy to a remarkable extent. The favourable effect of the preparation in photosensitivity could be confirmed. Also in generalised secondary epilepsy and in fits of partial epilepsy. Convulsofin is partly effective, so that it can recommended as a co-medication in the treatment of therapy-resistant forms of these epilepsies which are difficult to treat. Decrease in thrombocytes, transient increase in serum transaminases, gastrointestinal disorders, loss of hair and undesired increases in body-weight were the more frequently occurring side-effects.
6-Hydroxydopamine (6-OHDA) pretreatment of rats in the first 4 days of life considerably enhances the low hepatic ethylmorphine N-demethylation rate in 9 and 16 day old rats, whereas the higher rates in adult rats are not influenced. After 6-OHDA treatment the age differences in ethylmorphine N-demethylation rate disappear. The induction of ethylmorphine N-demethylation by phenobarbital is markedly enhanced by neonatal 6-OHDA pretreatment in 16 to 60 day old rats, whereas in 9 day old rats this synergistic action is not detectable. Unlike ethylmorphine N-demethylation, hepatic 7-ethoxy-coumarin 0-deethylation rate is only slightly or not at all influenced by 6-OHDA pretreatment. The basic activities in 9 day and rats are enhanced, the induction by phenobarbital in the same age-group is decreased. In other age-groups neither basic activities nor inducibility are influenced by 6-OHDA pretreatment. Hepatic cytochrome P-450 concentrations in 33- and 60 day old control rats do not differ from those in 6-OHDA-pretreated animals. Phenobarbital treatment increases the P-450 content; this induction effect is enhanced after 6-OHDA pretreatment, but not as markedly as with ethylmorphine N-demethylation.

The expertise and treatment documents of 173 invalid epileptics of the period from 1967 to 1977 have been analysed according to a catalogue of 117 characteristics. Furthermore, a catamnestic questioning has been carried out in 70 cases involving unclear facts. The analysis concerned in the main the social structure, treatment and appraisement centres, mode of treatment, reasons for the declaration of invalidity, classification of epilepsy, psychic findings and rehabilitation proposals. Suggestions for the improvement of both the treatment situation and the expertise situation are derived from the results obtained.

A case of metastasizing adenoid squamous cell carcinoma of the vulva in a 44 year old woman is described. The differential diagnosis of this rare tumour is discussed.
From 1976 to 1978 11, 197 women were examined clinically and mammographically. Biopsy material from 1,673 breasts were examined microscopically. In 536 cases, or almost every third case (32%), a carcinoma of the breast was detected. The cancer was bilateral in 19 cases and the total number of women was therefore 517. A clinically occult tumour was only found in 7.7% (40 of 517) of the cases. 5% of these patients were high risk patients and 2.7% preventive examinations. 5 women with occult carcinoma of the breast were under age 40 and 14 under age 50. Benign changes of the glandular tissue were found in 59.5% of the cases. Marked proliferative changes were found in 4.6% of the cases and carcinoma in situ was found in 3.8% of the patients. In the age group 45--54 benign and proliferative changes of the parenchyma occured almost twice as often as cancers. The ratio between benign and malignant findings was 1:1 in the age group 55--59 and was less than 1:2 in the age group over 70. A sophistication of the mammograhic technique must be obtained. A thorough microscopic examination of tissue from subcutaneous mastectomies and tissue obtained at the time of reduction mammoplasties showed occasionally unexpected malignant tissue in an unexpected location. Especially these cases are suitable for later comparison to the mammographies.

In 15 ambulatory patients with renal insufficiency (creatinine clearance, 9.9 +/- 3.0 ml/min) the effect of oral supplementation with alpha-ketoacids has been compared with that of placebo. The protein intake amounted to 0.55 g protein per kilogram body weight of high biological value, as estimated by dietary recordings. After a control period of 3 months the patients received, in a double-blind study, 1.05 g alpha-ketoacids/10 kg body weight per day or a placebo for 6 weeks with a subsequent cross-over. Fasting blood samples were analyzed at 3-week intervals for routine laboratory parameters and 17 proteins. Anthropometric and clinical data have been recorded every 3 weeks. While therapy with alpha-ketoacids diminished PO4 levels (P less than 0.05), no other significant effect could be demonstrated. No signs of protein deficiency existed either before or during alpha-ketoacid therapy. Therefore, supplementation with alpha-ketoacids appears to be superfluous in patients with renal insufficiency maintained on a 40-g protein diet.
By using cloned deoxyribonucleic acid fragments from the hemolysis determinant of the hemolytic plasmid pHly152 as hybridization probes, a deoxyribonucleic acid segment of about 3.8 megadaltons was identified as a common sequence in several hemolytic (Hly) plasmids of Escherichia coli belonging in four different incompatibility groups. This segment contained the genetic information for the synthesis and secretion of the extracellular toxin alpha-hemolysin of E. coli. With the exception of pSU5, representing a composite plasmid, one part of which seems to be very similar to pHly152, the overall sequence homology of these Hly plasmids with pHly152 seems to be rather restricted. However, the Hly plasmid pSU316 showed sequence homology with pHly152 that did not extend beyond the hemolysis determinant. The two other plasmids, pSU233 and pSU105, also shared homology with pHly152 in the hemolysis determinant as well as in various other parts of this plasmid which did not seem to be directly linked to the hemolysis determinant. This suggests that the hemolysis determinant has spread to presumably unrelated plasmids of E. coli.
In normally fed guinea pigs the effect of a sustained release pentagastrin preparation was studied on gastric mucosal weight, protein, DNA and RNA concentration and thymidine and uridine incorporation into DNA and RNA, respectively. Treatment with pentagastrin for 3 days significantly increased antral fundic mucosal DNA and RNA concentration and thymidine and uridine incorporation. It is concluded that pentagastrin stimulates gastric mucosal nucleic acid synthesis beyond the driving force elicited by food.
