A murine monoclonal antibody against blood group H type-1 and -2 structures.

A murine anti-H monoclonal antibody was produced. This antibody, called sp115, is an IgM that does not agglutinate classical Bombay. Sp115 may differentiate the two categories of H-deficient phenotypes. It detects H substance in both plasma and fresh saliva. Tissue antigen is found when using this antibody. The specificity of sp115 antibody was determined by adsorption with, and elution from synthetic oligosaccharide immunoadsorbents. Sp115 does not distinguish H type-1 and H type 2-structures.

Plasma protein polymorphisms in a puzzling Breton community, the Bigoudens.

A total of 386 and 393 members of a Breton community, in France, the Bigoudens, have been tested for polymorphisms of C3 and haptoglobin (Hp) plus Transferrin (Tf) respectively. An interesting observation was a gap between the observed and expected phenotype C3 frequencies. The Hp and Tf gene frequencies were not found to be closer to the other Celtic peoples than to the French and English populations.

Study of blood group B antigen with a specific monoclonal antibody (anti-B, b-183).

A murine anti-B monoclonal antibody was obtained by the hybridoma technique. This antibody called anti-B (b-183) is of IgM nature; it is capable of agglutinating normal B, B3, Bx, cis AB and some acquired B red cells. Its association constant is 1.1 X 10(8) l/mol, and appears high compared to those of the monoclonal anti-A. This monoclonal anti-B was used to determine the number of B sites on B3 and Bx red cells.

H-deficient blood groups of Reunion island. II. Differences between Indians (Bombay Phenotype) and whites (Reunion phenotype).

Two variants of recessive, H-deficient nonsecretor individuals (h/h, se/se) were identified on Reunion Island: (1) H-negative individuals corresponding to the classical Bombay phenotypes (OhO, OhA, OhB, OhAB) who lack completely the H antigen on their red cells; all of them were Indian and had strong anti-H antibodies reacting with normal O and Oh red cells from whites; and (2) H-weak individuals (Oh, Ah, Bh, ABh). This phenotype represented the majority (85%) of the H-deficient phenotypes on Reunion Island, and all of them were white. They had only a weak expression of the H antigen and showed small but detectable amounts of ABH antigens on their red cells. Their anti-H antibodies reacted with normal O erythrocytes, but failed to react with Oh red cells, regardless of the ethnic origin of the donor. They were all from the same geographical area on the Island (Cilaos) and showed homogeneous titers of anti-H antibodies in sera. We propose to call this particular variant of weak H phenotype, belonging to the so-called para-Bombay series, Reunion.

Pj variant, a new hybrid MNSs glycoprotein of the human red-cell membrane.

An unusual glycoprotein variant (Pj) was found inherited through a caucasian family exhibiting atypical N and Nvg blood-group reactivities. Pj erythrocytes are blood-group-MS homozygous and have a normal sialic acid content. On sodium dodecyl sulphate/polyacrylamide-gel electrophoresis the variant contains a new component Pj of 24kDa apparent molecular mass in the monomeric state which is sharply stained by periodic acid/Schiff reagent. Both blood-group-MN (alpha) and -Ss (delta) glycoproteins were present. Homodimers (Pj2) as well as heterodimers with MN-glycoprotein (alpha Pj) and the Ss-glycoprotein (delta Pj) were also identified. The new sialoglycoprotein Pj is trypsin- and chymotrypsin-resistant in situ and carries N- and Nvg- but not M- and S-reactivities. The Pj component is labelled by lactoperoxidase-catalysed radioiodination. A 3H label is also easily introduced into the sialic acid or the galactose and galactosamine of the Pj glycoprotein. It is proposed that the Pj is a hybrid glycoprotein containing the N-terminal end of delta-glycoprotein and the C-terminal end of the alpha-glycoprotein. This proposal is supported by the finding that Pj carries a leucine residue at its N-terminus and is not immunoprecipitated by a monoclonal mouse antibody (R18) reacting specifically with the external domain of glycoprotein alpha. The red cells from the proposita Pj were found positive for a very low frequency MN antigen named Sta.

[Sensitivity of antigen Y ta to proteolytic enzyme].

The study of the agglutinability of Yt(a+) red cells by several examples of anti-Yta showed that Yta antigen is destroyed by papaïn, chymotrypsin, ficin, bromelin. Neuraminidase and trypsin are not active.

Atypical transmission of ABO blood groups in a French family.

A group AB mother (Mrs P.D.) gave birth to a group O female baby (C.D.). Extensive study of the blood group genetic markers in both the parents and the child, carried out on several occasions, showed nothing unusual outside the ABO system. Mrs P.D. then, gave birth to a second female baby who was also group O. Mrs P.D. had normal amounts of A, B, H and Lewis antigens in her saliva. The H, A and B agglutinability of her red cells was in the range of normal A2B group. This A2B blood group was characterized by very low A gene-specified glycosyltransferase activity in serum. Moreover this activity was undetectable in red blood cell membranes. These results are discussed in the light of various hypotheses in order to explain this unusual transmission of ABO blood group.

[Effects of a gonadotropic hormone of teleost fish on the adenylate cyclase of rat ovary and on its stimulation by ovine gonadotropins].

Carp gonadotropin (cGTH) stimulated adenylate cyclase from 20-24 day old (but not 16-18 day old) rat ovaries; its subunits were inactive. Carp GTH (but not its subunits) increased the apparent KD for oFSH but not for oLH. These results suggest that cGTH mainly interacts with FSH sensitive adenylate cyclase-receptor systems, with an efficiency about 6 X 10(3) times lower than oFSH itself.

Study of granulocyte-macrophage progenitor (CFUc) preservation after slow freezing of bone marrow in the gas phase of liquid nitrogen.

Storage of human hematopoietic stem cells has been made possible through effective preservation of viability by freezing technique. We have studied the quantitative and qualitative aspects of granulocyte-macrophage precursor (CFUc) recovery from 29 bags of frozen bone marrow stored at - 160 degree C in the gas phase of liquid nitrogen for several months or years. it has been shown that with the freezing and preservation technique used, over 75% of the proliferative capacity of the cryopreserved CFUc was recovered. The influence of various factors on the quality of bone marrow preservation was studied and showed that dimethylsulfoxide (DMSO) appeared to cause substantial loss of CFUc at 4 degree C. Evaluation of CFUc in vitro is essential for determining the quality and richness of cryopreserved bone marrow with a view to using such marrow for autologous grafting intensive chemotherapy or total body irradiation in patients with hematological malignancies or solid tumors. No correlation was found between the number of nucleated bone marrow cells and CFUc content.

[Subunits of lutropins: an intrinsic gonadotropic activity of alpha and the role of beta in the determination of the zoological specificity of action].

Pieces of Eel ovary were incubated in presence of various gonadotropic hormones or their subunits (alpha, beta) and cyclic adenosine monophosphate tissue concentration was measured. Carp gonadotropin (c-GTH) and a hybrid (bovine lutropin alpha-c-GTH beta) exhibited a very similar, intense, dose-dependent activity in the range 0.002-1 microgram/ml. Mammalian lutropins (LH) and their subunits became active in the range 1-100 microgram/ml: a strong stimulation (#30X) was attained with bovine LH alpha whereas the maximal stimulation obtained with bovine or ovine LH was #3X. This the activity of LH alpha cannot be due to a contamination by the hormone and it is intrinsic. The comparison of c-GTH, LH and hybrid actions indicate that the beta subunit plays a major role in determining the zoological specificity of action of the dimer.
