Polymorphism of HLA class I genes after restriction by endonucleases Eco RI, Eco RV and Hind III.

After electrophoresis and transfer onto a hybridization membrane, the restriction endonuclease fragments obtained were probed with a cDNA carrying the nucleotide sequence encoding a class I HLA gene. Polymorphism for presence or absence of various Eco RI and Eco RV and Hind III fragments was noted in a panel of unrelated Parisian people: frequent polymorphism was described for 2 Eco RI restriction fragments of sizes 16.5 and 7.7 kb, for 5 Eco RV restriction fragments of sizes 25, 20, 11.5, 7.8 and 4 kb, and for 3 Hind III restriction fragments of sizes 27.5, 23.5 and 4.7 kb. The French population studied is in Hardy-Weinberg equilibrium for the different variants described.

[Demonstration of specific gonadotropin binding sites of chum salmon (Oncorhynchus keta) in membrane preparations of granulosa cells from the ovary of amago salmon (Oncorhynchus rhodurus)].

During late vitellogenesis, radioactive iodine labelled Salmon gonadotropin binds to theca--or granulosa--membrane preparations from amago Salmon ovary. In granulosa, native hormone inhibits iodinated GTH binding, thus indicating a specific binding (70% of total binding). In theca, a lower specific binding is found while non specific binding is higher: contamination by granulosa cells, on one hand, the possible heterogeneity of theca, on the other, could account for these results.

A monoclonal antibody directed against the homologous N-terminal domain of glycophorin A and B.

A monoclonal antibody (iB5, IgG1 kappa) reacting with human red cells was produced after immunization of BALB/C mice with cord red cells, followed by fusion of the spleen cells with the murine myeloma cell line Ag 8-653. The monoclonal antibody agglutinated blood group N+ much better than M + N-red cells but did not recognize erythrocytes from rare individuals typed as M + N-S-s-U- and those from an En(a-) individual (M.E.P.). However, S-s-U- donors typed as M + N + or M-N+ and En(a-) red cells from donor G.W. were agglutinated. The erythrocyte receptors for iB5 are completely destroyed by papain treatment and significantly decreased by neuraminidase. Interestingly also, the iB5 antibody failed to agglutinate trypsin-treated N+M-S-s-U- erythrocytes. Other investigations have shown that the monoclonal antibody precipitated glycophorin A and B from N+ red cells and only glycophorin B from M+N-erythrocytes. The reactivity of iB5 was further explored by immunostaining following the electrophoretic transfer to nitrocellulose sheets of membrane proteins from common (M and N) and rare erythrocytes [En(a-),S-s-U-, MgMg, McM, St(a+), Mi.V, Mi.III, Tn] separated by SDS-polyacrylamide gel electrophoresis. These studies have clearly demonstrated that the monoclonal iB5 antibody is directed against the homologous N-terminal domain of glycophorin A and B, a specificity which explains the serological reactivity of iB5 against common and rare erythrocytes.

[Non-Hodgkin's malignant lymphoma. Therapeutic value of autologous bone marrow transplantation].

Twelve patients with non-Hodgkin malignant lymphoma of poor prognosis were treated with heavy chemotherapy of the TACC type (cyclophosphamide 45 mg/kg/day i.v. X 4; cytosine arabinoside 200 mg/m2/12 hours i.v. X 7; 6-thioguanidine 100 mg/m2/12-hourly p.o X 7 and CCNU 200 or 250 mg/m2 p.o. single dose) followed by autologus bone marrow transplantation (853 to 20.000 CFUc/kg). The patients were divided into 2 groups depending on whether they received an induction treatment for large visible tumoral mass (group I: 3 initial presentations, 3 relapses) or a consolidation treatment for small residual tumour (group II: 6 complete and 1 partial remissions). The results show that autologous bone marrow transplantation shortens the duration of the therapeutic aplasia. White cell (greater than 10(9)/l) and platelet (greater than 50.10(9)/l) recovery was observed on days 12 (range 9-19) and 14 (range 8-27) respectively. In group I, 1 patient died of myocardial TACC toxity and acute renal failure on tumoral kidney; there were 2 failures and 3 complete remissions (8, 21, 45 + months). Remissions occurred in patients treated initially; the overall survival since diagnosis was 48+, 48+ and 60+ months. In group II patients there were 1 failure and 5 complete remissions persisting after a 2+ months to 30+ months follow-up; the overall survival was 23+, 24+, 27+, 42+ and 70+ months. The 3 failures in the series occurred in circumstances suggesting contamination of the cryopreserved bone marrow by tumoral cells. The toxicity, largely due to infection, of the TACC-bone marrow transplantation combination was tolerable. It was clearly lower in group II (6 patients, no septicaemia) than in group I (5/6 patients with septicaemia). These preliminary results confirm that there is room for autologous bone marrow transplantation in highly malignant non-Hodgkin lymphomas, particularly during complete remissions to facilitate the use of an aggressive consolidation chemotherapy.

Cytotoxicity of anti-PP1Pk antibodies and possible relationship with early abortions of p mothers.

IgG and IgM anti-PP1Pk antibodies of human sera from p individuals were examined for their ability to agglutinate red blood cells (RBC) of various P phenotypes and to mediate ADCC of these RBC by human effector cells. Agglutinating antibodies were found in either the IgG or the IgM fraction, while ADCC active antibodies were found mainly in the IgG fraction and were essentially cytotoxic with Pk RBC. The possible relationship of these antibodies with early abortions of p mothers is discussed.

Acquired B antigen: further studies using synthetic oligosaccharides.

The reactivity of acquired-B red cells with various antisera has been investigated by agglutination inhibition assays using four trisaccharides obtained by chemical synthesis. Two of these had the structure GalNAc alpha 1-3 (LFuc alpha 1-2). Gal and Gal alpha 1-3 (LFuc alpha 1-2) Gal which are characteristic of the A and B determinants respectively and were indeed strong inhibitors of human anti-A and -B antibodies. The other two sugars denoted B-OAc and B-NH2 are derivatives of the B-trisaccharide by substitution of the hydroxyl group on carbon-2 of the alpha-galactose residue with the O-acetyl group (B-OAc) or the amino group (B-NH2) respectively. We have shown that both B and B-NH2 trisaccharides inhibited strongly the agglutination of acquired B red cells by the anti-B reagents (crude or affinity purified) whereas sera containing "anti-acquired B" agglutinins were specifically inhibited by B-NH2 but not by the A, B or B-OAc structures. We have also shown that the agglutination of Tk-activated erythrocytes by the BS-II lectin is specifically inhibited by N-acetylglucosamine but not by B, B-OAc or B-NH2 structures. These results and the observation that anti-acquired B agglutinins cannot be adsorbed on Tk red cells suggest that (i) the "B-like" and the "acquired-B" determinants share a common structure best represented by B-NH2 and (ii) Tk and "acquired-B" antigens are not identical.

An analysis of the blood group composition of a population in Brittany, the Bigoudens.

Data are presented on the blood groups of the Bigoudens. Deviations from the expected Hardy-Weinberg distribution are interpreted as the result of a recent mixing of previously separated groups, in addition to the effect of silent alleles and inbreeding. Special reference to the historical evidence of their origin is made.

[Establishment of a DNA bank of human origin].

High molecular weight DNA extracted from 50 unrelated parisian individuals constitutes a "DNA-thèque" which can be used at the population level. This procedure allows preparation from a variety of tissues such as liver, placenta and leucocytes. The "quality" of the DNA obtained (about 50 Kb, RNA and protein free) and the efficiency of the method (in terms of the yield of DNA obtained) are considered. Blood is the easiest material to obtain and permits the obtaining of a reasonable amount of human and primate DNA, particularly when familial investigation is planned for polymorphism studies. One of the objects of this article is to encourage others to make use of our bank, for studies in population and evolutionary genetics.

The incidence of Gm and Km allotypes in a group of Bretons, the Bigoudens.

Immunoglobulin allotypes were studied in a peculiar Breton community, the Bigoudens. Results showed that the incidence of the common Gm and Km phenotypes fell into the ranges quoted for Caucasian populations, except for the Gm1;..;5,10,11,13,14 and Gm1,2;..;5,10,11,13,14. The frequencies were found to be significantly different from those of non-Bigouden Breton neighbouring controls.

[Demonstration, in a teleost fish, of the phenomenon of desensitization of the ovary to gonadotropin].

The in vitro maximal stimulation by gonadotropin of cAMP accumulation in the Eel ovary is three times less in in vivo treated Fish (75 micrograms gonadotropin 18h before killing) than in controls. The existence of this phenomenon of desensitization should be taken into account for an understanding of the mechanisms of oogenesis regulation as well as for the planning of the gonadotropic treatments applied to Fish in culture.

T cell subpopulations defined by monoclonal antibodies in autoimmune hemolytic anemia.

T cell subsets were studied using monoclonal anti-T cell antibodies in 10 patients with IgM cold agglutinins and 30 patients with IgG warm autoantibodies. Two of the 10 patients with cold agglutinin disease had abnormally low helper/suppressor T cell ratio. In the 30 patients with IgG warm autoantibodies this ratio was abnormally low in 7 and abnormally high in 7 other patients. Treatment by steroids or immunosuppressive agents tended to decrease OKT4/OKT8 ratio since high ratios were essentially found in untreated patients. This report documents the high incidence of T cell imbalance in autoimmune hemolytic anemia due to IgG warm autoantibodies and comments on its significance in the light of the great heterogeneity of this syndrome.
