[A new method for serologic detection of Treponema pallidum reinfection].

Sources of error in the detection of specific IgM-antibodies against Treponema pallidum could be overcome by the introduction of a new method for precise separation of immunoglobulins. The 19-S-IgM-FTA-ABS-test apparently permits a correct diagnosis of syphilitic reinfection. This method is not only specific but also earlier reactive than the VDRL-test. The short-time persistence of 19 S-IgM production after treatment and its renewed appearance enables for the first time a differentiation between treatment failure and reinfection.

[Practical significance of syphilis serodiagnosis].

Recent developments in syphilis serology are set down. The new specific tests for treponema pallidum are described. Reactivity in different stages of the disease including congenital and neurosyphilis is discussed. An outline is given of the possibilities of the IgM-FTA-ABS and 19 S IgM-FTA-ABS tests. Syphilis serology today requires only 3 tests: a screening procedure, where the specific AMHA-TP is the method of choice, FTA-ABS test should be used for confirmation, and the quantitative VDRL in order to follow up the effectiveness of treatment.

The major fraction of deoxyribonuclease activity from human urinary proteins. Purification and properties.

The major fraction of deoxyribonuclease activity from human urinary protein was purified 40-fold in about 14% yield. The enzyme shows an isoelectric point at pH 4.2 and has a molecular weight of 33,600+/-3,000. Optimum activity was shown at pH 6.8 in the presence of 12.5 mmol/l Mg2+ plus 1 mmol/l Ca2+. The enzymic reaction is inhibited by high ionic strength (greater than 300 mmol/l Na+). The purified enzyme readily hydrolyzes native DNA to oligodeoxyribonucleotides with an average chain length of 5.3+/-0.2 after exhaustive digestion. Therefore, this endonuclease may be designated as neutral deoxyribonuclease (deoxyribonucleate oligonucleotidohydrolase, EC 3.1.4.5).

Quantitative evaluation of the FTA-ABS-IgM and VDRL test in treated and untreated syphilis.

Observations made on fluorescent treponemal antibody absorption (FTA-ABS) immunoglobulin M (IgM) titres in patients with untreated early syphilis showed non-reactive or weakly reactive results in patients with primary (one of five cases) and secondary (two of 16 cases) lesions. In patients with primary (19.5%) and secondary (15%) syphilis sera remained reactive with increased titres for more than one year after treatment. The respective figures in the results of the Venereal Disease Research Laboratory (VDRL) test were zero in primary and 20% in secondary syphilis. The non-reactive FTA-ABS IgM results may possibly be explained by competitive inhibition of IgM by immunoglobulin G (IgG). The persistence of reactivity in a comparable percentage has been observed by other investigators (Grin et al., 1974; Wilkinson and Rodin, 1976). The current results therefore suggest that FTA-ABS IgM titres are less reliable for assessing the effect of treatment than the course of the VDRL titres. The phenomenon of a decrease in FTA-ABS IgM titres soon after treatment with a later rise before final non-reactivity is a matter for further investigation.

Interaction of oxidized and reduced N-methylphenazonium methosulfate (PMS) with photosystem II.

In 3-(3,4-dicholorophenyl)-1,1-dimethylurea (DCMU) poisoned chloroplasts, the restoration of the fluorescence induction is presumed to be due to a back reaction of the reduced primary acceptor (Q-) and the oxidized primary donor (Z+) of Photosystem II. Carbonylcyanide m-chlorophenylhydrazone (CCCP) is known to inhibit this back reaction. The influence of reduced N-methylphenazonium methosulfate (PMS) in the absence of CCCP and of oxidized PMS in the presence of CCCP on the back reaction was investigated and the following results were obtained: (1) Reduced PMS at the concentration of 1 muM inhibits the back reaction as effectively as hydroxylamine, suggesting an electron donating function of reduced PMS for System II. (2) The inhibition of the back reactionby CCCP is regenerated to a high degree by oxidized PMS which led to assume a cyclic System II electron flow catalysed by PMS. (3) At concentrations of reduced PMS higher than 1 muM it is shown that both the fast initial emission and more significantly the variable emission are quenched.

[Occurrence and frequency of orbital tumors in ethiopia (author's transl)].

Orbital tumors are more frequent in Africa than in Europe. Children are more often affected than adults. The most frequently observed tumors are the epithelioma of the conjunctiva in adults, and the retinoblastoma and Burkitt's lymphoma in children. Melanoblastomas and lid basal cell carcinomas are fairly rare. Findings from Ethiopia are demonstrated.

[Hamostasiological changes induced by platelet-aggregation-inhibitors, tested in children with congenital heart disease (in vitro and in vitro examinations) (author's transl)].

This report has delt with in vivo and in vitro examinations in the cases of sixteen children suffering from cyanotic congenital heart disease, and who had been placed under extended therapy with platelet-aggregation-inhibitors. The pharmacological effect of acetylsalicylic acid and dipyridamol has been discussed--as far as it is known. The discrepancy between in vivo and in vitro therapy-effect in the treatment with platelet-aggregation-inhibitors has been subjected to criticism. Consistant treatment of all children suffering from cyanotic congenital heart disease with aspirine and persantine has been advocated, however should only be carried out in collaboration with a competent hamostasiological laboratory.

The reabsorption of creatinine from the rabbit bladder.

Using a radioisotope technique the reabsorption of creatinine from normal, inflamed and chronically obstructed bladders of rabbits was investigated. The reabsorption of creatinine from normal bladders was minimal. Chronic obstruction lead to a rise of the reabsorption rate. The most marked reabsorption however was found with the inflamed bladders. This difference of creatinine reabsorption is statistically significant and it was detectable in the 14C-creatinine content of the blood, 14C-creatinine content of the renal pelvis urine and in the activity loss in the bladder urine. The vesical reabsorption of creatinine corresponds in principle with the urea reabsorption which was investigated earlier by the same method. The extent of reabsorption is however different and urea is reabsorbed to a substantially greater extent.

A 3'-deoxymononucleotide-producing nuclease from the marine sponge Verongia aerophoba. I. Purification.

A four-step procedure for purification of a nuclease from the keratinous sponge Verongia aerophoba is described. The extracted material is lyophilized, acidified, and subjected to chromatography on Sephadex, hydroxyapatite, and phosphocellulose. The nuclease is purified about 1,000-fold from the crude extract and approximately 1,600-fold from concomitant acid RNase. Phosphodiesterase is lost after chromatography on Sephadex. The purified enzyme solution contains one single activity as determined by polyacrylamide gel electrophoresis.
