Inhibition of antibody-dependent cellular cytotoxicity (ADCC) by normal and pathological sera from patients with advanced lung cancer.

An improved ADCC assay for the detection of cancer-associated serum blocking factors is described. Inhibition of ADCC by sera was performed without preincubation and washing of effector cells. Effects of individual susceptibility of normal effector cells to serum inhibitors and of the background inhibition by normal sera were also avoided. Based on their inhibitory effect on ADCC, normal and tumour sera from lung cancer patients could be distinguished with 81.4 to 100% accuracy, according to effector cell donors and to concentrations of the sera tested.

[Differences in the levels of serum immunoglobulin E determined by the competitive radio-immuno-sorbent and the paper-radioimmuno-sorbent methods: Removal of these differences by pretreatment of sera with polyethylene glycol (author's transl)].

The serum IgE of patients with chronic nonspecific lung diseases was determined with the competitive radio-immuno-sorbent and the paper-radio-immuno-sorbent methods (Phadebas). Some samples showed markedly higher values with the competitive radio-immuno-sorbent than with the paper-radio-immuno-sorbent method. These discrepancies were abolished by pretreatment of the sera: polyethylene glycol 6000 (final concentration 35 g/l) was added, followed by centrifugation at 0 degrees C for 30 min at 20 000 g, and IgE was determined in the supernatant. This treatment had no effect on the values of the paper-radio-immuno-sorbent method, but the values of the competitive radio-immuno-sorbent method were decreased so that the two values agreed within the experimental confidence limits.

Effect of changes in cyclic nucleotide levels on human antibody-dependent cellular cytotoxicity.

The effect of agents influencing intracellular cyclic nucleotide levels and that of exogenous cAMP was studied on the cytotoxicity of normal human peripheral blood mononuclear cells on antibody coated human and chicken erythrocytes. Elevation of the cAMP level resulted in a marked decrease in cytotoxicity, while basically similar in the allogeneic and the xenogeneic test systems, but more expressed in the allogeneic system.

Antibody dependent cellular cytotoxicity of lymphocytes in patients with renal diseases of immunopathological origin.

Antibody dependent cytotoxicity (ADCC) of lymphocytes from twenty-two patients with histologically proven chronic glomerulonephritis and eight patients with SLE nephropathy was studied in a xenogeneic assay on chicken red blood cells and in an allogeneic test system using Rh(+) human erythrocyte targets. ADCC activity of the lymphocytes of patients with chronic glomerulonephritis did not differ from that of normals, while all patients with SLE nephropathy showed significantly lower cytotoxicity than healthy controls. The possible role ADCC in the pathogenesis of renal diseases of immunopathological origin is discussed.

Effect of dialysable leukocytes extracts on antibody-dependent cellular cytotoxicity and lymphocyte Fc receptors.

Dialysable leukocyte extracts significantly inhibited the antibody-dependent cell-mediated cytotoxicity of normal human peripheral blood lymphocytes in both xenogeneic and allogenic assays. The inhibitory effect was dose-dependent. Fractions of dialysable leukocyte extracts separated on Sephadex G-25 column differed in their capacity to suppress cytotoxicity. Dialysable leukocyte extracts did not block EA-rosetting but inhibited the resynthesis of Fc receptors. Some explanations for the observed suppressive effect are discussed.

Separation of beta2-microglobulin from transfer factor in dialyzable leukocyte extracts.

Dialyzable leukocyte extracts prepared according to the original method of LAWRENCE contain 4 X 10(4) molecules of beta2-microglobulin per lymphocyte equivalent. The negative skin test converting biological activity (transfer factor) is separable by means of Sephadex G-25 gel chromatography from the beta2-microglobulin component of the extracts. This finding does not support the hypothesis of SHIFRINE and SCIBIENSKI on beta2-microglobulin being the nonspecific anchor of specific transfer factor to nonsensitized lymphocytes.

Demonstration of nuclear antigen binding lymphocytes by the resette method in SLE-patients.

A rosette method suitable for the demonstration of deoxyribonucleic acid and deoxyribonucleoprotein-binding peripheral lymphocytes is described. The results confirmed the existance of a human peripheral lymphocyte subpopulation capable of binding nuclear antigen specifically. The subpopulation was demonstrated in 63 out of 75 SLE-patients; 57 out of 59 SLE-patients being in the active period of the disease revealed nuclear antigenbinding lymphocytes, as against 6 our of 16 patients being in remission.

In vitro effect of transfer factor on active rosettes and leucocyte migration of patients with cancer.

The in vitro effect of transfer factor (TF) and its obtained by gel filtration was studied on the active rosettes and leucocyte migration inhibition. TF and one fraction (IV) restored the reactivity of lymphocytes from tumour-bearing patients to tuberculin and Candida antigens in the leucocyte migration assay. TF and fraction IV was also found to enhance the number of active rosettes of tumour-bearing patients, whereas no such effect was observed on lymphocytes of patients with active SLE or controls.

A simple allogeneic test system for the study of antibody-dependent cellular cytotoxicity (ADCC) in man.

A simple allogeneic test system for the study of ADCC activity of human mononuclear peripheral blood leucocytes (MPBL) is presented. The test uses human Rh-positive erythrocytes as target cells coated with specific human IgG antibody. The effector function of human MPBL is demonstrated after depletion of most of the monocytes and B lymphocytes on nylon wool column. The test system seems to be useful in the clinical study of ADCC activity in man.

In vitro effect of immune complex positive human sera on antibody dependent cellular cytotoxicity.

The effect of immune complex positive and negative (control) sera on the antibody dependent cytotoxicity of normal human lymphocytes was studied in a xenogeneic assay in vitro. Significant inhibitions were obtained with the sera of immune complex positive patients as compared with age and sex matched controls. The possible significance of these findings in autoimmune diseases and malignancies is discussed.
