Effect of peripheral blood mononuclear cells from aplastic anemia patients on the granulocyte-macrophage and erythroid colony formation in samples from normal human bone marrow in vitro--a cooperative work.

Thirty-six patients with aplastic anemia and three with aplastic anemia-PNH syndrome in eight institutes were studied for the presence of peripheral blood mononuclear cells having a suppressive effect on granulocyte-macrophage (G-M) and erythroid colony formation using a uniform protocol. In 11 cases of 29 (38%) and 9 cases of 29 (31%), the presence of mononuclear cells with a suppressive effect on G-M and erythroid colony formation, respectively, was demonstrated. However, the presence of mononuclear cells suppressive both to G-M and erythroid colony formation was demonstrated only in 3 of 19 cases (16%).

[A radioimmunoassay for plasma parathyroid hormone (PTH) using N-terminal PTH antiserum (author's transl)].

In order to investigate plasma bioactive PTH, we tried to assay the N-terminal portion of PTH by RIA. The antiserum to PTH was prepared by immunizing rabbits with a bovine 1-34 PTH conjugate BSA. A preparation of labeled PTH was radioiodinated by the chloramine-T or lactoperoxidase method. Labeled PTH was purified by means of adsorption by Quso G-32 powder or a sephadex G-50. The separation of the free and bound labeled hormone was performed by the dextran-coated charcoal method. The assay was carried out as follows: 0.2 ml diluted buffer (0.05 M, pH 8.6, veronal buffer), 0.1 ml standard PTH or sample to be tested, and 0.1 ml anti-PTH serum were mixed. After the first incubation at 4 degrees C for 4 days, 0.1 ml labeled PTH were added. After a second incubation at 4 degrees C for 12 hours, the assay tubes were centrifuged at 2,000 rpm for 30 min and the precipitates were counted. Various hypothalamic, pituitary and thyroid hormones did not interfere with the RIA for PTH. A dose response curve was obtained in a range from 100 pg to 5,000 pg per ml of standard PTH in this assay system. The serum immunoreactive PTH in healthy subjects values less than 290 pg per ml.

Reflex vasoconstriction to a cold stimulus for non-invasive evaluation of neurovascular function in man.

The response of limb blood flow to cold stimulus was determined by venous occlusion technique in 30 healthy subjects. The stress was applied by immersing one hand into ice-floating water for 35 sec, and the blood flow was measured serially in the contralateral upper limb. The change of blood flow at the 15th sec of stress was the largest among the intermittent measurements, and decreased by 48+/-8% of the control value. A significant rise in plasma dopamine-beta-hydroxylase was found in response to the same stress. This simple cold stress test may be used to evaluate the function of the reflex arc involved in reflex vasoconstriction. A significantly diminished vasoconstriction was observed in 12 uremic patients with a concomitantly smaller rise of plasma dopamine-beta-hydroxylase activity in the test.

Neurotensin--positive and somatostatin--positive cells in the canine gut.

By using immunoperoxidase and immunofluorescence techniques, the localization of neurotensin-positive cells and somatostatin-positive cells in the canine gut was examined on the same sections. Neurotensin-positive cells were found only in the jejunum and ileum, while somatostatin-positive cells were distributed throughout the stomach and all parts of the small intestine. These two types of cell in the jejunum and the ileum had no direct cellular contact with each other. Based on the hypothesis that somatostatin may inhibit the release of some peptide hormones through junctional complexes of cells, the possibility of functional interaction between neurotensin and somatostatin was discussed.

Does somatostatin in each organ act specifically on that particular organ?

Rats with hypercalcemia induced by injection of vitamin D2 had a decreased thyroid somatostatin content, whereas the somatostatin content in their pancreas was almost within the normal range. This suggests that somatostatin in different organs acts specifically on each particular organ as a local hormone or hormone-like substance.

Angiotensin-like immunoreactivity in the rat submaxillary gland.

Rat salivary glands were examined by an indirect immunofluorescence technique using anti-angiotensin II antiserum. Cells reacting with the antiserum were found in the convoluted granular tubules of the submaxillary gland. In view of the previous reports that renin was isolated from the rat submaxillary gland and that cells with a renin-like immunoreactivity were demonstrated in the convoluted granular tubules of the gland, the present study suggests that a set of substances forming the renin-angiotensin system may be present in this gland.

Effect of acetate upon arterial gases.

Following infusion of sodium acetate, a significant fall in arterial oxygen tension (PO2) was observed (p less than 0.001). A rise in PCO2 or bicarbonate was also statistically significant. pH was however remained unchanged. Though a rise in PCO2, bicarbonate, pH was observed following infusion of sodium bicarbonate, no significant fall in PO2 was observed. These results suggest a role of acetate ion for the fall of arterial oxygen tension; the hemodialysis-induced hypoxemia.

Changes of prekallikrein in the cases with disseminated intravascular coagulation syndrome.

Changes of prekallikrein in the cases with DIC were investigated, i.e., DIC cases including disseminated metastasis of gastric cancer, acute promyelocytic leukemia and endotoxin shock. Therefore, the trigger substances for this paper were the pathologic cells of the leukemia, the cultured well differentiated adenocarcinoma cells and endotoxin. (1) The lysates of the pathologic cells of the leukemia and the cultured cells showed prekallikrein activation. Endotoxin showed prekallikrein activation via factor XII. (2) Serine proteases (factor Xa, thrombin, plasmin and trypsin) activated prekallikrein in the plasma and the purified prekallikrein. (3) Antithrombin III, aprotinin and FOY inhibited prekallikrein activation. Antithrombin III was promoted by heparin in its inhibitory effect.
