Sources of extramitochondrial corticoidogenic cholesterol in the adrenal cortex.

Intracellular sources of extramitochondrial corticoidogenic cholesterol in bovine, rat and hamster adrenocortical cells were examined in vitro by comparing the species differences in the effects of various inhibitors on the adrenocorticotropic hormone (ACTH)-induced corticoidogenesis. The inhibitors were ML-236B (3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitor), W-7 (N-(6-aminohexyl)-5-chloro-1-naphthalene sulfonamide; calmodulin inhibitor), dichlorvos (O,O-dimethyl-2,2-dichlorovinyl phosphate; organic phosphorylation inhibitor), chloroquine [7-chloro-4-4-diethylamino-1-methyl-butylamino) quinoline; lysosomal enzyme inhibitor) and cycloheximide (protein synthesis inhibitor). During 2 to 3 hr incubation periods, the ACTH-induced corticoidogenesis was not inhibited by ML-236B (100 microM) in the bovine and rat adrenocortical cells. In the hamster adrenocortical cells, ML-236B (100 microM) did not affect the ACTH-induced corticoidogenesis during the initial 1 hr incubation periods; but thereafter, the ACTH-induced corticoidogenesis during the subsequent 2 hr incubation periods was completely blocked by ML-236B. The ACTH-induced corticoidogenesis was inhibited by W-7 (up to 25 microM) in the bovine and rat adrenocortical cells, but this was not the case in the hamster cells. Chloroquine (up to 400 microM) inhibited the ACTH-induced corticoidogenesis in the adrenocortical cells of three different species, but the hamster adrenal cells were much more vulnerable than the bovine and rat cells. The ACTH-induced corticoidogenesis in the adrenocortical cells of three different species were equally inhibited by cycloheximide (up to 1 mM).(ABSTRACT TRUNCATED AT 250 WORDS)

[Early invasive squamous cell carcinoma of the tongue--report of a case].

A 65-year-old woman with early invasive squamous cell carcinoma of the tongue is reported. Atypical but not definitely malignant epithelium was detected by cytology, although the gross appearance of the glossal lesion looked like early invasive squamous cell carcinoma. On the surgical material, the basal and para-basal cell layers were occupied by cancer, but the upper portion of the epidermis from the prickle cell layer through the surface of the mucosa was histologically benign. In such cases, to obtain the correct diagnosis on cytology, repeated examinations as well as using a sharp instrument like a curatte are necessary.

Endocytosis of human thyroglobulin (TG) in adenomatous goiter studied by an immuno-electron microscopic procedure.

Five cases of adenomatous goiter have been studied by an electron microscope using an immuno-reaction for thyroglobulin (TG) and focusing on the mechanism of endocytosis. Positive stain for TG was demonstrated in follicular lumina, large reabsorbed colloid droplets and small subapical vesicles. Endocytotic vesicles ranging from 320 nm to 1600 nm in diameter were observed in the cytoplasm as pits in the apical plasma membrane. Some of them showed direct connection with the positive stain for TG in the follicular lumen and the others were completely ingested in the cytoplasm. With statistic analysis, a majority of the vesicles showing the positive stain for TG in the cytoplasm distributed in the range of 200 nm to 1200 nm in diameter with the peak in 300 nm to 399 nm and was situated within an extent of the diameter measured from the endocytotic vesicles. Engulfment of colloid by pseudopods and fusion of the reabsorbed colloid droplets were encountered as extremely rare findings and appeared to play no major role for formation of large colloid droplets in adenomatous goiter.

Light and electron microscopic study of nonfunctioning parathyroid carcinoma. Report of a case with a review of the literature.

A case of nonfunctioning parathyroid carcinoma in a 69-year-old female has been studied by light and electron microscopy. The tumor, located on the left side of the anterior neck, was well encapsulated by connective tissue but showed invasion to the capsule and to the thyroid. The tumor cells exhibited a trabecular arrangement surrounded by capillary networks but focally showed several ductal structures. They were polygonal in shape, had a large nucleus showing frequent mitosis and poor cytoplasm containing glycogen. Some tumor cells had clear and abundant cytoplasm, and resembled water-clear cells of the parathyroid. Immunohistochemically, no thyroglobulin was demonstrated in the tumor tissue. Electronmicroscopically, the tumor cells with high N/C ratio contained poorly developed cell organelles and abundant glycogen particles. They were poor in secretory granules and had no conglomeration of lipid. Desmosomes and tonofibrils were observed. The ratio of the reported number of nonfunctioning parathyroid carcinoma to that of functioning one in Japan was compared with that in western countries. No difference of the ratio was found between these two, when identical criteria were employed.

Ultrastructural localization of endogenous peroxidase in the human thyroid gland under normal and hyperfunctioning conditions.

Ultrastructural localization of human thyroid peroxidase in normal and hyperfunctioning states has been studied. Peroxidase activity was visualized ultrastructurally with a cytochemical reaction using 0.05% diaminobenzidine-tetrahydrochloride, and hydrogen peroxide with a final concentration of 0.0025%, 0.005%, or 0.01%, respectively, and 2% osmium tetroxide. Reaction product demonstrating the enzyme was clearly observed in cell organelles with a final concentration of hydrogen peroxide at 0.0025% or 0.005%. In the normal portion of thyroid, follicular cells demonstrated the reaction product, located mainly in rough-surfaced endoplasmic reticulum and perinuclear cisternae and in addition a small amount of the product was found in Golgi cisternae and small vesicles dispersed throughout the subapical cytoplasm. The reaction product significantly increased in amount in the cell organelles described above and in addition, a characteristic heavy deposition of the product was observed at the external surface of the microvilli in the follicular cells of thyroid obtained from the patients with treated Basedow's disease.

[Study on the concentration of FT-207 and 5-FU in serum, lymph nodes and tissues after administration of FT-207 suppositories].

In 21 cancer patients receiving administration of FT-207 suppositories, we studied the concentration of FT-207 and 5-FU in the blood before surgery, and in the blood and tissues-tumor, normal and lymph node-sampled during surgery. These 21 cases were comprised of 12 cases of stomach cancer, 1 case of lymphosarcoma, 4 cases of rectal cancer, 2 cases of sigmoid cancer and 2 cases of breast cancer. Two hours after administration, the concentration of 5-FU reached its highest level and continued to maintain a level at which it could be expected to bring about clinical results even after four hours. The concentration of 5-FU in tumors and lymph nodes was high, especially in tumors compared with normal tissue (p less than 0.01). No difference could be found in the concentrations of FT-207 and 5-FU in serum and tissues from each 12 cases of stomach cancer, not only between of the early cancer and advanced cancer but also between patients more than 60 years old and those under 60 years old. No side-effects occurred in any case during suppository administration.

Immunoelectron microscopic localization of thyroglobulin (TG) in the thyroid gland in Basedow's disease.

An electron microscopic immunohistochemical localization of thyroglobulin (TG) using PAP methods from Epok-812 embedded tissues has been made in human thyroids obtained from 15 patients with treated Basedow's disease aged 13 to 43 years. Follicular cells vary considerably in shape and content of cytoplasmic organelles depending mainly upon the size of follicles. Reaction product for TG coincided well with previous investigations studied by electron microscopic autoradiography and is strongly positive in most follicular lumina having no relation to their size, reabsorbed colloid droplets or small subapical vesicles. The reabsorbed colloid droplets are rarely observed in cells surrounding distended follicles and more frequently appeared in small follicles. The reabsorbed colloid droplets after fusion with lysosomes show a variable amount of reaction products from intense deposition to a complete absence of product indicating hydrolysis of TG by lysosomal enzymes. There is no reaction product in rough endoplasmic reticulum or Golgi apparatus in spite of positive reaction product in the subapical vesicles. These subapical vesicles are composed of both endocytotic and exocytotic vesicles.

Cervical thymoma and incidence of cervical thymus.

A case of cervical thymoma in a 51-year-old female is reported. The tumor was located on the left side of the anterior neck just beneath the left lower lobe of the thyroid gland and there was no connection to the mediastinal thymus gland. It was well encapsulated and lobulated on the cut-surface. Histologically, the tumor showed a mixed proliferation of lymphocytes and epithelial cells. Mitoses were frequently seen. Several tiny fragments of ectopic thymic tissue were found around the tumor. Cervical thymic tissue, considered to be the original site of the cervical thymoma, was studied and found macroscopically in 12 out of 657 patients with Basedow's disease (1.8%).

Immunoelectron microscopic localization of thyroglobulin in human follicular adenoma.

An electron microscopic immunohistochemical localization of thyroglobulin (TG) using PAP methods has been made in 15 cases of cold follicular adenoma. All cases of follicular adenoma showed organ specific functions such as synthesis, storage, reabsorption, and hydrolysis of thyroglobulin except for an area composed of follicular cells with trabecular arrangement. Immuno-reaction product for TG was precisely demonstrated in follicular lumina, subapical vesicles and reabsorbed colloid droplets. The reaction product observed in the follicular lumen was clearly demarcated from the cytoplasm of the follicular cells by the apical plasma membrane. The subapical vesicles ranging approximately from 50 millimicrons to 300 millimicrons in diameter were rarely observed in follicular adenoma and some of them fused with the reabsorbed colloid droplets. The reabsorbed colloid droplets usually had the intense reaction product and hydrolyzed colloid droplets had a vacuole containing floccular low electron dense materials. There is no reaction product in rough endoplasmic reticulum and Golgi complexes.

Normal ontogeny of alpha 1-adrenergic receptor in rat liver is thyroid hormone dependent.

Postnatal ontogeny of rat liver alpha 1-adrenergic receptor was examined using alpha 1-specific radioligand [3H]prazosin in control and propylthiouracil-treated congenital hypothyroid rats at various ages. Partially purified rat liver membranes prepared by the Neville method had 8-fold purification of 5'-nucleotidase from the crude homogenates from postnatal day 5 to adulthood. [3H]Prazosin binding was typical of an alpha 1-adrenergic receptor, and (-)epinephrine affinity for the [3H]prazosin-binding sites was not altered in the presence of 10(-5) M guanylyl-imidodiphosphate. The receptor density was lower in 5- and 15-day-old rats than in 28-day-old or older rats in both control and hypothyroid groups. (P less than 0.01). At 28-34 days of age, hypothyroid pups had significantly lower alpha 1-receptor density than controls (399 +/- 10 vs. 869 +/- 40 fmol mg protein-1; P less than 0.01). Replacement therapy with daily T4 injection from postnatal days 16-27 restored 54% of the deficit in PTU-treated hypothyroid pups at 28 days. The dissociation constant of [3H] prazosin did not change with advancing age or with different treatment and was consistent at 0.1 nM. These findings indicate that the normal ontogeny of plasma membrane alpha 1-adrenergic receptors is dependent upon thyroid hormone and matures postnatally in rat liver.

[Combination chemotherapy (BEAM regimen) for head and neck cancer].

Twenty-eight patients with head and neck cancer were treated with a combination of bleomycin, cyclophosphamide, adriamycin, and methotrexate (BEAM regimen), consisting cyclophosphamide 600 mg/body and adriamycin 40 mg/body on day 1, bleomycin 15 mg/body and methotrexate 20 mg/body on days 1 and 5. Dose modifications were performed according to patient's conditions. Of 18 evaluable patients, 3 patients showed a complete response and 3 patients showed partial response with an overall response rate of 33.3%. In this study, the patients who had already been treated with surgery, radiotherapy, or chemotherapy produced good results. The median duration of response was 4.0 months in the complete responders, and 1.0 month in the partial responders. The median survival was 12.7 months in the complete responders 1.4 months in the partial responders, and 5.6 months in non-responders. The most frequent major toxic effects were leukopenia (66.7%), nausea (60%), and alopecia (57.3%) in the patients receiving the maximum dose of the drugs. Careful monitoring of patients is imperative during this treatment.

Ontogeny of alpha 1-adrenergic receptors in the rat myocardium: effects of hypothyroidism.

The effects of propylthiouracil (PTU) treatment on alpha 1- and beta-adrenergic receptors in neonatal and adult rat myocardium were examined with the alpha 1-selective antagonist [3H]prazosin and beta-antagonist (-)-[3H]dihydroalprenolol ((-)-[3H]DHA). Pre and postnatal PTU treatment (congenital hypothyroid state) decreased ventricular size and protein content at 15 and at 28 days (P less than 0.01). Similarly PTU treatment after maturation (acquired hypothyroid state) is associated with decreased ventricular size and protein content (P less than 0.01). PTU treatment did not alter ventricular DNA content at any age examined as compared to euthyroid controls. Specific [3H]prazosin binding to ventricular membrane increased with advancing postnatal age in both congenital hypothyroid pups and controls, reaching peak receptor density at 15 days of postnatal age (106 +/- 8 vs. 151 +/- 7 fmol X mg-1 protein). The density at 15 and 28 days was however significantly lower in hypothyroid pups (P less than 0.01, P less than 0.02). The effect of congenital hypothyroidism on (-)-[3H]DHA binding was more marked which significantly decreased from the first postnatal day. Acquired hypothyroidism is also associated with a decreased density of alpha 1- and beta-adrenergic receptors in rat myocardium. We conclude that alpha 1- and beta-adrenergic receptors are modulated by thyroid hormone in developmental as well as mature stages of the rat.

Ontogeny of alpha 1- and beta-adrenergic receptors in rat lung.

The binding characteristics of the alpha 1-selective adrenergic ligand [3H]-prazosin were determined in particulate membranes of rat lung from day 18 of gestation to adulthood. Specific binding was present at all ages studied, was reversible and inhibition of specific binding by agonists followed the order of potency: (-)-epinephrine = (-)-norepinephrine much greater than (-)-isoproterenol greater than (+)-norepinephrine. Inhibition by antagonists followed the order of potency: prazosin greater than WB4101, much greater than yohimbine. Binding capacity increased during the neonatal period from 52 +/- 9 fmoles x mg-1 protein in lung preparations on day 18 of a 21 day gestation increasing to 105 +/- 4 fmoles x mg-1 protein (mean +/- SE) by postnatal day 15. Binding activity decreased thereafter, reaching adult levels by 28 days of postnatal age, 62 +/- 3 fmoles x mg-1 protein. This pattern of alpha 1-adrenergic receptor density was distinct from that of beta-adrenergic receptors identified in rat lung membrane with the beta- adrenergic antagonist, (-)-[3H]dihydroalprenolol ((-)-[3H]DHA). (-)-[3H]DHA binding increased dramatically during this same time period, from 46 +/- 4 fmoles x mg-1 protein on day 18 of gestation to 496 +/- 44 fmoles x mg-1 protein in the adult lung. Affinity for [3H]-prazosin and (-)-[3H]DHA did not change with age. Pulmonary alpha 1-adrenergic receptors are present as early as 18 days of gestation in the rat and alpha 1-adrenergic receptor density is maximal by 15 days of postnatal age. The timing of the changes in alpha 1-adrenergic receptors correlates with the timing of increased sympathetic innervation of the developing rat lung and is distinct from that of beta-adrenergic receptor sites.
