Human serum thyroglobulin determination with monoclonal antibody one-step assay: minimum interference of autoantibody.

Combination of two thyroglobulin monoclonal antibodies (monoAbs) recognizing epitopes which are rarely recognized by an antibody enabled us to develop a rapid one-step enzyme immunoassay of serum Tg. Of 87 monoclonal antibodies, 20 were selected for the purpose. The method is a sandwich technique employing a monoAb covering microplate and horse-radish peroxidase monoAb conjugate. A combination of monoAb 7A7A solid phase and 31A2E for the conjugate gave the best results. The assay takes 60 min and the minimal detectable amount is 2 ng/ml. Intraassay variation is from 4 to 7%. Interassay variation is 5 to 12%. The recovery rate for Tg added to normal sera is between 89 and 111%. The correlation coefficient with the polyclonal antibody method in Tg hemagglutination negative sera is 0.98. The presence of autoantibody in sera up to 10 X 2(4) hemagglutination titer does not affect the recovery rate to a statistically significant extent.

Detection of antibodies to Sendai virus in mouse sera by enzyme-linked immunosorbent assay (ELISA).

Enzyme-linked immunosorbent assay (ELISA) for diagnosis of Sendai virus infection in mice was evaluated. A large-scale survey of infected mice showed that ELISA is approximately 100 times more sensitive than the hemagglutination-inhibition or complement-fixation test. Although a few SPF mice showed false-positive reactions at a serum dilution of 1:40, further dilution to 1:80 eliminated the non-specific reaction. It was shown that ELISA is a highly satisfactory method for examination of Sendai virus infection in mice.

Comparative study of mitogenic responses in man and Japanese monkeys (Macaca fuscata): responses of T-cell subsets, accessory cell dependency, and interleukin-2 receptor expression.

Responses of peripheral blood mononuclear cells to phytohemagglutinin-P (PHA-P), concanavalin-A (ConA), and pokeweed mitogen (PWM) were compared in man and Japanese monkeys. Both CD8+ and CD8- T subsets showed greater responses to ConA than to PHA-P in the Japanese monkey. Addition of macrophages to each T subset produced more effective augmentation of ConA response in the Japanese monkey than in man, and ConA induced more interleukin-2-receptor-positive blast cells than PHA-P did in Japanese monkeys.

Specific diagnosis of hepatocellular carcinoma by delayed hepatobiliary imaging.

For assessment of the value of delayed hepatobiliary imaging with technetium 99m (99mTc)-(Sn)-N-pyridoxyl-5-methyltryptophan (99mTc-PMT) for specific diagnosis of hepatocellular carcinoma, 88 patients with various malignant and benign liver diseases (49 with hepatocellular carcinoma, 4 with cholangiocellular carcinoma, 10 with metastatic liver carcinoma, 2 with liver cysts, 2 with liver hemangioma, 1 with liver abscess, 2 with intrahepatic lithiasis, 12 with liver cirrhosis, and 6 with chronic hepatitis) were studied. In 20 (41%) of the 49 patients with hepatocellular carcinoma, greater uptake of 99mTc-PMT by the tumor than by the surrounding liver tissue was seen in delayed hepatobiliary images, whereas in eight patients (16%), equilibrated uptake was seen. No increased uptake of the radioisotope by hepatic lesions was seen in 21 patients with localized liver diseases other than hepatoma. Moreover, in 18 patients with diffuse liver diseases, no focal accumulation of the radioisotope was seen in delayed 99mTc-PMT images. In addition, of 28 patients with hepatocellular carcinoma in whom the serum alpha-fetoprotein level showed little or no increase, 12 showed increased uptake of 99mTc-PMT by the tumor. In assessing delayed 99mTc-PMT images, however, it was necessary to consider following complications: accumulation of tracer in obstructed and dilated biliary trees; retention of radioactivity in nonneoplastic liver tissues; difficulties in evaluating 99mTc-PMT uptake by small hepatic tumors; overlapping of radioactivity in the gut and gallbladder in delayed 99mTc-PMT images of tumors. This study indicates that delayed 99mTc-PMT images can be useful in the diagnosis of hepatocellular carcinoma.

Effect of clofibrate on cholesterol metabolism in rats treated with polychlorinated biphenyls.

Serum and hepatic cholesterol content in rats treated with polychlorinated biphenyls (PCBs, KC-400) were increased compared to those of control rats. This increase of cholesterol content was reduced to control level by simultaneous administration of ethyl p-chlorophenoxyisobutyrate (CPIB). Also, when lecithin-cholesterol acyltransferase (LCAT) (EC. 2.3.1.43) activity was expressed as the net cholesterol esterification, the acyltransferase activity in rats treated with PCBs was elevated, while the elevated acyltransferase activity was brought to control level by simultaneous administration of CPIB. On the other hand, the amount of bile of rats treated with CPIB, PCBs and PCBs-CPIB was increased, but free and total cholesterol content in bile of these treated rats was decreased to 40-60% of those of control rats. Moreover, cytochrome P-450 content in liver microsomes of rats treated with CPIB, PCBs and PCBs-CPIB was increased. At the same time, cholesterol-metabolizing activity in liver microsomes of rats treated with CPIB, PCBs and PCBs-CPIB also was elevated. Similar results were obtained for drug metabolizing (aniline hydroxylation and aminopyrine N-demethylation) activity. In addition, the amount of bile acids excreted from rats treated with CPIB, PCBs and PCBs-CPIB was increased compared to that of control rats. These results suggest that hypercholesterolemia induced by oral ingestion of PCBs is recovered by CPIB treatment and that this hypocholesterolemic effect of CPIB may be related partly to the elevation of hepatic mixed function oxidase activity for cholesterol catabolism.

Sheep red blood cell receptor and the epitope detected by Leu-5 monoclonal antibody in primates.

The expression of the epitope detected by the monoclonal antibody Leu-5 and the rosette formation with sheep red blood cells was examined for the mononuclear cells of 25 primate species including human subject. The plot of rosette-forming cells against Leu-5-positive cells enabled us to classify these species into three large groups: hominoids--Old World monkeys (Leu-5-positive cells greater than or equal to rosette-forming cells), New World monkeys (Leu-5-positive cells less than rosette-forming cells), and prosimians (Leu-5-positive cells less than 5%, some of them with both Leu-5-positive cells and rosette-forming cells less than 5%).

Epitope expression on primate lymphocyte surface antigens.

The cross-reactivity of peripheral blood mononuclear cells from 28 nonhuman primates was investigated with ten kinds of Leu series of monoclonal antibodies specific to human T-, natural killer/killer-, and B-cells. The chimpanzees possessed all ten epitopes examined but the orangutan lacked Leu4 and Leu7 epitopes and the gibbons lacked Leu4, Leu7, and Leu12 epitopes. In addition to the above epitopes, the Old World monkeys lacked Leu1 and Leu10 epitopes. The Leu3a/Leu2a cell ratios varied from 0 to 1.56 among the 12 macaque species and this enabled classification of these species into three groups. In the New World monkeys, Leu2a epitope was absent, whereas Leu11a epitope was detected in several species and Leu3a epitope was found only in the owl monkeys. The prosimians expressed only HLA-DR epitope.

Late infantile tetany and secondary hyperparathyroidism in infants fed humanized cow milk formula. Longitudinal follow-up.

Five full-term infants with birth weights appropriate for gestational age presented with hypocalcemic tetany at 5 to 9 days of age. All infants had been fed Similac 20, a cow milk formula. Initial mean serum calcium (Ca), phosphorus (P), and magnesium (Mg) levels of the tetanic infants were 6.8, 9.5, and 1.6 mg/dL, respectively. The mean serum parathyroid hormone (PTH) level was elevated at 79 mu LEq/mL (adult normal values, less than or equal to 57 mu LEq/mL). Following restoration of normocalcemia with Ca supplements, feeding was reinstituted with Similac 20 in two infants and Similac PM 60/40 in three infants. Serum biochemical and hormonal values were compared with those of 18 exclusively breast-fed infants followed up from three weeks to six months and 14 Similac 20-fed full-term infants followed up from one week to six months. In tetanic infants, serum Ca concentrations became elevated (10.4 +/- 0.05 mg/dL; mean +/- SEM) by six weeks (vs 9.2 +/- 0.3 mg/dL in breast-fed infants) (P less than .001) and serum Mg concentrations (2.26 +/- 0.01 mg/dL) by four weeks (vs 1.92 +/- 0.07 mg/dL in breast-fed infants) (P less than .01). Mean serum P concentrations declined progressively. Mean serum PTH concentrations were elevated and ranged from 74 to 143 mu LEq/mL at two to 16 weeks (vs mean 28 to 35 mu LEq/mL in breast-fed infants (P less than .0001). In 14 formula-fed-nontetanic full-term infants, serum PTH concentrations were intermediate between formula-fed-tetanic and breast-fed infants, mean serum Ca concentrations ranged from 10.2 to 10.4 mg/dL, and mean serum P concentrations declined from 8.3 to 7.1 mg/dL. We speculate that acute hypocalcemic tetany in the study infants was induced by the relatively high P load in cow milk formulas (vs human milk); with the continued P load, secondary hyperparathyroidism continued, maintaining P, Ca, and Mg homeostasis.

Minute carcinoma of the thyroid and its development to advanced carcinoma.

The relationship between the size of carcinomas and their histological characteristics such as encapsulation and sclerosis was studied using 157 cases of thyroid minute carcinomas with diameters less than 10 mm. When the diameter of the tumor was less than 1 mm, encapsulated carcinomas were not found, but they were presented in a group with a larger diameter. Sclerosing carcinomas were more frequently distributed in the group with a diameter less than 5 mm. With increasing tumor size, the carcinomas were reduced markedly in number and only two cases of sclerosing carcinoma were observed with a diameter more than 8 mm. It was concluded that thyroid minute carcinomas arise as nonencapsulated carcinoma, either sclerosing or non-sclerosing, followed by encapsulation at a later time and that some of the non-sclerosing carcinomas become apparent clinically, whereas most of the sclerosing carcinomas remain clinically silent throughout the entire life of the host.

An immuno-electron microscopic study on intracellular localization of thyroglobulin (TG) in the thyroid gland in Grave's disease.

Intracellular localization of thyroglobulin (TG) using a pre-embedding diffusion technique and an indirect localization sequence has been made in human thyroid obtained from 20 patients with treated Grave's disease. Both antibodies, anti-human TG-rabbit IgG F(ab')2 and anti-rabbit IgG F(ab')2-goat IgG F(ab')2 fragments easily penetrated the cytoplasm of follicular cells which were dissociated by RPMI-1640 solution containing collagenase, dispase, and deoxyribonuclease. With light microscopic observation of semithin sections positive immuno-reaction for TG was demonstrated as fine granular deposits in the cytoplasm of the dissociated cells. In electron microscopic studies, intracellular antigen was well circumscribed within certain cell organelles in all cases with the positive immuno-reaction for TG being observed in perinuclear space, rough endoplasmic reticulum, Golgi complexes, secretory granules, and reabsorbed colloid droplets. Content of positive immuno-reaction product differed somewhat from one case to another and from one follicle to another even in the same case. There was no immunoreaction product in nuclei, mitochondria, lysosomes, and lipofuscin-like granules.

Thyroid carcinoma in benign thyroid diseases. An analysis from minute carcinoma.

The relative frequency of thyroid carcinoma in benign thyroid diseases such as toxic diffuse goiter (toxic goiter), adenomatous goiter (goiter), adenoma, and chronic thyroiditis (thyroiditis) was studied using 3,219 surgically removed thyroid glands. Coexistence of carcinoma and benign diseases was found in 257 glands. Among them, 157 glands had minute carcinoma (diameter of cancer nodule is smaller than 10 mm) and 100 glands had advanced carcinoma (larger than 10.1 mm in diameter). The incidence of carcinoma including minute carcinoma was 29.4% in goiter (98/333), 21.0% in thyroiditis (11/53), 8.6% in adenoma (55/638), and 5.3% in toxic goiter (98/1852). Chi-square test also revealed that the rate of carcinoma not only advanced carcinoma but also minute carcinoma was higher in goiter than in the other diseases (P less than 0.01, respectively). Among the patients aged under 39, the incidence of advanced carcinoma and minute carcinoma in thyroiditis were the highest, respectively (83% and 100% in thyroiditis, 9.0% and 11.4% in goiter, 2. 6% and 1.9% in adenoma, and 0.7% and 3.4% in toxic goiter), however, among the patients aged over 40, they were secondary lower and the lowest, respectively (19% and 16.8% in goiter, 5.2% and 7.5% in adenoma, 4.2% and 6.7% in thyroiditis, and 1.4% and 7.5% in toxic goiter). We concluded that adenomatous goiter accompanies carcinoma more frequently than other benign thyroid diseases and the high incidence of carcinoma in chronic thyroiditis is probably due to a preoperative selection of the patients.

cAMP-independent stimulation of glycogen phosphorylase in newborn rat hepatocytes.

Postnatal development of glycogen phosphorylase activation by the cAMP-independent pathway was examined in isolated rat hepatocytes from control and propylthiouracil-treated congenital hypothyroid rat pups. At 5 days postnatum there was complete phosphorylase activation by beta-adrenergic stimulation, glucagon, and the calcium ionophore A23187, but no activation by alpha-adrenergic stimulation. Activation of phosphorylase by angiotensin or vasopressin was less than in hepatocytes from adult rats (P less than 0.01). At 28 days postnatum activation by all of these hormones was complete. In the propylthiouracil-treated group hormone responsiveness was similar to the control at 5 days postnatum. However, alpha-adrenergic (P less than 0.025), angiotensin, and vasopressin (P less than 0.05) activation was decreased at 28 days postnatum, and beta-adrenergic, glucagon, and A23187 activation was complete. The attenuated responses were restored by thyroxine replacement from 15 days postnatum. [32P]Pi incorporation into phosphatidylinositol by epinephrine and vasopressin in 28-day propylthiouracil-treated rats was lower than the control (P less than 0.01). We speculate that the diminished phosphorylase response of hepatocytes to alpha-adrenergic, vasopressin, or angiotensin stimuli in the early neonatal period could be related to low receptor numbers and the weaker phosphoinositide response during this period. Also, the depressed phosphorylase response to alpha-adrenergic, vasopressin, and angiotensin stimulation in congenital hypothyroidism at 28 days postnatum could be related to a decrease in number of plasma membrane receptors for these agonists.

Age-related changes in the adrenergic control of glycogenolysis in rat liver: the significance of changes in receptor density.

The present study examined the developmental changes in the adrenergic control of glycogenolysis in the rat model. A relatively new beta-adrenergic radioligand, 125I-iodocyanopindolol (ICP), was examined in binding assays with rat liver plasma membrane (LPM). ICP demonstrated both a higher specificity and a greater affinity for beta-adrenergic receptors than any previously available beta-adrenergic radioligand used to study rat LPM. Utilizing this new ligand it was found that beta-adrenergic receptor density decreased from 114 +/- 4 fmol mg-1 in newborn LPM to 19 +/- 3 fmol mg-1 in adult male LPM. In contrast alpha-adrenergic receptor density examined using 3H-prazosin increased from 161 +/- 14 fmol mg-1 in the newborn to 554 +/- 59 fmol mg-1 in the adult male. The results of ICP displacement assays employing various beta-adrenergic agonists and antagonists indicated that ICP binding sites were beta 2-adrenergic receptors. Both guanosine triphosphate and its nonhydrolyzable synthetic analog guanylyl-imidodiphosphate lowered the affinity of epinephrine for ICP binding sites similarly in newborn and adult LPM. Thus the coupling of receptor to guanine nucleotide regulatory protein appeared to be the same in both age groups examined. In isolated hepatocytes glycogen phosphorylase activation was mediated by beta 2-adrenergic stimuli in the newborn and predominantly alpha-adrenergic stimuli in the adult male. These results suggest that the change in glycogen phosphorylase activation from beta- to predominantly alpha-adrenergic mechanisms seen with maturation is related to changes in receptor density.
