Flow cytometric analysis of the uptake of Hoechst 33342 dye by human lymphocytes.

Human peripheral blood lymphocytes have been shown to resist staining with the DNA binding fluorochrome Hoechst 33342 by the cellular membrane. The rate of uptake of the dye is strongly temperature-dependent with minimal uptake rate below 16 degrees C. The activation energy of dye transport was found to be 135 kJ/mol above 20 degrees C and about 20 kJ/mol below 16 degrees C. Metabolic inhibitors accelerated, instead of inhibiting, the transport of the dye. Dead cells have been shown to stain promptly in contrast with the gradually staining viable cells. The uptake process in the vital staining conditions is suggested to involve a carrier mediated mechanism. Application of Hoechst 33342 as a fluorescent indicator of viability is proposed.

Location of nodulation and nitrogen fixation genes on a high molecular weight plasmid of R. meliloti.

R. meliloti strain 41 (Rm41) was shown to harbour two indigenous plasmids with molecular weights of 140 Mdal (pRme41a) and more than 300 Mdal (pRme41b), respectively. Using a heat-treatment procedure, derivatives of Rm41 defective in nodulation (Nod-) or nitrogen fixation (Fix-) have been readily obtained. In some Nod- mutants the deletion of a segment of plasmid pRme41b was found. Based on the demonstrated homology between the nitrogen fixation (nif) genes of Klebsiella pneumoniae and of R. meliloti the Rhizobium nif region has been cloned into the cosmid vector pHC79, then recloned into pBR322 and the restriction map of the nif region has been determined. 32P-labelled nick-translated probe prepared from the cloned nif DNA fragment hybridized to pRme41b of Rm41 but for most Nod- mutants this hybridization was not detected. Hybridization of a cosmid containing Rm41 DNA to total DNA digest from the wild-type bacterium and from a series of Nod- mutants revealed that at least a 2 kb DNA fragment including the nif structural genes was missing from most of the Nod- mutants. These results, together with the genetic analyses of these symbiotic mutations suggest that some nod and fix genes are located on pRme41b.

Experimental model of leukemic infiltrative pericarditis reflecting a human disease.

In human leukemias, pericarditis may be associated with leukemic pericardial infiltration. In order to obtain more information about the pathogenesis of the leukemic involvement of the pericardium, sterile talc-induced pericarditis was produced in acute lymphoid leukemic male hybrid mice. The animals invariable showed leukemic pericardial infiltration within 14 days. In leukemic mice without any treatment or with intrapericardial physiological NaCl injection (instead of talc), leukemic involvement of the pericardium did not occur.

[Fractures of the medial humeral epicondyle in children (author's transl)].

Operation of this fracture, according to their experience and the literature, can be recommended only in 3 cases: 1. Impingement of the avulsed fragment 2. Paresis of the ulnar nerve 3. Displacement of a larger fragment down to the joint level. Conservative management is thought to be reasonable in any other forms of this lesion, as good results can be achieved without the risks of an operation.

Detection by radioimmunoassay of antigens related to the P30 polypeptides of primate type C oncoviruses in peripheral leukocytes from patients with chronic myelogenous leukaemia.

Leukocytes from patients with chronic myelogenous leukaemia were examined for antigens related to the p30 polypeptides of baboon endogenous virus (BaEV) and gibbon ape leukaemia virus (GaLV). Samples from patients with the quiescent phase of the disease proved to be negative or contained p30 of BaEV as the only viral antigen. In cases of blastoid crisis or acceleration, an antigen related to p30 of GaLV could be detected. In 5 of 6 patients, acceleration or blastoid crisis was preceded by expression of GaLV p30.

Cytogenetic observations in chronic myeloid leukemia (CML) correlations of the cytogenetic findings with morphological cytochemical features of blast cells in various types of blastic phase.

Forty-nine cases of chronic myeloid leukemia have been investigated using cytogenic methods. Twenty-six patients were studied in chronic phase, twenty-three during the transformation of the disease. Out of the second group of the patients, nine were myeloid, five myelomonocytic, two promyelocytic, two megakaryoblastic and five lymphoid in their morphological and cytochemical appearance. Some correlation could be observed between the clinical, morphological, cytological and cytogenetic findings of different subgroups of the blastic phase in CML. The results demonstrate the divergent clonal evolution of transformed blast cells, and emphasize the importance of chromosome investigations -- in addition to the early diagnosis of metamorphosis -- in distinguishing the different subgroups of the terminal stage of CML.

Distribution of catecholamines and serotonin in 17 portions of rat ventromedial nucleus and effect of acute immobilization stress.

The distribution of catecholamines (i.e. adrenalin -- A, noradrenalin -- NA, dopamine -- DA) and serotonin (5-HT) in 17 portions of hypothalamic ventromedial nucleus (VMN) and the effect of acute immobilization stress on their levels were studied in rats using of a special dissection technique and a sensitive radioenzymatic microassay. In unstressed rats distinct differences of biogenic amine concentrations between individual portions within the hypothalamic ventromedial nucleus subdivisions were found. The concentration of all catecholamines investigated, i.e. NA, A and DA was lower in the lateral half of VMN, while in the distribution of 5-HT we did not find such apparent regional differences. After acute immobilization stress for 30 min significant changes in all measured neurotransmitters, in NA and A decreases and in DA and 5-HT increases, were detected in several subdivisions of the nucleus. On the basis of the present data we suppose that not all subdivisions of the VMN are equally engaged in neuroendocrine processes activated during acute stress.

Lysosomal enzymes in sera and granulocytes of patients with chronic liver diseases.

The authors have measured the activity of acid phosphatase, beta-glucuronaidase and cathepsin-D in tthe sera of patients with different liver diseases, and the actvity of beta-glucuronidase in granulocytes and the rate of its release. Using the method for the releaseof beta-glucuronidase from the granulocytes they drew conclusions to thedegree of the membrane-damage occurring in the liver diseases. They provided that in the sera of patients with different liver diseases the increase in the activity of the acid phosphatase and the beta-glucuronidase is different. According to their in vitro studies the decrease in the beta-glucuronidase activity measured in the granulocytes, and the release of enzyme is in connection with the type of liver disease and with the different degree of lysosomal membrane alteration.
