Endotoxaemia following colonoscopy.

In 100 consecutive patients the occurrence of bacteraemia and endotoxaemia after colonoscopy was studied. The Limulus Amoebocyte Lysate Test was used to detect endotoxaemia. No bacteraemia was detected after colonoscopy. Growth of Staph, epidermidis was considered as contamination. In 9% of the patients endotoxaemia was discovered after colonoscopy. The occurrence of endotoxaemia was found to be unrelated to the duration of the procedure, the number of the biopsies taken, or the cleansing of the bowel prior to colonoscopy. In patients with liver cirrhosis endotoxaemia occurred more often than in patients without liver disease. No clinical consequences of endotoxaemia were found in patients who developed endotoxaemia after colonoscopy. We conclude that colonoscopy is followed by endotoxaemia quite frequently. The clinical implication of these conditions, especially in patients with liver cirrhosis merits further consideration.

Lymphocytes bearing both T and B markers in lymphoproliferative disorders.

The appearance of dual-marked (D) lymphocytes, i.e. lymphocytes having both T and B markers, was investigated in patients with acute (ALL) and chronic lymphocytic leukaemia (CLL) as well as with non-Hodgkin's lymphoma (NHL). E-rosette and immunobead-rosette techniques were combined and simultaneously administered, and the combination was found to be simple, fast and reproducible. D cells could be detected also in normal peripheral blood, however, only in a low percentage. In the blood of 4 patients suffering from ALL, in 2 with CLL and in 1 with NHL the number of D cells were increased significantly. In ALL the relative number of these cells decreased during remission and increased in relapses; thus the determination of the relative frequency of D cells might have prognostic value and could be useful in the choice of an optimal therapeutic protocol.

Distribution of catecholamines in individual hypothalamic nuclei of Japanese quail.

The dissection of eight hypothalamic nuclei or areas of Japanese quail brain (nucleus hypothalamicus medialis anterior, nucleus periventricularis magnocellularis, nucleus hypothalamicus lateralis anterior, nucleus hypothalamicus medialis posterior, nucleus hypothalamicus lateralis posterior, eminentia medialis, nucleus tuberis and area hypothalamica posterior) and a subsequent radioenzymatic measurement of the basic concentrations of noradrenaline, adrenaline and dopamine in these structures of female Japanese quail were demonstrated in this study. Individual hypothalamic nuclei were identified according to the histological map prepared. All the above mentioned structures were isolated from serial 200 micrometers thick frozen coronal sections of the hypothalamus under a stereomicroscope by means of a needle punch of 500 micrometers or 300 micrometers internal diameter. The highest levels of all biogenic amines measured were found in the area hypothalamica posterior which significantly differed from all other nuclei examined. Their lowest concentrations occurred in the nucleus hypothalamicus medialis posterior and in the nucleus hypothalamicus lateralis posterior. The results present the first picture of the quantitative distribution of catecholamines in the hypothalamus of the female Japanese quail.

Cytotoxicity of lymphocytes and antibodies against autologous tumor cells in patients with myeloid leukemias and preleukemic disorders. I. Blocking activity of gp70 antigens of primate type C viruses.

Cytotoxicity of lymphocytes or antibodies against autologous tumor cells could be demonstrated very frequently in patients with chronic myeloid leukemia, whereas the presence of such cytotoxic activities was very rare in patients with acute myeloid leukemia. Lymphocytes and antibodies cytotoxic against autologous granulocytes were found in persons with potentially preleukemic cytopenic disorders. Control subjects with nonpreleukemic hematological disorders and healthy persons exhibited no cytotoxic activity. In the majority of cases the lymphocyte- or antibody-mediated cytotoxicity could be blocked with gp70 antigen of gibbon ape leukemia virus and baboon endogenous virus.

Effect of acute and repeated immobilization stress on the ultrastructure of neurons of the rat hypothalamic ventromedial nucleus.

The fine structure of neuronal somata of the central subdivision of the hypothalamic ventromedial nucleus (VMN) was studied in acutely and repeatedly immobilized rats. Four categories of neurons were distinguished in control animals based mainly on ultrastructural appearance of rough endoplasmic reticulum (RER): 1. the neurons with longer segments of RER distributed throughout the cytoplasm which, at the same time, clearly dominated in this area, 2. the neurons with RER arranged into several groups located at different sites of the soma, 3. the neurons with only a few of short fragments of RER scattered in the cytoplasm, and 4. the neurons with an extensive parallel system of RER in the cell periphery. After acute stress only three categories were found (the 4th category being absent), whereby the number of the 2nd category of neurons evidently increased. Following repeated immobilization a further category of neurons occurred, which represented 9...11% of the neuronal population and showed a wide variety of RER modifications including whorls. The presented morphological evidence suggest that the central subdivision of rat VMN might be involved in neuroendocrine processes activated during increased pituitary-adrenocortical response under immobilization stress.

[Changes in the catecholamine content of the dopamine-synthesizing nuclei of the brain in rats under immobilization stress].

Changes of catecholamines (CA) contents in dopamine (DA)--synthetizing nuclei of the midbrain and hypothalamus were measured by means of radioenzymatic technique in Wistar rats after 6.5 hrs of immobilization (IMO), the blood pressure (BP) being recorded in the tail artery. After decapitation, the gastric mucous membrane was examined. In substantia nigra (A9) a tendency toward an increase of the noradrenaline level was observed both during the rise and fall of the BP. In n.arquatus (A12) and n. periventricularis (A14) the changes of noradrenaline and dopamine concentration were in good agreement with the gastric mucous membrane lesion: the CA content was higher in rats with hemorrhages than in ones with ulcers. A sharp decrease of adrenaline content was observed in DA--synthetizing hypothalamic nuclei and it was not correlated with BP changes or injury in gastric mucous membrane.

[Endemic goiter in Austria. Is iodine deficiency the primary cause of goiter?].

In spite of government-regulated iodide admixture to table salt, the incidence of goiter is still high in Austria. Iodine excretion and thyroid function were therefore investigated in 80 patients suffering from ordinary goiter in whom thyroid size and resulting symptoms had increased lately. 25 euthyroid non-goitrous subjects served as controls. 48% of the goitrous patients investigated presented with iodine excretion of less than 70 micrograms/24 h, suggesting an insufficient iodine supply. Thyroid I131 uptake, basal and TRH-stimulated plasma TSH concentrations, and serum T3 levels were higher, whereas serum T4 levels were lower in these patients than in goitrous patients with higher iodine excretion and non-goitrous controls. Iodine deficiency thus appears to be of pathogenetic relevance in about half of the goitrous Austrian population. Other factors enhancing goiter development seem to assume particular importance in goitrous patients with a sufficient iodine supply.

Permeabilization of lymphocytes with polyethylene glycol 1000. Discrimination of permeabilized cells by flow cytometry.

The toxicity of polyethylene glycol 1000 (PEG) used similarly as in cell hybridization experiments, has been studied by flow cytometry, measuring the light scattering and fluorescence distributions of PEG-treated human lymphocytes stained with propidium iodide, fluorescein diacetate and acridine orange (AO). The sensitivity of these tests to detect permeabilized, or potentially dead cells, was equal. In addition, PEG proved to interfere with AO staining most likely through the inhibition of its binding to nucleic acids. The decrease of AO fluorescence in cells killed by PEG was unexpected since intercalation of propidium iodide was the same as in alcohol fixed cells. Permeabilization of cells by PEG appears to be an all-or-none phenomenon, accompanied by entrance of PEG into the cells. The findings are described in the context of a review of the currently used flow cytometric techniques to discriminate viable and lethally affected cells; also, the problems of interpretation are discussed.

Separation of Neurospora crassa myo-inositol-1-phosphate synthase from glucose-6-phosphate dehydrogenase by affinity chromatography.

The purification of Neurospora crassa myo-inositol-1-phosphate synthase (EC 5.5.1.4) was studied by affinity chromatography using the substrate (glucose-6-phosphate), the inhibitor (pyrophosphate), the coenzyme (NAD+) and the coenzyme analogues (5'AMP and Cibacron Blue F3G-A) of the enzyme as adsorbents attached to agarose gel. Myo-inositol-1-phosphate synthase could be separated completely from the contaminating substance, glucose-6-phosphate dehydrogenase (EC 1.1.1.49), on Blue Sepharose CL-6B and on pyrophosphate-Sepharose. The purified enzyme had a specific activity of 16 400 U/mg. The sodium dodecyl sulfate/polyacrylamide gel electrophoresis of the 60 micrograms of this purified enzyme gave a homogenous band. The enzyme was found to be composed of four identical subunits having a molecular weight of 65 000.

Studies on antibodies reactive with glycoproteins of primate type C viruses in patients with myeloid leukemias and with potential preleukemia.

Human blood plasma from patients with myeloid leukemias, potentially preleukemic disorders and healthy individuals contains antibodies which react with purified glycoproteins of the baboon endogenous virus and the gibbon ape leukemia virus. Experiments are presented which illustrate characteristic distributions of antiviral antibodies in the plasma of different investigated groups. The presence of high-titer antibodies is associated with remission of acute myeloid leukemia and longer survival of patients with preleukemia.

The neurotoxicity of organotin: behavioural changes in rats.

The neurotoxic action of the fungicide Tinestan 60 WP (containing 60% triphenyltin acetate) was examined in male rats. Due to daily doses as small doses as 6 and 0.6 mg/kg daily administered over six weeks the exploratory activity in maze was diminished, the animals made significantly more errors. There was no difference between the poisoned and the control groups in the speed of acquisition of the conditioned avoidance responses, however the extinction was significantly slower, as a sign of slower development of active inhibition. Tested in stress situation the behaviour of poisoned animals altered, as a sign of decreased habituation. It is concluded that Tinestan crosses the blood-brain barrier, and induces diminished plasticity of the CNS.

Morphological study on the central subdivision of the rat hypothalamic ventromedial nucleus.

Central subdivision of the rat hypothalamic ventromedial nucleus (VMN) was examined with the transmission electron microscope. This portion of VMN consists of densely packed neuronal somata which are characterized by a large pale nucleus with a prominent nucleolus, with clear cytoplasm containing many free ribosomes and randomly distributed fragments of rough endoplasmic reticulum, with small mitochondria and homogeneous lysosomes, with well developed Golgi complex surrounded by a few clear and largely 2-3 dense-core vesicles of 90-110 nm in size and with nucleotid bodies. Besides neuronal somata and their processes also astrocytes, oligodendrocytes and microglial cells and their processes are often seen in this portion of VMN. In the neuropil axo-somatic and axo-dendritici synapses, capillaries, myelinated nerve fibers, nerve profiles filled with dense-core vesicles and distal parts of ependymal tanycytic processes are usually found in the central subdivision of VMN.

Acute carbon tetrachloride induced lysosomal membrane damage and the membrane protecting effect of a new dihydroquinoline-type antioxidant.

The authors examined the damage of lysosomal membrane caused by acute CCl4 intoxication by in vitro methods. They measured the acid phosphatase as well as beta-glucuronidase enzyme levels and determined the rate of release of these two enzymes. The in vivo changes in enzyme activity were extrapolated from the in vitro results. The CCl4 causes a significant increase in the permeability and rigidity of the lysosomal membrane. By oral and/or intraperitoneal administration of MTDQ the state of permeability can be improved or even corrected. On the basis of their results, the authors conclude that the lysosomal damage caused by CCl4 is mediated by peroxidation of lipids and the lysosomal membrane can be stabilised by MTDQ.
