Phenytoin, tetrodotoxin, and acetylcholine release.

To determine if phenytoin reduces depolarization-linked acetylcholine release from synaptosomes solely by interacting with Na+, the effect of phenytoin (100 to 200 microM) and/or tetrodotoxin (1 microM) on ACh release was assayed in synaptosomes depolarized with either KCl 56 mM or veratridine 10 microM. Phenytoin reduced release in both depolarizing solutions. In KCl the effect of phenytoin was greater at 200 microM than 100 microM but with veratridine, phenytoin 200 microM was not more effective than 100 microM. Tetrodotoxin failed to affect release induced by KCl but the effect of tetrodotoxin and phenytoin on veratridine-stimulated release was much greater than that with phenytoin alone 100 microM. In conclusion, the inhibition of acetylcholine release by phenytoin must be independent of any effect of the drug on Na conductance.

Regulation of [3H]mazindol binding to subhypothalamic areas: involvement in glucoprivic feeding.

The distribution of low-affinity sodium-sensitive binding sites of [3H]mazindol were studied in rat hypothalamic nuclei. Using microdissection methods, it was demonstrated that the highest level of [3H]mazindol binding is localized to the paraventricular nucleus (PVN) and the lowest binding is observed in the lateral hypothalamus. Following food deprivation, a significant decrease in [3H]mazindol binding in the PVN and ventromedial hypothalamus (VHM) were observed. Refeeding food-deprived rats resulted in restoration of the level of binding in the PVN, and this was correlated with changes in blood glucose levels. Thus, changes in the binding of [3H]mazindol in the PVN may reflect local changes in glucose levels. In related studies, the involvement of the PVN in the regulation of food deprivation or 2-deoxyglucose (2-DG)-induced food intake was studied. Application of amphetamine (20 micrograms) into the PVN had no effect on food deprivation induced feeding, but significantly inhibited 2-DG induced (glucoprivic) feeding. The PVN may play an important role in the glucostatic regulation of feeding and in mediating the anorectic action of amphetamine and related anorectic drugs on glucoprivic feeding.

Overproduction of the Bacillus sphaericus R modification methylase in Escherichia coli and its purification to homogeneity.

A DNA fragment containing the information coding for the GGCC-specific Bacillus sphaericus R modification methylase, BspR, was inserted into plasmid vector pKK223-3 under the control of the strong and inducible tac promoter, and transformed into Escherichia coli HB101. Upon induction this strain accumulated the methylase enzyme (while cell growth was inhibited) up to several percent of total cellular protein. Homogeneous methylase could be prepared in three purification steps.

Oesophageal and gastric motility disorders in patients categorised as having primary anorexia nervosa.

UNLABELLED: Gastrointestinal motor function in patients with primary anorexia nervosa has rarely been investigated. We studied oesophageal motor activity in 30 consecutive patients meeting standard diagnostic criteria for primary anorexia nervosa (Feighner et al; DSM III). Seven were found to suffer from achalasia instead of primary anorexia nervosa, one from diffuse oesophageal spasm and one from severe gastro-oesophageal reflux and upper oesophageal sphincter hypertonicity, while partly non-propulsive and repetitive high amplitude, long duration contractions prevailed in the lower oesophagus of another six. In four patients with oesophageal dysmotility not responding to therapy and in 12 of 15 patients with normal oesophageal manometry, gastric emptying of a semisolid meal was studied. Emptying was normal in only three but markedly delayed in 13 cases (half emptying times 97-330 min, median: 147 min, as compared with 21-119 min, median: 47 min, in 24 healthy controls). In eight patients, the effects of domperidone 10 mg iv and placebo were compared under random double blind conditions. Half emptying times were shortened significantly (p less than 0.01) by domperidone. CONCLUSIONS: symptoms of disordered upper gastrointestinal motor activity may be mistaken as indicating primary anorexia nervosa; clinical evaluation of patients with presumed primary anorexia nervosa should rule out the possibility that disordered oesophageal motor activity underlies the symptoms; delayed gastric emptying is a frequent feature in primary anorexia nervosa and might be returned to normal with domperidone.

Differentiation between chronic lymphocytic leukaemia (B-CLL) and non-Hodgkin lymphomas in the leukaemic phase by the mouse red blood cell rosette assay.

A distinction between B-CLL and other malignant B-cell lymphomas in the leukaemic phase may be difficult. Mouse red blood cell rosette formation of lymphocytes from 97 patients with B-CLL and 19 patients suffering from other B-cell lymphoproliferative disorders was examined together with lymphocyte rosette formation of healthy controls. The majority of circulating lymphocytes of B-CLL patients formed rosettes with mouse red cells, whereas there was no relationship between the number of peripheral neoplastic B-cells and that of rosette forming cells in other lymphoproliferative diseases. The relatively simple mouse red blood cell rosette assay proved to be of value in the differentiation of otherwise nearly related conditions.

Platelet function studies in myeloproliferative disorders.

Platelet functions were studied in 64 patients with various myeloproliferative diseases. The characteristic alterations were prolonged bleeding time, decreased platelet aggregation (but normal results induced by ristomycin), elevated level of BTG, high production of MDA, increased level of TXB2 with almost normal level of 6-keto-PGF1. However, considering the bleeding time and the amount of BTG in relation to the whole blood platelet count, no differences could be detected.

A novel method for the isolation of carboxypeptidase B.

Carboxypeptidase B was isolated from porcine pancreas by selective heat treatment of the autolyzed tissue at 60 degrees C and pH 6.0 the presence of phosphate ions. The heat treatment was followed by ammonium sulfate fractionation and ion-exchange in a batch system. The method could also be used for the isolation of carboxypeptidase B from beef pancreas.

History of patients with chronic granulocytic leukaemia observed in 1974-1984.

138 CGL-patients were treated in 1974-1984. The female-male ratio was 1.26. In the majority (82.3%) of the 68 patients having died at the blastic phase, all three phases of the disease could be followed. The chronic phase lasted for 31 months. The accelerated disease was detectable in 56 patients, with a mean duration of 5.3 months. The blastic phase, lasting for 4.1 months, was morphologically heterogeneous. Using adequate methods, several groups could be distinguished, i.e. myeloid in 38.3, promyelocytic in 2.9, myelomonocytic in 19.1, megakaryoblastic in 14.7, lymphoblastic in 13.2 and mixed blastic in 11.8%. 23 patients died prior to the development of the blastic phase (9 in the chronic phase, 12 in the accelerated phase and two with chronic neutrophilic leukaemia). The mean survival was in accordance with that in the literature, i.e. 40.6 months. The shortest survival was shown by the lymphoblastic group with the difference manifesting already in the chronic phase. Females survived 13-14 months longer than did males.

Cytotoxicity of lymphocytes and antibodies against autologous tumor cells in patients with myeloid leukaemias and preleukaemic disorders. III. Stage-dependence of oncovirus-specific immune response.

Lymphocyte and plasma samples from the quiescent and blastic phase of chronic granulocytic leukaemia (CGL) and from the blastosis and remission of acute myeloid leukaemia (AML), were compared for cytotoxic activity. Target cells were collected from the blastic phases of diseases. 51Cr-release tests showed that the lymphocytes and plasma samples from blastic crisis of CGL had no cytotoxic activity for autologous blast cells. In contrast, cryopreserved lymphocytes and plasmas from the quiescent phase of CGL proved to be cytotoxic for the autologous tumor cells, and their effect could be blocked by native gp70 antigens of gibbon ape leukaemia virus (GaLV) and baboon endogenous virus (BaEV). A blocking effect was less frequently exerted by carbohydrate-free gp70 and p15(E) antigens. A similar relationship was found between the blastosis and remission stage of AML, however, out of the antigens of BaEV only the native gp70 showed a marked blocking effect.

[Recurrent disseminated infundibulofolliculitis].

A white male patient with disseminated, pruritic, follicular papules on the skin is reported. Histologically, an inflammatory infiltration around the infundibulum, perifollicular oedema and spongiosis of the follicular epithelium were demonstrated. The lesions failed to respond to therapy. On the basis of the clinical and histological findings, we suggest that this is a case of disseminated and recurrent infundibulofolliculitis, which was first described by Hitch and Lund in 1968.

Nucleotide sequence of the BsuRI restriction-modification system.

The genes of the 5'-GGCC specific BsuRI restriction-modification system of Bacillus subtilis have been cloned and expressed in E. coli and their nucleotide sequence has been determined. The restriction and modification genes code for polypeptides with calculated molecular weights of 66,314 and 49,642, respectively. Both enzymes are coded by the same DNA strand. The restriction gene is upstream of the methylase gene and the coding regions are separated by 780 bp. Analysis of the RNA transcripts by S1-nuclease mapping indicates that the restriction and modification genes are transcribed from different promoters. Comparison of the amino acid sequences revealed no homology between the BsuRI restriction and modification enzymes. There are, however, regions of homology between the BsuRI methylase and two other GGCC specific modification enzymes, the BspRI and SPR methylases.

[Clinical aspects and course of Crohn disease].

The influence of clinically observable criteria on the course of disease was investigated in 150 patients, 72 men and 78 women, with morbus Crohn who could be followed for an average period of 6.6 years (8-348 months). In 29 patients (19%), the small intestine only was involved, in 31 patients (21%) the colon, and in 90 patients (60%) both the small intestine and colon. During the observation period 71 patients (47%) developed fistulae and 55 (37%) extra-intestinal manifestations. The patients were admitted to hospital on a total of 466 occasions (3.1 times per patient), and 199 operations (1.3 times per patient) were carried out. There were significantly more extra-intestinal manifestations in the colon cases (P less than 0.01) and of admissions and operations in the small intestine-colon cases (P less than 0.001), whereas patients with isolated involvement of the small intestine underwent significantly fewer operations (P less than 0.001). In addition, significant positive correlations existed between the appearance of fever and fistulae (P less than 0.001) and between the number of operations within the first 2 years and the number of subsequent operations (P less than 0.001).

Behavioural effects of prenatal exposure to carbon disulphide and to aromatol in rats.

The neurotoxic effects of prenatal organosolvent inhalation were studied in rats, because of the expectation that a developing organism may be more sensitive than the adult to the induction of functional deficits. The aim was to determine whether prenatal exposure to the new organosolvent mixture, Aromatol, and the well known neurotoxic carbon disulphide, would impair reflex ontogeny or produce neurobehavioural dysfunctions in the offspring. Development of gait, motor coordination, and activity, avoidance learning and swimming were tested in the offspring of CFY rat mothers, exposed to CS2 inhalation (0, less than 10, 700 and 2000 mg/m3) and to Aromatol (0, 600, 1000 and 2000 mg/m3) on days 7-15 gestation. Prenatal CS2 inhalation induced dose related perinatal mortality of pups. Eye opening and the auditory startle were retarded. There were immature gait, motor incoordination, diminished open field activity and altered behavioural patterns on day 21 and 36 but they were nearly age-appropriate on day 90. As signs of disturbed learning ability, there were diminished performance and lengthened latency of the conditioned avoidance response, related to the concentrations administered. Contrary to expectations, prenatal Aromatol inhalation had no effect on maturation of gait, behaviour patterns, or learning ability.
