Acetylcholinesterase and choline acetyltransferase activity in some hypothalamic nuclei under immobilization stress in rats.

Rats were subjected to acute immobilization (IMO) for 1 or 2 h and killed by decapitation. The hypothalamic supraoptic and paraventricular nuclei were isolated by dissection technique and the activity of acetylcholinesterase (AChE) and choline acetyltransferase (ChAT) was assayed. A period of 1 or 2 h of IMO resulted in a significant increase of AChE activity only in the supraoptic nucleus, while no change of ChAT activity occurred. Acute IMO of 1 or 2 h duration resulted in an increase of AChE activity in supraoptic nucleus.

Retrovirus-neutralizing antibodies in AML and AMMoL patients: stage-specific distribution.

Plasma samples of patients with AML or AMMoL were tested for antibodies reacting with gp70 antigens of BaEV and GaLV as well as for antibodies neutralizing BaEV or GaLV. Both frequency and titer values of antibodies were higher in remission than in blastosis. Neutralizing activity could be detected only in those plasma samples which contained antibodies to the appropriate gp70 antigen. The data suggest the presence of retroviruses in humans as antigenic stimuli for the immune system in AML and AMMoL.

[The significance of psychosocial complaints in the initial interview--a study of unselected patients of an internal medicine/polyclinic university ambulatory service].

In clinical practice one is repeatedly confronted with patients, who report psycho-social problems during the first interview. In this study 128 patients have been examined in the outpatient clinic at the Department of Medicine 1 of the University Hospital in Vienna. The most important issue investigated was to clarify whether or not the psycho-social complaints expressed at the initial interview could be correlated with either functional disorders or somatic disease. Further we tried to determine whether or not the complaints influenced the course of the disease and gave any indication for the presence of a depressive disorder or complicated a somatic disorder by additional psychosomatic complaints. Our results indicate that the expression of psycho-social problems indeed correlates with stronger psychosomatically conditioned body dysfunctions, but do not sufficiently explain whether or not functional disease can be expected. However, the reported psycho-social problems result in a stronger inclination towards depressive mood and lead the patient to his/her own explanation how the illness has developed. Both facts are of importance for the patient-physician relationship and constitute an integral part of the (non-)compliance.

[Microcomputer-assisted system for photographing the movements of biological valve prostheses].

The aim of the study was to develop a clinically usable system of photographic imaging of the movements of biological valve transplants and of evaluating temporal and spatial relationships. The system has been established by means of a microcomputer and associated peripheral electronic equipment so that there is no need to use a high-speed camera. Functioning of the system was verified by testing 37 aortal valve allotransplants and the Björk Shiley mechanical valve prosthesis.

Characterization of a mutation that causes overproduction of inositol in Neurospora crassa.

Slow-growing (inl+/-) spontaneous mutants have been isolated from an inositol requiring (inl) strain of Neurospora crassa that produces defective myo-inositol-1-phosphate synthase (MIPS), the enzyme responsible for the production of inositol-1-phosphate from glucose-6-phosphate. The defective enzyme has some residual activity. In the inl+/- strain the synthesis of the defective enzyme is enhanced, which enables the strain to grow slowly on minimal medium. The mutation (opi1) responsible for the partial inositol independence segregates independently from the inositol locus, and suppresses the inositolless character by overproduction of defective MIPS. opi1 acting upon the wild type (inl+) allele increases MIPS production and causes inositol excretion.

Light microscopic triple-colored immunohistochemical staining on the same vibratome section using the avidin-biotin-peroxidase complex technique.

The sequential application of the avidin-biotin-peroxidase complex technique was used to localize multiple tissue antigens on a single free floating section of rat brain. Sequential visualization of individual antigens was achieved by the silver-gold-intensified diaminobenzidine (DAB) in the first step, nickel-intensified DAB in the second step, and the DAB alone in the third step of the immunostain procedure. For the demonstration of this method, tyrosine hydroxylase (TH), corticotropin-releasing factor (CRF), and vasopressin (VAS) antisera were used. Sections from the hypothalamic paraventricular nucleus (PVN) of rats pretreated with colchicine were stained. Black TH containing cell bodies were clearly distinguished from blue stained CRF cells and from yellow stained VAS-containing cell bodies in the PVN on the 25-30 micron thick vibratome sections. The sequential immunostaining procedure presented here results in superior staining of multiple antigens as compared to that achieved by the sequential application of the peroxidase-antiperoxidase (PAP) technique.

Lack of association between HLA antigens and anorexia nervosa.

Studies of the human leukocyte antigen (HLA) system were performed in 37 patients with anorexia nervosa to confirm an alleged association with HLA-B16 and haplotype HLA-A26,B38. No correlation between anorexia nervosa and HLA, including HLA-DR, could be found.

Interferon production in myelo- and lymphoproliferative diseases. I. Spontaneous interferon production in acute and chronic leukaemias.

Blood plasma interferon (IFN) of patients with acute myeloid (AML) and chronic granulocytic (CGL) and with acute and chronic lymphoid leukaemia (ALL and CLL) was measured by bioassay and characterized by neutralization with anti-human-IFN-alpha treatment. Elevated IFN-alpha level (60-125 IU/ml) was found in the quiescent phase of CGL as compared to the control samples (15 +/- 10 IU/ml). No significant differences could be found between plasma IFN of patients suffering from the blastic crisis of CGL and control persons. Analogous results were obtained in experiments with plasmas of AML patients, regardless the stage of the disease. Elevated IFN-alpha production (60-100 IU/ml) was found in patients being in the remission phase of T-cell ALL but only in one case in the progressive phase of the disease. No significant elevation of plasma IFN level was demonstrated in patients with O-cell ALL and B-cell CLL, as compared to the control samples.

Defective myo-inositol-1-phosphate synthase production in an inositolless double mutant Neurospora crassa strain.

A slow growing inl+/- mutant was isolated from an inositol dependent (inl) Neurospora crassa strain. The latter strain produces defective myo-inositol-1-phosphate synthase which has residual activity. Inositol, similarly to that found in wild and inl mutant strains, represses the enzyme production in the inl+/- strain as well. Withdrawing inositol from the medium results in derepression of the enzyme synthesis. Derepression is hindered by cycloheximide. Inl+/- character in the double mutant is brought about by overproduction of the defective myo-inositol-1-phosphate synthase.

Development of the central nervous system functions in rat pups prenatally exposed to alcohol (study on the behavioural teratology of ethanol in CFY rat pups).

As a model of fetal alcohol syndrome (FAS) the rate of the maturation of the functions of the central nervous system was studied in the offspring of pregnant CFY rats receiving (from the 7th-15th day of gestation) either oral ethanol treatment or liquid diet containing ethanol. Both types of exposure caused numerous behavioural impairments, besides high perinatal mortality also the opening of the eyes and ears, and the appearance of postural reflexes were delayed. The newborn rats could be characterized by hyperactivity and weak motor coordination. The learning capacity, the avoidance conditioned reflexes was the poorest in the case of the offspring of mothers kept on liquid diet, containing alcohol, the latency of the conditioned response was significantly lenghtened. During reconditioning, in the case of the sexually already mature pups, the weakest performance was observed in the offspring of mothers having received oral alcohol treatment. This findings indicated, on one hand, that the retardation ceased and, on the other, that the learning and memory impairments caused by oral alcohol exposure was persistent. Following prenatal alcohol exposure carried out by different methods the neurotoxic effect, the retardation of the rate of maturation of the central nervous functions, and the adaptive mechanisms were all affected to different extent. Besides alcohol exposure also other factors (relative protein insufficiency, malnutrition) may be involved in the pathomechanism of the above mentioned phenomena.

Cytotoxic activity of lymphocyte subpopulations against autologous tumour cells in patients with myeloid leukaemias and preleukaemic disorders.

Autologous lymphocyte populations from different phases of chronic granulocytic leukaemia (CGL), acute myeloid leukaemia (AML) and preleukaemic disorders were compared for cytotoxic activity. 51Cr-release tests showed that T lymphocytes from the quiescent phase of CGL and from the remission phase of AML exerted cytotoxic activity against autologous tumour cells. Such activity was also found in patients with potentially preleukaemic haematological disorders characterized by cytopenia, but not in polycythaemia vera patients. In the majority of cases cytotoxic activity of T lymphocytes could be blocked by native gp70 antigens of gibbon ape leukaemia virus (GaLV) and baboon endogenous virus (BaEV). Blocking effect of carbohydrate-free gp70 as well as p15(E) antigens could be observed less frequently. The role of cell-mediated immune response to oncovirus antigens in the course of myeloproliferative diseases is discussed.

Left ventricular diastolic function in diabetics.

Left ventricular diastolic function was studied in 29 young diabetic patients (aged from 14 to 44 years) without any clinical sign of heart disease. The metabolic state, the presence and the degree of microvascular and neuropathic complications have been established. Age and sex matched 32 healthy subjects served as controls. The parameters of left ventricular diastolic function were determined by means of phonomechanocardiography. By this method in diabetic patients impaired diastolic function of the left ventricle was found. This alteration could be best characterized by the values of normalized relaxation index referring to the isovolumetric relaxation of the left ventricle. A close correlation was found between the microvascular and neuropathic complications and the left ventricular diastolic dysfunction, while no correlation could be demonstrated between the metabolic state and the diastolic cardiac disorder.
