Estrogen receptor-negative breast cancer cells transfected with the estrogen receptor exhibit increased RAR alpha gene expression and sensitivity to growth inhibition by retinoic acid.

We and others have shown previously that retinoic acid (RA) selectively inhibits the growth of estrogen receptor (ER)-positive human breast carcinoma (HBC) cells and ER-negative cells are refractory to RA inhibition of growth. The ER-negative cells inherently express lower levels of RAR alpha and retinoic acid response element (RARE)-mediated RA-induced CAT activity. In this study we report that when ER-negative MDA-MB-231 cells were transfected with the ER gene they not only expressed higher levels of RAR alpha and RARE-mediated RA-induced CAT gene expression, but their growth was not inhibited by RA. Estrogen enhanced RAR alpha gene expression not only in established ER-positive cell lines but also in ER-transfected MDA-MB-231 cells. The estrogen effect appears to be direct and at the gene transcription level since it did not alter the stability of RAR alpha mRNA and cycloheximide failed to block estrogen-mediated enhancement of RAR alpha gene expression. Our data strongly suggest that ER-mediated enhancement of RAR alpha levels plays an important role in RA inhibition of HBC growth. In addition, we also report here that HBC cells appear to express a unique isoform(s) of RAR alpha which was detected only when the full-length RAR alpha cDNA was used as a probe; the RAR alpha 1 and RAR alpha 2 specific probes failed to hybridize with the HBC specific RAR alpha message.

Regulation of insulin-like growth factor-binding-protein-1, 2, 3, 4, 5, and 6: synthesis, secretion, and gene expression in estrogen receptor-negative human breast carcinoma cells.

Insulin-like growth-factors I and II (IGF-I, II) are potent mitogens for breast carcinoma proliferation. In extracellular fluids, most of the IGF-I and II is associated with specific IGF-binding proteins (IGFBPs). The role of these IGFBPs in IGF action is still not clear, but it has been demonstrated that these proteins may either enhance or inhibit IGF-mediated cellular effects. Synthesis and secretion of IGFBPs have been demonstrated in breast carcinoma cells. In this study, we examined retinoic acid (RA) and IGF-I modulation of IGFBP mRNA and IGFBP levels in two ER-negative human breast carcinoma cell lines. Treatment of MDA-MB-231 and MDA-MB-468 cells with RA increased the levels in conditioned media of a M(r) 42-46-kDa IGFBP, which was immunoprecipitated by an IGFBP-3 antibody. IGF-I also increased the accumulated levels of IGFBP-3 in the conditioned media of both cell lines. Both cell lines expressed high basal levels of IGFBP-3 mRNA; the addition of RA increased IGFBP-3 mRNA levels by 1.5-fold, whereas the addition of IGF-I had no effect on IGFBP-3 mRNA levels in either cell line. The difference in the magnitude of the RA enhancement of IGFBP-3 mRNA levels (1.5-fold) and RA stimulation of IGFBP-3 levels in conditioned media (3.5-4-fold) suggests that some of the effect of RA is at a posttranscriptional level. IGF-I increased the levels of IGFBP-2 and IGFBP-5 in conditioned media by greater than tenfold but had no effect on IGFBP-2 and IGFBP-5 mRNA levels, again suggesting the involvement of posttranscriptional controls. Pretreatment of MDA-MB-468 and MDA-MB-231 cells with IGF-I receptor antibody (alpha IR3) blocked the IGF-I effect on IGFBP-3 levels in the media in both cell lines and IGFBP-2 and IGFBP-5 secreted levels in MDA-MB-468 cell conditioned media. The addition of RA also blocked IGF-I stimulation of IGFBP-2 and IGFBP-5 levels. Cycloheximide treatment completely blocked the RA and/or IGF-I-mediated modulation of these binding proteins, suggesting that these agents enhance IGFBP-3, IGFBP-2, and IGFBP-5 synthesis and consequent secretion. MDA-MB-468 cells expressed IGFBP-5 mRNA, whereas both MDA-MB-231 and MDA-MB-468 expressed IGFBP-6 mRNA. RA enhanced IGFBP-6 gene expression by threefold in MDA-MB-231 cells, whereas IGF-1 had no effect on IGFBP-6 gene expression in either cell line.(ABSTRACT TRUNCATED AT 400 WORDS)

Determinants of night blindness in Bangladesh.

A cross-sectional study was conducted to evaluate the effect of a community-based health education intervention programme and to study the determinants of night blindness in Bangladesh. The intervention programme was implemented to reduce the morbidity of nutritional blindness (night blindness) in the northern part of Bangladesh (Ranjpure district) during 1986-1989. A baseline study in 1986 covered 2010 households with a total population of 11,600, and the evaluation study in 1989 covered 2011 households with a total population of 10,456. Prevalence of night blindness was studied among children aged < 9 years in these households. The prevalence of night blindness per 1000 children was reduced significantly during the intervention period from 50.7 in 1986 to 26.7 in 1989. However, the post-intervention prevalence varied significantly between areas. Multivariate analyses showed that consumption of fish, meat, milk or eggs, dark green leaf vegetables, yellow fruits and vitamin A capsules were significant predictors of night blindness. In addition, family income, mother's literacy, family size and area of residence exhibited strong and statistically significant associations with night blindness in the 1989 cross-sectional study. The prevalence of night blindness was highest among 4-6 year old girls and 7-8 year old boys. The sex difference was, however, not statistically significant.

Tubed, folded radial forearm free flap for pharyngeal reconstruction and voice rehabilitation.

Reconstruction after total laryngectomy ideally includes restoration of voice with protected swallowing. Local flaps and tracheo-esophageal puncture with a prosthesis are widely used to accomplish this. Persistent problems, perhaps inherent to this solution, have led to the recent use of a tubed, folded radial forearm free flap for postlaryngectomy reconstruction. This flap has produced a good voice with low phonation pressures and minimal aspiration in 7 patients. A new configuration for this flap is described and 3 patients reported. Our flap's advantages include a simplified pedicle path, ease of construction, remote microanastomosis, and safer revision. Both patients with surviving flaps rapidly developed a good voice after reconstruction. Mild aspiration in 1 delayed oral intake and forced a successful revision. Our reconfigured flap may have some technical advantages, survives to length, and restores a good voice without significant aspiration. This and its simple care make it popular with patients. Development of this flap holds promise of more complete reconstruction after total laryngectomy.

Supportive and emergency use of prostaglandin in the neonate with congenital heart disease.

A retrospective study of 28 neonates, admitted as emergencies with hypoxia due to inadequate pulmonary blood flow caused by congenital lesions in the pulmonary and/or tricuspid valves, showed a marked improvement in the degree of oxygenation after treatment with Prostaglandin E1 (PGE1) infusion. The side effects related to the PGE1 administration were observed, and were easily managed. The study demonstrates the efficacy of PGE1 in maintaining the patency of the ductus arteriosus; we therefore propose that this drug should be used, though with caution, routinely in peripheral hospitals before such patients are transferred to specialised centres for further diagnostic and therapeutic intervention.

Coupled expression of Ca2+ transport ATPase and a dihydrofolate reductase selectable marker in a mammalian cell system.

Stable expression of a full-length cDNA encoding chicken fast muscle Ca2+ transport ATPase was obtained in a Chinese hamster lung cell line (DC-3F), using a dual-promoter expression vector (pH beta FCaA3) in which the ATPase was cloned downstream of a human beta-actin gene promoter, and a mutant dihydrofolate reductase cDNA (A3/DHFR) was cloned downstream of an SV40 promoter-enhancer. Owing to its essentially normal catalytic activity and modest (20-fold) resistance to the antifolate methotrexate (MTX), the A3/DHFR mutant enzyme served as an efficient dominant selection marker in transfected cell populations challenged with MTX and, within a broad range of drug concentrations, allowed subsequent amplification and overexpression of vector sequences. In stable transfectants, the expressed ATPase was targeted to intracellular membranes, and the microsomal fractions from those cells exhibited high rates of Ca2+ transport. In comparative experiments using transient expression in COS1 cells, the level of ATPase per transfected cell was greater, but less than 5% of the transfected population exhibited ATPase expression. Furthermore, as opposed to the stable lines, the transiently expressing cells could not be propagated. Overall, the yield of ATPase was 12-16 and 4-6 micrograms per milligram of microsomal protein in the stable and the transient expression systems, respectively. The advantages of the stably transfected cell lines therefore lie in the homogeneity of ATPase expression and its distribution in cells and microsomes, in the large yield of microsomes obtained by continuous cell propagation, and in the reproducible functional characteristics of the microsomes. Moreover, the microsomes derived from stably transfected cell lines provide a convenient system for studies of Ca2+ transport and ATPase partial reaction, eliminating the need to conduct repetitive transient transfections to obtain sufficient amounts of enzyme for functional studies.

Construction of a dominant selectable marker using a novel dihydrofolate reductase.

Simian virus 40 promoter-enhancer-based mammalian expression plasmids using dihydrofolate reductase (DHFR)-encoding cDNA sequences originally isolated from two methotrexate (MTX)-resistant, DHFR-overproducing Chinese hamster lung cell lines were constructed. One, designated pSVA75, contains a DHFR cDNA that encodes leucine (Leu22) and corresponds to the wild type (wt), MTX-sensitive form of the enzyme [Melera et al., J. Biol. Chem. 263 (1988) 1978-1990]. The other plasmid, pSVA3, contains a cDNA that encodes a novel mutant form of the enzyme in which Leu22 has been changed to Phe [Melera et al., Mol. Cell Biol. 4 (1984) 38-48]. The resulting DHFR displays a 20-fold-enhanced resistance to inhibition by MTX, but maintains the catalytic activity of the wt enzyme [Albrecht et al., Cancer Res. 32 (1972) 1539-1546]. Transfection of DHFR- Chinese hamster ovary cells with either plasmid demonstrated that both were able to reconstitute the DHFR+ phenotype with equal efficiency (i.e., greater than 2.5 x 10(-3), indicating that both the wt and mutant enzymes were catalytically active in transfected cells. In addition, the mutant form of the enzyme was found to act as a dominant selectable marker when transfected into diploid DHFR+ cells, and to allow selection of resistant clones at low MTX concentrations (125 nM MTX) with a frequency of greater than 8 x 10(-4). Moreover, transfected clones were found to amplify their exogenous DHFR sequences to reasonably high levels (42-fold) at relatively low (888 nM) MTX concentrations, suggesting that substantial amplification of DHFR DNA and cotransfected sequences as well, can be achieved with this vector.

Vectors with segmented multiple cloning sites (SMCS) for easy monitoring of restriction digests and post-cloning orientation reversal of genes.

A novel approach is described, through which the transcriptional orientation of cloned genes is manipulated without further subcloning. This is achieved through a restriction/ligation process (without changing the test tube) leading to approx. 1:1 mixture of both possible orientations. The plasmid containing the reversed orientation is easily distinguished from the wild type by built-in restriction-indicator sites for SpeI. Additionally, a large marker fragment is incorporated into the segmented multiple cloning site (SMCS) regions to facilitate monitoring the progress and/or efficiency of vector restrictions. The marker fragment is released upon appropriate double digestion, thus acting as an indicator for complete restriction-enzyme digestion. We have equipped some existing plasmids, commonly used in molecular biology, with these features and demonstrated their superiority to plasmids having regular MCS sequences.

Kinetics and mechanism of chlorine exchange between chloramine-T and secondary amines.

The kinetics and mechanisms of chlorine transfer from chloramine-T (CAT) to several amines are second order and independent of p-toluenesulfonamide concentration; thus, the reaction does not involve disproportionation of CAT to dichloramine-T. From the profile of pH versus rate, the following mechanisms were proposed: (1) reaction of the ionized species of CAT with the ionized amine (ionic mechanism) and (2) reaction of the un-ionized species of CAT with the un-ionized amine (nonionic mechanism). The second-order, pH-independent rate constants calculated for the ionic and nonionic mechanisms were 1.6 and 5 x 10(6) M-1 s-1, respectively. Although these two mechanisms are kinetically indistinguishable, the rate constant for the nonionic mechanism is of the same order of magnitude as those calculated for similar chlorination reactions involving nonionizable chloramines, such as N-chlorosuccinimide, N-chloroquinuclidine, and N-chloro-N-methylbenzenesulfonamide. The proposed mechanism for the chlorine exchange involves a molecule of water in a cyclic, six-membered transition state.

Structure-activity considerations in kinetics and mechanism of chlorine exchange between chloramine-T and secondary amines.

To study the mechanism of N-chlorination of secondary amines by chloramine-T, the kinetics of the reactions of some aromatic-substituted analogues of N-chlorobenzenesulfonamide with various secondary amines were determined. The importance of amine basicity and reactivity of the N-Cl bond of the N-chlorobenzenesulfonamide was also assessed. The results indicate that a mechanism involving the un-ionized species of both reactants (i.e., a molecular mechanism), rather than an ionic mechanism, is operating and that the reaction most likely proceeds via a six-membered-ring transition state that incorporates a water molecule.
