Trypan blue has previously been shown to interact with the C3 receptor, but not with the Fc receptor. In the present work the effects of Trypan blue on mouse erythrocyte (ME) and C3 binding receptors of the peripheral blood mononuclear cells have been studied in healthy individuals and in patients with chronic lymphocytic leukemia (CLL). The resulting dose-response curves have been compared with each another. The results of statistical analysis indicate that there is no significant difference between the effects of trypan blue on the two receptors in healthy individuals. In contrast ME and C3 binding receptors differ significantly in trypan blue sensitivity in CLL patients. These data suggest that the two receptors are similar, or are situated on the cell membrane in such a way that the trypan blue bound to one of them inhibits the functioning of the other one too, but they are not the same. In the course of leukemic transformation, the trypan blue sensitivity of the ME binding receptor does not vary, whereas that of the C3 receptor is enhanced significantly.

When peripheral blood mononuclear cells separated at 4 degrees C were incubated at 37 degrees C, they shed their surface receptors. Total absorption of the released mouse erythrocyte binding receptors from the supernatant of the cell suspension with mouse erythrocytes did not lead to a change in the IgG Fc and C3 receptor content of the supernatant. On removal of the C3 or IgG Fc receptors, the quantity of mouse erythrocyte binding receptor remained unchanged. These observations indicate that the three receptors are mutually independent structures, or undergo dissociation in the course of the shedding.
An increase in the production of macrophage migration inhibitory factor, chemotactic factor for neutrophils, and skin reactive factor, was observed in lymphocyte cultures if the cells were allowed to age in culture for 24 h. The increased lymphokine production was reduced by adding concanavalin A-stimulated and mitomycin C-treated suppressor cells. It is suggested that the lymphokine production could be regulated by suppressive mononuclear cells.
1. Myofibrillar protein breakdown was calculated from the urinary excretion ratio of NT- methylhistidine (3-methylhistidine) to creatinine in newborn premature and full-term infants. Representative values were obtained from single voidings provided that the infant's metabolic status was stable. 2. NT- Methylhistidine in infant urine was measured by a rapid Auto Analyser method and shown to give similar values to those obtained by ion-exchange separation techniques. 3. The molar excretion ratio of NR- methylhistidine to creatinine averaged 0.0159 in urine samples obtained within 12 h after birth. A similar ratio was found in amniotic fluid collected at birth. It is argued that this ratio does not reflect a low rate of myofibrillar protein breakdown in the foetus, but rather a more effective transplacental passage of NT- methylhistidine than of creatinine. 4. The urinary ratio increased during the first 2 days after birth to a plateau at 0.0372. This represents a myofibrillar protein degradation rate of 3.40% day-1 in full-term infants. 5. The molar excretion ratio during the period 40-120 h after birth increased in premature infants and reflects a fractional degradation rate of 5.34% day-1 in those infants weighting less than 1 kg at birth. 6. Lower excretion ratios were found in some infants of diabetic mothers and in athyroid infants. 7. The urinary excretion ratio of NT-methylhistidine to creatinine is presented as a useful method for evaluating the breakdown rate of myofibrillar protein in neonates and can be applied to a number of abnormal nutritional or hormonal states.

Polymorphonuclear leukocytes from patients with psoriasis demonstrated a significantly enhanced chemotactic responsiveness to zymosan-activated human serum as compared to granulocytes from healthy volunteers. Furthermore, psoriatic peripheral blood mononuclear cells stimulated with concanavalin a produced an increased amount of lymphocyte derived chemotactic factor (LDCF) as compared to that in the case of healthy persons. The LDCF proved to be chemokinetic for the psoriatic granulocyte. It is postulated that these two phenomena may play a role in the pathogenesis of psoriasis.

In 40 patients suffering from diabetes mellitus and in 18 healthy volunteers the phagocytic function was investigated using a quantitative NBT test. The NBT reduction was significantly lower in diabetes than in healthy donors irrespective of affliction with other dermatoses. A significant correlation was found between the severity and hence the insulin dependency of diabetics and the NBT-reductive capacity.
The suppressor activity of peripheral blood mononuclear cells was investigated in 25 patients with psoriasis vulgaris. In the psoriatic patients the suppressor activity was found to be significantly lower than in the control group, which suggests that the weak suppressor activity may play a role in the pathogenesis of this disease.
The proportion of T lymphocytes in the peripheral blood, their phytohaemagglutinin reactivity, and the leucocyte migration inhibitory effect of purified tuberculin protein were found to be equal in 25 recurrent corneal herpes simplex patients and 17 healthy individuals who had never suffered from clinically manifest herpes simplex virus (HSV) infection. Conversely, the HSV vaccine inhibited the leucocyte migration in the healthy group, but not in the recurrent herpes patients when the clinical symptoms were manifest. In 14 recurrent corneal herpes simplex patients, the examinations were repeated after a 3-month symptom-free period, and in these periods the patients corresponded equally to the healthy group in every respect.
The shedding process of the mouse erythrocyte binding receptors of human peripheral lymphocytes of a healthy donor group was compared to that of patients with chronic lymphocytic leukemia. The shedding process exhibited significant differences with respect to the groups tested, suggesting that the mouse erythrocyte binding receptors were altered during leukemic transformation or that the leukemic cells were fixed in a stage of maturation.

Significantly increased suppressor activity of peripheral blood mononuclear cells was demonstrated in patients suffering from chronic discoid lupus erythematosus.
Lymphoid cells from 4 of 5 patients diagnosed as pemphigus vulgaris (PV) and 6 of 7 patients diagnosed as bullous pemphigoid (BP) demonstrated specific cell-mediated immunity by the production of migration inhibitory factor (MIF) in the presence of autologous epidermal saline extracts. Clinical treatment of these patients with immunosuppressive agents resulted in a state of unresponsiveness of their lymphoid cells to similar concentrations of the antigen. Controls consisted of lymphoid cells from patients with bullous burns or various drug allergies which failed to show significant MIF production in the presence of autologous skin extract. These studies suggest that both PV and BP patients posses cell-mediated immunity (CMI) to their own autologous tissue antigens and this CMI may play a role in the pathogenesis of these diseases.
A significant decrease of serum prealbumin levels and an increased proportion of prealbumin bearing lymphocytes in peripheral blood have been found in patients of hapten-induced diseases. Prealbumin deposition in the vessel walls, and/or prealbumin positive cells (granulocytes, lymphocytes, macrophages) in the dermis have been observed in patients with proved drug allergy as well as allergic contact dermatitis using immunofluorescence technique. The eluates from membrane of lymphocytes of patients with drug allergy contained a mitogenic substance, probably antigen and sometimes prealbumin as well. These results also support the earlier suggested hypothesis that the prealbumin may act as a hapten carrier protein.
