An hepatic osmoreceptor mechanism in the rat: electrophysiological and behavioral studies.

Rat liver was perfused with Ringer solution through the portal vein by use of a perfusion system which was designed to switch from standard Ringer solution to hypertonic or hypotonic Ringer solution. Neural responses to the osmotic change in the perfusion solutions were analyzed. They showed that two different types of osmosensitive afferent fibers exist in the hepatic vagus; one is characterized by increasing the frequency of spike discharges responding to higher osmotic pressure, while the other shows the same response to lower osmotic pressure. Behavioral changes caused by hepatic vagotomy were also observed. Though no differences could be detected in routine behavior (e.g., daily intakes of food and water, body-weight increase) between the vagotomized and the sham-operated rats, the former lost the ability to adjust urine concentration immediately in response to osmotic changes in the internal environment. These results provide evidence for the hypothesis that hepatic osmoreceptors exist in the rat.

Gas-liquid chromatographic determination of vitamin D in multivitamin preparations containing excess amounts of vitamin E.

Gas-liquid chromatographic (GLC) determination of vitamin D in multivitamin preparations containing excess amounts of vitamin E (a more than 2,500 weight ratio of dl-alpha-tocopheryl acetate to vitamin D) was investigated and a simplified routine method was established because the method reported previously (1) could not be applied to such special preparations. After applying the unsaponifiable matters of a sample to a phosphate-treated alumina column chromatography prepared according to MULDER et. al. (2), the eluate was evaporated and the residue was subjected to thin-layer chromatography (TLC) and GLC. When this method was applied to model preparations made by mixing vitamin D2 and dl-alpha-tocopheryl acetate (excess amounts), good results were obtained. Since the results on a commercial multivitamin preparation containing excess amounts of vitamin E were also satisfactory, it was confirmed that the proposed method could be used for simplified routine determinations.

Gas-liquid chromatographic determination of vitamin D in tuna liver and vitamin D resin oils.

Gas-liquid chromatographic (GLC) determination of vitamin D3 in tuna liver and vitamin D3 resin oils was investigated and a routine method slightly modified from the previously reported methods (1, 2) was established. Since both tuna liver and vitamin D3 resin oils contained large amounts of sterols, digitonin-Celite column chromatography according to SHEPPARD et al.(4) was used to remove the sterol influence from the unsaponifiable matters of the oils. After collecting the eluate and evaporating the solvent, the residue was subjected to thin-layer chromatography (TLC) using Kieselgel GF254 as an adsorbent and a mixture of n-hexane-ethyl acetate (4: 1) as a developing solvent. The scraped zones corresponding to vitamin D3 and pre-D3 were trimethylsilylated and then applied to the GLC using 1.5% OV-17 packed on Shimalite W (80-100 mesh) as a stationary phase. Trimethylsilylation of the gas chromatograms was an essential procedure, because the peaks of unknown substances in tuna liver oils and lumisterol in vitamin D3 resin oils could not be separated from the peak of pyro-D3 without trimethylsilylation. When the proposed method was applied to the samples, satisfactory results were obtained.

Gas-liquid chromatographic determination of vitamin D in multivitamin preparation.

Gas-liquid chromatographic (GLC) determination of vitamin D in multivitamin preparations was investigated and a simple routine method was established. The unsaponifiable matters of a sample was applied to a thin-layer chromatography (TLC) plate using Kieselgel GF254 as an adsorbent and a mixture of n-hexane-ethyl acetate (4:1) as a developing solvent. The scraped zones corresponding to vitamin D and previtamin D were extracted and determined by GLC using 1.5% OV-17 packed on Shimalite W (80-100 mesh) as a stationary phase. When the proposed method was applied to model preparations made by mixing vitamin D, A and E, it was confirmed that the determination of vitamin D was possible when the ratios of vitamin A and E (DL-alpha-tocopheryl acetate) to vitamin D were within 104 (I.U. ratio) and 2,500 (weight ratio), respectively. Since the ratios of most of commercial multivitamin preparations on sale in Japan are within the limitations and the results on multivitamin preparations were also satisfactory, the proposed method was confirmed to be suitable simple routine.

Intravascular hemolysis: a complication of midtrimester abortion: a report of two cases.

Intravascular hemolysis occurred twice in a series of 1582 consecutive intraamniotic midtrimester saline abortions. The hemolysis was associated with consumption coagulopathy and acute renal failure. Both patients made a complete recovery after a prolonged hospital stay; one required peritoneal dialysis. The pathogenesis of this serious complication of saline abortion is discussed but no satisfactory explanation has been reached.
