HLA-DQA1 and HLA-DQB1 haplotypes in aborted fetuses and couples with recurrent spontaneous abortion.

HLA haplotypes may be associated with spontaneous abortion through a variety of mechanisms, including maternal hyporesponsiveness to fetal alloantigens, maternal autoimmunity, and HLA-linked t-locus homologues. HLA-DQA1 and HLA-DQB1 haplotypes were determined in 37 couples with a history of recurrent spontaneous abortion (RSAB), 40 of their abortuses, and 20 fertile control couples. The distribution of haplotype frequencies did not differ between control subjects and RSAB wives, RSAB husbands, or abortuses. The frequency of the HLA-DR5-linked haplotype, DQA1*501/DQB1*301, which was considered a marker for immune hyporesponsiveness, did not differ between RSAB wives and control subjects (P = 0.353). The frequency of the autoimmune-associated HLA-DR3-linked haplotype, DQA1*0501/DQB1*0201, did not differ significantly between RSAB wives and control subjects (P = 0.103). The frequency of the DQA1*0201/DQB1*0201 haplotype in RSAB husbands was greater than the 95th percentile confidence limit of the frequency of this haplotype in control subjects. Among seven RSAB husbands who were heterozygous for this haplotype and did not share a DQA1*0201 allele with his wife, the haplotype was transmitted to 6 of 7 abortuses (3.5 expected). Although the small size of this sample precludes drawing conclusions regarding HLA transmission biases in RSAB couples, these data have generated a specific hypothesis regarding the DQA1*0201/DQB1*0201 haplotype that can be investigated in future studies.

Bovine leptospirosis: microbiological and serological findings in aborted fetuses.

Leptospiral infection was diagnosed in 41.6 per cent of 245 randomly selected aborted bovine fetuses and 68.9 per cent of 103 aborted fetuses from farms with abortion problems. Diagnosis was based on combined leptospiral isolates from fetal kidney and eye, and fetal serology and immunofluorescence. Fifty-six of the 58 strains of leptospira isolated belonged to the Hebdomadis serogroup and were similar to serovar hardjo while one of the remaining two strains belonged to the Icterohaemorrhagiae serogroup and the other to the Canicola serogroup. These findings indicate that serovar hardjo infection is a major factor in the aetiology of bovine abortion and that it is possible to isolate this organism from fetal kidney by the careful application of dilution culture technique.

Neosporosis in the aborted fetus and newborn calf.

Protozoal encephalitic lesions were found in four aborted fetuses and one dead newborn calf. The organism was identified as Neospora caninum by immunoperoxidase. The brain lesions were of two forms. One was observed in three fetuses of 5 months gestation and was characterized by multifocal necrosis. The other was found in a 7-month fetus and in a newborn calf, and showed severe infiltration with macrophages and plasma cells containing IgG. This association, between the age of fetus and inflammation, may reflect development of the immune system in bovine fetuses.

MHC class II compatibility in aborted fetuses and term infants of couples with recurrent spontaneous abortion.

Maternal-fetal histocompatibility for alleles at HLA class II loci, HLA-DQA1 and HLA-DQB1, was examined in 40 abortuses and 31 liveborn children of 68 couples with a history of idiopathic recurrent spontaneous abortion (RSAB) who underwent leukocyte immunization prior to the index pregnancy. Significantly more couples with RSAB shared two HLA-DQA1 alleles as compared with fertile control couples (0.18 vs. 0.03, respectively; P = 0.031). There were no differences in HLA sharing between couples with RSAB who experienced a repeat abortion in the index pregnancy as compared with couples with RSAB who were delivered of a liveborn child. Non-significant deficits of abortuses who were compatible for alleles at the HLA-DQA1 (6 observed vs. 8.5 expected; P = 0.225) and the HLA-DQB1 (7 observed vs. 9.2 expected; P = 0.254) loci were observed. A significant deficit of HLA-DQA1 compatible liveborn children was observed (1 observed vs. 5.5 expected; P = 0.0069). The overall deficit of HLA-DQA1 compatible fetuses (7 observed vs. 14.0 expected; P = 0.0018) after approximately 8 weeks gestation suggests that HLA-DQA1 compatible fetuses may be aborted early in pregnancy, prior to the time when fetal tissue can be recovered for genetic studies.

Isolation of equine herpesvirus-2 from the lung of an aborted fetus.

This study describes the isolation of equine herpesvirus-2 (EHV-2) from the lung of an aborted equine fetus in Argentina. The isolated virus was confirmed as EHV-2 by indirect immunofluorescence using a rabbit anti-EHV-2 polyclonal antiserum and by virus-neutralization test using an equine polyclonal antibody against EHV-2. Restriction endonuclease DNA fingerprinting with BamHI also confirmed the identity of the virus as EHV-2. Furthermore, viral nucleic acid was detected by polymerase chain reaction from the original lung sample and from the DNA obtained from cells infected with the virus isolate. This work constitutes the first reported isolation of EHV-2 from an aborted equine fetus. The presence of EHV-2 in the lung of the aborted fetus would indicate that this virus is capable of crossing the placental barrier. However, no cause-effect relationship was established between the EHV-2 isolate and the abortion.

Survey of Neospora caninum infection by nested PCR in aborted fetuses from dairy farms in Aguascalientes, Mexico.

The aim of this study was to determine by nested PCR probe the presence of Neospora caninum in fetal brain tissue from aborted dairy fetuses in Aguascalientes, a state in the central part of Mexico. Forty-four fetal brains from eight dairy farms were analyzed using single tube nested PCR probe with primers NF1, NS2, NR1 and SR1, with histopathology as the reference technique. From the 44 bovine fetuses studied, 35 (80%) were diagnosed as being infected by N. caninum using PCR probe and 20 (45%) were considered positive by histopathology, because they had lesions suggestive of bovine neosporosis. Fair agreement was observed (31%) between both probes. The age of aborting cows ranged from 1.6 to 2.9-year-old in 17 (49%) of the positive cases diagnosed by PCR. Gestational ages of aborted fetuses ranged from <3 to 9 months, with average of 5.6 months, and no statistical significant difference was observed when ages of infected and no infected fetuses were compared (P > 0.05). The frequency of N. caninum positive cases observed could be considered to be higher than other areas of Mexico and other countries.

Placental cultures for cytogenetic assessment in saline-aborted fetuses.

Confirmation of cytogenetically abnormal fetuses following saline abortion has been shown to be possible with the placenta as the source of viable fetal cells. The method is described in detail. In one third of cultures, only female cells were present. Differentiation between maternal and female fetal tissue when no numerical or structural cytogenetic disorder is present requires detailed analysis of fluorescently stained chromosomes for polymorphisms.

[2 strains of swine parvoviruses isolated from aborted fetuses].

Two hemagglutinating virus strains were isolated (in primary cell cultures of pig kidneys) from viscera of aborted swine fetuses. A number of serologic, cytologic, physico-chemical, and laboratory investigations with the strains revealed that they belonged to the group of porcine parvovirus (PPV). The isolation of SPV from aborted fetuses pointed to the fact that the disease had been widespread among the swine population and plays a part in reproduction disturbances that have come to be known recently. The isolated strains did not produce a clear and distinguishable cytopathic effect in inoculated cell cultures. They, however, could be demonstrated in the cultures indirectly through cytologic investigation (the demonstration of intranuclear inclusion bodies, type B after Cowdry, through hemagglutination tests, and via immunofluorescent microscopy.

Antibody isotypes in sera of equine fetuses aborted due to Leptospira interrogans serovar pomona-type kennewicki infection.

Leptospira-specific antibody isotypes in sera of late term equine fetuses aborted due to Leptospira interrogans serovar pomona-type kennewicki infection were characterized and compared with those of their dams. IgM was the dominant Leptospira-Specific isotype in both fetuses and mares. However, IgGa was the isotype in highest concentration in petal sera and strong Leptospira-specific IgGa but no IgGb and little or no IgG(T) were detected. In contrast, although IgGb was quantitatively the dominant isotype in mares serum, Leptospira-specific serum IgG in aborting mares was dominated by IgG(T) but also included large amounts of IgGa and IgGb. IgGa and IgGb were quantitatively the dominant isotypes in sera of fetuses and mares, respectively. Affinity purified IgGa from fetuses did not agglutinate leptospires but serum devoid of IgGa did, suggesting that IgM is the principal agglutinating antibody. It is concluded that the equine fetus is deficient in IgGb and IgG(T) synthesis.

[Pathomorphological studies of aborted fetuses from sheep infected with R. burneti].

Serologic investigations were carried out with 441 samples of blood sera from ewes, coupled with histopathologic studies of 18 aborted fetuses. Positive serologic Q fever results were obtained by the CF-test with 14.4 per cent of the sheep. Abortions were ascertained with 4 to 8 per cent of the animals per flock in the final month of pregnancy. Nonviable lambs were also obtained that died in the course of the first hours after lambing. The gross lesions observed consisted in the swelling and hyperemia of the cotyledons, with gray-whitish deposits; the chorion was diffusely thickened, was folded, with a dirty-reddish colour, and in some cases also presented gray-whitish deposits. Histopathologically, featuring in the fetuses were the lympho-leukocytic accumulations and the lytic micronecrotic foci in the liver, kidneys, adrenals, and lymph nodes, while the central nervous system showed lympho-leukocytic meningoencephalitis.
