Adrenalectomy does not prevent the ability of 8-OH-DPAT to decrease the ejaculatory threshold in male rats.

Abstract

The present experiments demonstrate that 8-OH-DPAT (0.25 mg/kg SC, - 15 min) produced a decrease in the ejaculatory threshold to the same extent in adrenalectomized male rats as in sham operated controls. Both groups of animals displayed a marked and statistically significant decrease in number of mounts and penile intromissions preceding ejaculation and in the ejaculation latency, as a result of treatment with 8-OH-DPAT. Adrenalectomy per se did not affect any aspect of the male rat's sexual behavior (latency to first intromission, number of mounts or penile intromissions, ejaculation latency or the postejaculatory interval). The surgical removal of the adrenals was verified by measurements of plasma corticosterone levels. It is concluded that well documented effects of 8-OH-DPAT on adrenal secretions do not contribute to its ability to decrease the ejaculatory threshold in male rats.

Serotonin 5-HT1D and 5-HT1A receptors respectively mediate inhibition of glutamate release and inhibition of cyclic GMP production in rat cerebellum in vitro.

The K(+)-evoked overflow of endogenous glutamate from cerebellar synaptosomes was inhibited by serotonin [5-hydroxytryptamine (5-HT); pD2 = 8.95], 8-hydroxy-2-(di-n- propylamino)tetralin (8-OH-DPAT; pD2 = 7.35), and sumatriptan (pD2 = 8.43). These inhibitions were prevented by the selective 5-HT1D receptor antagonist N-[4-methoxy-3-(4-methyl-1-piperazinyl)phenyl]-2'-methyl-4'-(5-methyl-1, 2,4 - oxadiazol-3-yl) (1,1-biphenyl)-4-carboxamide (GR-127935). The three agonists tested also inhibited the cyclic GMP (cGMP) response provoked in slices by K+ depolarization; pD2 values were 9.37 (5-HT), 9.00 (8-OH-DPAT), and 8.39 (sumatriptan). When cGMP formation was elevated by directly activating glutamate receptors with NMDA or alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA), the inhibition of the cGMP responses displayed the following pattern: 5-HT (pD2 values of 8.68 and 8.72 against NMDA and AMPA, respectively); 8-OH-DPAT (respective pD2 values of 9.15 and 9.00); sumatriptan (0.1 microM) was ineffective. The 5-HT1A receptor antagonist (S)-(+)N-tert-butyl-3-[4-(2- methoxyphenyl)piperazin-1-yl]-2-phenylpropionamide dihydrochloride [(+)-WAY 100135] did not prevent the inhibition of glutamate release by 5-HT but blocked the inhibition by 8-OH-DPAT of the NMDA/AMPA-evoked cGMP responses. It is suggested that presynaptic 5-HT1D receptors mediate inhibition directly of glutamate release and indirectly of the cGMP responses to the released glutamate; on the other hand, activation of (postsynaptic) 5-HT1A receptors causes inhibition of the cGMP responses linked to stimulation of NMDA/AMPA receptors.

8-Hydroxy-2-(di-n-propylamino)tetralin

A series of N4-imidoethyl derivatives of 1-(2,3-dihydro-1,4-benzodioxin-5-yl)piperazine as 5-HT1A receptor ligands: synthesis and structure-affinity relationships.

A series of unsubstituted and substituted succinimido, maleimido, and glutarimidoethyl derivatives of eltoprazine (3) was synthesized and tested for affinity for the 5-HT1A receptor in rat brain homogenates. The unsubstituted compounds have a moderate affinity for the receptor, while the affinity considerably increases by substitution at or enlargement of these cyclic ring systems. A good correlation was found between the inhibition constant Ki (expressed as pKi) and the lipophilicity (clogP). No correlation was observed between the pKi or pKi+ (local inhibition constant) and the basicity of the N4-nitrogen atom.

8-Hydroxy-2-(di-n-propylamino)tetralin

Serotonin-induced calcium signaling via 5-HT1A receptors in human leukemia (K 562) cells.

In this study, serotonin (5-HT) was found to diminish the intracellular calcium, [Ca2+]i, concentrations in a dose-dependent manner in Fura-2-loaded human leukemia (K 562) cells. Prior addition of verapamil (a calcium channel blocker) to the cells abolished the serotonin-induced response, suggesting that the diminution of free [Ca2+]i contents was due to the opening of calcium channels. Thapsigargin (an agent which increases [Ca2+]i via its action on endoplasmic reticulum) augmented the [Ca2+]i contents. Addition of serotonin before or after thapsigargin (THAP) curtailed the THAP-stimulated increases in [Ca2+]i, supporting the notion that 5-HT acts on the calcium channels and that it empties, in part, the THAP-stimulated calcium contents. Spiperone and NAN-190, antagonists to 5-HT1A receptor subtype, abolished the serotonin effects on calcium signaling, whereas an agonist to 5-HT1A receptor, 8OHDPAT, mimicked the serotonin-like action on the diminution of the intracellular calcium contents. These results indicate that the 5-HT response is mediated via 5-HT1A serotonergic receptors in these cells. Furthermore, we characterized the 5-HT receptors in K 562 cells. The specific binding of serotonin to these cells was saturable and hyperbolic. The Kd and the Bmax of serotonin binding were 100 nM and 1690 fmol/10(6) cells, respectively. 8-[3H]OH-DPAT also labeled these cells with a Bmax of 2.8 fmol/10(6) cells and a Kd of 20 nM. The specific binding of 8-[3H]OH-DPAT to K 562 cells was displaced by serotonin, 8OH-DPAT, and NAN-190. These results suggest that K 562 cells possess functional 5-HT1A receptors coupled with calcium signaling.
