Revised cobalt-60 depth dose data.

Abstract

It has long been suspected that the depth-dose characteristics of modern 60Co teletherapy units are no longer well represented by the traditional data of The British Journal of Radiology Supplement 11. Extensive measurements have been undertaken on three modern 60Co units in an attempt to clarify this position. Three different values of source-to-skin distance were studied, namely 50, 80 and 100 cm. Systematic deviations from the traditional data are unambiguous, amounting to up to 3% at 80 cm SSD and as much as 7% at 50 cm SSD. Comparison with calibration results at a large number of institutions lends confidence that the present results are representative of modern treatment units.

Mechanisms of heparin transport through expanded poly(tetrafluoroethylene) vascular grafts.

Thrombosis and neointimal hyperplasia limit the utility of small-caliber artificial vascular grafts. Surface modifications and adjunctive pharmacological therapy might mediate these complications. We examined the mechanisms by which a model vasoactive compound, heparin, transverses porous graft materials and how material modifications alters this drug's transport. The effective permeance of [(3)H]heparin was measured after application of a uniform concentration of drug to either the internal or external surface of the graft and in the presence or absence of pressure-driven physiologic hydraulic flows. Transgraft permeance was equivalent to those observed in normal arteries and, while enhanced by convection, was mediated in major part by diffusion. Peclet numbers under the various conditions examined ranged from 0.05 to 1.2, indicating that diffusive forces were equal to or exceeded convective forces in governing transmural heparin motion. Heparin traversed the graft even when applied from the outer perivascular surface, against adverse hydraulic flows. Modifications of the grafts that included a yarn barrier of spun poly(tetrafluoroethylene) or chemical modification of surface tension energy altered permeances as well. A unifying model for interpretation of these data incorporates the concept of entrapped air and surface tension energy in the graft. These characterizations allow for the design of vascular grafts that are optimized for pharmacotherapy to help prolong graft patency, especially in small-caliber vascular beds.

Respiratory muscle blood flows during physiological and chemical hyperpnea in the rat.

Whether the diaphragm retains a vasodilator reserve at maximal exercise is controversial. To address this issue, we measured respiratory and hindlimb muscle blood flows and vascular conductances using radiolabeled microspheres in rats running at their maximal attainable treadmill speed (96 +/- 5 m/min; range 71-116 m/min) and at rest while breathing either room air or 10% O(2)-8% CO(2) (balance N(2)). All hindlimb and respiratory muscle blood flows measured increased during exercise (P < 0.001), whereas increases in blood flow while breathing 10% O(2)-8% CO(2) were restricted to the diaphragm only. During exercise, muscle blood flow increased up to 18-fold above rest values, with the greatest mass specific flows (in ml. min(-1). 100 g(-1)) found in the vastus intermedius (680 +/- 44), red vastus lateralis (536 +/- 18), red gastrocnemius (565 +/- 47), and red tibialis anterior (602 +/- 44). During exercise, blood flow was higher (P < 0.05) in the costal diaphragm (395 +/- 31 ml. min(-1). 100 g(-1)) than in the crural diaphragm (286 +/- 17 ml. min(-1). 100 g(-1)). During hypoxia+hypercapnia, blood flows in both the costal and crural diaphragms (550 +/- 70 and 423 +/- 53 ml. min(-1). 100 g(-1), respectively) were elevated (P < 0.05) above those found during maximal exercise. These data demonstrate that there is a substantial functional vasodilator reserve in the rat diaphragm at maximal exercise and that hypoxia + hypercapnia-induced hyperpnea is necessary to elevate diaphragm blood flow to a level commensurate with its high oxidative capacity.

A new alpha-particle irradiator with absolute dosimetric determination.

A new experimental setup for uniform alpha-particle irradiation of cells in vitro is described. The alpha-particle irradiator is based on a radioactive (212)Pb/(212)Bi source. In the experimental setup proposed, cells are grown directly on a polylysine-coated track-etch material that forms the base of custom-made cell dishes. Alpha-particle irradiation is done through the base of the dish. Immediately prior to irradiation, the cell dish is scanned under a microscope, and images of cells with the corresponding coordinates are saved. After irradiation and after the biological end point under study has been determined, the cell dish is etched to develop alpha-particle tracks in the dish base. A microscope image series of alpha-particle track images is obtained by accurately revisiting every original (preirradiation) cell position in the track-etched dish. The number of alpha-particle traversals of each individual cell is scored by mapping images of alpha-particle tracks onto the images of cells recorded prior to irradiation. The uncertainty of the alpha-particle hit determination is 0.9 microm. The procedure described thus presents a method for radiobiological experiments with absolute, rather than statistical, cell dosimetry.
